Abstract

The  Tripa  Peat  Swamp  forest  is  located  in  Nagan  Raya  and  Aceh  Barat  Daya districts. The forest has been deforested and converted to agricultural lands. Hence, the objective of the present study was to evaluate the community structure of macrozoobenthos in Tripa peat swamp. Five sampling locations were determinated using purposive random sampling which was focussed in the main river mouths of Tripa, Suak Tripa, Seuneam, Batee and Taduu Rivers. The sediment was collected in three plots (1m x 1m) at every sampling location. The results showed that, there were 34 species of macrozoobenthos recorded during the sampling, it is belonging to 4 classes; Gastropoda, Bivalvia, Crustacea and Insecta.  The species richness were ranged between 1.12 to 1.58, indicates low category. The diversity and evenness indices were ranged between 0.62 to 2.44 and 0.19 to 0.73, respectively; indicates low to moderate values. It was concluded that the macrozoobenthos community of Rawa Tripa was in unstabile condition. Rawa  Gambut  Tripa  terletak  di  Kabupaten  Nagan  Raya  dan  Aceh  Barat  Daya Provinsi Aceh. Penelitian ini bertujuan untuk mengetahui komunitas makrozoobentos di kawasan Rawa Gambut Tripa. Penelitian dilakukan pada bulan Juni dan Juli 2013. Penentuan area dan titik sampling menggunakan metode purposive random sampling berdasarkan  pertimbangan  topografi  kondisi  lingkungan,  yaitu  pada  4  muara sungai utama di kawasan ini. Sampel sedimen diambil menggunakan skop dengan plot (1m x 1m) sebanyak 3 plot pada setiap stasiun, kemudian disaring dan disortir menggunakan tangan (hand sorting). Makrozoobentos yang tertangkap diawetkan dengan  formalin  10%,  selanjutnya  diidentifikasi  di  Laboratorium  Biologi  Laut Jurusan Ilmu Kelautan Fakultas Kelautan dan Perikanan. Hasil penelitian ditemukan 34 spesies makrozoobentos yang tergolong dalam 4 kelas makrozoobenthos yaitu Gastropoda, Bivalvia, Krustacea dan Insekta. Spesies yang paling banyak ditemukan adalah Faunus ater dari kelas Gastropoda. Nilai indeks kekayaan jenis berkisar 1,12- 1,58 tergolong rendah. Nilai Keragaman (H') makrozoobentos di semua stasiun berkisar antara 0,62-2,44 dan tergolong rendah sampai sedang. Nilai indeks keseragaman berkisar antara 0,19-0,73 tergolong rendah sampai dengan sedang, dan indek similaritas makrozoobentos disetiap wilayah tergolong rendah. Dengan demikian  dapat  disimpulkan  komunitas  makrozoobentos  di  kawasan  ini  dalam kondisi labil.

Abstract

The objective of the present stady was to determine the optimum level of feeding rate for peres fish (Osteochilus vittatus). The experimental fish fed with a commersial diet contains 48% crude protein. The fish sample was 1.0 cm in average length and 0.066 g in average weight at stocking density of 20 fish per aquarium (45 cm x 45 cm x 35 cm). The fish was fed three times a day at 8 AM, 12 AM and 5 PM for 60 days. The ANOVA test showed that the feeding rate gave a significant effect on weight gain, length gain, and specific growth rate (p0.05), but it not gave a significant effect on survival rate and hepatosomatic index (p0.05). In general the higher growth rates were found at 8% feeding rate, while the higher hepatosomatic index and survival rate were found at 5% feeding, but these values were not different significatly with 8% feeding rate. It is coucluded that the optimum level at feeding rate for peres fish (O. vittatus) was 8% body weight per day.Penelitian ini bertujuan untuk menentukan jumlah ransum harian yang optimal kepada benih ikan peres (Osteochilus vittatus). Pakan uji yang digunakan adalah pakan komersil yang mengandung protein 48% dengan perlakuan jumlah ransum harian yaitu 3%, 4%, 5%, 6%, 7%, 8% dan 9% dari bobot tubuh perhari. Akuarium yang digunakan dalam penelitian 45x45x35 dengan padat tebar ikan 20 ekor/akuarium. Benih yang digunakan berukuran rerata panjang 1 cm dan berat 0,066 g. Rancangan percobaan yang digunakan adalah Rancangan Acak Lengkap (RAL) dengan 7 perlakuan dan 3 kali ulangan, pakan diberikan 3 kali sehari pada pukul 08:00, 12:00, 17:00 WIB selama 60 hari. Hasil uji ANOVA menunjukkan bahwa jumlah pemberian ransum harian yang berbeda berpengaruh nyata terhadap pertambahan panjang, pertambahan bobot, pertumbuhan spesifik (0,05), namun tidak berpengaruh terhadap kelangsungan hidup dan hepatosomatk indeks (P0,05). Hasil penelitian menunjukkan pertumbuhan tertinggi dijumpai pada ransum harian 8%, hepatosomatik indeks dan kelangsungan hidup tertinggi pada ransum harian 5%. Dengan demikian dapat disimpulkan bahwajumlah ransum harian yang optimal untuk benih ikan peres (O. vittatus) adalah 8% bobot tubuh per hari.

