Abstract

Breast cancer incidence is still in the second position after cervical cancer within womenmalignancy. Fine needle aspiration biopsy (FNAB) has been widely known and used in diagnosing breastlesions last few decades. FNAB diagnostic procedure is considered less invasive and relatively lessexpensive than surgery biopsy. This research was diagnostic study with cross sectional design. Researchdone by reading FNAB cytology and histopathology slides from 86 patients with breast tumor betweenJanuary 2004 until December 2007. WHO classification and Elston & Ellis method was used to diagnosebreast tumor variant and grading. While fnab cytology used the NHS Breast Screening Program(NHSBSP) guidelines. Diagnostic tests of FNAB showed sensitivity 91%, specificity 79%, and accuracy87%. The sensitivity and specificity values of FNAB on this research was still quite high, because in manyliterature explained that the sensitivity and specificity rates of FNAB ranged between 66% and 92%depending on the experience of diagnostic and hospital procedure

Abstract

Colorectal carcinogenesis induced by cigarette smoke requires at least 30-40 years. This long time duration causes an animal research conducted becomes relevant. This research was carried out to observe colorectal carcinogenesis due to cigarette smoke exposure in Wistar Rat. The observations focused on changes in epithelial morphology and expression of APC, MSH2 and MLH1. Twenty male Wistar rats inbreed strain were randomly allocated into control group and experimental group exposure to cigarettes smoke for 14 weeks and 28 weeks sequentially. Colorectal epithelial morphology was assessed on the histopathology examination, whereas the expression of APC, MSH2 and MLH1 was assessed on aspect of immunohistochemistry. The comparative analysis between the two groups was performed using non-parametric Mann-Whitney U test. Histology of colorectal epithelium showed pattern of colitis associated cancer that was significant both in 14 weeks and 28 weeks of treatment. This research indicated negative expression of APC, MSH2 and MLH1 in the colorectal cancer that were significant at 28 weeks of exposure. This research implies that chronic exposure to cigarette smoke can induce colitis associated colorectal cancer through decreased expression of APC, MSH2 and MLH1.

Abstract

Intercellular adhesion plays a role in cancer formation and protein has a key potential in maintaining cell adhesion, including syndecan-1. Meanwhile, oral cancer originates from the oral epithelium, which has an invasive and metastatic level. Its treatments involving chemotherapy and radiotherapy commonly leave unfavorable side effects, hence, suitable alternatives are needed. Natural ingredients are widely used as an alternative treatment for cancer, for example, Ganoderma lucidum (G. lucidum) which has anti-cancer and anti-angiogenic properties, induces apoptosis, stimulates an immune response, inhibits the degradation of Extracellular matrix (ECM), reduces inflammation, affects cell cycles, cytotoxic, and acts as an antioxidant.This study aims to determine the effect of ethanol extract from Ganoderma lucidum Cianjur isolate on syndecan-1 expression in KB CCL-17 oral cell cancer. This was an experimental study with a post-test only control group design, the treatment group used G. lucidum ethanol extract with a concentration of 2.12 μg/ml (P1), 4.24 μg/ml (P2), and 8.49 μg/ml (P3), while the positive control group used cisplatin with a concentration of 11.5 μg/ml (K1). In contrast, the negative control used aquadest (K0), while syndecan-1 expression was observed using the immunohistochemical examination.The highest syndecan-1 expansion rate was found in the treatment group with a concentration of 8.49 μg/ml. A significant difference was indicated by one-way ANOVA (p<0.05) between K0 - K1, K0 - P1, K0 - P2, K0 - P3, K1 - P1, K1 - P2, K1 - P3, P1 - P2, as well as P1 and P3. The administration of ethanol extract from G. lucidum Cianjur isolate increases syndecan-1 expression in KB CCL-17 oral cell cancer.

