Abstract

This study aims to describe the process of making audio-visual media and to determine the feasibility of it based on the assessment of material experts, media experts and student responses. This research is a descriptive study that adopts the ADDIE development step which has five stages. However, this research is limited to only three stages, the analysis, design, and development stages. The population used in this study were all majoring students of Biology in the 2018 at Unimed and the sample of this study were 40 majoring students of Biology in the 2018 who had taken microbiology courses at Unimed. Theinstrument used in this study was a questionnaire using a Likert scale. The results showed that videos of sterilization and media making, videos of observing bacterial colonies and counting the number of bacteria using the pour plate dilution method, isolation videos of pure bacteria cultures, videos of bacterial staining, videos of bacterial biochemical activity, and video of testing of antibacterial activity on sponge symbionts against bacteria MDR (Multi Drug Resistance) is categorized as very feasible based on the assessment of material experts, media experts and student responses.

Abstract

This study aims to determine the feasibility of worksheets based on guided inquiry using Articulate storyline on fungi material. This study uses the ADDIE development model which consists of five steps, namely analysis, design, development, implementation, and evaluation. The results showed that teachers and students needed worksheet so that the worksheet  was designed based on guided inquiry. The validation of material experts, learning experts, and design experts are in the very good category with an average score of 96.87%, 96.87%, and 88.46%. The biology teacher's assessment obtained an average of 98.4% which was included in the very good category, and the student response to the worksheet obtained an average of 96.07% which was included in the good category. The product developed is effective in achieving learning indicators on fungi subject matter in terms of the N-gain test with a medium category of 0.65%. So, it can be concluded that worksheet is suitable for use in the learning process.

Abstract

This study elucidates the role of secondary metabolites in green betel leaves (Piper betle) as antibacterial agents against Bacillus cereus, a bacterium causing various foodborne illnesses in humans. The experimental research was conducted at the Biology Laboratory, Faculty of Mathematics and Natural Sciences, Universitas Negeri Medan, utilizing Bacillus cereus bacterial isolates and extracting betel leaves using water. The measurement of inhibition zones was performed to evaluate the antibacterial activity of the betel leaf extract. The research results indicate the presence of an inhibition zone against Bacillus cereus growth, albeit categorized as weak. The measurements varied across repetitions, namely 2.6 mm, 3.35 mm, and 1.05 mm. The average of these three results was 3 mm.

Abstract

This study aims to determine the presence of antibacterial-producing bacteria from sponges from Bakar Island, Central Tapanuli Regency and to determine the antibacterial activity of methanol extracts from potential bacterial isolates against Staphylococcus aureus and Escherichiacoli. 21 pure isolates were found and it was known that 9 isolates produced antibacterial against S. aureus and 4 isolates against E. coli. Bacterial isolates with the highest inhibition zone against bothS.aureus and E. coli were found in isolates with code SP20 with each large inhibition zone are 14.5 mm and 16.04 mm. Then microscopic observations were carried out, namely gram staining and were obtained 8 isolates were gram positive, and 1 gram was negative. The biochemical test showed that SP20 showed positive results for the Motility, TSIA and Catalase Test but negative for Citrate Test. Bacterial isolates withthe code SP20 had the largest inhibition zone so that these isolates were to be extracted.The resultsof the screening test showed that the secondary metabolitesof the sponge symbiont bacterial extract from Bakar Island were positive for flavonoids and saponins. And then test the activity of secondary metabolites with 3 concentrations, 10%, 20%, and 30%.The results showed that the best concentration of the test bacteria was S. aureus at a concentration of 30% with an inhibition zone of 3.08 mm and E. coli at 5.04 mm.

Abstract

The use of enzymes in Indonesia is increasing every year, so action is needed to harmonize the production and needs of enzymes in Indonesia. Enzymes can be produced from various sources such as plants, animals and microorganisms. The advantage of utilizing microorganisms in enzyme production is that they can be produced in large quantities with a shorter time and can be produced continuously. One of the sources of bacterial screening to produce enzymes is figs. This study aims to look at the extracellular enzyme activity of fig endophytic bacteria and identify endophytic bacteria capable of producing extracellular enzymes such as proteases, amylase, cellulase and lipase. Isolation and purification of bacteria using NA media for 24 hours. Test of extracellular enzyme activity using NA media enriched with amylum, tween 80, skim milk, CMC. Biochemical tests are performed to identify endophytic bacteria of figs producing extracellular enzymes. The results of the study were 21 isolates of fig endophytic bacteria, as many as 14 isolates were able to produce extracellular enzymes. The identification results of fig endophytic bacteria producing extracellular enzymes obtained 8 isolates suspected to be from the genus Bacillus, 3 isolates suspected to be from the genus Micrococcus, 1 isolate suspected to be from the genus Cellulomonas, 2 isolates suspected to be from the genus Acetobacter

Abstract

Antibiotic resistance has the greatest impact on human health, at least 2 million people are infected by antibiotic-resistant bacteria. The use of natural medicines is increasingly in demand by the public, one of which is often used to treat diseases is trigona honey. This study aims to determine the antibacterial activity of Trigona honey against Escherichia coli and Klebsiella pneumoniae bacteria. The test bacteria used were Multidrug Resistant Escherichia coli and Klebsiella pneumoniae bacteria obtained from the collection of the Microbiology Laboratory of the University of North Sumatera Hospital. Antimicrobial activity testing was carried out using the hole / well diffusion method on Mueller Hinton Agar (MHA) media with chloramphenicol as a positive control, acetone as a negative control and trigona honey with a concentration of 500 ppm. Antibacterial activity was seen by measuring the inhibition zone formed. The results showed that Trigona honey was able to inhibit the growth of Escherichia coli and Klebsiella pneumoniae bacteria with inhibition zone activity included in the weak category, while the positive control showed antibacterial activity in the strong category.