Abstract

Seurukan fish (Osteochilus vittatus) has many advantages, besides the fish also has disadvantages which are the slow growth, so the temperature shock of triploidization technique was expected to solve the problem. The objective of the present study was to obtain an effective temperature to increase of triploidization success of seurukan fish (Osteochilus vittatus). The experimental method and completely randomized design model were used in this study. Five levels of temperature shocks at three replicates were tasted: 4? (cold), 6? (cold), 28? (normal), 35? (heat) and 37? (heat). The sperms and eggs were fertilized in the plastic jar then a total 100 of fertilized eggs (zygotes) were taken randomly 3 minutes after fertilization and soaked in respective temperature for 90 seconds, and then incubated in incubation jars at the water temperature of 28-29?. The results showed that the temperature shock gave the significant effect on the hatching and the success of triploidization success (P?0.05), but did not give the significant effect the fertility and survival rates (P>0.05). The triploid fish can be achieved using cold and heat shock, but the higher triploid fish was recorded at 37? was the best temperature recommended for triploidization of Seurukan fish.

Abstract

The objective of present study was to determine the optimum level of papain in the diet of keureling fish (Tor tambra). The complete random design was utilized in this study. Six levels of papain dosage were tested in triplicates, i.e. 0 (control); 17.5 mg kg-1, 20.0 mg kg-1, 22.5 mg kg-1, 25.0 mg kg-1 and 27.5 mg kg-1 of feed. The experimental fish were fed the experimental diet two times a day at 8 AM and 5 PM at feeding level of 5% body weight for 90 days. The Anova test result showed that papain enzyme gave a significant effect on the weight gain, daily growth rate, specific growth rate, survival rate, feed conversion ratio and feed efficiency (P<0.05). The Duncan multi-rage test result showed that the higher values for all measured parameters were obtained at the dosage of 27.5 mg kg-1. Abstract

The cryopreservation process might reduce the quality of spermatozoa due to an increase in the production of reactive oxygen species (ROS) compounds during cooling, freezing, and thawing. The quality of spermatozoa can be maintained by adding glutathione as an exogenous antioxidant into cryo-diluent. This study aimed to examine the effect of the addition of different doses of glutathione in cryopreservation of Aceh Local catfish (Clarias gariepinus) spermatozoa after freezing. The method used was a completely randomized design (CRD) with four treatments and four replications. Fresh semen was diluted in Ringer, 15% DMSO, and 20% Fetal Bovine Serum (FBS) and then added with glutathione antioxidants of 0.0 mgL-1, 0.5 mgL-1, 1.0 mgL-1, and 2.0 mgL-1. The parameters observed in this study were motility, integrity of the plasma membrane, fertility, and DNA integrity. The results showed that the concentration of glutathione had no effect on motility, integrity ofthe plasma membrane, or fertility, but had an effect on DNA integrity. The highest percentage of motility and plasma membrane integrity respectively was 40.50% (P3) and 70.87% (P2). Furthermore, the assessment of DNA integrity showed that there was no DNA fragmentation both treatments and fresh spermatozoa. This research is the first study regarding glutathione supplementation in cryo-diluent of Aceh Local catfish spermatozoa. Finally, the results obtained can provide information about the exact concentration of glutathione in the extender on the quality of spermatozoa of Aceh Local catfish (C. gariepinus) after the cryopreservation process. These results can also increase the success of fertility be used by the seed hall unit and the aquaculture industry to increase the productivity and supply high quality seeds.