Abstract

Imiquimod menginduksi terjadinya aktivasi sel Th17 dan sel dendritik yang berperan dalam terjadinya psoriasis. Ekstrak ciplukan ( Physalis angulata L.) mengandung senyawa steroid, flavonoid, alkaloid, dan saponin seperti Physalin B, Physalin F, Physalin G yang memiliki aktivitas antiinflamasi dalam patofisiologi psoriasis. Penelitian ini merupakan penelitian eksperimental dengan post test only with control group design. Tiga puluh lima ekor mencit betina dibagi dalam 7 kelompok. Kelompok A: kontrol negatif, kelompok B: kontrol positif, kelompok C yang diinduksi imiquimod dan diberikan ekstrak ciplukan 400 mg/kgBB selama 7 hari, kelompok D yang diinduksi imiquimod dan diberikan ekstrak ciplukan 800 mg/kgBB selama 7 hari , kelompok E yang diinduksi imiquimod dan diberikan ekstrak ciplukan 1.200 mg/kgBB selama 7 hari, kelompok F yang diinduksi imiquimod dan diberikan methotreksat 1 mg/kgBB selama 7 hari, dan kelompok G yang diinduksi imiquimod dan diberikan kombinasi ekstrak ciplukan 1.200 mg/kgBB dan methotreksat 1mg/kgBB selama 7 hari. Jumlah sel fibroblas dihitung pada hari ke lima belas dengan mengambil sampel kulit mencit dan dilakukan pembuatan preparat histologi kemudian dihitung secara manual menggunakan mikroskop. Rerata jumlah sel fibroblas pada kelompok A,B,C,D,E,F,dan G masing- masing adalah 21,6±2,3; 39,2±5,5; 30,6±1,3; 24,0±2,8; 24,8± 2,9; 28,4±3,0;28,2±3,2. Hasil uji One Way ANOVA menunjukkan nilai p=0,000(p<0,05) sehzingga menunjukkan hasil signifikan.

Abstract

Gamal leaves (Gliricidia sepium) well known as liquid organic fertilizer, are also known to have potential as vegetable larvicides because there are flavonoids, triterpenoids, coumarin, coumaric acid, mitotic acid, and stigmastanol glucoside which can be identified and isolated from various parts from Gamal. The aim of this study was to examine the larvicidal effect of Gamal (G. sepium) leaf extract against larvae of Aedes aegypti mosquitoes. The study was conducted using an experimental design with a posttest-only control group design and a completely randomized design consisting of 6 treatment groups. Two groups as control (positive and negative control), while 4 other groups received treatment of Gamal leaf extract (G. sepium) at doses of 50 ppm (P1), 150 ppm (P2), 250 ppm (P3), and 350 ppm (P4). There was no mortality of larvae Aedes aegypti from pure lines after giving Gamal leaf extract, but mortality only occurred in Aedes aegypti larvae positive control group originating from the field. Unknown LC50 and LC90. A total of 13 (52%) Ae. aegypti strain classified as tolerant, while as many as 19 (76%) larvae of Aedes aegypti originating from the field are classified as resistant.

Abstract

Smoking cigarette is one of risk factors for developing colorectal carcinoma. Various researches have been made to suppress the toxicity of cigarette smoke. One of such effort is to reduce the size of cigarette smoke particles using nano water solution containing aromatic groups and EDTA. This study aims to assess the effects of this cigarette smoke nanoparticle regarding colorectal carcinogenesis. The experimental study was carried out with sequential post test only control group design. Thirty Wistar rats were divided into 3 groups: exposure to cigarettes smoke, exposure to smoke of cigarettes containing nano water solution and control for 14 weeks and 28 weeks. Colorectal epithelial morphology was assessed on the histopathology examination, whereas the expression of APC, KRAS, MSH2, MLH1, and p53 was assessed on immunohistochemistry procedure. Our results showed that cigarette smoke nanoparticles had better effects regarding colorectal epithelial morphology, especially through the increased expression of APC.

