Abstract

Type 2 Diabetes Mellitus (T2DM) is the more common type of diabetes results from the ineffective use of insulin. Improvement of the metabolic system in T2DM patients can be done through the regulation of gut microbiota balance. Gut microbial improvement can be modulated directly by probiotic food consumption. Soygurt is probiotic food with a low glycemic index (GI) and glycemic load (GL) value and rich in isoflavones, which has a potential effect in reducing diabetes risk. The aim of this study is to determine the effect of soygurt consumption in blood glucose levels and body weight of albino wistar rats (Rattus norvegitus). Reseach using a completely randomized design for experimental study. Subjects of this research are 30 male rats (R. norvegistus) aged 2-3 months with average body weight 150-200 gr. Abstract

Perilaku merokok berkorelasi positif dengan meningkatnya angka kejadian infeksi oleh Methicillin Resistant Staphylococcus aureus atau dikenal dengan MRSA. Perokok dilaporkan memiliki tingkat kolonisasi MRSA lebih tinggi dibandingkan non-perokok. Hal tersebut akan sangat berbahaya karena infeksi MRSA dapat menyebabkan infeksi parah seperti infeksi aliran darah, infeksi paru-paru atau necrotising fasciitis (kerusakan jaringan). Penelitian ini bertujuan untuk melakukan deteksi awal keberadaan MRSA yang berasosiasi dengan perokok aktif. Responden pada penelitian ini adalah perokok aktif yang secara rutin melakukan konseling di Klinik Berhenti Merokok (KBM) Puskesmas 1 Denpasar Utara yang memenuhi kriteria inklusi dan eksklusi. Pengujian bakteriologi dilakukan di Laboratorium STIKES Wira Medika Bali dengan melakukan kultur terhadap spesimen apus tenggorokan responden. Identifikasi MRSA dilakukan dengan melakukan pengujian sensitivitas dengan metode Kirby Bauer menggunakan cakram antibiotik oxacillin OX 1 µg. Hasil isolasi dan identifikasi S. aureus pada sampel apus tenggorokan 13 responden didapatkan hasil sebanyak 4 isolat (30,8%) teridentifikasi sebagai S. aureus sedangkan sebanyak 9 (69,2,8%) sampel menunjukkan hasil negatif. Hasil uji sensitivitas terdapat 2 isolat yang memiliki daya hambatan intermediate yaitu SA1 dan SA2 dengan diameter daya hambatan rata-rata sebesar 12 mm dan 11,83 mm. Berdasarkan hasil tersebut disimpulkan bahwa tidak ditemukan bakteri MRSA pada sampel usap tenggorokan perokok aktif pasien konseling di KBM Puskesmas 1 Denpasar.

Abstract

Intestinal microbial dysbiosis is one of the triggers for metabolic syndrome. The increase in pathogenic bacteria triggers the high release of lipopolysaccharides which underlie the incidence of insulin resistance in diabetes mellitus. It is important to modulate the intestinal microbiota towards eubiosis through consumption of probiotics. A potential probiotic product is soy yogurt which contains live bacteria and high protein and isoflavones. The aim of this study was to examine the effect of soy yogurt supplementation on the abundance of cecum microbes (Lactic Acid Bacteria (LAB) and Escherichia coli) in diabetic rats. The research was conducted at the Laboratory of Stikes Wira Medika Bali. Type of research is experimental with a completely randomized design consit of 6 groups. The conditioning of diabetes mellitus was done by induction of nicotinamide (NA) 230 mg / kgBW and streptozotozin (STZ) 65 mg / kgBW. Abundance of cecum microbes analysis using Total Plate Count. The results showed soya yogurt supplementation in rat with diabetes mellitus had a positive correlation in improving the abundance of microbial cecum. The total abundance of LAB in diabetic rat was 7.36 ± 0.15 Log10 CFU/mL and was significantly different (p <0.05) compared to diabetic rat treated with soy yogurt supplementation, namely as much as 8.08 ± 0.1 Log10 CFU/ml. Meanwhile, the abundance of E. coli in diabetic rat was 9.42 ± 0.12 Log10 CFU/mL and was significantly different (p <0.05) compared to rats treated with soy yogurt supplementation, namely 8.14 Log10 CFU/ml.

