Abstract

Gracilaria verrucosa is a one of the seaweeds with a red pigment in the form of phycoerythrin. Phycoerythrin pigment contains phenol which is an antioxidant. The pigment can be obtained by extraction. One of the extraction methods that can be done is freeze thaw. This study uses the Response Surface Methodology to obtain the optimum sample. The purpose of this study was to determine the optimization model, the optimum point and the phenol content of the phycoerythrin Gracilaria verrucosa using the freeze thaw using Response Surface Methodology. Extraction of phycoerythrin pigment was carried out using the freeze thaw with a factor of freeze time, thawing time and solvent ratio, then the phenol content test was carried out. The results obtained are that the mathematical model used for optimization is the quadratic method with the equation y = 148.71 – 3.47(A) + 20.94(B) + 32.36(C) + 10.94(AB) + 9 .90(AC) + 6.25 (BC) + 6.52(A2) – 15.57(B2) + 30.46(C2). The optimum response obtained is the freeze duration 121,964 minutes; thawing duration 144,281 minutes; and the solvent ratio is 62.556%, and the phenol content obtained is 163.742 g GAE/g.

Abstract

Nanas madu memiliki umur simpan yang pendek yakni hanya 4-6 hari. Alternatif memperpanjang umur simpan nanas adalah dilakukan pelapisan lilin lebah didahului dengan pencucian menggunakan disinfectant agent. Penelitian ini bertujuan untuk 1) Mengetahui pengaruh kinerja jenis disinfectant agent; 2) Mengetahui pengaruh variasi konsentrasi emulsi lilin lebah; 3) Mengetahui interaksi antara jenis disinfectant agent dengan variasi konsentrasi emulsi lilin lebah; 4) Mengetahui kombinasi antara kinerja disinfectant agent dengan variasi konsentrasi emulsi lilin lebah terhadap atribut mutu nanas madu. Rancangan percobaan yang digunakan adalah Rancangan Acak Kelompok (RAK). Faktor yang diteliti meliputi jenis disinfectant agent (L) terdiri dari air kran (L1), larutan klorin 1% (L2), dan larutan garam 15% (L3), serta konsentrasi emulsi lilin lebah terdiri dari 6% (S1), 9% (S2), dan 12% (S3). Variabel yang diamati yaitu variabel fisikokimia (kekerasan, susut bobot, dan kadar air. Analisis data menggunakan analisis ragam (ANOVA) dan uji lanjut DMRT pada taraf 5%, serta perlakuan terbaik menggunakan uji indeks efektivitas. Hasil penelitian menunjukkan bahwa disinfectant agent berpengaruh nyata terhadap tekstur, susut bobot, sedangkan konsentrasi emulsi lilin lebah berpengaruh nyata terhadap susut bobot dan kadar air. Interaksi antara keduanya tidak berpengaruh nyata

Abstract

Research in recent years has shown the antioxidant activities of kecombrang and roselle flowers, as well as the antibacterial properties of kecombrang, so these materials have potentials to be developed as a functional food product. With an easy manufacturing process and basic ingredients that are easy to obtain, jelly candy is one of the preparations that can be applied to kecombrang and roselle flower extracts. This research aims 1) to determine the effect of variations in microwave-assisted extraction time and power on the physicochemical characteristics of the resulting extract and 2) to determine the effect of variations in the ratio of kecombrang and roselle flower extracts as ingredients for jelly candy products on the physical, chemicall, and sensory characteristics of the product. Research on the extraction of kecombrang and roselle flower powder used a randomized block design (RBD). Treatment consisted of two factors: 1) extraction power (150, 200, and 250 watts); 2) extraction time (3, 5, and 7 minutes). The data obtained were analyzed using the SPSS program. The analysis results showed that the best extraction of kecombrang and roselle flowers were at 250 watts of power and 5 minutes of extraction time. The best extraction results are then processed into jelly candy with the best sensory results in the 15%:25% sample. This jelly candy product contains reduced sugar levels of 12.41%, vitamin C levels of 23.58 mg/100g, total phenol levels of 47.58 mg/100g, total flavonoid levels of 18.80 mg/100g, and antioxidant activity amounting to 28.24 mg/100g. Abstract

