Abstract

ABSTRAKCallose, β-1,3-glukan, merupakan senyawa alami yang terdapat pada tanaman dan terlibat dalam sistem pertahanan tanaman. Callose merupakan bentuk pertahanan tanaman yang penting karena dapat menjadi penghalang awal masuknya patogen ke dalam tanaman. Deposisi callose termasuk ke dalam sistem pertahanan aktif karena akan terinduksi jika tanaman terserang patogen. Molekul patogen yang dilaporkan dapat menginduksi deposisi callose adalah lipopolisakarida (LPS), komponen utama membran luar bakteri gram negatif. Telah dilakukan penelitian untuk mempelajari induksi deposisi callose pada tanaman tembakau oleh LPS yang diekstraksi dari bakteri Pseudomonas syringae pv. tabaci (Pta). Bakteri Pta merupakan fitopatogen penyebab penyakit wildfire pada tembakau. Untuk pengamatan deposisi callose, daun tembakau diinfiltrasi dengan LPS Pta 400 μg/ml dan 800 μg/ml serta diinkubasi selama 24 jam dan 48 jam. Selanjutnya, klorofil daun diluruhkan menggunakan larutan laktofenol dan daun diwarnai dengan aniline blue. Hasil pengamatan menunjukkan deposisi callose yang telah diwarnai dengan aniline blue tampak berpendar warna hijau ketika diamati menggunakan mikroskop fluoresensi. Intensitas deposisi callose, hasil induksi LPS bakteri Pta, dalam satu daun tampak tidak merata di semua bagian daun, namun dan cenderung terdeposisi di sekitar pertulangan daun.

Abstract

Lipopolysaccharide (LPS) is a major component of outer-membrane gram-negative bacteria, and it can act as a Pathogen-Associated Molecular Pattern (PAMP) for perception of pathogens by plants. LPS can be recognized by plants, triggering certain plant defense-related responses, including callose deposition. This study investigated induction of callose deposition by bacterial LPS in tobacco. Tobacco leaves were infiltrated with 400 µg/mL and 800 µg/mL LPS extracted from Pseudomonas syringae pv. tabaci (Pta) and Pseudomonas syringae pv. glycinea (Pgl) and incubated for 24 h or 48 h. To detect callose deposition, tobacco leaves were cleared in lactophenol solution, stained with aniline blue, and visualized by fluorescence microscopy. Results showed that LPS from Pgl induced more callose deposition in tobacco leaves than did that from Pta. In addition, a Pearson correlation test revealed that incubation period was the most significant factor in callose deposition, followed by the type of LPS bacteria. However, LPS concentration was not significantly corelated to callose deposition in tobacco leaves

Abstract

Penelitian ini bertujuan untuk menghasilkan LKS (lembar kerja siswa) berbasis CTL (Contextual Teaching and Learning) sebagai bahan ajar siswa SMA/MA kelas XII subkonsep teknik kultur in vitro. Metode penelitian yang digunakan adalah metode penelitian dan pengembangan (Research and Development) dengan model 3D yang terdiri dari pendefinisian (define), perancangan (design), dan pengembangan (develop). Lembar kerja siswa teknik kultur in vitro berbasis CTL ini dikembangkan berdasarkan penelitian pendahuluan terkait pengamatan pertumbuhan tanaman cocor bebek terhadap berbagai media kultur alternatif, yang dijadikan konten (isi) LKS, serta menerapkan tujuh komponen CTL yaitu constructivisme, inquiry, questioning, learning community, modelling, reflection, dan authentic assesment yang digunakan untuk meningkatkan aktivitas dan keterlibatan siswa dalam proses pembelajaran. Teknik pengumpulan data dalam penelitian menggunakan angket penilaian untuk uji kelayakan LKS. Instrumen penilaian LKS menggunakan skala penilaian 1 sampai 5 yang didalamnya terdapat 8 kriteria. Uji kualitas LKS dilakukan oleh 2 orang dosen jurusan Pendidikan Biologi Untirta dan 3 orang guru Biologi SMA. Berdasarkan hasil uji kelayakan LKS berbasis CTL (Contextual Teaching and Learning) sebagai bahan ajar siswa SMA/MA kelas XII subkonsep kultur in vitro memperoleh nilai rata-rata 4,3 (skala likert) yang termasuk kedalam katagori sangat layak.

