Abstract

ABSTRACTProfile of Small mammals from Gunung Slamet, Central Jawa. Research of small mammals atGunung Slamet, Central Jawa was conducted. Three types of habitats as representation ofprimary forest, secondary forest and plantation were examined at Kalipagu, Kaliwadas andBambangan in order to record its small mammals biodiversity. Combination of trapping anddirect observation recorded 31 species of small mammals from the areas observed. Shannon-Wiener index was the highest in secondary forest (3.8) compared to primary forest (3.6) andplantation (3.4). The results indicated that most small mammals of Gunung Slamet use secondaryforest for their home range. However data on microchiropterans that were more frequentlyrecorded in plantations indicated that agricultural plants were their primary foraging areas. Onthe other hand, most of small mammals species were rely on both secondary forest and primaryforest as an important food resources. The dendogram resulting from cluster analysis wasconcordant with this view. Primary forest and secondary forest clustered closely together,while plantation was recognized as a distinct group.Key words: Small mammals, Gunung Slamet, biodiversity, habitat.

Abstract

ABSTRACTA total of 781 base pairs of mitochondrial DNA ND2 gene was analyzed to investigate their divergence in the Arachnothera longirostra (Nectariniidae) birds in Jawa and Sumatera islands of Indonesia. Blood and tissue samples were collected from 27 birds consisted of 8 samples from T.N.G. Halimun, 6 samples from T.N.G. Gede Pangrango, 6 samples from T.N. Ujung Kulon, 1 sample from Dieng (Jawa), and 6 samples from Jambi (Sumatera). Total genomic DNA was extracted from each sample and used in PCR to amplify a single fragment of mDNA ND2 gene. Their DNA sequence data were compared with those from Johor (Malaya Peninsula) and Sabah-Sarawak Borneo (GenBank). There were 61 variable sites included 34 parsimony sites and 27 sequence haplotypes. Mean genetic distance or sequence divergences within bird populasions were ranged from the lowest (0.25 %) for Ujung Kulon to the highest (0.76 %) for G. Gede Pangrango; while mean genetic distance between populations were of 1.67% to 2.82 % for Java island vs Jambi (Sumatera), and 1.06 to 2.09 % for Java vs Borneo. Phylogenetic NJ tree showed that there were two main clades, i.e. 1) birds of Jambi (Sumatra), Johor (Malay Peninsula), Sabah-Sarawak (Borneo), and Ujung Kulon (Java), and 2) birds of G. Gede Pangrango, G. Halimun, and Dieng (Java). Past geologic event may influenced genetic divergence occurred within the populations. Moreover, geographic features such as mountain may played role for population isolation as showed on the Little Spiderhunter bird from G. Halimun, G. Abstract

Triploid banana breeding could be achieved by crossing tetraploid to diploid banana cultivars. The availaibility of tetraploidbananas is naturally very limited. One of methods to produce tetraploid bananas is inducing tetraploid plants usingchemicals that inhibit spindle fiber formation during mitosis such as oryzalin or colchicine. The present research is aimed toproduce and characterize tetraploid Pisang Mas Lumut (Musa acuminata, AAAA genome) induced by in-vitro oryzalintreatment from diploid Pisang Mas Lumut. Fifteen in vitro-cultured shoots were treated wih oryzalin at of 60μM for 7days in a liquid MS basal medium with addition of 2 mg/l BA. After 6 sub-cultured, 54 shoots and 104 plantlets wereproduced. Forty seedlings were then survived and grew well and healthy. Ploidy identification of induced Pisang MasLumut was conducted using Flowcytometer at seedling stage and repeated at reproductive phases for 2 cycles. The resultsshowed that oryzalin treatments produced tetraploids at a frequency of 30%, diploid at 65% and mixoploid at 5%. Thetetraploid plants have longer and thicker pseudostems, produce fewer suckers, drooping and larger leaves, and broader fruitscompared to its diploid control at harvest time. The tetraploid plants have similar genetic properties with their diploidcontrols as shown by genetic identity of 0.90-0.97.Key words: Pisang Mas Lumut, Musa acuminata, AA, oryzalin, tetraploid

