Abstract

Garcinia merupakan tanaman khas daerah tropis yang memiliki banyak manfaat. Manfaat ini telah diperoleh dari pengalaman empiris masyarakat setempat yang telah menggunakan buahnya sebagai makanan dalam bentuk buah-buahan, bumbu dapur, maupun sebagai obat untuk penyakit-penyakit tertentu. Penelitian terhadap kandungan senyawa kimia dari tanaman tersebut telah banyak dilakukan. Bahkan, beberapa komposisi maupun formulasinya telah menghasilkan paten, dan produknya telah banyak beredar di pasaran. Untuk menghasilkan paten yang dimulai dari penelitian memerlukan waktu yang cukup panjang.

Abstract

Diabetes mellitus (DM) is a disease that can be identified by high levels of blood glucose. Garcinia plants have been widely used for many traditional medicines as antioxidant, anticancer, antidiabetics, and antiinflammation. The antioxidant and antidiabetic activities of (Garcinia picrorrhiza Miq.) or sesoot fruit extract were evaluated in this study and compared with xanthone. The antioxidant and antidiabetic of ethanolic ripe sesoot (G. picrorrhiza Miq.) fruit extract (GpKar) was evaluated by (ABTS) reducing activity, α-glucosidase, β-glucosidase, and α-amylase inhibitor activity. GpKar showed higher ABTS-reducing activity (IC50 = 49.30 µg/mL) than xanthone (IC50 = 404.30 µg/mL). GpKar showed IC50 = 109.32 µg/mL for α-glucosidase inhibitory activity, while xanthones had a better activity (IC50 = 33.97 µg/mL). GpKar also showed lower α-amylase inhibitory activity and  β-glucosidase (IC50 = 126.01 and 9432.09 µg/mL) compared to xanthone (IC50 = 44.32 and 405.03 µg/mL, respectively). The compounds of GpKar are proven to have antioxidant and antidiabetic activities. Therefore, it will be industrially relevant to develop a natural medicine for decreasing DM risk, thus evaluating the antioxidant and antidiabetic effect of G. picrorrhiza by a pre-clinic study is needed.

Abstract

Oxidative stress has been known to contribute to Alzheimer’s disease. Acetylcholinesterase (AChE) enzyme may lead to Alzheimer’s disease as a neurotransmitter. Antioxidants may have protective activities against oxidative damage and Alzheimer’s disease. Acetylcholinesterase inhibitors also can be used in the treatment of various neurological disorders for management of Alzheimer’s disease. This study aimed to determine antioxidant and anticholinesterase effects of Garcinia picrorrhiza Miq. fruit extract (GpKar) and its compounds, xanthone. Antioxidant activity was measured by H2O2 scavenging inhibitory activity, while anticholinesterase activity was measured using modified Ellman method. GpKar has higher H2O2 scavenging inhibitory activity (IC50= 967.28 µg/ml) compared to xanthone (IC50= 1198.95 µg/ml). In the anticholinesterase inhibitory activity, GpKar has lower activity (IC50= 70.25 µg/ml) compared to xanthone (11.80 µg/ml). In summary, GpKar has higher antioxidant activity but lower anticholinesterase activity compared to its compounds, xanthone. However, GpKar has potency as antioxidant agent to prevent Alzheimer’s disease.

Abstract

Obesity has a role in the development of diseases such as diabetes, cardiovascular disease and hyperlipidemia which is characterized by the increase of adipose tissue mass due to an imbalance of energy intake and expenditure. Freeze-dried fruits are well known to possess antiobesity activity. In this study, we have evaluated the antiobesity activity of freeze-dried fruit (M. domestica, Canarium sp., and A. bilimbi) using CHOL, G6PDH, TG level, and Oil Red O assay. The viability of 3T3-L1 cell in the Canarium sp. freeze-dried in the concentration of 12.50 µg/ml has a higher value compared to M. domestica and A. bilimbi. The measurements of CHOL, G6PDH, TG level, and Oil Red O assay of the M. domestica freeze-dried in the concentration of 75 μg/ml has higher inhibitory activity compared to the Canarium sp. and A. bilimbi freeze-dried. In the CHOL assay, M. domestica freeze-dried has a higher value compared to A. bilimbi and Canarium sp. In the G6PDH assay, the freeze-dried of M. domestica has the value of 49.56%, Canarium sp. (45.22%), and A. bilimbi (47.13%), while in the Oil Red O assay, M. domestica has inhibition activity of 62.63%, A. bilimbi 50.01% and Canarium sp. 44.13%. The level of TG showed that M. domestica has higher activity with the value of 60.61%, A. bilimbi 57.54%, while Canarium sp. 55.03%. The freeze-dried of M. domestica in the concentration of 75 μg/ml has good inhibitory activity of lipid compared to A. bilimbi and Canarium sp.