Abstract

CO2 emissions continue to increase every year. The increase in CO2 emissions in Indonesia is caused by an increase in greenhouse gases and the production of CO2 emissions is inseparable from human activities. One of the efforts to reduce CO2 emissions can be done through the Biodiversity Park (Kehati) development program. This study aims to determine the estimation of biomass, carbon stocks and carbon dioxide absorption in PT Chandra Asri's biodiversity park as one of the efforts to reduce carbon emissions. The method used in collecting data on biomass is a non-destructive sampling method using allometric equation analysis to extrapolate biomass. The results showed that the total estimated biomass of all standing categories was 2745 tons/ha, the total carbon stock was 1290.15 tons/ha and the total CO2 absorption was 4734.85 tons/ha. This shows that the carbon stock in Taman Kehati PT. Chandra Asri Cilegon is in the high category because it spans more than 100 tonnes/ha so that PT. Chandra Asri Cilegon can maximize its function as a carbon absorption and storage area to reduce CO2 emissions. Keywords: Allometric equations, biodiversity park, biomassa, carbon dioxide absorption, carbon   Abstrak Emisi CO2 terus mengalami peningkatan di tiap tahunnya. Meningkatnya emisi CO2 di Indonesia diakibatkan oleh meningkatnya gas rumah kaca (GRK) dan produksi emisi CO2 ini tidak terlepas dari aktivitas yang dilakukan oleh manusia. Salah satu upaya penurunan emisi CO2 dapat dilakukan melalui program pembangunan Taman Keanekaragaman Hayati (Kehati). Penelitian ini bertujuan untuk mengetahui perkiraan estimasi biomassa, cadangan karbon dan serapan karbon dioksida di taman kehati PT Chandra Asri sebagai salah satu upaya dalam penanganan untuk mengurangi emisi karbon. Metode yang dipakai dalam pengambilan data biomassa yaitu metode nondestructive sampling menggunakan analisis persamaan allometrik untuk mengekstrapolasi biomassa. Hasil penelitian menunjukan jumlah estimasi biomassa semua kategori tegakan sebesar 2745 ton/ha, jumlah cadangan karbon sebesar 1290,15 ton/ha dan jumlah serapan CO2 sebesar 4734,85 ton/ha. Hal ini menunjukkan bahwa cadangan karbon pada Taman Kehati PT. Chandra Asri Cilegon berada dalam kategori tinggi karena berada direntang lebih dari 100 ton/ha sehingga Taman Kehati PT. Chandra Asri Cilegon dapat memaksimalkan fungsinya sebagai salah satu kawasan penyerapan dan penyimpanan karbon untuk untuk mengurangi emisi CO2. Abstract

The rate of transpiration in plants is affected by morphological characteristics and environmental conditions. This research aimed to examine how leaf surface area influences the transpiration rate of three tropical plant species: taro (Colocasia esculenta), starfruit (Averrhoa carambola), and water guava (Syzygium aqueum). The study measured the initial and final mass of leaves over a defined observation period to calculate the transpiration rate per unit area. Findings indicated that taro leaves exhibited the highest transpiration rate (0.0628 mg·cm⁻²·min⁻¹), followed by starfruit (0.0481 mg·cm⁻²·min⁻¹), while water guava recorded the lowest value (0.0350 mg·cm⁻²·min⁻¹), despite its relatively large leaf area. These results suggest that leaf area alone is not the primary determinant of transpiration rate; other factors such as cuticle thickness, stomatal density, and environmental parameters (including humidity, temperature, and light intensity) also play significant roles. Future research is recommended to explore a wider range of plant species and controlled environmental conditions to achieve more comprehensive insights.

Abstract

Pemanfaatan teknologi laboratorium virtual dapat memberikan pengalaman praktikum yang mendukung peningkatan keterampilan proses sains siswa. Akan tetapi, diperlukan panduan berupa E-LKPD agar eksperimen lebih terarah dan efektif dalam mengembangkan keterampilan siswa. Penelitian ini bertujuan untuk mengembangkan E-LKPD berbasis guided discovery dengan memanfaatkan OLabs pada submateri fotosintesis untuk melatih keterampilan proses sains siswa serta mengetahui kelayakannya berdasarkan validasi ahli, uji coba perorangan pada guru, dan uji coba kelompok kecil pada siswa. Metode penelitian yang digunakan adalah Research and Development (R&D) dengan mengadaptasi model ADDIE, yang mencakup tahap analisis, desain, dan pengembangan sampai subtahap evaluasi formatif. Penelitian dilaksanakan pada Mei 2024–Mei 2025 di Universitas Sultan Ageng Tirtayasa dan SMAN 1 Cikeusal. Hasil validasi ahli materi menunjukkan skor 92% (sangat layak), sedangkan validasi ahli media memperoleh skor 95% (sangat layak), dengan uji reliabilitas memperoleh percentage agreement (PA) sebesar 93% (reliabel). Adapun hasil uji coba perorangan pada guru memperoleh skor 85% (sangat layak), sementara uji coba kelompok kecil pada siswa kelas XII IPA memperoleh skor 80% (layak). Berdasarkan hasil penelitian, E-LKPD berbasis guided discovery yang memanfaatkan OLabs termasuk dalam kategori sangat layak untuk dilanjutkan ke tahap implementasi sebagai bahan ajar untuk memahami konsep fotosintesis dan melatih keterampilan proses sains siswa.

