Abstract

The rate and risk factors on protective immunity against rabies were conducted in Makassar city. Theaims of this study were to determine the level of immunity of dog and find out the association betweenmanagement factors and the level of maintenance of the dog immunity against rabies. Two hundred andforty-six serum samples were collected for measurement of dog immunity level with a method of enzymelinked immunosorbent assay (ELISA). Maintenance management data were gathered throughquestionnaires and direct observation. Analysis of data obtained by the level of immunity was donedescriptively. The significance association between the risk factors and the strength of immunity level wascalculated by using Chi square (X2) and odds ratios. The results showed only 12.2% of dogs had a protectiveantibody against rabies at > 0.5 IU/ml. Factors affecting the level of protective immunity against rabieswere the first vaccination at 1-6 months age (OR = 18.6), post-vaccination after 1-6 months (OR = 6.0), themaintenance location of dogs in urban areas (OR = 4.5), dog caring management in house (OR = 3.8), andowner income above Rp. 2.000.000/month (OR = 3.0). It is concluded that the low level of protective immunityagainst rabies and the dog bites reported each year indicates that Makassar, as endemic areal of rabies,is a threat to the outbreak of the disease in animals and humans to the surrounding areas.

Abstract

The purpose of this study was to determine the characteristics of blood flow which passed through theaortic valve of normal Indonesian mongrel dogs using trans-thoracic Pulsed Wave Doppler (PWD)echocardiography. Eight (8) adult dogs (three male dogs and 5 female dogs) with an average age 3.31 ± 0.59years and average body weight 12.45 ± 1.30 kg were used in the study. Trans-thoracic PWD echocardiographyexaminations were performed in conscious/unsedated animals at the left lateral recumbence position. Theinstruments used in this study were: i) two dimensional ultrasound device and ii) phased array transducersmall foot print with 3.7 – 5.0 MHz frequency. Six elements of PWD were measured: i) heart rate (HR); ii)peak velocity (Vpeak); iii) velocity time integral (VTI); iv) mean pressure gradient (MPG); v) pulsatilityindex (PI); and vi) ratio of systole-diastole (S/D). Results showed the typically characteristics of blood flowin semilunar aorta valve is demonstrated by the only one peak wave (peak velocity = Vpeak) upon examinationby placing the Doppler scan volume right on the aorta valve. In general, all six elements of PWD measuredwere found to be higher in female dogs compared to male dogs. However, these differences were notstatistically significant (P>0.05).

Abstract

The aim of this study was to investigate the expression of cytokines profiles interferon-? (IFN-?),interleukin-5 (IL-5) and IL-10 in Rasciola gigantica resistant Indonesian thin-tailed (ITT) sheep comparedto susceptible merino sheep infected with Fasciola gigantica. A total of ten ITT and merino sheep wererandomly allocated into infected (n=5) and control (n=5) groups, sheep were infected with 250 viablemetacercariae of F. gigantica. The cytokines were determined by isolated mRNA from hepatic lymph nodeby semi-quantitative RT-PCR (Reverse Trancriptase -Polymerase Chain Reaction). The result showedITT sheep produced significantly higher IL-5 and 10 ( P<0,05 ) than merino sheep, while ITT sheepproduced less IFN-? ( P<0,05) than merino sheep at 10 weeks post infection. It could be concluded thatmerino sheep tend to develop T type 1 cells, while the ITT sheep tend to develop T type 2 cells whicheffectively killed F. gigantica.

Abstract

Aeromonas hydrophila is a gram negative and opportunistic bacteria, which could cause fish mortalityin a short time from 80%-100%. One virulent factor of A. hydrophila on common carp (Cyprinus carpio L)that could cause fish mortality is aerolysin. This research used a synthetic primers of oligonukleotide todetect aerolysin, a specific genomes of A. hydrophila on common carp (Cyprinus carpio L). The commoncarps have been feed a woof that contain garlic extract during 30 days before they challenged with A.hydrophila. Polymerase Chain Reaction (PCR) was used to detect an aerolysin gen from A. hydrophila. Theelectrophoresis result showed aerolysin gene of Aeromonas hydrophila from Veterinary Faculty of GadjahMada University (FKH-UGM) isolate was amplified with 462 bp of molecule weight. While the aerolysingen was detected in the fish kidney with 900 bp of molecule weight. Further, DNA sequence analysis of thePCR product of A. hydrophila from FKH – UGM isolate showed homolog with isolate A. hydrophila subsphydrophila ATCC 7966 complete genome with score 55.4 (71%).

Abstract

Sorghum is a cereal crop that could be use as an alternative source in food diversification strategy. Ithas high carbohydrate content similar to rice. Its protein content is higher than that of rice and this crophas the potential to be produce in increasing quantities in Indonesia. Nowadays, the demand of foodindustry is not only concern for nutritious food but also for health benefits. Several research reports haveshowed that sorghum contains bioactive compounds that are beneficial to human health such as in decreasingthe risk of degenerative diseases. In vitro studies have shown that this cereal crop has the capacity toincrease lymphocytes proliferation. The objective of this study is to study the biological potency of sorghumin lymphocytes cell proliferation in rats. Rats were grouped into three: i) control group animals; ii) animalswere fed 100% of sorghum diets, and iii) animals were fed 50% of sorghum diets. The results showed thatan increase in lymphocytes cells proliferation activity by 63% and 70% were seen in animals that were fed100% and 50% sorghum diets, respectively.