Abstract

ABSTRACTPyrene is a high molecular weight chemical compound belongs to polycyclic aromatic hydrocarbon (PAHs) group that are difficult to degrade by environment. Biodegradation techniques using indigenous marine bacteria are used to be as an effort to reduce pollutants that are carsinogenic. The objectives of this research are to screen of 18 marine bacteria isolates qualitatively by sublimation method and quantitatively by growth test and to optimize degradation activity of marine bacteria isolates by pyrene concentration and cell concentration. Identification by 16S rDNA and phylogenetic tree analysis were conducted to determine the molecular basis of bacterial identity. The result of sublimation showed that 15 isolates were positive result for pyrene degradation and classified to 3 groups. The first group consisted of 5 isolates that can produce clear zone, while the second group are 5 isolates with isolate color changes. The third group have both of activities. Growth test showed that isolate LBF-1-0132 has high potency to degrade pyrene compound. Isolate LBF-1-0132 is capable of degrading pyrene compounds optimally at concentration of 600 ppm and optimum cell concentration of 20. Based on 16S rDNA gene analysis, isolate LBF-1-0132 is Pseudomonas aeruginosa with 98% identity.Keywords :pyrene, marine bacteria, optimization, 16S rDNA identification

Abstract

The aims of this research is to isolation of bacteria that potential to produce of milk clotting protease enzymes fromfermented food that will be used as a substitute for rennet in cheese making. There are five food fermentations suchas tauco, tempeh, red oncom, sticky tape, and pickled mustard greens that are used as a source for isolation of bacteriathat could produce milk clotting protease. The results obtained four isolates proteolytic bacteria from two fermentedfood samples, three isolates bacteria from tauco (TCN 1, TCN 2, TCN 3) and one isolate from pickledmustard greens (DSN 1). Based on 16S rDNA, these isolates were identified as Bacillus sp. Bacterial isolate TCN 1has a milk clotting activity of 29.17 U/mL, whereas bacteria isolates of TCN 2, TCN 3 and DSN 1 have activities of70 U/mL achieved at the 24 hours incubation, respectively. The proteolytic activities of bacteria isolates TCN 1,TCN 2, TCN 3 and DSN 1 at the 24 hours fermentation process were 0.0117 U/mL, 0.0021 U/mL, 0.0150 U/mL,and 0.200 U/mL, respectively. The ratio of milk clotting protease activity and the proteolytic activity for bacteriaisolates TCN 1, TCN 2, TCN 3 and DSN respectively were 5402, 175000, 7292, and 3333. This showed that theenzyme from bacterial isolates TCN 2 can be used as an alternative to rennin in cheese making.Keywords: milk clotting protease, cheese, calf rennet, fermentation food

Abstract

Dinamika Komposisi Spesies Struktur Pohon dan Biomasa di Hutan Terganggu di Kalimantan Timur. Kajian dinamika komposisi spesies dan biomasanya dilakukan selama 4,3 tahun (Desember 2004 - April 2009) di hutan terganggu Kalimantan Timur. Untuk melakukan kajian di gunakan 6 plot permanen (100x100 m2), yang berada di hutan penelitian Samboja. Semua pohon berdiameter > 10 cm dbh (diukur pada posisi 1,3 m diatas tanah). Pada Desember 2004,tercatat 2143 pohon tersebar di plot tersebut terdiri dari 39 famili, 82 genus dan 111 spesies. Pada pengamatan April 2009 tercatat 2466 pohon terdiri dari 40 famili, 86 genus dan 123 jenis.Sebagian besar spesies yang mendominasi adalah Garcinia nervosa dan Trigonostemon laevigatus tetapi setelah pengamatan pada tahun 2009, keduanya tidak lagi mendominasi. Selama kurun 4,3 tahun terjadi penambahan kepadatan 15,1%, basal area 12,9% dan biomasa 11,6%. Penambahan kepadatan tercatat meningkat dari 357 menjadi 411 pohon/ha. Basal area meningkat mulai dari 20,09 menjadi 22,67 m2 ha-1, sedangkan biomasa meningkat dari 286,3 menjadi 319,4 ton ha-1.Kata kunci: Komposisi, spesies, biomas, hutan terganggu

