Abstract

Pengabdian masyarakat tentang “Pengembangan Usaha Rumah Tangga Kripik Singkong di Amal Aman Rempung Lombok Timur” bertujuan untuk memberikan wawasan dan pengetahuan kepada pelaku usaha mikro kecil yang ada di desa Rempung Lombok Timur. Kegiatan dilakukan pada tanggal 16 Januari 2025 bertempat di Musholla Haqqul Mubin Rempung Lombok Timur. Dengan menggunakan metode ceramah, diskusi grup serta tanya jawab intensif, menunjukkan hasil yang cukup memberikan harapan untuk kemajuan usaha para peserta. Dengan jumlah peserta sekitar 20 orang yang didampingi aparat kampung dan tokoh masyarakat diperoleh hasil antaranya; Peserta sangat berterima kasih dan antusias mengikuti kegiatan yang diadakan tim Unram; para peserta terbuka wawasan untuk mengelola usaha dengan cara yang lebih profesional, baik dalam proses produksi, pengemasan, promosi dan pemasaran; adanya kebutuhan untuk kerjasama sesama pelaku usaha dalam bentuk kegiatan kelompok; serta para peserta berharap adanya pendampingan yang berkesinambungan dari tim pembina Unram dalam rangka memberi arahan dan bimbingan tehnis. Akhirnya dengan harapan dan semangat yang tinggi para peserta, kiranya acara/kegiatan pengabdian masyarakat ini bisa terus dilanjutkan dan ditingkatkan kualitas maupun kuantitasnya dengan dukungan dana yang lebih besar untuk mencapai harapan yaitu kemajuan usaha-usaha masyarakat kecil khususnya.

Abstract

Breast cancer is a serious disease that occurs in women and contributes to the highest mortality compared to other types of cancer. This study aims to synthesize curcumin analog compounds ((3E,5E)-1-benzyl-3,5 bis (3 (benzyloxy)benzylidene) piperidin-4-one), test them in vitro against various breast cancer cells (T47D, HER-2, MCF-7, and 4T1) and normal cells (vero cells), and study their molecular docking. Synthesis was carried out by reacting 3-benzyloxybenzaldehyde with N-benzyl-4-piperidone catalyzed by 5% KOH at room temperature; in vitro testing was carried out using the Microculture Tetrazolium Technique Assay method, ADMET analysis with an online database server, and molecular docking studies in Autodoc Vina. The synthesis results obtained yellow solid powder with a yield of 65.85%, characterization with TLC gave black fluorescence (Rf 0.63), melting point 114-116oC, TLC scanner one peak (100%), retention time 0.65 minutes, 1H &13C-NMR analysis showed the molecular formula C40H35NO3, moderate activity against 4T1 breast cancer cells and inactivity on T47D, HER-2, and MCF-7 cancer cells, and did not show cytotoxicity to normal cells (vero cells). ADMET predictions from Lipinski's five rules contained two parameters that did not meet, namely molecular weight and log P value. Molecular docking studies were carried out on estrogen receptor protein (ER)-α (PDB ID: 2ERT), which showed a binding affinity energy of -8.7 kcal/mol and -7.1 kcal/mol of the native ligand. Further research and development is needed on synthetic curcumin analog compounds to increase their activity value against breast cancer by paying attention to Lipinski's five rules to obtain compounds with better potential activity and ADMET.

Abstract

Goat’s milk proteins can undergo hydrolysis during digestion, producing peptides that may inhibit α-amylase and help treat type 2 diabetes with minimal side effects. Identifying the amino acid composition of these peptides is essential for determining their inhibitory potential. Recent in silico digestion methods have been developed to generate specific peptides. This study aims to identify α-amylase inhibitory peptides from goat’s milk casein hydrolyzate using in silico digestion, followed by peptide synthesis and activity assay. Peptides were derived from goat’s milk casein hydrolyzed using in silico digestion. Molecular docking was employed to predict protein-peptide interactions utilizing the HADDOCK2.4 server, CABS-dock, and PepSite 2 server. Peptides EDVPSER and TNAIPYVR could inhibit α-amylase with IC50 values of 14.16 ± 0.65 and 76.58 ± 2.13 µM, respectively. In vitro evaluation confirmed that EDVPSER from αS1-casein exhibited α-amylase inhibitory potential. This peptide could be developed as a potential therapeutic agent for type 2 diabetes, offering a natural and targeted approach to α-amylase inhibition. Peptide EDVPSER may serve as a basis for further research and development of antidiabetic treatments derived from goat’s milk proteins.