Abstract

Kesalahan tahap pra-analitik memberikan kontribusi paling besar pada kesalahan laboratorium. Kesalahan pada tahap pra-analitik yang sering terjadi adalah hemolisis (53,2%), volume spesimen kurang (7,5%), tulisan tangan yang tidak bisa dibaca (7,1%), kesalahan identifikasi pasien, ada bekuan, vacum container yang salah/antikoagulan, volume antikoagulan yang tidak sesuai, spesimen diambil dari jalur infus, dan kesalahan waktu dalam pengambilan spesimen. Penelitian dilakukan untuk menganalisis tahapan pengambilan dan pengelolaan spesimen darah di laboratorium RSUD Wangaya Denpasar. Teknik pengambilan sampel dilakukan secara total sampling sebanyak 18 orang petugas ATLM di laboratorium Patologi Klinik. Data primer dikumpulkan dari hasil wawancara dan kuisioner untuk menganalisis pengetahuan terkait pengambilan dan pengelolaan spesimen darah. Hasil dengan skor tertinggi dan dalam kategori sangat baik yaitu pada pengambilan spesimen (97,8%). Skor terendah dan dalam kategori baik adalah pemberian identitas pasien (82,5%). Petugas laboratorium di RSUD Wangaya telah melakukan tahapan pengambilan dan pengelolaan spesimen darah sesuai peraturan Menteri Kesehatan RI tahun 2013 dan masuk dalam kategori sangat baik yaitu 91,48%.

Abstract

ABSTRAKKalsium adalah mineral penting yang paling banyak dibutuhkan bagi tubuh manusia. Defisiensi kalsium rentan terjadi pada kelompok vegetarian khususnya vegan vegetarian yang disebabkan oleh keterbatasan jenis makanan yang dikonsumsi. Berbeda dengan kelompok lacto-ovo vegetarian yang masih mengonsumsi susu dan telur sehingga masih mendapat asupan kalsium secara tidak langsung dari produk hewani. Tujuan penelitian ini untuk mengetahui perbedaan kadar kalsium darah pada vegan vegetarian dan lacto-ovo vegetarian. Sampel sebanyak 30 orang vegan vegetarian dan 30 orang lacto-ovo vegetarian. Hasil penelitian ini menunjukkan rerata kadar kalsium darah keseluruhan pada kelompok responden vegan vegetarian sebesar 8,94±0,8677 mg/dL dan pada kelompok responden lacto-ovo vegetarian sebesar 9,64±0,3234 mg/dL. Hasil uji Independent Sampel T-Test menunjukkan p value sebesar 0.000 yang berarti terdapat perbedaan kadar kalsium darah pada vegan vegetarian dan lacto-ovo vegetarian. Hal ini dapat disebabkan oleh perbedaan pada asupan makanan kelompok vegan vegetarian dan lacto-ovo vegetarian.Kata kunci : Kalsium darah, Vegan vegetarian, Lacto-ovo vegetarianABSTRACTCalcium is an important mineral that is most needed for the human body. Calcium deficiency is susceptible to vegetarian groups, especially vegan vegetarian due to the limited types of food consumed. In contrast to the lacto-ovo vegetarian group who still consume milk and eggs so that they still get calcium intake indirectly from animal products. The purpose of this study was to determine differences in blood calcium levels in vegan and lacto-ovo vegetarian. A sample of 30 vegan vegetarians and 30 lacto-ovo vegetarians. The results of this study showed the average overall blood calcium level in the respondents group are as follows: vegan vegetarian was 8.94±0.8677 mg/dL and in the lacto-ovo vegetarian group was 9.64±0.3234 mg/dL. Independent T-Test Sample test results showed p value of 0.000 which means that there were differences in blood calcium levels in vegan and lacto-ovo vegetarian. Abstract

Cholesterol is a factor that influences cases of coronary heart disease. One of the factors that influence blood cholesterol is physical activity. Each type of occupation has different physical activities and will affect blood cholesterol. In the group of office workers, less physical activity and more dominant work just sit and cause an increase in blood cholesterol levels. For groups of menial workers, more physical activity is carried out so that blood cholesterol levels can be controlled. This study aims to determine differences in total blood cholesterol levels between office workers and menial workers in Sukawati Gianyar. This type of research is analytic with cross sectional approach. Samples were 30 office workers and 30 menial workers. The results showed the mean total cholesterol level of office workers blood was 194.40 ± 30.56 mg / dL and menial workers were 158.63 ± 35.28 mg / dL. Based on the results of the Independent Sample T-Test, p value of 0.000 is obtained where p <0.05 which means there are differences in blood cholesterol levels in office workers and menial workers. The results showed the average level of total cholesterol in office workers was higher than in menial workers. This is due to lack of physical activity every day in office workers. Suggestions in this study are expected to further researchers can examine the lipid profile of office workers and menial workers.

