Abstract

Proper splicing is important for the functioning of a gene, and any interruption in splicing causes several deleterious events. Triggering receptors present on myeloid cells, TREM-1, are implicated in inflammation and act as amplifiers by mediating the release of proinflammatory chemokines and cytokines in response to fungal and bacterial infections. In bovines, mastitis is an inflammatory disease in which mammary gland inflammation is generally caused by bacteria. We found rs109937179 and rs208224995 SNPs in the splicing and miRNA binding region of TREM-1 gene in Chinese Holstein cows. The genotype distribution of the alleles for TREM-1 (rs109937179 and rs208224995) gene polymorphisms was investigated in 364 and 320 Chinese Holstein cows, respectively. We found that the GG genotype of the rs109937179 polymorphism and rs208224995 genotype of CA within the TREM-1 gene were associated with an increased risk of mastitis. Importantly, rs109937179 was found in the splicing region of TREM-1, and rs208224995 has a miRNA binding region for bta-miR- 2329-3p in the 3'UTR, which determines its effective roles in gene expression regulation.

Abstract

Kami mengamati nilai hemoglobin 11,78-12 gr/dL; hematokrit 32,7-34,52 %; leukosit 9,75-14,25 10 3 /μL; neutrofil 1,93-2,54 10 3 /μL, eosinofil 0,32-0,48 10 3 /μL, limfosit 7,38-10,12 10 3 /μL.

Abstract

Abstract

This study aimed to evaluate the effects of energy and organic zinc supplements, specifically zinc palm oil soap (ZPOS), on digestibility and unsaturated fatty acid profiles in vitro. The study used a completely randomized design with 4 treatments and 5 replications. The treatments were: T0= basal diet without supplementation, T1= basal diet + 5% palm oil (PO), T2= basal diet + 5% partial ZPOS (3.75% ZPOS + 1.25% PO), and T3= basal diet + 5% ZPOS. The inoculum source was rumen liquid from three fistulated female dairy goats and was homogenized. The goats were fed ration consisting of corn straw, soybean hulls, and concentrate containing total digestible nutrients (TDN) 63%, crude protein (CP) 14%, and neutral detergent fiber (NDF) 35%. Results showed that both 5% partial ZPOS and 5% ZPOS supplementation (T2 and T3) resulted in the increase of total volatile fatty acids (VFA), acetate, propionate, butyrate, unsaturated fatty acids (USFA) and a decrease in the ratio of acetate/propionate (A/P) compared to the control and supplementation of 5% PO (p<0.05). Supplementation of 5% partial ZPOS (T2) is better than 5% ZPOS because increased the digestibility of ether extract (EE), crude fiber (CF), NDF, and acids detergent fiber (ADF) (p<0.05) and decreased of methane compared to the control (p<0.05). In conclusion, adding 5% partial ZPOS (3.5% ZPOS and 1.5% PO) increases fiber digestibility, VFA, LCFA, and USFA concentration, and decreases methane production in the rumen liquid.

Abstract

Abstract. Pandupuspitasari NS, Sugiharto S, Agusetyaningsih I, Lestari DA, Khan FA, Setiaji A, Raza MA. 2025. PCA-based metagenomics reveals functional and taxonomic diversity in Kedu chicken gut microbiota. Biodiversitas 26: 4035-4041. The Gastrointestinal Tract (GIT) microbiota plays a central role in poultry health, productivity, and resilience, influencing digestion, nutrient absorption, immune development, and disease resistance. This study provides the first multi-database Principal Component Analysis (PCA)-based metagenomic characterization of the gut microbiota in Kedu chickens (Gallus gallus (Linnaeus, 1758)), an indigenous Indonesian breed. Digesta from five GIT segments (crop, gizzard, jejunum, colon, and cecum) of 21 traditionally reared Kedu chickens were sequenced on the Illumina NovaSeq X Plus platform, yielding 1,523,876 unigenes. Functional annotation against Non-Redundant Protein Database (NR), Clusters of Orthologous Groups (COG), Kyoto Encyclopedia of Genes and Genomes (KEGG), Carbohydrate-Active enZYmes Database (CAZy), Antibiotic Resistance Genes Database (ARDB), Comprehensive Antibiotic Resistance Database (CARD), and Virulence Factor Database (VFDB) revealed enrichment of genes involved in metabolism (38.2%), cellular processes (15.7%), environmental information processing (12.6%), and genetic information processing (11.0%). PCA separated GIT segments by functional specialization: The jejunum clustered distinctly (PC1 variance = 26.48% for NR, 30.04% for COG, 67.16% for KEGG) due to nutrient absorption-related functions, while the cecum exhibited the highest diversity and carbohydrate-active enzyme profiles (PC1 = 24.98% for CAZy). ARDB/CARD analysis showed that distal GIT segments (colon, cecum) contained higher densities of antimicrobial resistance genes, and VFDB revealed elevated virulence factor abundance in ileum and cecum. These findings highlight segment-specific microbiota roles, supporting the development of targeted probiotics and feed strategies, and contribute to conserving the functional biodiversity of Kedu chickens in sustainable production systems.

Abstract

The study examined the effect of lyophilized fecal microbiota transplantation (FMT) from healthy chicken donors on growth performance, haematological indices and intestinal morphology of Kedu chickens during the starter phase. A number of 120 Kedu chickens (0-6 weeks old) were randomly as-signed to T0 (orally inoculated with 1 mL PBS), T1 (inoculated with 0.5 mL FMT) and T2 (1.0 mL FMT). Body weight and feed intake were weekly recorded, while blood and intestinal segments were collected at week 6. Results showed that live body weight and body weight gain were highest (p<0.001) in T1, and the lowest in T0 group. Feed conversion ratio (FCR) was better (p=0.001) in T1 and T2 compared to T0 group. Feed intake was lower (p=0.014) in T2 than in T0 and T1 groups. The FMT administration tended to increase erythrocytes (p=0.054) and haemoglobin levels (p=0.056) of Kedu chickens. The relative weight of duodenum, jejunum, ileum, caecum and colon were lower (p<0.05) in T1 and T2 compared to those in T0 group. The FMT administration had no effect (p>0.05) on duodenal and jejunal villi height, crypt depth and the ratio of villi height to crypt depth of Kedu chickens. In conclusion, daily low-dose lyophilized FMT was associated with improved growth perfor-mance and feed efficiency in starter Kedu chickens, while haematological and intestinal morphological responses were limited.
