Abstract

Intercellular adhesion plays a role in cancer formation and protein has a key potential in maintaining cell adhesion, including syndecan-1. Meanwhile, oral cancer originates from the oral epithelium, which has an invasive and metastatic level. Its treatments involving chemotherapy and radiotherapy commonly leave unfavorable side effects, hence, suitable alternatives are needed. Natural ingredients are widely used as an alternative treatment for cancer, for example, Ganoderma lucidum (G. lucidum) which has anti-cancer and anti-angiogenic properties, induces apoptosis, stimulates an immune response, inhibits the degradation of Extracellular matrix (ECM), reduces inflammation, affects cell cycles, cytotoxic, and acts as an antioxidant. This study aims to determine the effect of ethanol extract from Ganoderma lucidum Cianjur isolate on syndecan-1 expression in KB CCL-17 oral cell cancer. This was an experimental study with a post-test only control group design, the treatment group used G. lucidum ethanol extract with a concentration of 2.12 μg/ml (P1), 4.24 μg/ml (P2), and 8.49 μg/ml (P3), while the positive control group used cisplatin with a concentration of 11.5 μg/ml (K1). In contrast, the negative control used aquadest (K0), while syndecan-1 expression was observed using the immunohistochemical examination. The highest syndecan-1 expansion rate was found in the treatment group with a concentration of 8.49 μg/ml. A significant difference was indicated by one-way ANOVA (p<0.05) between K0 - K1, K0 - P1, K0 - P2, K0 - P3, K1 - P1, K1 - P2, K1 - P3, P1 - P2, as well as P1 and P3. The administration of ethanol extract from G. lucidum Cianjur isolate increases syndecan-1 expression in KB CCL-17 oral cell cancer.

Abstract

Abstract

Introduction: Hyperuricemia is a significant risk factor for the development of chronic kidney disease. Excessive uric acid can deposit in the kidneys, triggering inflammatory reactions and oxidative stress, leading to structural damage in the glomeruli. Black garlic, with its antioxidant and bioactive compounds, has the potential to protect the kidneys from hyperuricemia-induced damage. Our objective is to evaluate the effect of black garlic supplementation on the histopathological features of glomeruli in a hyperuricemic rat model. Methods: This study was an analytical observational study using Biological Remnant Materials (BRM) from a previous study. The sample consisted of 30 male Sprague-Dawley rats divided into 5 groups: disease control, drug control (allopurinol), and 3 black garlic dose groups (240 mg/day, 480 mg/day, and 960 mg/day). Data were collected through histopathological examination of kidney tissue using the glomerular damage scoring method (0-3). Data analysis was performed using one-way ANOVA and post hoc LSD tests. Results: Black garlic demonstrated protective effects on the histopathological features of glomeruli in hyperuricemic rats. The 240 mg/day dose showed the most significant improvement in kidney health, reducing glomerular damage scores compared to the disease control group (p < 0.05). These findings suggest that black garlic can serve as a preventive agent against kidney complications caused by hyperuricemia. Conclusion: Black garlic provides protective effects on glomerular histopathology in hyperuricemic rats, with the 240 mg/day dose being the most effective. These results suggest potential use for black garlic in preventing kidney complications in hyperuricemia.

Abstract

Abstract

Introduction: Clear cell papillary renal cell carcinoma (ccpRCC) was recently identified, which acts like a benign tumor entity introduced by WHO in 2016. It shares morphologic features in both clear cells and papillary cells. It is characterized by its unique immunohistochemical, morphologic, and genetic features and indolent behavior with no reported recurrence or metastasis before. This paper presents a case of ccpRCC in adulthood with spinal metastasis.Case Presentation: A 16-year-old female patient presented to the urology polyclinic of Prof. Dr. Margono Soekarjo Hospital with an abdominal lump and hematuria. Subsequently, the patient underwent radical nephrectomy. Histopathological examination confirmed ccpRCC. After six months, the patient exhibited weakness in the lower extremities, sensory disturbances, and urinary retention. She was referred to the Neurology department, where physical examinations revealed paraplegia inferior and segmental hypoesthesia at the vertebrae T12 level. MRI examination showed signs of vertebral metastases. After receiving immediate treatment from the neurology department, the neurosurgery department scheduled the patient for a surgical procedure. The case was followed for eight months. Conclusions: ccpRCC is a benign kidney tumor exhibiting a combination of clear cell and papillary cell carcinoma characteristics. While patients with ccpRCC are typically asymptomatic, cases are presenting with symptoms such as hematuria. This case demonstrates the occurrence of hematogenic metastases in ccpRCC, highlighting the need for surgical intervention to preserve existing functionality
