Sorghum is a multifunctional crop that can be used as an alternative food for the people of East Nusa Tenggara. However, one of the challenges in cultivating sorghum in this region is the presence of the Rhizoctonia solani pathogen. This fungus causes a devastating root rot disease in sorghum crops, resulting in loss of crop yields and potentially disrupting food security in the region. Using resistant varieties is one of the most effective ways to prevent the growth of R. solani biologically. Therefore, it is important to study the resistance of local sorghum varieties. The purpose of this study was to determine different sorghum varieties' resistance responses to the R. solani fungus that causes root rot disease. This study used a completely randomized design (CRD) with 2 levels of treatment: the treatment of sorghum varieties and the treatment of fungi inoculums, which were inoculated separately and simultaneously. Each treatment was repeated 3 times. Observations and data collection were carried out on the 7th day, and some of the parameters observed included germination percentage, radicle length, coleoptile length, sprout length, GTI value, and disease incidence value. The sorghum variety with the highest resistance value was found in the red variety. This red variety had a disease incidence value of 63.3%, much lower than the other four test varieties. These results provide important insights for developing a more root rot disease-resistant local sorghum plant variety.

Ammonium enrichment reduces the diversity and changes the composition of ammonia-oxidizing microbial communities in agricultural soil media. Biodiversitas 25: 916-923. Nitrification is the process of ammonium oxidation to form nitrites and nitrates. Ammonium availability in the soil is one of the factors influencing the activity, abundance, and diversity of ammonia-oxidizing microorganisms (AOM). This study aimed to determine the effect of enriching a soil media with different ammonium concentrations on the abundance and diversity of AOM as well as the potential for nitrification. Soil as the media was prepared from an agricultural land receiving cow dung sewage. The media was then placed in the microcosms, and was added to a nitrification medium containing ammonium at three levels; 0 (control soil), 200 (low ammonium-enriched soil), and 500 mg L-1 (high ammonium-enriched soil). The microcosms were further incubated for 42 days at room temperature. The nitrification potential determination was based on the formation of nitrite from ammonium oxidation. The abundance of AOM and the potential nitrification were measured every 14-day interval, including day 0 as the initial condition. The AOM composition was analyzed based on the similarity level of the amoA gene sequence to the NCBI BLAST GenBank database. However, this analysis was only conducted for the control and high ammonium-enriched soil. This study indicated that ammonium enrichment increased the nitrification activity and the abundance of AOM, but it decreased the diversity of AOM communities. There were positive correlations between nitrification and nitrate potential, as well as between ammonium content and AOM abundance. Negative correlations appeared between pH and nitrification potential, nitrate concentrations, ammonium concentrations, and MPN. The diversity of ammonium-oxidizing archaea (AOA) in the control soil was higher than in the high ammonium-enriched soil. The enrichment with ammonium also changed the AOA composition. The Candidatus Nitrosocosmicus oleophilus strain of the MY3 chromosome was the most dominant archaea in the control and high ammonium-enriched soil. This implies that the high diversity of AOA in this soil may be beneficial for further use of the soil as a source for inocula in the development of nitrifiers-based biofertilizers.

Biodiversitas 25: 957-963. Water Pollution caused by Cr6+ contamination is difficult to remove from the environment. One of the efforts to reduce Cr6+ contamination in water is by bioremediation. Bioremediation using the interaction of rhizosphere bacteria with aquatic plants can be employed to decrease the content of Cr6+ in water environment. This study aimed to determine the effect of adding Microbacterium sp. SpR3, Micrococcus luteus RT-9, and a mixture of both bacteria on the ability of Eichornia crassipes to reduce the concentration of Cr6+ in the growth media and the plant's growth potential. This study was conducted in the Microbiology Laboratory of the Faculty of Biology at Satya Wacana Christian University. Eichornia crassipes plans of similar size were grown in 450 mL of Hoaglands solution with Cr6+ 5 mg.L-1 in a container. The treatment in this study involves the addition of 50 mL (10% of the working volume) of Microbacterium sp SpR3 culture, Micrococcus luteus RT- 9, and a combination of both Cr6+-reducing bacteria. The reduction rate of Cr6+ concentration in the mixture of both types of bacteria reached 88.06%, which was higher compared to the control without the addition of the inoculum (87.08%). The inoculation of both Cr6+ reducing bacteria did not hibit the growth rate of the plants. The ability of Microbacterium sp. SpR3 and Micrococcus luteus RT-9 to reduce Cr6+ in liquid media can be employed in the bioremediation of Cr6+ contaminated wastewater.