Abstract

The genetic variation of Rasbora group in Lake Laut Tawar has been reported previously, however information on morphometric variations of this Genus was not available. Hence, the objective of the present study was to evaluate the morphometric characters of Rasbora group in Lake Laut Tawar, in order to contribute useful information on the biology of this important fish. The truss morphometric method was utilized in this study. A total of 45 samples of Depik, 42 samples of Eas and 44 samples of Relo were used in this evaluation. Eight homologous landmarks were determined along the outline of the fish, and based on these landmarks, 14 characters or linear measurements were recognized. Discriminant function analysis was employed to distinguish the fish sample. The results showed that the truss morphological characters could highlight the high differentiation between Relo and the other two groups, and the closeness of Eas and Depik. The morphometric data strongly indicated that Eas and Depik should be regarded as a same species, Rasbora tawarensis, and Relo may be considered as a cryptic species. This finding is in agreement to the genetic data  which was published previously.

Abstract

 Abstract –The objective of the present study was to determine the best concentration of urea solution and rinsing time to remove adhesiveness of African catfish eggs. Four different concentrations of rinse solution were used in this experiment comprising of 2 g, 4 g, 6 g and 8 g of urea diluted in 4 g NaCl/L of water producing a concentration of 0.486 mM, 0.973 mM, 1.458 mM, and 1.946 mM of urea concentration respectively, while 4 g NaCl/L of water without urea were used as control. The fish eggs were rinsed with the solutions at different durations namely; 1, 5 and 10 minutes. Each rinsing solution concentration and rinsing time were conducted in three replicates. The fertilization and hatching rates increased and peaked from the control (0 mM of urea) to 0.466 mN urea and decreased when concentration of urea was increased to 0.973 mM urea and 1.458 mM urea, then increased again at 1.946 mM urea, but these values were lower than at 0.486 mM urea. Thus, the best concentration for fertilization and hatching rates were 0.486 urea of urea. However 0.973 mM of urea was optimum in reducing clumping of eggs. Keywords: Clumping; Fertilization; Hatching; Brood fish

Abstract

The artificial breeding method for depik fish Rasbora tawarensis was not available, and the extenders and its dilution ratio are the crucial factors in artificial breeding of fishes. Herein, we tested several common sperm extenders and its dilution ratio  in the artificial breeding for depik fish. Two  series of experiments were performed to find the best extender and its dilution ratio. In the first experiment, five types of extenders, namely: Ringer’s solution, physiological solution, coconut water, sugarcane water, and roomie water. The second experiment studied four dilution levels of sperm and extender (v/v), namely: 1: 20, 1: 30, 1: 40, and 1: 50. Every treatment was conducted in four replicates and experiments were conducted in series so that the best results in the first experiment were used in the second.  The first experiment showed that Ringer’s is the best extender for depik sperm, and the second experiment revealed that Ringer’s at dilution ratio 1: 40 gave higher sperm motility, fertilization and hatching rates. Hence,  the best formula for the dilution solution of depik sperm is Ringer’s at the dilution ratio of 1: 40, sperm: extender (v/v).

Abstract

Groupers are among the fish groups that are difficult to recognize due to their high morphological similarities. Therefore, molecular techniques, particularly DNA barcoding, are extensively utilized to differentiate this fish group. This study aimed to analyze and validate six grouper species belonging to the Epinephelidae family that were harvested from Langsa district waters in Aceh province, Indonesia, based on DNA barcode data. It was conducted from June to December 2021, with the fish specimens collected from fishers at fish landing sites and the fish market in Langsa City, Aceh province. A total of 22 grouper sequences belonging to six species were generated, namely Epinephelus coioides, E. bleekeri, E. malabaricus, E. erythrurus, E. sexfasciatus and Mycteroperca poecilonotus (formerly Epinephelus poecilonotus). Genetic distance within these species ranged from 0.10 to 0.73% (average: 0.40%). Notably, E. malabaricus and E. coioides exhibited the closest genetic kinship (4.07%), while E. sexfasciatus and M. poecilonotus displayed the greatest genetic distance (19.33%). This study provides the first DNA reference for grouper in Langsa district, Indonesia, with significant implications for future sustainable grouper management.