Abstract

BACKGROUND: The number of obesity cases is still increasing worldwide and has reached an epidemic scale. Plants are known to have a protection role in the development of obesity, however their antioxidant and anti-obesity activities have not widely known. This study was conducted to assess the in vitro antioxidant and anti-obesity activities of different types of freeze-dried fruits cultivated in Indonesia, especially Canarium sp., Averrhoa bilimbi L. and Malus domestica.METHODS: Total phenolic content of freeze-dried fruits was identified by Folin-Ciocalteu method, while the total flavonoid content was measured by aluminium chloride colorimetric assay. To assessed the antioxidant activity, 2,2-diphenyl 1-picrylhydrazyl (DPPH) scavenging activity assay and 2,2’-azino-bis (3-ethylbenzothiazoline6-sulphonic acid) (ABTS) reducing activity assay were performed. The α-amylase and lipase inhibitory activity assay were performed to assess the anti-obesity activity. For comparison, hydroxycitric acid (HCA) compound was also assessed with DPPH, ABTS, α-amylase and lipase assays.RESULTS: A. bilimbi had the highest total phenol content (6.35 µg GAE/mg), meanwhile M. domestica had the highest total flavonoid content (2.06 µg QE/mg). A. bilimbi also showed the highest antioxidant activity both in DPPH and ABTS assay, with inhibitory concentration (IC50)=279.99 µg/mL and 631.78 µg/mL, respectively. The freeze-dried M. domestica had the highest anti-α-amylase activity (IC50=258.85 µg/mL), while Canarium sp. had the highest anti-lipase activity (IC50=118.66 µg/mL).CONCLUSION: Freeze-dried fruits demonstrate in vitro benefits toward obesity. A. bilimbi has potent antioxidant activity and is beneficial against obesity-related adverse health effect by relieving oxidative stress. M. domestica and Canarium sp. hamper the fat accumulation by reducing the carbohydrate absorption and dietary lipid.KEYWORDS: antioxidant, anti-obesity, Canarium sp., Averrhoa bilimbi L., Malus domestica, hydroxycitric acid

Abstract

Latar Belakang: Senyawa radikal bebas sering kita jumpai pada kehidupan sehari-hari yang dapat berdampak buruk terhadap kulit. Antioksidan dibutuhkan untuk meredam senyawa radikal bebas terhadap kulit. Buah delima merah merupakan buah yang terdapat dalam Al-Qur’an dengan antoksidan berupa flavonoida, asam fenolat dan tanin. Penuaan kulit dapat dicegah dengan menggunakan masker wajah yang mengandung antioksidan. Tujuan: Mengetahui aktivitas antioksidan pada kulit delima merah dalam bentuk masker wajah bubuk melalui reaksi antara ekstraknya dan mengetahui tinjauannya menurut pandangan Islam terhadap kulit buah delima merah yang diolah menjadi masker wajah dan diuji aktivitas antioksidannya dengan metode DPPH. Metode: Kulit delima merah dikeringkan, kemudian digerus dengan blender agar mendapatkan bubuk kulit delima merah. Bubuk kulit delima merah yang telah diperoleh direndam dengan etanol 70%, lalu disaring dan filtratnya dipekatkan menggunakan vacuum rotary evaporator sehingga didapatkan ekstrak kental yang selanjutnya dikeringkan dengan menggunakan oven sehingga didapatkan ekstrak bubuk kulit delima merah. Penetapan aktivitas antioksidan ini dilakukan melalui pendekatan pengukurannya terhadap ekstraknya karena masker wajah yang merupakan bubuk kulit delima merah tidak dapat ditetapkan dengan metode DPPH. Hasil: Aktivitas antioksidan dinyatakan dengan nilai IC50 pada kulit delima merah yang dijadikan masker wajah bubuk mempunyai nilai IC­­50 sebesar 8,33 ppm sedangkan asam askorbat sebagai kontrol positif memiliki nilai IC­­50 sebesar 2,98 ppm. Kesimpulan: Ekstrak kulit delima merah mempunyai aktivitas antioksidan dan termasuk kategori antioksidan yang sangat kuat dan penelitian ini sejalan dengan perintah Allah SWT yang menyarankan umatnya untuk menjaga kesehatan dan kecantikan diri.

Abstract

Garcinia is a plant belonging to the family of Guttiferae or Cluciaceae and has more than 180 famous species. Most of Garcinia species contains many prenelated xanthones, biflavonoids, and benzophenones. Using Liquid Chromatography - Mass Spectrometry (LC-MS), the development of electrospray ionization (ESI) could be detected and could be applied to a wide range of molecular biology such as xanthones, biflavonoids, and benzophenones and their derivatives. This method with a high speed of scanning at once could be measured a high degree of multiplexing and many components can be measured with a single analysis. These results were separated and analyzed using literature software of Dictionary of Natural Product 2006. The ethanolic extract of ripe fruit (IC), the n-hexane fraction of half ripe (IIB), and the water fraction of half ripe fruit (IVB) of sesoot (G. picrorrhiza Miq.) showed that the major compounds of them were xanthones, benzophenones with their derivatives,  furanone and pyranone which were proven as cytotoxic to the brine shrimp nauplii using Brine Shrimp Lethality Test (BSLT) and to the doxorubicin-resistant breast cancer cell line of MCF-7, the doxorubicin-sensitive breast cancer cell line of T47D.

