Abstract

Lactobacillus plantarum S34 dilaporkan mempunyai aktivitas antibakteri yang terkait dengan produksi bakteriosin. Bagian dari gen yang menyandikan salah satu lokus bakteriosin yang diproduksi oleh L. plantarum S34, disebut dengan plantarisin W (plnW), diamplifikasi dari plasmid dan dikloning menggunakan sistem vektor pGEM®-T Easy ke dalam Escherichia coli DH5?. Sekuens nukleotida plnW (± 405 pb) diidentifikasi sebagai protein integral membran. Lebih lanjut, plnW diekspresikan secara heterologus sebagai fusi protein dengan His(6)-tag tioredoksin menggunakan vektor ekspresi pET-32a(+) ke dalam E. coli BL21 (DE3) pLysS. Protein fusi rekombinan plnW terdapat dalam sitoplasma sel, tetapi selain fraksi terlarut terdapat juga fraksi tidak terlarut berupa badan inklusi. Purifikasi parsial dilakukan menggunakan kromatografi afinitas ligan Co2+ untuk fraksi terlarut dan metode elektroelusi gel poliakrilamid untuk fraksi tidak terlarut. Massa molekul berukuran kurang lebih 33 kDa terdeteksi berdasarkan pemisahan SDS-PAGE dan dikonfirmasi dengan Western blot sebagai protein fusi rekombinan plnW. Protein yang sudah terpurifikasi bermanfaat untuk mengetahui kaitan antara struktur dan fungsi bakteriosin.

Abstract

Virus was isolated from infected koi Cyprinus carpio koi with KHV and confirmed by polymerase chain reaction (PCR) and transmission electron microscope (TEM). Fish that were naturally living near koi pond such as catfish Clarias batrachus, nila Oreochromis niloticus, koki Carrasius auratus, and komet Macropinna microstoma did not infected after the cohabitation test. Based on the results KHV was the agent responsible for mortality of million cultured koi in East Java Province Indonesia. Absolute mortality was occurred in koi between 48-72 h post infection, while other cyprinids family was not caused in mortality. Other fresh water fish such as catfish Clarias batrachus, nila Oreochromis niloticus, koki Carrasius auratus, and komet Macropinna microstoma was not infected. Virus-like particles as found in gill ofinfected fish with KHV.

Abstract

A total of 10 mungbean (Vigna radiata (L.) Wilczek) genotipes including check variety (Vima) were evaluated on rainfed after rice at Ngale Experimental Farm, Ngawi, East Java during early dry season of 2010. The experiment was conducted in randomized complete block design with three replications. Plot size was 4 m x 5 m, plant spacing was 40 cm x 20 cm, each hill contained two plants. A correlated and path coefficient analyses were used in order to determine the association among characters and quantify the direct and indirect effects of agronomy traits on seed yield. The study has identified 2 promising lines, i.e. MMC342d-3-4 and MMC342d-kp-3-3 produced the highest yield of 2.2 t and 2.0 t/ha respectively and had good adaptation on rainfed. Two varieties (Kutilang and Kenari) having large seed size (>7.5g/100 seed). Therefore, these varieties can be used as source of parents on breeding program. Among the agronomic characters, number of pods per plant, number of seeds per pod and seed size were positively and significantly correlated with grain yield. The direct effect of number of pods per plant, number of seed per pod and seed size to grain yield as indicated by path coefficient were the highest, while other causal effects to grain yield were small or negative. Based on both analyses, mungbean genotipes with high grain yield should have high number of pods per plant, number of seeds per pod and large seed size. Therefore, number of pods per plant, number of seeds per pod and seed size can be suggested for selection criteria in selecting high yield mungbean genotipes on rainfed.

Abstract

The exploitation of pitcher plants (Nepenthes spp.) for economics interest and forest degradation make a higher extinction risk for Nepenthes globally. This research was aimed to examine anatomical structure of stomata of Nepenthes gracilis, N. reinwardtiana, N. ampullaria. The result showed that the type of stomata of N. gracilis and N. reinwardtiana has the actinocitic type while, N. ampullaria has anomocitic stomata type.

Abstract

Raistonia solanacearum,the bacterial wilt pathogen, has a wide host range and genetic variability.Rapid and sensitive molecular techniques need to be developed for eariy detection and strain differentiation of the pathogen.Molecular techniques such as PCR and DNA hybridization have been succesfully used to detect and identify bacterial plant pathogens including R.solanacearum.These techniques were adopted under Indonesian condition, using purified and crude DNA from infected plant samples.An R.solanacearum specific DNA primer (OH/Y2) was used in the PCR test,and a DNA probe 5a67 were used in the non-radioactive hybridization test.The PCR techniqe could be used to detect R.solanacearum from infected plant samples in less than 5 hours.The DNA hybridization technique was applicable to differentiate strains ofR.solanacearum into three groups based on their DNA profiles.

