Abstract

Antigen-antibody reaction is an important tool for the analysis of localization of target molecules, including antigenic protein within worm tissues. The purpose of the present research was to demonstrate the ability of immunoglobulin yolk (IgY) anti-excretory/secretory recognized the antigen in the tissue of Ascaridia galli by mean of immunohistochemistry method. The excretory/secretory protein was procured from A. galli and concentrated by mean of vivaspin 30,000 MWCO. IgY was produced by egg yolks of immunized chickens with excretory/secretory, and purified using fast protein liquid chromatography (FPLC) method. A. galli adult worms were cut in transversal and longitudinal section of the center and anterior region. Slides were incubated with both primary IgY for overnight at 4 oC and secondary antibody rabbit anti-chicken IgY HRP-conjugate for one hour at room temperature. The slides were stained with 3-amino, 9-ethylcarbazole (AEC) chromogen, counterstained with Lillie Mayer Haematoxylin, and mounted in glyserin aqueous mount.  Antigen-antibody reaction was investigated under a microscope. The result showed that antigen was appeared in the tissues such as cuticle, epicuticle, buccal cavity, and eggs inside the uterine of A. galli. This research concluded that IgY stimulated by the excretory/secretory was able to recognized the antigen scattered in the tissues of A. galli so the IgY could be applied for immunodiagnostic.

Abstract

Intestinal defense mechanism against helminthes parasitic nematode to be associated with mucosal mast cells reaction. The aim of this research was to examine the effect of infection by Ascaridia galli parasite to trigger mucosal defense based on mucosal mast cells response in laying hens. Amount of ten head laying hens 12-wk old were divided into two groups containing five chickens of each. The first group, chickens were left as un-infected controls. The second group, chickens were infected orally with 1,000 embryonated eggs of A. galli.  Mucosal mast cell responses were assayed by in situ jejunal mast cell counts in stained serial histological sections with Alcian blue (pH 0.3) and Safranin-O (pH 0.1) of the jejunum. Mucosal mast cells response were observed and counted on days 14 post infection. The result showed that A. galli infection was able to increase significantly (P<0.05) mast cells response. This research concluded that the A. galli infection can trigger the involment of mucosal mast cells response in jejunal defense of laying hens against parasitic diseases caused by A. galli.

Abstract

Generally, Bacteria in the intestine consist of beneficial bacteria and bacteria that have the potential to disrupt animal health such as pathogenic bacteria. The presence of pathogenic bacteria is one of the factors that cause gastrointestinal dysfunction, but can be suppressed by maintaining the presence of beneficial bacteria for the digestive tract, especially the type of Lactic Acid Bacteria (LAB) (Astuti, 2016). LAB has antagonistic activity because it is able to inhibit pathogenic bacteria so it can compete to maintain normal flora balance in the digestive tract.  The main inhibitory activity of LAB is caused by the accumulation of primary metabolites such as lactic acid, acetic acid, ethanol, and carbon dioxide. In addition, LAB is also capable of producing antibacterial compounds such as bacteriocins (Furtado et al., 2014). Naturally LAB has existed in the digestive tract of chickens, but it is easy to experience changes in the amount due to the influence of given the feed (Widodo et al., 2015). Feed and the environment can affect the composition of microbes in the digestive tract in chickens(Ghadban, 2002; Apajalahti et al., 2004). Provision of fermented feed can improve the composition of intestinal microflora and increase the number of LAB. Fermented feed is generally easily biodegradable and has a higher nutritional value than the original ingredient and it can reduce the pH of the digestive tract.AKBISprob is an alternative product to increase poultry production in the form of supplement made from a mixture of soybean wate, and palm kernel meal which is fermented with Aspergillus niger.  The fermentation process using Aspergillus niger can reduce the crude fiber contained in AKBIS, because the mold can produce amylase, pectinase, amyloglucosidase and cellulase enzymes which can degrade cellulose so that it is easier to digest. In addition, Aspergillus niger also produces metabolites in the form of citric acid which is a component that can reduce intestinal pH so that it is suitable for LAB growth. Based on research conducted by Nurliana et al. (2016; 2017), supplementation of 4% AKBISprob can maintain the chickens production and health as well increase the number of LAB and reduce the number of pathogenic bacteria such as Escherichia coli and Salmonella sp. in chickens intestine. Thus creating a balance of microflora in the digestive tract of laying hens. Based on the above reasons, it is necessary to conduct research on antibacterial detection of BAL in laying hens that have been given AKBISprob.