Rumput laut berdasarkan pigmen warna dapat dikelompolkkan menjadi tiga yaitu chlorophyta, phaeophyta dan rhodophyta. Rumput laut Gracilaria Salicornia merupakam golongan rhodophyta yang mengandung berbagai senyawa aktif seperti pigmen dan antioksidan. Salah satu senyawa bioaktif yang berpotensi menjadi sumber antioksidan alami adalah fikobiliprotein yang terkandung pada rumput laut Gracilaria Salicornia. Permasalahan yang dihadapi adalah belum optimalnya proses ekstraksi fikobiliprotein yang terkandung dalam rumput laut Gracilaria Salicornia. Tujuan penelitian ini adalah melakukan proses optimasi, mendapatkan formula optimum dan mendapatkan karakteristik fisikokimia hasil ekstraksi fikobiliprotein Gracilaria salicornia dengan metode Freeze Thaw menggunakan Response Surface Methodology (RSM). Hasil penelitian ini menunjukkan bahwa kondisi optimum ekstraksi diperoleh pada penambahan pelarut aquades 55,737 ml, lama freezing 172,114 menit dan lama thawing 90,120 menit. Hasil prediksi kadar fikobiliprotein berdasarkan proses optomasi sebesar 54,507 µg/ml, sedangkan hasil validasi diperoleh sebesar 49,794 µg/ml yang sesuai dengan rentang prediksi modelnya. Karakteristik ekstrak fikobiliprotein berdasarkan hasil validasi mengandung fikobiliprotein sebesar 49,794 µg/ml; kadar protein 46,67%; total fenol 465,76 µg GAE/g; aktivitas antioksidan 8,40%; warna L* (20,20), a* (-1,93), b* (6,71).ABSTRACTSeaweed based on color pigments can be grouped into three, namely chlorophyta, phaeophyta and rhodophyta. Gracilaria Salicornia seaweed is a group of rhodophyta that contains various active compounds such as pigments and antioxidants. One of the bioactive compounds that has the potential to be a source of natural antioxidants is phycobiliprotein contained in Gracilaria salicornia seaweed. The problem faced is the less than optimal extraction process of phycobiliprotein contained in Gracilaria salicornia seaweed. The purpose of this study was to carry out the optimization process, obtain the optimum formula and obtain the physicochemical characteristics of the extraction results of Gracilaria salicornia phycobiliprotein with the Freeze Thaw method using Response Surface Methodology (RSM). The results of this study indicate that the optimum extraction conditions were obtained by adding 55.737 ml of aquadest solvent, freezing time 172.114 minutes and thawing time 90.120 minutes. The predicted results of phycobiliprotein levels based on the optimization process were 54.507 µg/ml, while the validation results were 49.794 µg/ml which was in accordance with the model prediction range. The characteristics of the phycobiliprotein extract based on the validation results contained phycobiliprotein of 49.794 µg/ml; protein content 46.67%; total phenol 465.76 µg GAE/g; antioxidant activity 8.40%; color L* (20.20), a* (-1.93), b* (6.71).

Abstract

Forest Farmer Group/ Kelompok Tani Hutan (KTH) Tani Jaya located at RT 03 RW02 Blok Blanakan, Wonodadi Village, Buayan District, Kebumen Regency, Central Java. This KTH manages a coffee plantation with Robusta coffee under a pine forest of 7 hectares. Currently, the coffee production of KTH Tani Jaya is 1000 kg/year and is sold exclusively to the local market in Kebumen Regency. The coffee products of KTH Tani Jaya currently do not meet the quality and food safety standards. The products produced are often inconsistent, for example, the roasting results are sometimes too burnt. The solution that can be used is the implementation of Good Manufacturing Practices (GMP). The method used is training and guidance through pre and post-tests. The results of the pre-test showed that the knowledge and basic knowledge of farmers about GMP was still limited, with a low average score in the areas of raw material control and documentation. After intensive training and guidance, the post-test results showed that participants had a significantly improved understanding of GMP, especially with regard to the control and documentation of raw materials.