Abstract

Penelitian ini bertujuan untuk menganalisis pengelolaan dana desa dalam meningkatkan kesejahteraan masyarakat di Desa Mambalan, Kecamatan Gunungsari, Kabupaten Lombok Barat. Penelitian ini menggunakan pendekatan kuantitatif dengan teknik analisis regresi linear berganda. Populasi dalam penelitian ini adalah Seluruh Kepala Keluarga (KK) yang ada di Desa Mambalan, Kecamatan Gunungsari, Kabupaten Lombok Barat. Jumlah sampel yang digunakan dalam penelitian ini sebanyak 43 KK. Teknik pengumpulan data yang digunakan adalah wawancara, dokumentasi dan menyebarkan kuesioner yang berisi pertanyaan tentang Pengelolaan Dana Desa dan Kesejahteraan Masyarakat. Pengolahan data dilakukan dengan menggunakan program IBM SPSS (Statistical Package for Social Science) versi 26, dengan uji validitas, uji reliabilitas, uji asumsi klasik, serta uji hipotesis (uji t dan uji F). Hasil penelitian secara parsial menunjukkan bahwa variabel perencanaan dan pelaksanaan berpengaruh signifikan terhadap kesejahteraan masyarakat. Artinya, semakin baik perencanaan dan pelaksanaan dalam pengelolaan dana desa, maka semakin tinggi tingkat kesejahteraan masyarakat. Sementara itu, variabel pertanggungjawaban tidak memiliki pengaruh yang signifikan secara parsial. Namun, secara simultan, ketiga variabel pengelolaan Dana Desa (perencanaan, pelaksanaan, dan pertanggungjawaban) berpengaruh signifikan terhadap kesejahteraan masyarakat. Temuan ini menegaskan bahwa penguatan dalam aspek perencanaan dan pelaksanaan perlu terus ditingkatkan, serta optimalisasi pertanggungjawaban perlu dilakukan agar pengelolaan Dana Desa dapat berdampak lebih maksimal bagi kesejahteraan masyarakat. Kata Kunci: Pengelolaan Dana Desa, Perencanaan, Pelaksanaan, Pertanggungjawaban, Kesejahteraan Masyarakat.

Abstract

This study aims to analyze the role of fish pond farming in improving the welfare of fish pond farmers in Peteluan Indah Village, Lingsar District, West Lombok Regency. Fish pond farming is one of the rapidly growing local economic sectors with great potential to increase community income. The research method used is a qualitative approach with data collection techniques including interviews, observations, and documentation. The results of the study show that fish pond farming not only contributes to increasing farmers’ income but also encourages the creation of job opportunities, improvement of housing conditions, and better access to education and healthcare services. In addition, support from the government and local institutions further strengthens the sustainability of fish pond farming. Therefore, fish pond farming plays an important role in improving the living standards and welfare of pond farmers in the region.

Abstract

Peptides have been reported as a potential alternative for antidiabetic therapy by inhibiting α-amylase, one of the key enzymes involved in carbohydrate metabolism. This study aimed to identify α-amylase inhibitory peptides derived from trypsin hydrolysate of Caulerpa racemosa protein. The protein was extracted using trichloroacetic acid (TCA)/acetone. The resulting peptides were fractionated using a strong cationic exchanger-solid phase extraction (SCX-SPE) column. The peptide fractions were tested for α-amylase inhibitory activity, and the peptides in the most active fraction were identified using liquid chromatography-high resolution mass spectrometry (LC-HRMS). Seven fractions were obtained from SCX-SPE elution at pH 3 to 9. The fraction eluted at pH 5 exhibited the highest α-amylase inhibitory activity, with an IC50 value of 43.70 µg/mL. Four peptides with the sequences VQKEKR, MNFYCISSK, DLCDYIHNK, and ISICYEK were identified from this fraction. Molecular docking studies revealed that the peptides with sequences MNFYCISSK and VQKEKR had the strongest interactions with α-amylase. The peptide binding affinity energies of MNFYCISSK and VQKEKR were −10.3 and −9.4 kcal/mol, respectively, with RMSD values of 0.3 ± 0.0 and 0.3 ± 0.2 Å. It can be concluded that the peptides with sequences MNFYCISSK and VQKEKR could be proposed as potential antidiabetic peptides with an α-amylase inhibition mechanism.