Abstract

The present study aimed to isolate gram-negative bacteria infected metacarpal of Panthera tigris sumatrae trapped in Subulussalam, Aceh Province. Swab sample was obtained from metacarpal injury of P. tigris sumatrae. Swab sample was cultured to nutrient broth media using sterile cotton swabs or Pasteur pipettes, and incubated at 37°C temperature for 24 hours. Culture was spared on MacConkey media and incubated again at 37°C temperature for 24 hours. Determination of bacteria colony growth on the surface of MacConkey media based on shape, colour, surface, size, and viscosity (consistency). The bacteria colony stained with Gram staining, and tested biochemically.  The result showed that gram-negative bacteria such as Citrobacter sp., Proteus sp., Providencia sp., Pseudomonas sp.,and Salmonella sp. isolated from metacarpal injury of P. tigris sumatrae.

Abstract

This study was aimed at finding out the histopathological changes of chicken intestines that were naturally infected by Ascaridia galli. Ten intestines were obtained from wet market in Banda Aceh then measured in length and divided into three sections: duodenum, jejunum and ileum, of which histologicslides were made. The number of Ascaridia galli founded in the lumen and ingesta were counted. Parameter picture of histopathologycal changes were analized descriptively based on desquamation, hiperplasia, and villi fussion occure. Duodenum, jejunum and ileum infected by A. galli showed istopathological changes of villi the cover of desquamation, hyperplasia and fusion. Demage level of the small intestine chicken was largely determined by the number of infecting by A. galli. It was concluded that the more number of A galli infected to chickens, the higher level of damage would be occured in the small intestine of chicken

Abstract

Enzyme catalyse a broad spectrum of important biological reactions, including protein metabolism and immune reactions. A study was carried out to partial characterize enzyme from exretory/secretory of A. galli L3 stage based on influence of inhibitor. A. galli L3 were recovered from intestines of 100 heads chickens 7 days after oesophagus inoculation with 6000 L2. L3 recovered in this manner were cultured (5 – 10 ml-1) in flasks containing rosswell park memorial institute (RPMI) 1640 media, pH 6.8, without phenol red. Cultures were incubated at 370C in 5% CO2 and culture fluid was collected after 3 days in culture. Excretory/secretory was prepared from metabolic product of L3 released in culture medium. The enzyme activity was assayed against casein 2%. The amount of concentration was determined from the absorbance at 595 nm. Inhibitor sensitivity on enzyme activity was studied in phenil methanyl methane sulfonyl fluoride (PMSF) 0,5 and 1 mM.    The result showed that enzyme activity of A. galli larvae was inhibited by PMSF 0.5 and 1 mM, which enzyme activity remainded 1,8 and 0%, respectively. The results indicate that the excretory/secretory secreted by L3  A. galli contained serine protease

Abstract

This research was carried out to investigate the infiltration of inflamatory cells in the intestines that were naturally infected by Ascaridia galli. Ten intestines were obtained from wet market in Banda Aceh. Inflamatory cells were assayed by in situ jejunal mast cell counts in stained histological sections of the duodenum, jejunum and ileum, of which histologic slides were made. Duodenum, jejunum and ileuminfected by Ascaridia galli showed hyperemia, and inflammatory cell infiltration in part of A. galli infection