Abstract

Breast cancer is the most common cancer in woman. Accumulation of free radical in the body is one of cancer risk fac-tors. Since 2009, Research Institute for Free Radicals Shedding Malang has been developing balur nano divine kretek method, combination of balur and divine tobacco smoke to decay free radicals. It has been tested on volunteers with cancer, including breast cancer. This study aimed to assess the effect of balur nano divine kretek method to breast can-cer tissue and CD44 expression. The results showed that 66.67% tumor samples suffered extensive damage (grade 3) and high CD44 expression. The high expression of CD44 in most of the samples may indicate better prognosis. The augmentation for the effectiveness consistency of balur nano divine kretek should be continuously conducted with greater samples and more complete molecular markers such as CD24 and ALDH.

Abstract

Sweet potato (Ipomoea batatas L) leaf is one of well known vegetables among Indonesian people.  It is also often used as traditional medicine for diabetes mellitus.  This research aimed to investigate the protective effect of I. batatas L leaves extract on the pancreas of IDDM animal model.  Twenty five male Sprague-Dawley rats were divided into 3 treatment groups, and 2 control groups.  Streptozotocin (STZ) was injected at multiple low doses (40 mg/kg BW) intraperitoneally for 5 consecutive days,  I. batatas L leaves extract (doses 0.25, 0.8, and 2.5 g/ kg BW) were administered for 14 days after the first STZ injection.  Fasting blood glucose was analyzed after complete STZ induction (day 6), and after 14 days treatment.  At the end of the study, rats were terminated, and pancreas were removed for histological examination and immunohistochemical procedure using anti-insulin antibody.  Diabetic rats treated with 2.5 g/kg BW I. batatas L leaves extract showed lowest fasting blood glucose among treatment groups, and had approximately 50% normal Langerhans islets with functional beta cells.  These results suggest that I. batatas L leaves extract has anti diabetic activity through its protection effect on the pancreas.

Abstract

A giant cell tumor of soft tissue (GCT-ST) is a rare tumor composed of mononuclear and osteoclast-like multinucleated giant cells.  It is a low-grade malignant potential tumor morphologically analog to a giant cell tumor of bone.  The superficial and deep soft tissue of extremities is the most frequent site of this tumor which is cured by complete or radical resection.  GCT-ST as a primary tumor in the female Breast is exceedingly rare.  Presentation of osteoclast-like multinucleated giant cells in the breast tumor has been described in association with the variant of breast carcinoma.  We report a case of primary GCT-ST arising in the female Breast without a relationship with the epithelial component.  To the best of our knowledge, this is the fifth such case report, and only the second one occurred in a female under 50 years.

Abstract

BackgroundHypercholesterolemia due to a high cholesterol diet can increase free radicals resulting in oxidative stress. Superoxide dismutase (SOD) and malondialdehyde (MDA) have been used as the study markers of oxidative stress in cases of hypercholesterolemia. Purple passion fruit contains various compounds that may reduce free radicals. This study aimed to determine the effect of purple passion fruit juice on SOD and MDA levels in hypercholesterolemic rats. MethodsAn experimental analysis with post-test only control group design involving 28 male Wistar rats. They were divided into 4 groups: normal control (K1), hypercholesterolemic control (K2), purple passion fruit juice treatment at 4.2 mL/200 gBW/day (K3), and simvastatin treatment at 0.018 mg/200 gBW/day (K4). The purple passion fruit juice at 4.2 mL/200 gBW/day was administered for 14 days. SOD levels were examined by enzymatic colorimetric methods using the Ransod kit and MDA levels by the TBARS method. ResultsThe Kruskal-Wallis test showed a significant difference in SOD levels between the tested groups (p<0.05). One-way ANOVA test for MDA levels showed a significant difference (p<0.05). Post Hoc test (Mann-Whitney for SOD and LSD for MDA levels) also showed significant differences: K1 vs. K2, K2 vs. K3, K2 vs. K4, and K3 vs. K4 (p<0.05). ConclusionThis study demonstrated that purple passion fruit juice significantly increases the SOD and lowers the MDA level in hypercholesterolemic male Wistar rats. Consumption of purple passion fruit juice may help to modulate oxidative stress caused by hypercholesterolemia in rats.