Phytohormone-producing microorganisms are an essential component of biofertilizers. One example of a phytohormone is Indole Acetic Acid (IAA). IAA-producing microorganisms can be originated from various habitats. In this study, IAA-producing bacteria will be isolated from eco-enzyme, a liquid-fermented organic material rich in benefits and contains numerous microorganisms and IAA. The research involves processes of isolation, detection of cell and IAA production, and molecular identification. Through the processes of isolation and purification, 14 bacterial isolates were obtained. After testing their ability to produce IAA using a medium containing L-tryptophan and Salkowski's reagent, only 11 isolates were found to produce IAA. The DNA of these 11 isolates was isolated, amplified, sequenced, and identified through molecular analysis. The nucleotide sequences of these 11 bacterial isolates have been registered in the gene bank and assigned accession numbers PQ095569 to PQ095579. Based on alignment and phylogenetic tree analysis, the 11 isolates were grouped into three categories: the Bacillus group, consisting of Bacillus altitudinis, Bacillus subtilis, Bacillus licheniformis, Priestia megaterium, and Paenibacillus sp.; lactic acid bacteria, including Lacticaseibacillus paracasei and Lactiplantibacillus plantarum; and vibrio-shaped bacteria, including Vibrio sp. and Vibrio diazotrophicus. The Bacillus group (including Paenibacillus megaterium) could produce high levels of IAA. However, among the members of this group, P. megaterium exhibited the highest cell production capability and IAA production, with values of 2982.208 mg·L?¹ and 35.49 mg·L?¹, respectively. This high growth ability and IAA production make P. megaterium a promising candidate as an inoculum for use as a PGPR (Plant Growth-Promoting Rhizobacterium).

Abstract

Bird’s eye chili (Capsicum frutescens L.) is a high-value horticultural crop, yet yields in Indonesia remain low due to long-term use of chemical fertilizers, which reduce soil fertility. Organic fertilizer alternatives, such as eco-enzyme and Trichoderma viride, offer potential for sustainable agriculture. This research aimed to study the impact of eco-enzyme and T. viride, applied individually and in combination, on the growth and yield of Bird’s eye chili, as well as on soil N, P, K, organic carbon, and IAA levels. The study employed a Completely Randomized Design (CRD) with four treatments: control, eco-enzyme, T. viride, and a combination of eco-enzyme and T. viride, each with eight replications. The research was conducted in a polybag over 20 weeks. Parameters observed were plant height, leaf number, root length, number of flowers and fruits, fruit weight, and soil N, P, K, C-organic, and IAA. Compared with the control treatment, the application of eco-enzyme and T. viride improved several growth and soil quality parameters. The combined treatment increased vegetative growth and enhanced soil N, P, K, and IAA contents, whereas T. viride alone produced the highest fruit yield. These results suggest that eco-enzyme and T. viride can be used as biological inputs to improve chili production while helping maintain soil fertility and reduce dependence on chemical fertilizers. The findings are relevant to SDG 2 (Zero Hunger), SDG 12 (Responsible Consumption and Production), and SDG 15 (Life on Land), particularly regarding sustainable food production, resource recycling, and soil conservation.

Abstract

Telah dilakukan kegiatan Pengabdian kepada Masyarakat berupa pendampingan siswa SMA Kota Salatiga. Kegiatan bertujuan untuk peningkatan pengetahuan dan minat generasi muda terhadap jamu melalui inovasi Minuman Jamu Herbal kekinian guna melestarikan kearifan lokal jamu. Siswa SMA di Kota Salatiga sebagai mitra kegiatan mewakili generasi muda. Kegiatan terdiri dari penggalian pengetahuan mitra tentang Minuman Jamu dan Herbal (MJH), sosialisasi tentang pemanfaatan MJH sebagai upaya meningkatkan imunitas tubuh, sosialisasi tentang mengembangkan peluang bisnis MJH; Pendampingan mitra dalam praktik inovasi produk MJH kekinian; dan (5) Evaluasi kegiatan. Sebelum pendampingan Sebagian besar siswa mitratidak terlalu paham dan berminat terhadap produk jamu. Pendampingan berhasil peningkatan pengetahuan dan minat generasi muda terhadap jamu. Siswa mitra sangat kreatif dan inovatif dalam menciptakan produk Minuman Jamu Herbal kekinian yang menarik generasi muda antara lain minuman segar masa kini, minuman krim, pudding, jelly, permen, minuman dalam bentuk serbuk, dan es krim