Abstract

Obesity is excessive accumulation of fat due to an imbalance between energy intake and energy expenditure for a long time. Someone is categorized as obese if their body mass index is more than 25. Obesity triggers various diseases such as coronary heart disease, stroke, cancer and diabetes. Obesity is also one of the factors that affect the increase of triglyceride levels. Beside of increased triglyceride levels, obese patients there is also get increase in hs-CRP levels above normal limits. This study aims to determine the relationship between hs-CRP and triglycerides in obese respondents. This type of research used is correlational. The study was conducted at Siloam Hospital Bali from February to March 2023. The sample in this research was 30 obese respondents who were examined for hs-CRP and triglyceride levels. The results showed that 13 people (43,3%) were in the high category and 1 person (3,3%) was in the very high category. Hs-CRP levels in the respondents were 25 people (83%) with normal hs-CRP levels or less than 5 mg/l and 5 people (17%) with hs-CRP levels of 5 mg/l. Based on the results of the correlation test, it was found that the p-value was 0,505 which indicated a p-value greater than 0,05, meaning that there was no correlation between hs-CRP levels and triglyceride levels. This is probably because the respondent has not carried out strenuous activities, no inflammation and has received treatment related to obesity status.

Abstract

Dengue Hemorrhagic Fever (DHF) is a viral infection marked by a significant decrease in platelet count. Accurate platelet count testing is crucial for diagnosing and monitoring DHF patients. The pre-analytical phase is a critical stage in laboratory testing, where errors can significantly impact test outcomes. Anticoagulants, such as K3EDTA and 3.2% sodium citrate, are commonly used in routine hematology tests to prevent blood clotting by binding calcium ions and inhibiting clotting factor activation. This study aimed to evaluate the difference in platelet counts between blood samples collected with K3EDTA and 3.2% sodium citrate anticoagulants in DHF patients. An analytical observational study with a cross-sectional design was conducted involving 50 purposively sampled DHF patients. Statistical analysis yielded a p-value of 0.615 (p > 0.05), indicating no significant difference in platelet counts between the two anticoagulants. It can be concluded that the use of K3EDTA and 3.2% sodium citrate does not significantly affect platelet count results in DHF patients. Future studies are recommended to involve larger sample sizes and consider microscopic examination methods, such as peripheral blood smears, to assess platelet aggregation visually.

Abstract

Laboratory staff knowledge is one of the key factors in ensuring the success of internal quality control in blood glucose testing. This study aims to determine the relationship between laboratory staff knowledge and the implementation of internal quality control at the Laboratory of RSUD Bahteramas, Southeast Sulawesi Province. This research used a quantitative approach with a correlational design. Data collection was carried out through questionnaires distributed to laboratory staff. Instrument testing was conducted through pilot testing, and data normality was assessed using the Shapiro-Wilk test. The relationship was analyzed using the Spearman correlation test. The results showed a significant relationship between staff knowledge and the implementation of internal quality control (p < 0.05). High knowledge contributed to more consistent and accurate application of quality procedures. These findings are supported by previous studies showing that staff knowledge competence positively correlates with the quality of laboratory services. This study is expected to serve as a basis for hospital management to improve technical training and supervision of laboratory staff.

Abstract

Errors in the pre-analytical phase are a dominant factor causing inaccurate laboratory test results, including blood serum cholesterol analysis. Pre-examination specimen handling is crucial to ensure the quality of measurement results. This study aimed to compare serum cholesterol levels in blood frozen before centrifugation with those immediately centrifuged. The research method was analytical using a cross-sectional design. A total of 35 respondents were involved, consisting of laboratory staff, emergency room staff, pharmacists, and administration at Sinar Kasih GKST Tentena Hospital. Blood samples were obtained via venipuncture after participants had fasted for 10–12 hours. Each sample was then separated into two groups: those immediately centrifuged and those frozen 20–30 minutes before centrifugation. Data collection was carried out by examining serum cholesterol levels using an Erba Mannheim Chem-7 photometer. The results showed that the average cholesterol level in frozen samples was higher (161.4 mg/dL) compared to samples immediately centrifuged (140.45 mg/dL). The Mann-Whitney statistical test yielded a p-value of 0.006 (p<0.05), indicating a significant difference between the two treatments. This confirms that pre-analytical handling of blood specimens significantly impacts the validity of cholesterol test results. Therefore, careful attention to the pre-analytical stage is essential to ensure the accuracy of laboratory results.
