Abstract

“If we want to overturn the bleak situation of our current food system, we need to attack theroots of the problems. Injustice in land, water and seeds must be addressed. Stop land andwater grabbing and also patents of seeds. Genuine agrarian reform must be implemented. Weneed to focus to rural areas to address our hunger problem by restructuring our mode ofproduction to a more sustainable-agroecology farming. This is also to protect and restore ourMother Earth.”(Henry Saragih, former Global Coordinator of La Via Campesina, from his address delivered tothe 9th Asia-Europe People’s Forum held in October 2012). Abstract

Cassava Brown Streak Disease (CBSD) is a major disease in cassava plants which have the serious problems in cassava plantations in the world, especially in Africa, Tanzania and India (Wassawaet., al, 2010). In Indonesia, the virus is still not optimal yet in the handling. The disease is present in plants that can destructive cassava leaves, stems and tubers.  It was greatly reduces the quality and production in the world such as India.  The decrease of cassava yield can reach 100% due to disease of CBSD (Lopez, 2003). The primer was designed from the coat protein gene of CBSD with a distance of 380 bp (Abarshiet.,al, 2012). The primers designed the forward primer (GGARCCRATGTAYAAATTTGC) and Reverse (GCWGCTTTTA  TYACAAAMGC). The RNA isolation have been used Plant Virus RNA Kit (Geneaid).  The CBSD RNA concentration around 55,2ng/ul.  The RT PCR program were one cycle of RT PCR reaction (45oC for 30 min), denaturation (45oC for 5 min) and 30 cycles for denaturation (94oC for 1 min), annealing (52oC for 30 sec), extention  (72oC for 1 min).  The results showed a single band of about 380 bp which is the  distance between the two primers were tested.  The multiplication shoot around 5 shoots per meristem explants with a combination of 0.5 ppm and 0.1 ppm BAP GA3 Keyword : Cassava Brown Streak Disease (CBSD), CASSAVA (Manihotesculentum), RT PCR

Abstract

Duffy-binding like (DBL) domains of Plasmodium falciparum are believed to be involved in erythrocytes invasion and infected erythrocytes cytoadhesion during the blood stage of malaria infection. In Plasmodium falciparum, DBL domains found in the two different protein families; Erythrocyte Binding Ligand (EBL) including EBA-175, EBA-140, EBA-181 and EBL-1, and Plasmodium falciparum Erythrocyte Membrane Protein-1. The study aimed at investigating the specific sequence of Plasmodium falciparum involved in severe malaria outcome.Blood samples from severe and uncomplicated falciparum malaria cases from Papua and South Kalimantan province, Indonesia, were collected for DNA extraction. A dried blood on filter paper were used for RNA extraction. PCR was performed using UNIEBP primers and directly sequenced. Internal var D primers were designed according to the sequencing of the ~550 bp band produced by UNIEBP primers. The nucleotide sequences were analyzed by NCBI BLAST. Multiple bands ranging from nearly 250 bp to 1 kb were resulted from gDNA in all samples. Two isolates yielded bands of 450 and 525 bp, three isolates showed three bands additionally 250 bp, one isolate presented four bands additionally 800 bp and one isolate resulted one band additionally 1 kb.Amplification of cDNA from severe malaria cases produced one to four bands ranging from 250 bp to 700 bp, and no band observed from cDNA of uncomplicated malaria. Sequencing of the 418 bp bands matched with the eba-175 gene, the 316 bp determined as DBL1a domain and 486 bp band matched with the DBLg domain isolated from placenta of PAM’s Malawian woman. The expression of a 237 bp sequence corresponding to var D gene, was detected solely in severe malaria patients, implicating an association of gene expression and manifestation of severe malaria. Further characterization of the var D gene with a larger sample size is required to draw a definite conclusion. Keywords: DBL domains, Plasmodium falciparum, severe malaria, var D gene.

Abstract

Durian seeds can be utilized as a producer of Water Soluble Polysaccharide (WSP) that can be applied to food products because crude extract of WSP is hydrocolloid. WSP interacts with other components, directly and indirectly affect processing applications, food quality, and acceptance.Water binding, solubility, viscosity and surface activity are important properties, determining usefulness and final product quality in a food system. The absence of extraction conditions that support so that the need for improved methods to variations in temperature and long extraction. This study aim to determine the effect of temperature and long extraction that produces the highest yield value and to determine the functional properties of the WSP.  The results of this study are the highest yield values in the 50 oC temperature extraction ang long extraction 2 hours amounted to 2,26%, solubility in water of 51,61%; lightness 34,14; water holding capacity (WHC) 2339,35%;  oil holding capacity (OHC) 425,36%; emulsion power 55,77m2/g; emulsion stability 585,50 minutes, foaming power 157,39 ml/g; foaming stability 25,43%. Long extraction increase the value of WHC and OHC but discrease the value of water solubility, foaming power and lightness. Abstract

Isolation and the physicochemical properties of rice bran protein concentrate (RBPC) produced from unstabilized and heat-stabilized Pandanwangi and Ciherang rice bran were studied. Rice bran stabilization process optimization done on previous research resulted in the extrusion conditions at a temperature of 130.96oC and screw speed 26.65 Hz. Kjeldahl analysis showed that protein content of unstabilized rice bran protein concentrates (URBPC) and stabilized rice bran protein concentrates (SRBPC) of Pandanwangi and Ciherang were 60.76%, 61.38%, 60.19%, and 60.23% respectively. Amino acid composition showed that polar amino acid composition of RBPC Ciherang was higher than that in Pandanwangi leading to its solubility. The protein percentage of acid-soluble glutelin of Pandanwangi was higher than that in Ciherang rice bran protein concentrate. The molecular weight were in range from 11.19 to 60.29 kDa. Glutelin differentiated into α-glutelin (30-39 kDa) and β-glutelin (19-25 kDa). The  RBPCs from two varieties had similar denaturation temperatures (77.22 - 77.99oC) with enthalpy ranged between 109.72 J/g and 200.98 J/g. Foaming stability and emulsion activities had similiar pattern with solubilities profile and showed no significant difference between varieties (p> 0.05). This finding shows potential protein concentrate of  both heat-stabilized and unstabilized rice bran as food ingredient.
