Abstract

Abstract A popular natural resource of Sumbawa Island is macroalgae. Chlorophyta, Phaeophyta, and Rhodophyta are types of macroalgae that have been cultivated and utilized by the local community. However, its potential to treat many diseases is still under-explored. Based on the content of bioactive compounds, macroalgae is believed to have antioxidant activity that could scavenge free radicals and thus potentially prevent and treat various diseases caused by excessive free radicals. This study aimed to analyze the antioxidant activity of Sumbawa macroalgae in various solvent fractions. Samples were macerated in n-hexane, ethyl acetate, and ethanol solvents. Antioxidant activity was tested using 1,1-Diphenyl-2-picryl Hydrazil (DPPH) method and ascorbic acid as a positive control. The percentage yield of extracts from various solvent fractions varied between 0.25% to 0.62%. The result of antioxidant activity testing showed that the IC50 (Inhibition Concentration) value of macroalgae extract was moderate to weak in the range of 108.1 ± 1.068 to 665.2 ± 0.479 g/mL and positive control was 6.156 ug/mL ±0.563g/mL. The relationship between antioxidant activity and the content of bioactive compounds and their pharmacological effects were discussed in this study. This study concluded that the ethyl acetate fraction of macroalgae had higher antioxidant activity than the n-hexane and ethanol fractions. Further analysis was needed to determine the composition of the bioactive compounds and identify their pharmacological effects at the molecular level.   Keywords: Macroalgae, antioxidant, Sumbawa, bioactive compounds, pharmacological effects   Abstrak Sumber daya alam yang populer di Pulau Sumbawa adalah makroalga. Abstract

An analytical method based on combination UV-Vis spectroscopy and chemometric was developed for detecting commonly listed adulterants such as paracetamol and piroxicam simultaneously in traditional medicines. No complex sample preparation and separation are required except grinding, dissolving, and filtering. The spectral interferences were resolved by multivariate techniques. Wavelengths selection and number of components optimization were performed by a combination of Genetic Algorithm and Partial Least Square (GA-PLS) followed by backward elimination through Jack-Knife Partial Least Square Regression (JK-PLSR). The capability PLSR model for quantitative analysis was assessed from the coefficient of determination (R2) and root mean square of error prediction/cross-validation (RMSEP/RMSECV) dan predicted residual sum of square (PRESS). Classification performance of PLS Discriminant Analysis (PLS-DA) was evaluated from the area under the receiver operating characteristic curve (AUROCC). For ensuring the sensitivity of the method, the detection limits from the two pseudo-univariate lines were estimated. The R2, RMSEP, RMSECV, AUROCC, and detection limit obtained from the selected models of paracetamol and piroxicam were 0.999, 0.25 mg/L, 0.15 mg/L, 100%, and 0.4 mg/L respectively. Therefore, the proposed method is suitable for the rapid screening of adulterated herbal medicine.

Abstract

The Passiflora edulis f. flavicarpa, a yellow passion fruit, is known for its distinctive aroma and tart flavor. The pulp of the yellow passion fruit can be used to make tea, jam, and syrup commercially. Commercial items are processed using oven heating, which involves prolonged exposure to high temperatures. As a result, fruit content, particularly water content, may alter. The benefit of reducing the water content is that less microbial growth medium means the product has a longer shelf life. However, fruit flesh’s nutritional composition will also change due to the ascorbic acid and phenolic compounds' thermolability and other factors. In this study, the yellow passion fruit pulp juice is heated in a controlled manner at 60°C, 70°C, and 80°C for 7 hours using an oven. The purpose is to assess changes in the amounts of ascorbic acid and total phenol that occurred. A paired T-test was used to assess the association between weight loss, ascorbic acid levels, and phenolic levels in the control and treatment groups. The amount of ascorbic acid was determined using the 2.6 dichloroindophenol titrimetry method, and the amount of phenol was determined using the Folin-Cioceltau method and UV-Vis spectrophotometry. According to this study, increasing the temperature to 60 °C or 80 °C significantly reduces weight and ascorbic acid levels. Also, the heating temperature unaffected the amount of phenol in each group.

Abstract

The effervescent tablet is an alternative dosage form to deliver the active pharmaceutical ingredients or natural products for therapeutic effects. Moreover, this dosage form is also used in the food industry for nutritional drinks. The tablet is preferred because of its practicality, improve water solubility, refreshing taste, and more easily absorbed than the conventional tablet. Tablet’s water dissolving time in the medium will affect the rate and amount of active ingredients. The disintegrating time of effervescent tablets is affected by the concentration of both acid and base elements, the tablet binder’s concentration (PVP), the interaction between citric acid and tartaric acid, and the concentration of the base element, the water content over the tablet, and the lubricant’s variation. The acid will affect the disintegration time however its imbalance will decrease the freshness. Therefore, an optimal balance of the base and acid element is required to make an effervescent tablet with a good disintegration time and an appropriate refreshing taste.

Abstract

The Indonesian government recommended sunbathing for 15 to 30 minutes in the morning to deal with the Covid-19 pandemic. Exposure to sunlight will activate the formation of vitamin D, which the body needs for defense against Covid-19 infection. However, excessive sun exposure will cause skin problems, including psoriasis, sarcoidosis, sunburn to skin cancer. One of the protections against sun exposure is the use of sunscreen. Sunscreen protects the skin from UV exposure by two mechanisms, namely, absorbing UV rays or reflecting UV rays. Currently, many sunscreens are developed with active substances derived from nature in various dosage forms. Sunscreen protection is stated using a photoprotector claim in the form of an SPF value or other supporting claims. This SPF value can be claimed using in vivo test, which previously could be predicted by on paper and in vitro approaches. Sunscreen safety is carried out by ensuring that the raw materials used comply with the requirements and penetration tests.

Abstract

The Bajakah root (Spatholobus littoralis), a native plant of Kalimantan, is recognized for its phenolic content and antioxidant activity. This study aims to compare the effects of three extraction methods—maceration, soxhlet, and reflux—on the total phenolic content and antioxidant activity of Bajakah root extracts. The plant material was extracted using 96% ethanol, followed by phenolic content analysis via the Folin-Ciocalteu method and antioxidant activity evaluation using the DPPH assay. Results revealed that the reflux method produced the highest phenolic content (17.6 ± 0.1 mg GAE/g extract) and the strongest antioxidant activity (IC50 of 26 ± 1 µg/mL). The soxhlet method yielded a phenolic content of 17.4 ± 0.2 mg GAE/g extract with an IC50 of 30 ± 0 µg/mL, while maceration resulted in the lowest phenolic content (14.6 ± 0.7 mg GAE/g extract) and an IC50 of 34 ± 2 µg/mL. The heating processes involved in the reflux and Soxhlet methods likely facilitated the degradation of glycosides, thereby enhancing phenolic content. All methods demonstrated exceptionally strong antioxidant activity (IC50 < 50 µg/mL). This study underscores the significant influence of extraction methods on the phenolic quality and antioxidant capacity of Bajakah root extracts.