Abstract

Wisata Taman Anggur K.D Garden, yang terletak di Karangsalam, Baturaden, Banyumas, merupakan sebuah destinasi agrowisata yang menawarkan pengalaman unik bagi pengunjung. Keunikan tersebut dapat dilihat dengan kebun anggurnya yang indah. Penelitian ini bertujuan untuk menganalisis kelayakan investasi pada taman wisata ini dengan mempertimbangkan berbagai faktor ekonomi dan biaya operasional yang digunakan melalui pendekatan studi kasus, mengola data mengenai biaya investasi awal, operasional, serta pendapatan yang dihasilkan untuk dianalisis dan memberikan gambaran lengkap mengenai kelayakan investasi. Adapun perhitungan ini menggunakan Net Present Value (NPV), Profitability Index (PI), Internal Rate Return (IRR), dan Payback Period (PP). Kelayakan investasi Wisata Taman Anggur K.D Garden dapat diketahui berdasarkan perhitungan tersebut. Berdasarkan hasil perhitungan didapatkan hasil nilai NPV > 0 yaitu sebesar Rp. 34.140.639,00 sehingga investasi tersebut layak untuk dijalankan. Nilai PI sebesar 1,41 (PI > 1), IRR sebesar 19% (IRR > discount factor), dan PP sebesar 2,63 tahun. Berdasarkan hasil analisis menunjukkan bahwa usaha ini layak untuk dijalankan dengan nilai investasi akan kembali setelah usaha beroperasi selama 2,63 Tahun.

Abstract

Small Rice mill or called “Selipan” in Indonesian, is a series of rice mill machines with various engine capacities, generally found in agricultural production centers. Rice mills require relatively large investment costs for producers in the production area. A feasibility study on the rice mill business at Selipan Riyanto is needed to be a reference for pioneering similar businesses. A business feasibility study is a comprehensive assessment to assess the success of a business. Rice mills are industrial facilities designed to convert paddy into milled rice. The milling process includes a series of operations, including rice cleaning, hulling, polishing, and packaging. This study aimed to evaluate financial feasibility of Selipan Riyanto based on NPV (Net Present Value), PI (Profitability Index), IRR (Internal Rate of Return), and PP (Payback Period). The study's results obtained a value, NPV (Net Present Value) of Rp. 266.613.794, PI (Profitability Index) of 2.54, IRR (Internal Rate of Return) of 59%, and PP (Payback Period) of 1.46 years.

Abstract

The productivity of pineapple in Karangjengkol Village, Purbalingga Regency is 110 million/Ha/year. Based on the situation analysis, currently pineapple sales use the slashing system and the existing processed products are jam, jam and pineapple dodol with low quality. As a result, the price obtained is low and the shelf life of pineapple and its processed products is relatively short, which is 3 days for fresh pineapple and 14 days for its processed products. The objectives of this community service are: 1) To find out the understanding and implementation of pineapple dodol and instant powder processing techniques. 2) To find out the level of mastery of target group members regarding simple post-harvest technology, namely grading, coating harvest wounds with rosella antimicrobial bioactive compounds. After the technology transfer, the increase in the number of people who know the proper and correct processing methods regarding dodol, instant powder and grading increased, respectively: 17%, 22% and 34%. Grading and rinsing wounds with rosella extract disinfectant can prevent contaminants and extend the shelf life of fresh pineapple. In the manufacture of instant powder, stirring after caramelization and stopping the heat source must be done quickly and without pause until small crystals are formed. In the processing of dodol, the right stirring and temperature affect the quality attributes of the resulting dodol.

Abstract

Karakteristik fisikokimia dan atribut sensori kopi arabika dipengaruhi oleh lingkungan tempat tumbuh tanaman kopi dan proses pengolahannya. Tujuan penelitian mencari pengaruh kombinasi tingkat ketinggaian tempat tumbuh tanaman kopi dengan variasi proses sangrai terhadap atribut fisikokimia dan sensori kopi arabika Kabupaten Banjarnegara. Penelitian ini merupakan penelitian eksperimental yang menggunakan metode Rancangan Acak Lengkap (RAL) dengan faktorial 2 faktor. Faktor yang diujikan pada penelitian ini adalah: 1) Ketinggian tempat tanaman Kopi dengan taraf: 1000 mdpl (Kecamatan Wanayasa), 1050 (Kecamtan Karangkobar) dan 1100 mdpl (Kecamatan Kalibening). 2) Level sangrai dengan taraf: light, medium dan dark. Faktor tersebut disusun kedalam rancangan faktorial sehingga diperoleh 9 kombinasi perlakuan. Perlakuan diulang tiga kali sehinggga diperoleh 27 unit percobaan. Pengujian atribut sensoris kopi arabika menggunakan metode analisis mutu skroring pembeda. Hasil penelitian ini adalah level penyangraian berpengaruh terhadap kadar air, pH, total fenol dan aroma kopi arabika sedangakn ketinggian tempat tanaman kopi tidak berpengaruh terhadap kadar air, pH, total fenol dan aroma kopi.

