Abstract

The Covid-19 pandemic has hit the whole world, including Indonesia. This pandemic has greatly impacted all sectors, including education. Online learning process has many obstacles and brings serious impacts. As a result, the policy of implementing face-to-face learning during pandemic was issued. Mitigation training is very important. This training attempts to discuss face-to-face learning at schools and obtain representative opinions regarding the preparation and consideration based on health reviews in relevant with the latest research and recommendations. This online training was held with the participants involving teachers and parents (n=219) from Elementary and Junior High School of Al-azhar SummareconSerpong, and has been shared on YouTube platform. Several opinions were obtained from parents who disagree with face-to-face learning’s idea. It also increased the knowledge regarding mitigation of face-to-face learning at schools. The education aspect is indeed something that must become a priority, but without compromising the student’s health aspect.

Abstract

Background: Prolonged hyperglycaemia can make the pancreatic beta cells work harder and cause fatigue. When this happens, it can trigger oxidative stress reactions, which can produce free radical compounds that can damage pancreatic beta cells. The body compensates by activating protective mechanisms such as the production of antioxidant compounds to reduce the levels of free radicals in the cells. One such compound is glutathione (GSH). Insulin Gaussia Luciferase (iGL) cells are a cell line derived from rat pancreatic β (beta) cells. These cells can be used as a model of oxidative stress in hyperglycaemia to measure GSH levels and there are no studies using iGL cells to measure GSH levels. Therefore, in this study, the iGL cells are used as the object of research.Objective: To investigate the effect of GSH levels on glucose toxicity condition through in vitro experiments on iGL cells.Methods: The study used 5 different glucose concentrations of 11, 16.5, 22, 33, and 44 mM with the addition of iGL cell growth medium exposed for 7 days. We measured the amount of intracellular GSH using a colourimetric method at a wavelength of 405 nm. The analysis used in this study was a one-way ANOVA test. Differences between groups were tested using SPSS.Results: The results of this study showed that there was an increasing trend in total GSH levels on the third and seventh day.Conclusion: there was a daily decrease in GSH/cell in the iGL sample cells exposed to different concentrations of glucose for 7 days. This can be due to an increase in the oxidative stress reactions in the cells, which can lead to a decrease in the levels of antioxidants.

Abstract

Background. Vitamin B12 deficiency is a significant problem, particularly in developing countries. The increasing complexity of the Vitamin B12 molecule, the deficiency condition, causes a complex cell level disorder. The clinical manifestation of vitamin B12 deficiency is anemia, but it is believed that due to the complexity of vitamin B12, there are still other molecular disorders. This study aims to establish the pathway of damage caused by vitamin B12 deficiency in the Nrf2 molecule. Methods. An animal experiment involving 12 male Sprague Dawley rats were randomly divided into control and 16-week treatment. There are two research groups, namely the control group and the treatment group. The control group was given a standard diet, while the treatment group was assigned a vitamin B12 deficiency diet. At the end of treatment, The levels of vitamin B12 in the liver tissue and plasma NRF2 were measured. Statistical Analysis. Nrf2 and vitamin B12 levels were statistically analyzed using the Student's T-test, and the correlation between vitamin B12 and Hb was analyzed using the Spearmen test. Data that cannot use by Independent T-test were tested using the Mann Whitney test. Results/Conclusion. There was no significant difference between the control and treatment groups on plasma Nrf2 levels (4.02 ± 0.59 pg/ml vs 3.34 ± 0.55 pg/ml; p = 0.07) and tissue vitamin B12 (0.0035 ± 0.0016 µg/ml vs 0.0030 ± 0.0006 µg/ml ; p = 0.8). Abstract

Penyakit Jantung Koroner (PJK) merupakan salah satu spektrum penyakit jantung yang paling umum di dunia, dengan tingkat morbiditas dan mortalitas yang tinggi. Pada tahun 2020, PJK diperkirakan menjadi penyebab utama kematian global, menyumbang 36% dari seluruh kematian, angka ini dua kali lipat dibandingkan kematian akibat kanker. Di Indonesia, PJK (yang termasuk dalam kategori penyakit sistem sirkulasi) tercatat sebagai penyebab utama kematian, dengan angka 26,4%, atau empat kali lebih tinggi dibandingkan kematian akibat kanker yang hanya sebesar 6%. Dengan demikian, sekitar satu dari empat kematian di Indonesia disebabkan oleh PJKKader puskesmas berfungsi sebagai perpanjangan puskesmas dalam menjangkau dan melayani masyarakat di area kerjanya. Berdasarkan permasalahan ini, kegiatan pengabdian masyarakat bertujuan memberikan edukasi kepada kader puskesmas tentang deteksi dini, pencegahan PJK, dan faktor risikonya. Dengan demikian, kader diharapkan dapat mengedukasi masyarakat dan mendorong mereka untuk melakukan deteksi dini serta pencegahan PJK guna mengurangi angka kematian akibat penyakit ini. Setelah pelaksanaan edukasi, terjadi peningkatan pengetahuan kader tentang PJK, yang terlihat dari kenaikan nilai pretest ke post-test sebesar 19,3%.