Abstract

Penelitian ini  bertujuan untuk mengetahui pengaruh radiasi sinar gamma terhadap pertumbuhan dan produksi dari tanaman bawang putih (Allium sativum L.) kultivar Doulu pada generasi Mutasi Vegetatif ke 5. Penelitian ini dilakukan selama 4 bulan dari tanggal 3 September 2022 - 2 Januari 2023. Penelitian ini dilaksanakan di Desa Sionggang Utara, Kabupaten Toba Samosir, Sumatera Utara menggunakan 240 sampel siung bawang putih (Allium sativum L.) kultivar Doulu generasi MV4. Rancangan yang digunakan dalam penelitian ini yaitu Rancangan Acak Kelompok (RAK), menggunakan uji AVONA dan memakai uji lanjut BNJ 5%. Dosis perlakuan dari sinar gamma ini adalah G0 (kontrol), G1 (2 Gy), G2 (4 Gy), G3 (6 Gy), G4 (8 Gy), dan G5 (10 Gy). Parameter yang diamati yaitu umur tumbuh, jumlah daun, diameter umbi, berat umbi, jumlah siung, berat siung, dan diameter siung. Susunan daun pada tanaman bawang putih sama yaitu rendah atau low. Struktur umbi bawang putih nya regular two-fan groups, regular multi-cloved radial, regular multi-fan groups, dan irregular. Pertumbuhan bawang putih pada dosis 4 Gy yang lebih baik untuk digunakan. Sedangkan untuk umur tumbuh (4 Gy), jumlah daun (10 Gy), diameter umbi (4 Gy), berat umbi (4 Gy), jumlah siung (8 Gy), berat siung (4 Gy), dan diameter siung (4 Gy).

Abstract

This study aims to determine the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) against pathogenic bacteria and to determine secondary metabolites in kirinyuh leaves (Chromolaena odorata) using the GC-MS (Gas Chromatography-Mass Spectroscopy) method. The test bacteria used were Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa. The extraction method was carried out by maceration using 96% ethanol solvent. The crude ethanol extract of kirinyuh leaves in this study used concentrations of 30%, 40%, 50%, 60% and 70%. Using a UV-VIS spectrophotometer, the most effective concentration in inhibiting and killing the growth of Escherichia coli, Staphylococcus aureus and Pseudomonas aeruginosa bacteria was a concentration of 70%, with an average OD (absorbance) value of -0.2029, -0.055, and -0.0523. Then secondary metabolite identification was carried out using GC-MS (Gas Chromatography-Mass Spectroscopy) ethanol-water (9:1). The test results showed that a concentration of 70% was the most effective in inhibiting and killing Escherichia coli, Staphylococcus aureus and Pseudomonas aeruginosa bacteria. The content of secondary metabolites in the ethanol extract of kirinyuh leaves contains 33 secondary metabolite compounds, 6 of which are dominant compounds as antibacterials.

Abstract

This study aims to determine the application of e-worksheet with a scientific approach in class XI IPA SMA Unggulan Al-Manar in the learning process as measured by students' critical thinking skills. The instrument for measuring the question is administered both before and after the implementation of the learning. The data analysis technique employs quantitative analysis, specifically the calculation of the average value, to assess the students' critical thinking skills. The results of the data analysis indicate that the implementation of learning using e-worksheet with a scientific approach can enhance the critical thinking skills of students in class XI IPA at SMA Unggulan Al-Manar. This is evidenced by the change in the cognitive value of students during the pretest and posttest, which fell within the very good category. The average value of students during the pretest was 54.13, while the average value increased to 86.75 during the posttest, indicating an improvement in critical thinking skills.

Abstract

This research employed the experimental method with descriptive data analysis. It aimed to isolate and identify sponge symbiont bacteria with antibacterial potential against Multi-Drug Resistance Organisms (MDRO) with the 16S rRNA gene marker. Based on the study results, two sponges were obtained, namely Clathrina sp and Agelas sp, each having 11 and 7 isolates of symbiont bacteria, respectively. Based on the antibacterial activity test results, out of 18 isolates of symbiont bacteria, only 4 had antibacterial potential against MDRO (Klebsiella pneumoniae ESBL, Pseudomonas aeruginosa ESBL, or Staphylococcus lugdunensis MRSA) with inhibition zones of 9.3 mm, 8.3 mm, and 8.1 mm, respectively. The results of bacterial identification using the 16S rRNA gene sequence isolate S1I3, S1I9, and A1I3 belong to the Bacillus cereus and Bacillus paramycoides types. This research is expected to provide important information about sponge symbiont bacteria with antibacterial potential against MDRO.