Abstract

The use of Duddingtonia flagrans as the biological control of nomatode infections has been widelyreported. However, no report is available on the use of yeast Saccharomyces cerviciacae for such purpose.The aim of this study was to ivestigate the use of both fungi to reduce the number of Heamoncus contortusinfective larvae. Agar and fecal media containing the spore of the fungi was inoculated with infected H.contortus larvae (3rd stage). Fecal media containing the fungi was prepared by oral inoculation of sheepwith liquid containing 106, 107 spores of D. flagrans, and 106, 107 spores of D. flagrans, and 106, 1012 sporesof S. cerviciae. The number of larvae trapped in the fungi was counted. The result showed both fungi wereable to reduce the number of infective lave. However, for D. flagrans, beside it able to kill the larvae, it alsoable to trap the larva which did not occur in S. cerviceae. The combination of both fungi can be used to reduceof the number of invected H. contortus larvae.

Abstract

Study in order to develop molecular techniques using specific primers for the early diagnosis oftoxoplasmosis have been conducted. Detection of Toxoplasma gondii genome was performed usingpolymerase chain reaction (PCR) technique. The primers used in this study were rDNA, P30, and B1. ThePCR products were further run using gel electrophoresis (gel 1.5% – 2.0%) and the band was documented.Toxoplasma was detected at 500 bp and 600 bp using primer P30 and B1, respectively. Whereas usingprimer rDNA no band was observed. It was assumed that primer rDNA was not sensitive since the targetamplification was 88 bp.

Abstract

Noni fruit (Morinda citrifolia) has been used extensively as a drug, which is advertised with a varietyof benefits, such as can increase endurance, improve productivity. Those statements have not been provedscientifically. The results of this study, are expected to provide some information to the public regardingthe true benefits of Morinda citrifolia. The aims of this research were to study the effect of Noni (Morindacitrifolia) fruit extract given to broilers to their antibody titers and the counts of lymphocytes, basophils,eosinophils, monocytes, heterophils, and leukocytes. Production performance of these chickens was alsostudied. One hundred and sixty Cobb’s strain of broilers were used in this researh and were divided intofour groups of treatments which were divided into four experiment units which each comprised of tenchickens. Mengkudu fruit extract was given for 14 consecutive days orally using doses 0.0 g, 0.1 g , 0.5 g, and1.0 g per kilogram of body weight (kbw). The chickens utilized as control (0.0 g /gbw) were given only thesame plain water which was used to extract the fruits. The blood sampling were collected at the 3rd, 5th, 14th,and 21st days of the research. The study showed mengkudu fruit extract given at dose of 0.5 g/kbw was ableto elevate immune response (antibody) significantly if given in seven succesive days, and maintain toperform as an antiallergic when the extract given at dose 0.1, 0.5, dan 1.0 g/kww for 5 consecutive days. Butthe effect on the production performance had never been observed at all level of the dose and duration ofobservation.

Abstract

Isoflavone compounds have been known to have biological effects in improving health. The purpose ofthis study was to determine the bioavailability of isoflavone in plasma and its role as lowering bloodglucose levels in hyperglycemia rats. Bioavailability of isoflavones in plasma was performed in 18 ratsstrains Spraque Dawley age 3 months with 200 g of body weight on average and were given 300 ppm oralof isoflavone single dose. The analysis of isoflavon as done at 0, 1, 6, 12, 18 and 24 hours using HPLCmethod. Study effect on blood biochemical values used 25 male rats, which was divided into 5 treatmentgroups. One group as a negative control (K1) was given aquades; positive group hyperglycemia (K2), and 3other groups (K3, K4 and K5) were a positive groups of hyperglycemia and given isoflavone different doses,i.e. 1.0, 1.5 and 2.0 mg/200g bw per oral. Blood glucose were analyzed by using glucose oxidase biosensor,while cholesterol, HDL (Hight-density lipoprotein) and triglycerides were analyzed enzymatically using aspectrophotometer. The results showed the analyzes isoflavone genistein was more efficiently absorbedthan daidzein and glisitein. Isoflavone as found at highest concentration in plasma at 6 hours after oraladministration i.e: genistein of 11,17 ppm, daidzein of 2,86 ppm and glisitein of 2,47 ppm. Theadministration isoflavones at dose 2 mg/200g bw could decrease blood glucose levels in hyperglycemiarats, and also has hipocholesterolemia effect.

Abstract

The aims of this study was to determine the presence of Toxoplasma gondii in free-rangechicken using Polymerase Chain Reaction (PCR) method based on the tachyzoite and bradyzoitestage specific genes. SAG1 and BAG1 are the tachyzoite and bradyzoite stage-specific gene respectively.The primers for SAG1 and BAG1 were designed using Web-base Program Primer 3. Genomic DNAfrom free-range chicken heart and brain was isolated using Pure-Link Genomic Isolation Kit.DNA amplification by PCR using primers for SAG1 and BAG1 genes was used for diagnosis ofT.gondii. The results showed that the DNA amplification using primers for SAG1 and BAG1 geneswas successfully applied to determine of Toxoplasma gondii in free-range chicken.
