Abstract

Background: Increased activity of Adenosine Monophosphate-Activated Protein Kinase (AMPK) can inhibit cell growth and biosynthetic pathwaysin some cancer cells. Recently, several endogenous ligands have been identified to activate AMPK including Adenosine Monophosphate (AMP).Virtual screening is one of the recent technique, which is used for drug discoveries. Therefore the aim of this study was to identify active compounds of Indonesian herbal plants that activate AMPK by using molecular docking. Methods: Samples used in this study wereall of active compounds which were derived from Indonesian herbal plantsand had three-dimensional structures. Molecular docking was performed by using AutodockVina version 1.1.2 to evaluate interaction between AMPK and active compounds. The AMP structure obtained from protein data bank (www.rcsb.org/pdb)was used as a molecule standard of AMPK interaction. Docking score indicated affinity of molecule interaction and was determined in Cal/mol. To visualize docking results that fit to the AMPK molecule, chimera software version 1.9 was used. Results: Curcumin has docking scores as same as docking score of AMP (-7.5 Cal/mol) and has hydrogen bound in ? subunit of AMPK (Thr86, Thr88, and Arg117). Conclusions: Curcumin might be an activator of AMPK. Another molecular docking which has a flexible substrate-enzyme algorithm is required for confirming the docking result. Keywords: AMPK, Herbal plant, Molecular Docking.

Abstract

Background: Young women can suffer from anemia due to monthly menstruation and by having many activities without adequate iron-filled nutrition. Otherproblems found on them are overweight and obesity. The data show that one with overweight or obesity can lead to iron deficiency anemia (IDA) along with increasingproinflammatory cytokines, which will cause the transcription of hepcidin. The increaseof hepcidin transcription will inhibit the absorption of iron from the intestine into the circulation, inhibiting spending iron reserves into circulation, and inhibitingthe release of iron recycled from splenic macrophages. This can cause IDA. The purpose of this study was to obtain the proportion of iron deficiency anemia in overweight or obese young women. Methods: The design of this study was cross sectional using purposive sampling. The subjects of this study were the student from fiveselected Senior high schools inBoyolali, Central Java, Indonesia. The determination of IDAis done by using hemoglobin and indexes of erythrocytes. Results: This study was the follow-upof the previous study, which was conducted to 14 obese and/or overweightyoung women withanemia From 14 subjects, there wasone subject suffered from other anemia and 13 subjects with IDA (using index erythrocytes examination). Their mean of MCV was 72.68±4.29fl, MCH was 23.08±2.63pg, and MCHC was 35.21±0.65 %.Conclusions: From this study, almost all overweight or obeseyoung women with anemia were indicated as IDA (92.8%).

Abstract

Patients with obesity often fail to control their body weight through diet and physical activity, while the existing anti-obesity drugs have some side effects for long-term use. Banana is Indonesia's tropical fruits with the highest production, and the banana peel has become soil waste. This study aimed to analyze nutrients and active compounds of methanol extracts of raja and kepok banana peels (ERP and EKP respectively). Macro and micro-nutrient compositions of the ERP and EKP were determined using the proximate analysis, while Thin Layer Chromatography (TLC) was used to detect the ERP and EKP active compounds. The ERP contained 0.11% crude fibers, 4.44% fat, and 58.08% carbohydrates, different from the EKP (4.25% protein, 20.95% water, and 14.45% minerals). By contrast, both the ERP and EKP contained flavonoids, phenols, tannins, alkaloids, and saponins. In conclusion, ERP is potentially developed as a nutraceutical for obesity treatment in the future. Further study is needed to measure specific fat levels and to analyze specific active compounds in the ERP which can modulate gene expression and interact with protein targets in lipid metabolism.

Abstract

Anemia remains a global nutritional problem, which prevalence continues to increase every year. Children, female adolescents, and pregnant women are more susceptible to anemia than other age groups. Prolonged anemia has negative impacts on their life such as premature birth, low birth weight (LBW) babies and intrauterine fetal death. Snake fruit Pondoh (Salacca edulis Reinw) is an original Indonesian fruit, which is well cultivated at the Sleman regency, Yogyakarta. Snake fruit seeds contribute to 25-30% of the total fruit, which has not been established their health effect. Therefore, this study aims to analyze the nutritional content of aqueous extract of snake fruit seeds (AES). The powder of snake fruit seeds was made using a previous method, which was adopted from our research group. The powder was then extracted using hot aqueous with 65°C. A proximate analysis was used to determine the AES nutritional content. A hundred g AES contained 21.92% water, 41.10% carbohydrates, 12.66% protein, 4.51% fat, 0.62% tannins, 32.8 mg Iron, 10.25 mg zinc, 276.42 mg magnesium, and 495.48 mg vitamin C. In conclusion, the AES has complete macronutrients and contains high levels of Iron, Magnesium, and vitamin C, which becomes a potential nutraceutical for anemia treatment. Further research is required to confirm the beneficial effect of AES administration in female rats with anemia.

Abstract

Protein-energy deficiency (PED) is a major infant health problem in developing countries with high mortality rate. Inadequate food intake and infectious diseases are the main factors for PED. In clinical setting, branded foods are commonly used to treat the PED but these foods are expensive. Canna (Canna edulis Ker.), catfish (Pangasius sp.), and red beans (Phaseolus vulgaris) are easily found in Indonesia and rich carbohydrates and protein. Therefore, this study aimed to compare the effect between pellets and porridge from canna, catfish, and red beans (GANIME) on the body weight (BW) of rats with PED.Ten male wistar rats, which aged 3 weeks and weighed 50-100 g, were used in this in vivo study. The rats model with PED was made by using the existing method through 40-60% gradual feed restriction for 14 days. Selected rats were randomly divided into 2 groups: G1 was given GANIME pellets and G2 was given GANIME porridge for 14 days. The rat BW each group routinely weighed every 7 days. The average BW of G1 rats was 78,00 ± 15,52 g, which was not significant different from the average BW of G2 rats was 70,50 ± 2,12 g (p= 0,64). In group G1 there was a greater increase than in group G2, namely 12.67 grams.Administration of GANIME pellets increases more body weight in rats with PED, compared to administration of GANIME porridge.