Abstract

ABSTRAKAnalisis Kebutuhan Nutrien dan Efisiensi Penggunaan Pakan pada Lutung Merah (MaroonLeaf Monkey Mueller, 1838). Penelitian ini bertujuan untuk mempelajari kebutuhan nutriendan efisiensi penggunaan pakan pada lutung merah (Presbytis rubicunda). Penelitian telahdilakukan di Penangkaran Mamalia Kecil Bidang Zoologi, Pusat Penelitian Biologi ? LIPI selama38 hari yang terdiri dari 10 hari masa adaptasi pakan dan 28 hari (4 minggu) masa pengumpulandata. Obyek yang digunakan adalah dua ekor lutung merah terdiri dari satu ekor jantan dansatu ekor betina. Pakan alternatif yang diberikan selama penelitian adalah daun beringin(Ficus benyamina), daun selada (Lactuca sativa), kangkung (Ipomoea aquatica), buncis(Phaseolus vulgaris), markisa (Passiflora quadrangularis), ubi jalar (Ipomoea batatas), danpisang lampung (Musa sp.). Parameter yang diamati adalah konsumsi pakan, pendugaankebutuhan nutrien, dan efisiensi penggunaan pakan. Jenis pakan yang paling palatabel bagilutung merah jantan adalah ubi jalar sedangkan bagi lutung merah betina adalah buncis. Rataankonsumsi pakan berdasarkan bahan kering adalah 78,09 gram/ekor/hari. Rataan konsumsinutrien adalah abu 4,60 gram/ekor/hari, protein kasar 7,87 gram/ekor/hari, lemak kasar 3,05gram/ekor/hari, serat kasar 10,56 gram/ekor/hari, bahan ekstrak tanpa nitrogen 51,40 gram/ekor/hari, dan bruto energi 2948,50 kal/ekor/hari. Rataan pertambahan bobot badan lutungmerah selama penelitian adalah 6,25 gram/ekor/hari dengan efisiensi penggunaan pakan 15,84%.Data konsumsi nutrien tersebut dapat digunakan sebagai dasar penyusunan ransum dan pakanbagi lutung merah di penangkaran.Key words: Consumption, feed utilization, Presbytis rubicunda, captive breeding

Abstract

ABSTRACTA green house experiment were conducted to study the application of ?compost plus? on the growth of Orthosiphon aristatus. The experiment was designed in Complete Randomized Design with factorial and five replicates. The first factors were fertilizer application (compost plus, compost, and control). The second factors were 3 kinds of soil (i.e. soil from Cibinong, Ciomas, and Sukabumi). The result showed that compost plus application increased 113.90 gram/pot of dry weight of Orthosispon aristatus leaves compared with compost application and control in 3 kinds of soil.Key words : Compost plus (compost + microbes), Orthosispon aristatus

Abstract

The current study is the first report on intraspecies analysis of L. plantarum from sayur asin in Indonesia using molecular approach. Three molecular techniques, i.e., restriction fragment length polymorphism (RFLP) 16S-23S rDNA intergenic spacer region (ISR), random amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR) and enterobacterial repetitive intergenic consensus (ERIC-PCR) were used to determine the intraspecies diversity of L. plantarum responsible for spontaneous fermentation in sayur asin. These methods were aimed to discriminate 46 isolates of L. plantarum isolated from sayur asin, including the type strain. PCR amplification of the 16S-23S rDNA ISR revealed two-bands profile of 800 and 600 bp specific to lactobacilli. RAPD-PCR and ERIC-PCR were very valuable in discriminating genetic polymorphism among L. plantarum isolates by producing bands ranged from 4-10 bands (360-2620 bp) and 6-12 bands (160-2900 bp), respectively. Dendograms generated from UPGMA cluster analysis based on RAPD-PCR and ERIC-PCR data showed that all isolates were grouped into three major clusters with 74% and 68.6% genetic similarity thresholds, respectively.The study indicated that strains belong to L. plantarum isolated from sayur asin were divided into three genotypic groups. Keywords: ERIC-PCR, Intraspecies, Lactobacillus plantarum, RAPD-PCR, RFLP 16S-23S rDNA ISR
