Abstract

In order to develop yeast Candida tropicalis as a model cell for evaluation of substance having anti- or pro-oxidant activity in cell level, the effect of hot water tea {Camellia sinensis (LJ Kuntze) extract on peroxidized lipids, a marker of oxidative stress and cell mortality in the yeast caused by paracetamol has been evaluated. Incubation of yeast cell in the presence of 1.38 % green tea for 2 h decreased cell viability followed with increasing of peroxidized lipids, whereas 0.27 % green tea caused increasing of peroxidized lipids without causing cell death.Yeast cell was not affected by 0.1 % green tea. Incubation of yeast cell with presence of 0.15 % (g/v) paracetamol for 2 h did not cause cell mortality, however content of peroxidized lipids increased significantly. In the presence of higher dose of paracetamol (0.3 %) cell viability remarkably decreased and followed with increasing of peroxidized lipids significantly. Green tea of 0.1 % increased cell viability of cells treated with 0.3 % paracetamol while peroxidised lipids decreased. The data indicated that high dose of paracetamol caused oxidative stress in yeast cells, while green tea with lower concentration might protect paracetamol toxicity due to its antioxidant property. Although the antioxidant property, high dose of green tea could cause oxidation due to its pro-oxidant activity. In conclusion, yeast C. tropicalis could be potentially used as a model cell to evaluate substances having antioxidant property in cell level.

Abstract

Urban forest is one of the green open area consisted of natural forest-like vegetation. It has suitable microclimate for understory habitat, including terrestrial ferns. The diversity of fern in three urban forest in DKI Jakarta is reported, i.e the urban forest at The University of Indonesia (UI) Campus, The Cibubur Arboretum and PT JIEP (Jakarta Industrial Estate Pulogadung). Vegetation analysis was conducted by purposive sampling method with a transect line of 200 m in each location with 15 plots of (3x3) m2 each. Fern determination was done based on herbarium specimens and literatures. Fern diversity was analyzed based on Shannon diversity index, and equality index. Canonical correspondence analysis (CCA) was performed to determine the correlation between fern diversity and abiotic factors. There were 18 fern species classified into 11 genera and eight families, at the UI urban forest and Cibubur Arboretum, but there was no fern in PT JIEP. Fern with the highest importance value index was Sphaerostephanos sp. (161.33%) in The UI urban forest, and Lygodium microphyllum (Cav.) R.Br. (71.63%) in Cibubur Arboretum. The CCA result showed that the fern diversity correlated to four abiotic factors i.e temperature,humidity, light intensity and soil pH.

Abstract

The effect of cryoprotectants glycerol and sucrose on cell viability and fermentation rate of Saccharomyces cerevisiae after freezing (-30 °C) and Chawing (30 °C) were studied.Both freezing and thawing were done rapidly. The mortality of cells treated with low concentrations of cryoprotectans (2.5, 5, and 10 %) after 15 and 30 days of cryopreservation, was remarkably higher than that of control and higher concentration (20% and 40%).Glycerol or sucrose with concentration of 20 % and 40 % protected cells from severe mortality only after 90 days of cryopreservation.Fermentation rate of cells treated with 20 % or 40 % of the two cryoprotectants were higher than that of control after 60 and 90 days of cryopreservation.The data indicated that in certain circumstance cryoprotectant could be toxic for the cells during freezing and thawing.Since biomembrane is not permeable to sucrose, therefore we proposed that target of sucrose toxicity may be extracellular, whereas glycerol, which penetrate cells,targets of the toxicity may be both extracellular and intracellular domains.Interaction between cryoprotectant and cell membranes is discussed.

Abstract

The research was conducted to study the structure and composition of herbaceous and seedling communities (understorey species)occurred on herbivore habitat within Cikepuh wildlife sanctuary. A series of 25 plots of 1 m x 1 m each with an interval of 5 m were used to establish a species-area relationship. The proportion (percentage) of canopy (leaf) coverage of each species occurred within the sampling plots to the sampled soil surface area was measured. A total of 54 species belonging to 47 genera and 29 families occurred in the 25 plots develoved, with the species diversity index (value) of 2.09 using the Shannon Wiener formula. Nine species were categorized as herbaceous plants (16.67 %) while the other 45 species were categorized as seedlings (83.33 %). The average density measured was 39 individuals/m or c. 386.400 individuals/ha. The most dominant family is Euphorbiaceae (possessing 5 diffrent species) while the most dominant species is Urochloa subquadripara (Poaceae) with and importance value of 96.08%.

Abstract

Kelabau fish (Osteochilus sp.) is an endemic fish to Kalimantan inland waters that is potential to be developed. The aim of this research was to characterize Kelabau fish and to study the data base (genetic character) of kelabau as well as its relationship. The result showed that the highest polymorphism and heterozigosity was on Kelabau Sintang and the lowest was on Kelabau Kapuas Hulu. The closest genetic distance value were between Kelabau Pontianak-Kapuas Hulu (0.5351) and the furthest were between Kelabau Pontianak-Sintang (0.6852).