Abstract

Kelopak bunga rosella adalah sumber senyawa fenolik yang baik. Dalam penelitian ini ekstraksi dibantugelombang mikro digunakan untuk mengekstrak senyawa fenolik dari kelopak bunga Rosella. Denganmenggunakan metode respon permukaan, efek daya gelombang mikro, konsentrasi etanol, dan waktu ekstraksipada hasil total fenol yang diselidiki dan kondisi optimal ditentukan sebagai berikut: daya gelombang mikro 250W, konsentrasi etanol 78,36% dan waktu ekstraksi 4,91 menit. Nilai perkiraan untuk total hasil fenol, 24,61 mg/gdiperoleh pada kondisi tersebut. Percobaan verifikasi pada kondisi optimum, terdiri dari 4 putaran, dilakukan danhasil praktis 23,77 + 0,25 mg/g jumlah fenol lebih tinggi dari pada metode konvensional 19,84 + 0,46 mg/g.Dibandingkan dengan metode konvensional, antosianin, vitamin C dan hasil ekstraksi dibantu gelombang mikroadalah 14,80 + 0,08, 10,74 + 0,14 mg/g dan 22,09 + 3,3% yang lebih tinggi dan berbeda secara signifikan dalamtingkat kepercayaan 95% dari pada ekstraksi konvensional, yaitu 9,25 + 0,04, 9,99 + 0,16 mg/g dan 16,18 + 1,9%.Aktivitas antibakteri yang dinyatakan dengan nilai zona hambat adalah 12,6 + 0,6 mm pada E. coli dan 11,6 + 0,3mm terhadap S. aureus. Nilai IC50 ekstrak adalah 202,47 μL/mL. Dibandingkan dengan metode konvensional,zona hambatan 10,2 + 0,4 mm pada E. coli dan 9,8 + 0,6 mm pada S. aureus. Aktivitas antioksidan dinyatakandengan nilai IC50293,09 μL/mLKata kunci: kelopak bunga Rosella, ekstraksi berbantu gelombang mikro, fenolABSTRACTRoselle calyx is an excellent source of phenolic compounds. In this study, microwave-assisted extractionwas applied to extract the phenolic compounds from Roselle calyx. By using response surface methodology, theeffects of microwave output power, ethanol concentration, and extraction time on total phenolic yield wereinvestigated and the optimal conditions were determined as follows: microwave output power 250 W, ethanolconcentration 78.36% and extraction time 4.91 min. The estimated values for total phenols yield, 24.61 mg/g wasobtained at those conditions. A verification experiment at the optimum condition, consisting of 4 runs, wasperformed and the practical yield 23.77 + 0.25 mg/g total phenols higher than using conventional method 19.84+ 0.46 mg/g was obtained. Compared with conventional method, anthocyanin, vitamin C and yield of microwaveassisted extraction were 14.80 + 0.08, 10.74 + 0,14 mg/g and 22.09 + 3.3 % which were higher and significantlydifferent within 95% confidence level than conventional extraction , which were 9.28 + 0.04, 9.99 + 0.16 mg/gand 16,18 + 1,9%, respectively. Antibacterial activity expressed with inhibitory zone value was 12.6 + 0.6 mmagainst E coli and 11.6 + 0.3 mm against S aureus. IC50 value of the extract was 202.47 μL/mL.Compared withconventional method, with inhibitory zone of 10. 2 + 0.4 mm against E coli and 9.8 + 0.6 mm against S aureus.Antioxidant activity expressed with IC50 value of 293.09 μL/mL.Key words: roselle calyx, extraction, microwave, phenols
