Abstract

Angina pektoris merupakan salah satu gejala awal yang menjadi penanda masalah kardiovaskuler berat seperti penyakit jantung koroner. Sementara gen PLA2G10 dipercaya memiliki peran, baik dalam progresi aterosklerotik yang menyebabkan deposit plak dan penyumbatan pada arteri koroner, atau justru sebaliknya yaitu antiaterogenik. Polimorfisme PLA2G10 pada titik SNP T512C (rs36072688) dan T-123/in1C (rs4003232) mampu mengubah level ekspresi gen PLA2G10 sebagai gen pengode enzim sPLA2-GX. Penelitian ini bertujuan untuk mengetahui genotip dari PLA2G10 pada titik SNP T512C (rs36072688) and T-123/in1C (rs4003232) para penderita angina pektoris dan mengkaji apakah penyakit disebabkan karena mutasi. Sampel penelitian berupa peripheral blood mononuclear cell koleksi Divisi Litbang Rumah Sakit Jantung dan Pembuluh Darah Harapan Kita. Sampel berjumlah 113, berasal dari penderita angina pektoris dengan dan tanpa plak pada arteri koroner. Hasil penelitian menunjukkan bahwa gen PLA2G10 pada titik SNP T512C (rs36072688) memiliki genotip homozigot wildtype (TT) dan T-123/In1C (rs4003232) memiliki genotip heterozigot (TC) melalui metode TaqMan® SNP Genotyping Assay, dan tidak ada polimorfisme yang ditemukan pada kedua grup sampel tersebut.

Abstract

Macaca nemestrina has been shown to respond to infectious disease agents, such as HIV, and is more sensitive compared to other species of macaques such as rhesus (M. mulatta) and cynomolgus monkeys (M. fascicularis). To evaluate M. nemestrina for the ability to support dengue (DEN) viremia and serve potentially as an improved model for testing DEN vaccines, a series of experiments were conducted using primary viral isolates from individuals with DEN virus infections. This study shows that M. nemestrina develops consistent, measurable viremia with all four DEN serotypes and produces immune responses sufficient to protect against homologous virus. Anti-dengue antibodies generated after infection are predominately IgG1. This species of monkey therefore appears to be a suitable model for testing DEN virus vaccine candidates.

Abstract

Background: Prolonged hyperglycaemia can make the pancreatic beta cells work harder and cause fatigue. When this happens, it can trigger oxidative stress reactions, which can produce free radical compounds that can damage pancreatic beta cells. The body compensates by activating protective mechanisms such as the production of antioxidant compounds to reduce the levels of free radicals in the cells. One such compound is glutathione (GSH). Insulin Gaussia Luciferase (iGL) cells are a cell line derived from rat pancreatic β (beta) cells. These cells can be used as a model of oxidative stress in hyperglycaemia to measure GSH levels and there are no studies using iGL cells to measure GSH levels. Therefore, in this study, the iGL cells are used as the object of research.Objective: To investigate the effect of GSH levels on glucose toxicity condition through in vitro experiments on iGL cells.Methods: The study used 5 different glucose concentrations of 11, 16.5, 22, 33, and 44 mM with the addition of iGL cell growth medium exposed for 7 days. We measured the amount of intracellular GSH using a colourimetric method at a wavelength of 405 nm. The analysis used in this study was a one-way ANOVA test. Differences between groups were tested using SPSS.Results: The results of this study showed that there was an increasing trend in total GSH levels on the third and seventh day.Conclusion: there was a daily decrease in GSH/cell in the iGL sample cells exposed to different concentrations of glucose for 7 days. This can be due to an increase in the oxidative stress reactions in the cells, which can lead to a decrease in the levels of antioxidants.

Abstract

Background: The number of COVID-19 cases in Indonesia has continually increased since the first cases appeared in March 2020. This disease is due to SARS-CoV-2 virus infection in the respiratory system that induces an immune response. The innate and adaptive immune response triggered the secretion of an excessive pro-inflammatory cytokine-caused cytokine storm that became one of the mechanisms of acute respiratory distress (ARDS). The anti-inflammatory cytokines (IL-10, IL-13, and IL-4) were secreted as the immune response in the ARDS condition. Purposes: This study aims to determine the ratio of the IL-6/IL-10 profile as basic information for the therapeutic approach to prevent ARDS. Methods: This cross-sectional study used stored biological material in plasma form from COVID-19 patients in Jakarta Islamic Hospital – Pondok Kopi and Dr. M. Goenawan Partowidigdo Hospital, Cisarua. The plasmas were from severe (n=20) patients and mild to moderate severity (n=25). The negative control sample was collected from 13 healthy subjects. Assessment of IL-10 levels in plasma using ELISA technique. Results: Our analysis showed that IL-10 has no statistical difference between negative control, mild to moderate, and severe categories (p=0.629). Meanwhile, the ratio IL-6/IL-10 presented statistical differences between mild to moderate and severe categories (p=0.011). The average ratio of IL-6/IL-10 in severe categories is two-fold higher than in mild-moderate categories. Conclusion: We conclude that there is a cytokine storm condition in severe COVID-19 patients with an imbalance ratio of pro-and anti-inflammatory cytokines and could be used as basic information for drug development in cytokine storm conditions to prevent ARDS.

Abstract

Background: Fase remaja merupakan fase perubahan fisik, psikologis dan sosial, yang diekspresikan melalui perilaku meniru dan eksplorasi seperti mencoba jenis kuliner baru. Banyak remaja yang melupakan kesehatan giginya, selain itu, dalam kondisi psikis yang masih labil, usia remaja menjadi periode rawan akan pengaruh pengedar narkoba. Kegiatan ini diharapkan dapat meningkatkan pemahaman dan kewaspadaan mengenai bahaya narkoba, serta meningkatkan kebersihan dalam pemeliharaan fisik terutama gigi. Metode: Sebanyak 89 siswa SMA 4 Muhammadiyah Jakarta Timur, menjadi peserta dalam kegiatan ini. Tahapan kegiatan ini dilakukan dengan pendataan tingkat pengetahuan, pemberian edukasi, evaluasi hasil edukasi, dan pendampingan. Hasil: 100% siswa-siswi SMA Muhammadiyah 4 Jakarta Timur tidak mengkonsumsi Narkoba jenis apapun, dan, 92% tidak menggunakan behel gigi. Terdapat 18% siswa-siswi perokok, 29% rutin melakukan cek kesehatan gigi ke dokter gigi, dan 38% pernah sakit gigi. Kesimpulan: Siswa SMA Muhammadiyah 4 Jakarta Timur tidak menggunakan Narkoba jenis apapun. Perlu dilakukan bimbingan dan pendampingan kepada 18% siswa-siswi yang merokok, 71% yang tidak melakukan kunjungan rutin ke dokter gigi agar mereka dapat memeperbaiki gaya hidupnya dan terhindar dari hal-hal yang kurang baik pada dirinya.