Abstract

Pendahuluan: Perilaku hidup masyarakat modern dapat memicu stres akibat pola makan yang tidak sehat, kurangnya aktifitas fisik, tidur yang tidak cukup, dan beban kerja yang semakin banyak. Upaya pengendalian stres dapat dilakukan dengan cara mendengarkan musik, mengobrol dengan teman, menonton film komedi, minum teh hijau, berolahraga, tidur dengan cukup, teknik pernapasan dalam, meditasi, menjalani hobi, bahkan membaca Al-Qur’an. Program ini bertujuan sebagai upaya pengendalian stres di masyarakat dengan cara membaca Al-Qur’an melalui metode DEMAJI. Metode: Edukasi dilakukan dengan metode ceramah. Peserta diminta mengisi pre test sebelum edukasi dan mengisi post test setelah edukasi dengan soal yang sama. Hasil: Kegiatan diikuti oleh 46 peserta dengan terbanyak pada kelompok usia 41-50 tahun (37%), jenis kelamin perempuan (74%), dan tingkat pendidikan SMA (46%). Hasil uji t berpasangan terdapat perbedaan signifikan (p-value=0,000) antara mean skor pre-test (78,26) dengan mean skor postest (89,13). Kesimpulan: Edukasi pengendalian stres dengan mengaji terbukti efektif meningkatkan pengetahuan peserta dalam mengendalikan stres.

Abstract

Background: Patent Ductus Arteriosus (PDA) occurs when the blood vessels connecting the aorta and pulmonary artery do not close after birth. Transcatheter closure using a PDA occluder is a first-line treatment, but hemodynamic monitoring is essential as patients may experience sudden declines during the procedure. Methods: A case study involving a 2-year-old female patient with patent ductus arteriosus who underwent transcatheter closure at the Dr Cipto Mangunkusumo National Central Public Hospital (RSCM). Results: Hemodynamic changes were found in patients who were planned for transcatheter closure. Pre-operatively the patient's hemodynamics were stable, but during the action the hemodynamic monitoring decreased and the patient's ECG experienced a change in bradycardia rhythm. Because there is no flow in the pulmonary arteries and reduced blood supply to the lungs when a PDA Occluder will be developed. The technician immediately informed the doctor and team that there was a decrease in hemodynamics and EKG. Doctor gives epinephrine 0.02 mcg IV to patient. Hemodynamics stabilized, the procedure was continued and the PDA occluder was successfully installed. Conclusion: Despite stable pre-operative hemodynamics, hemodynamic monitoring during transcatheter closure in patients with patent ductus arteriosus is essential and should not be neglected.

Abstract

Connexin-36 (Cx36) merupakan salah satu protein kanal antar sel. Protein ini paling banyak ditemukan pada otak. Gen Cx36 mengekspresikan protein penghubung Cx36 yang membentuk sinaps listrik. Penelitian ini bertujuan untuk memperoleh desain primer terbaik untuk menganalisis ekspresi Cx36 pada otak tikus dengan metode Quantitative Real Time PCR (qRT-PCR). Jenis penelitian ini adalah penelitian true experimental menggunakan mRNA dari 4 otak tikus. Analisis in silico menggunakan tools Primer 3 dan OlygoAnalizerTM by IDTdengan menggunakan database gen pada NCBI. Tiga primer dengan kriteria yang paling optimum kemudian dievaluasi dengan metode qRT-PCR. Berdasarkan hasil penelitian ketiganya mampu mengamplifikasi gen Cx36 dengan menggunakan qRT-PCR. Hasil primer terbaik menunjukkan melt curve tunggal pada primer no.5 gen Cx36 pada kisaran Tm 80-81oC. Hasil perbandingan uji in silico primer Cx36 yang sudah pernah dipublikasi sebelumnya menunjukkan bahwa primer no. 5 juga memiliki nilai optimal pada setiap kriteria. Maka hasil desain primer yang memiliki spesifisitas dan efektifitas yang optimum untuk mengamplifikasi gen Cx36 pada otak tikus Rattus norvegicus ialah primer no.5 (F: 5’-ATTTCCCGCTTCTACATCATCCAAG-3’ dan R: 5’-CACAGCAAACATGAACACCAGAAAG-3’).

Abstract

Objectives: To determine the effectiveness of Diazepam in comparison with Phenobarbital.Methods: Twenty-seven male Swiss Webster mice were used and randomly divided into three groups of negative control (NS), positive control (Phenobarbital), and diazepam group. Two tests were performed on these group: the traction Test and the Fireplace Test. Pupillary diameter was also observed.Results: A significant difference based on the Kruskal - Wallis statistical test was observed between the positive control and the diazepam group (<0.05) in the traction test, which was also true for the fireplaced test (p<0.05). The pupillary diameter in the test animals in the positive control and diazepam group was not statistically significant (p>0.05).Conclusion: Diazepam has a better sedative effect than Phenobarbital. The sedative effect produced by Diazepam is stronger, with faster onset and longer half-life than the Phenobarbital the positive control. However, different test methods and comparisons should be sought to support this conclusion.