Abstract

Rawa Danau Nature Reserve is a conservation area that protects various remaining flora and fauna on Java island, including birds. However, at present, Rawa Danau Nature Reserve 's existence is threatened due to changes in the structure and habitat function by the activities of the surrounding community. Therefore, it is necessary to have a solution in the form of efforts to preserve and preserve the diversity of species and their ecosystems. One form of this program is conservation education. Conservation education is a form of effort to protect biodiversity and natural resources by increasing knowledge, skills, participation, attitudes, and awareness of the importance of natural resources to sustain life. Information media is needed to facilitate the achievement of conservation education goals, one of which is the development of a birdwatching field guidebook. This study aimed to develop a birdwatching field guidebook as a medium for conservation education in the Rawa Danau Nature Reserve area The development method was used to create the field manual. The developed guidebook contains information on bird diversity, bird conservation, observation techniques, and bird observation locations, as well as bird identification sheets accompanied by bird distribution patterns in the form of descriptions and illustrative pictures or photographs of research results. The book was also developed to meet the principles of environmental education, namely to develop awareness, knowledge, attitudes, skills, and participation in protecting the environment.

Abstract

The establishment of Taman Kehati Asri by PT Chandra Asri is a form of protection of biodiversity to improve the quality of biodiversity, including fauna. The existence of fauna is very important in ecosystem activities because fauna is a consumer in the food pyramid. If a population decreases, it will have a negative impact on the continuity of food webs and hinder the flow of energy cycle. This study aimed to identify the diversity of fauna in the Taman Kehati Asri area of PT. Chandra Asri Petrochemical, in Gunung Sugih Village, Ciwandan District, Cilegon City, Banten. The research was conducted in October 2022. The methods used were point count and cruise method. Based on the research results, it was found that there were 37 species of fauna belonging to 4 classes, namely birds, mammals, herpetofauna and insects. The result of total species diversity index was 2.98 which was categorized as in the medium category. Furthermore, the fauna dominance index was 0.075 which indicated that one or several species of fauna were not dominated in that area.

Abstract

Lahan yang semakin sempit berdampak pada berkurangnya pasokan pangan di Indonesia. Menanam penggunaan sistem hidroponik dapat menjadi salah satu solusi mendukung ketahanan pangan karena sistem hidroponik dapat dilakukan di lahan yang terbatas. Tim Edu Hydro Vers menawarkan jasa pelatihan dan edukasi mengenai budidaya hidroponik berbasis media metaverse di SMA Negeri 1 Ciruas untuk mendukung ketahanan pangan di Indonesia dan dapat menjadi salah satu solusi bagi pendidik untuk mendukung kegiatan Proyek Penguatan Profil Pelajar Pancasila (P5) dengan memanfaatkan limbah botol plastik . Kegiatan pelatihan ini bertujuan untuk mewujudkan kegiatan P5 pada tema gaya hidup berkelanjutan yang disesuaikan dengan karakteristik sekolah. Pelatihan ini berbasis metaverse sebagai pengenalan peserta didik kepada teknologi terkini. Pelatihan ini menggunakan metode pelatihan terbimbing. Berdasarkan hasil kegiatan yang telah dilakukan, kegiatan pelatihan ini dapat menjadi salah satu metode yang dapat dipilih dalam tema gaya hidup berkelanjutan pada kegiatan P5 sebagai implementasi kurikulum merdeka. Setelah kegiatan pelatihan, siswa dapat melakukan pengamatan pertumbuhan dan perkembangan pada tanaman hidroponik.

Abstract

Bivalvia is an invertebrate animal that has a high level of diversity and a marine biological resource that has significant economic value. This research was conducted in November 2021, aiming to determine bivalve diversity, mapping of manrove land and the potential for bivalve diversity, the relationship between the results of analysis of the presence of water on bivalves diversity in the Pulau Dua Nature Reserve and the implications of research results in the field of education. The method used is the roaming method to determine bivalves diversity and remote sensing methods to map areas of potential bivalve diversity in Pulau Dua Nature Reserve. The results of observations found that the diversity of bivalves in Pulau Dua Nature Reserve was included in the medium diversity category with a value of 1.085. The results of the relationship between bivalve diversity and MNDWI are classified as having a strong relationship, namely the correlation coefficient (r) is 0.668. In the Pulau Dua Nature Reserve, it was found that areas with a high potential for bivalve diversity were found at coordinates 106.198956 6.018374. The results of this research were then carried out to analyze the material at KD 3.10 and KD 4.10 for class X SMA at school.