Abstract

High total cholesterol levels are a major risk factor for cardiovascular disease. Consumption of red meat and offal, which are high in saturated fat and cholesterol, can increase total blood cholesterol levels. Dietary patterns in urban communities, including academic staff, show a tendency toward increased consumption of high-fat animal products. From an Islamic perspective, adhering to a halal and thayyib diet and avoiding excess are essential principles in maintaining health, including regulating the intake of red meat and offal. This research used a quantitative method with a prospective cohort design involving 14 days of consumption recording using an estimated food record among 38 male respondents aged < 60 years. Total cholesterol examinations were conducted before and after the study using the enzymatic method with the Mindray BS series chemistry analyzer. Normality tests were performed using Shapiro–Wilk and Kolmogorov–Smirnov. The correlation between red meat consumption and total cholesterol was analyzed using Pearson’s correlation test, while offal consumption was analyzed using Spearman’s correlation test. The study results indicated a significant positive relationship between red meat consumption and changes in total cholesterol levels (r = 0,385; p = 0,017). Offal consumption also showed a significant positive relationship with changes in total cholesterol levels (rₛ = 0,419; p = 0,009). Most respondents experienced a decrease in total cholesterol levels after the study, although higher consumption of red meat and offal remained associated with increases in cholesterol levels. There is a significant relationship between the consumption of red meat and offal and total blood cholesterol levels. The higher the consumption of both, the greater the increase in total cholesterol. These findings align with Islamic principles emphasizing the importance of consuming food that is halal, thayyib, and not excessive in order to maintain physical health.

Abstract

Probiotik dapat memodulasi sumbu sumbu mikorobiota usus-otak, sehingga mendapat perhatian dalam penelitian kesehatan neurosains. Produksi asam lemak alifatik rantai pendek (SCFA), yaitu fraksi asetat, propionat, dengan butirat merupakan mekanisme kunci yang menjadi dasar terjadinya efek biologis tersebut. SCFA mengatur inflamasi, integritas sawar usus, dan fungsi sistem saraf pusat. SCFA diketahui mempengaruhi ekspresi faktor neurotrofik dari otak (BDNF). BDNF adalah suatu neurotrofin penting yang terlibat dalam neuroplastisitas, neurogenesis, dan fungsi kognitif. Selain itu, SCFA berkontribusi pada patofisiologi neuropsikiatri dan gangguan neurodegeneratif. Tujuan dari review literatur sistematis ini adalah untuk mengevaluasi peran probiotik dalam pengaturan SCFA dan efeknya terhadap pengaturan BDNF. Dengan mengikuti pedoman PRISMA, Penelusuran eksplorasi literatur ditempuh secara metodologis melalui berbagai basis repsositori ilmiah yaitu PubMed, Scopus, dan Web of Science. Studi yang disertakan meliputi penelitian eksperimental pada hewan, uji klinis terkontrol pada manusia, yang menelaah hubungan antara suplementasi probiotik, perubahan kadar SCFA, dan BDNF. Hasil penelusuran menunjukkan bahwa peningkatan produksi SCFA, baik tunggal maupun multi-strain, dapat dicapai dengan suplementasi probiotik Namun demikian, keanekaragaman desain penelitian dan karakteristik intervensi tetap menjadi keterbatasan.Akhirnya, dalam memodulasi SCFA dan BDNF, probiotik dapat berfungsi sebagai strategi eskalasi kesehatan neurosains.

Abstract

Sarkopenia merupakan sindrom geriatri terkait penuaan yang ditandai oleh penurunan massa, kekuatan, dan fungsi otot rangka. Beberapa jalur molekuler, termasuk FSTL1, mTOR, dan BDNF, berperan penting dalam regenerasi, metabolisme, inflamasi, dan fungsi neuromuskular otot. Studi ini bertujuan untuk secara sistematis mengkaji hubungan keempat biomolekul tersebut dalam proses patogenesis sarkopenia serta keterkaitannya dengan intervensi latihan fisik, terutama latihan ketahanan dan latihan fleksibilitas. Pencarian bahan literatur dilakukan melalui PubMed, Scopus, dan ScienceDirect dengan memilih penelitian eksperimental, klinis, dan tinjauan yang relevan. Hasil analisis menunjukkan bahwa berkurangnya latihan fisik berhubungan dengan terganggunya miogenesis, sementara peningkatan FSTL1 akibat latihan berfungsi sebagai myokine yang melindungi melalui aktivasi jalur PI3K/Akt dan AMPK. Disregulasi mTOR berkontribusi pada resistensi anabolik pada orang lanjut usia, sementara penurunan BDNF memperburuk masalah neuromuskular dan metabolisme pada mitokondria. Latihan ketahanan terbukti paling ampuh dalam meningkatkan mTORC1, sintesis protein otot, serta merangsang pelepasan myokines, sedangkan latihan fleksibilitas membantu meningkatkan aliran darah, elastisitas jaringan, dan ketersediaan bio NO. Sebagai kesimpulan, interaksi FSTL1, mTOR, dan BDNF berperan penting dalam patogenesis sarkopenia. Latihan fisik terstruktur berpotensi menjadi terapi nonfarmakologis untuk memperlambat progresi kondisi ini.