Abstract

Ethanol 95% and water extracts of Kaempferia spp. (K. galanga, K. angustifolia, K. pandurata and K. rotunda) were tested for their influences in in-vitro phagocytosis activity and phagocytosis capacity of mouse peritoneum macrophage cells induced by Staphylococcus epidermidis. Extracts were tested at a series concentration in logarithmic order (0.1-1000) ug/ml. Imboost and distilled water were employed as positive and negative controls, respectively. The results shows that almost all extracts were capable to increase on phagocytosis activity (SPA - Screening for Phagocytosis Activity) and phagocytosis capacity (IP • Index Phagocytosis) of macrophage cells compared to positive and negative controls significantly (p<0.05).The phagocytosis activity and capacity macrophage cells were increased by increasing extract concentration. K.rotunda extracts shows reveal better on above activities in low concentration (0.1-100) ug/ml then other species.

Abstract

Research on the effect of soil moisture on agronomic characters, yield and nicotine content of Temanggung tobacco has been conducted in the Research Institute for Tobacco and Fibre Crops at Malang, East Java from March to October 2010. The treatment has been arranged in a Splitplot design with 3 replications. The experimental plot consisted of three types of soil: Complex Eutrudepts-Hapludalfs, Vitraquands, and Complex Dystrudepts-Hapludalfs. The subplot comprised 5 levels of soil moisturation: 60, 70, 80, 90, and 100% of field capacities. The results showed that the dry slice yield, content of nicotine, and agronomic characters of the Temanggung tobacco were affected by the result of interaction between soil moisture and type. The decreasing of soil moisture up to 40% from field capacity in the Complex Eutrudepts- Hapludalfs had decreased dry slice yield, nicotine content, plant height, root length, dry weight of plant, shoots, roots, stems, leaves, flowers, and suckers accounted for 54.3, 19.3, 33.7, 17.1, 74.1, 71.4, 82.3, 80.1, 54.3, 60.5 and 86.5% respectively, in the Vitraquands 72.7,20.2, 37.0, 13.2, 79.8, 78.5, 84.3, 80.7, 72.7, 63.3 and 87.8% respectively, and in the Complex Dystrudepts-Hapludalfs 74.2, 23.0, 39.6, 15.7, 79.3, 78.0, 83.6, 80.3, 74.2, 79.1 and 80.6% respectively.

Abstract

Osteochilus jeruk is one of the two new species of Osteochilus occuring in Gunung Leuser National Park, Aceh, Sumatra, Indonesia.It has a big size and beautiful colour, so it is potential as food and aquarium fish. Notes on growth, food and reproduction were examined as base for the culture of this fish.

Abstract

Nitrogen fixing dan phosphate solubilizing bacteria are important PGPR bacterial in addition of plant nutrients by increasing N and P, especially in saline soils. Experiments were conducted to observe the effects of  soil salinity and bacterial inoculant on growth eggplant (Solanum melongena) at green house of  Microbiology division, Research Center for Biology, The Indonesian Institute of Sciences,  Cibinong. Experiment laid out as factorial based randomized complete design  with three replications. Five levels of watering consisted of (1). Fresh water, (2). NaCl 0.1%, (3). NaCl 1%, (4). Sea water 75 %+25% fresh water, (5). Sea water 100%  as first factors. Five levels of inoculations/biofertilizers consisted of (1).control without fertilizer (K), (2). NPK (P), (3). Nitrogen Fixing bacteria (BPN1), all treatments was repeated 3 times  BPN2, mixed inoculation (BPN1 +  BPN2 + Phosphate Solubilizing Bacteria/BPF) as second factors.  Results showed that biofertilizer application increased growth and yield of eggplant in various levels of salinity at about 21.56%-53.35%  compared to uninoculated plants

Abstract

The trace element Selenium is toxic at high concentration.Most of organisms living in selenium rich environment are selenium resistant.One of the resistance mechanisms is methylation,in which selenium is methylated and transformed to non-toxic selenium compound.The methylation is catalyzed by seleno methyltransferase (SMT) coded by smt gene. The gene are expressed by selenium tolerant plants. However, there was no available report yet on such specific gene in the bacterial genome. This study was carried out to determine smt homologous gene on selected selenium accumulator bacteria, Geobacillus sp. 20k, The smt gene of was determined by amplifying target DNA and analyses its sequences through homology search (BLAST). The result showed that the DNA and its protein part of thermophilic enzyme involved selenium metabolisms.