Abstract

Biodiversity of Basidiomycetes was studied in Kelila, Jayawijaya, Papua.The aim of this observation was to identify species diversity of Basidiomycetes and their utilities. Sixty one species were found in the location. The 19 species were edible mushroom while 7 species have been used as traditional medicines and some species have a common odours (smell) and 1 species (Clavatia sp.) contains clavatin which could be used as arm tumour agent.

Abstract

Nine foodstuffs from Enggano island were processed to flour as wheat flour alternative. To increase flour quality and its derivative product, Lactobacillus-plantarum B110 was added those flour and talam cake made from such flour. They were forest cassava (Dioscorea sp.), ararut sago (Marantha arundinacea), tacca (Tacca Leontopetaloides), egg taro (Alocasia sp.), oil taro (Alocasia sp.), gadung (Dioscorea hispida), gogo rice (Oryza sativa), corn (Zea mays), and belinjo (Gnetum gnemon).  The quality assessment consisted of HCN detection (qualitative), nutrition contents (proximate analysis), acid  (titration method), glucose (GOD-POD kit), and organoleptic tests (20 panelists). The results show that acid and glucose contents of the nine flours increased after L. plantarum B110 addition. Acid content of  the flours was 0.0144-0.2475%, while  glucose was  0.056-0.449%.  Carbohydrate, energy and acid contents of L. plantarum B110 talam cake were higher than control, which those were 34.13%, 190.33 cal/100gr, 0.00082%, while protein, lipid, water, ash were 1.75%, 5.22%, 57.9 %, 1.01%, respectively. The talam cake was accepted by panelists with values: 5.50 (taste), 5.6 (colour), 4.55 (flavour), 4.00 (texture), 4.35 (homogeneous). It was concluded that the flour and talam cake quality increased with addition of L. plantarum B110.

Abstract

Phosphorus (P) is an essential element for plants, deficiency and excess of P in soil will limit plant growth. It is required to applicate P fertilizer in appropriate dosage for promoting plant growth on the soil with different properties. The purpose of experiment was to study the respon of maize (Zea mays L.) for phosphorus fertilization on Regosol and Latosol soils. A glass house experiment was conducted to study maize response on P fertilizer application in Regosol and Latosol soils. The treatment 1was 0, 112, 336, 1008, 3024 and 9072 kg superfosfat ha . P fertilizer tend to increase the biomass of maize but exceed of P fertilizer reduced maize biomass. The optimum fertilization and vailability of P for plant affected by soil properties.

Abstract

Bacterial leaf blight (BLB) caused by Xanthomonas oryzae pv. oryzae (Xoo) is an important rice disease due to its high intensity that and  the mereased durability of its resistant variety. This study was aimed  to determine the pathogenicity of Xoo isolates, which obtained from four regions/districts in West Java on three rice near isogenic lines (NILs) containing a single resistance (R) gene; and  to study reaction of rice NILs/ differential genotypes containing mixture of resistant genes to Xoo population under endemic areas. Out of 22 Xoo isolates have been collected and further determined by ELISA assay. Ten Xoo isolates were selected and inoculated to identify their pathogenicity on three NILs i.e., IRBB5 (xa5), IRBB7 (Xa7) and IRBB21 (Xa21). Pathogenicity test showed that most of isolates produced large lesion, and four virulence groups were identified. Phylogenetic analysis suggested that the dominant virulent isolates were widely distributed at several district in West Java. It was shown that 11 NILs exhibited high levels of resistant reaction to the predominant Xoo pathotypes in Cianjur, while 10 lines were susceptible. The single R gene (IRBB7, IRBB21 and Java14) exhibited lower BLB severity. The IRBB50, IRBB51, IRBB52, IRBB53 lines (carrying two R genes), IRBB56, IRBB57 (carrying three R genes); IRBB64 (carrying four R genes), and IRBB66 (carrying five R genes) showed lower severity and thus produced higher resistance to Xoo. This study may implied further work to deploy effective R genes against certain Xoo pathotypes in differ  region.

Abstract

Shorea henryana - a member species of Dipterocarpaceae known as white meranti. Ecologically distributed on lowland Dipterocarp forest and flourishing in coastal areas. A fairly large tree up to 40 m tall with bole up to 115 cm diameter. Seeds were dispatched from Thailand and showing 72% germination and initial moisture content of 20.3%. First lot of seeds were dried to moisture content 12.5%, 9.4% and 6.1% respectively. Germination of seeds dried to 6.1% m.c. was still relatively high (68%). Contrary to the desiccation resistance, however, the dried seeds seemed not to withstand relatively low temperature for Shorea species, i.e. 14-16 C. Further study will be needed as to investigate a proper storage treatment for this important woody species.
