Abstract

Staphylococcus epidermidis is a common pathogen responsible for peri-implantitis, an infection localized around dental implants. Its virulence is largely attributed to biofilm formation, which enhances resistance to antibiotics and immune defenses. The challenge of treating S. epidermidis is further compounded by its resistance to several antibiotics, including methicillin. This study aimed to evaluate the antibacterial and anti-biofilm properties of a copper-zinc (Cu-Zn) metal alloy and its efficacy when integrated with beta-tricalcium phosphate (β-TCP) as a graft medium against S. epidermidis using in vitro methods. This research employs Staphylococcus epidermidis ATCC 12228. Antibacterial activity was assessed using Direct contact test and viability count via Colony-Forming Unit (CFU) analysis, while biofilm formation was evaluated using the tube adherence method. Increasing copper and zinc concentrations enhanced antibacterial efficacy, with Cu-Zn-β-TCP suspensions showing significantly greater antibacterial activity (p = 0.01) and improved anti-biofilm properties compared to Cu-Zn suspensions without β-TCP. The incorporation of copper and zinc effectively inhibited bacterial growth and biofilm formation of S. epidermidis. Furthermore, the Cu-Zn alloy demonstrated higher efficacy when used in conjunction with β-TCP as a graft medium, suggesting its potential for managing peri-implantitis.

Abstract

Periodontal bone defects require bone grafting materials that provide structural stability, biocompatibility, and reliable biological performance. Synthetic alloplastic scaffolds composed of biphasic calcium phosphate, integrating hydroxyapatite (HAp) and β-tricalcium phosphate (β-TCP), offer a promising alternative to other graft materials. Polycaprolactone provides flexibility and support for pore formation, while copper (Cu) and zinc (Zn) ions contribute to their antimicrobial and osteogenic benefits. This study aimed to investigate the physicochemical, morphological, mechanical, and biological properties of a synthetic HAp40/β-TCP60 alloplastic scaffold reinforced with polycaprolactone and to compare them with those of a commercial xenograft. The scaffold was fabricated using a freeze-drying method with dimethylformamide, and characterization included Fourier-transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD) for chemical and crystalline confirmation, Archimedes principle for porosity, light microscopy, and scanning electron microscopy (SEM) for morphological assessment, compressive testing, blood adhesion, as well as water and blood absorption analysis. FTIR and XRD verified the successful incorporation of polycaprolactone and the formation of a well-ordered biphasic calcium phosphate structure. Light microscopy showed that the synthetic scaffold had smaller macropore dimensions (324±49 µm) than the control (1410±541 µm), while SEM demonstrated markedly higher microporosity (8.184±2.581 µm) compared to the control (0.287±0.091 µm). Water absorption was lower in the synthetic scaffold (174.7%) than in the control (1172.5%), whereas blood absorption was comparable (300% vs 316.6%). The synthesized scaffold also exhibited superior blood adhesion, as evidenced by the absence of turbidity after agitation. Mechanical testing revealed that the control had superior compressive and yield strengths (0.980 MPa and 0.537 MPa, respectively) compared to the synthesized scaffold (0.287 MPa and 0.126 MPa, respectively). In conclusion, the synthetic HAp40/β-TCP60 scaffold exhibited properties within optimal ranges, highlighting its potential as a promising synthetic alloplastic material for clinical bone regeneration.

Abstract

Karies atau gigi berlubang merupakan permasalahan rongga mulut yang paling utama di dunia, dengan anak-anak dan remaja sebagai populasi yang paling berisiko terkena karies. Bukti klinis menunjukkan bahwa karies dapat dicegah, salah satunya adalah dengan mempromosikan cara menjaga kebersihan gigi dan mulut. Kegiatan pengabdian masyarakat ini bertujuan untuk mencegah karies sejak dini, dengan memberikan penyuluhan kesehatan gigi dan mulut kepada masyarakat sekolah TK Cikal, Penjaringan. Upaya untuk menilai tingkat pemahaman dan perilaku menyikat gigi anak TK dilakukan dengan metode partisipatif. Para peserta yang berjumlah 106 orang dibagi menjadi beberapa kelompok kecil dan dirotasi ke pos-pos penyuluhan dengan topik mengenai penyebab gigi berlubang, dampak tidak menjaga kesehatan gigi dan mulut, makan makanan yang tidak sehat dan merusak gigi, serta demonstrasi cara menyikat gigi dengan baik dan benar. Peserta dipanggil secara acak untuk menilai tingkat pengetahuan mengenai materi penyuluhan dan keterampilan menyikat gigi. Hasilnya menunjukkan bahwa sebagian besar peserta (96,3%) memahami materi penyuluhan, tetapi hanya sebagian peserta (46,7%) yang dapat mendemonstrasikan seluruh metode penyikatan gigi dengan sempurna. Kesimpulannya adalah peserta memahami materi penyuluhan, tetapi keterampilan menyikat gigi masih belum dikuasai seluruhnya, sehingga perlu diadakan kegiatan penyuluhan secara berkala disertai dengan pemeriksaan indeks kebersihan rongga mulut untuk menilai efektivitas penyuluhan yang sudah dilaksanakan.
