Abstract

Backgrounds. Diabetes mellitus is a degenerative disease that has various complications. One of neglected diabetes mellitus complication is skeletal muscle atrophy. Turmeric and java turmeric extracts were need to be clarified due to those effect as antihyperglycemic agent which might affected muscle atrophy as one of diabetes mellitus complication. Aim. To compared the efficacy of turmeric and java turmeric extract on reducing the progression of skeletal muscle atrophy in streptozotocin-induced diabetes mellitus. Methods. We conducted experimental animal studies with a post test only control group design using 25-30 grams male Swiss mice aged 12 weeks. Diabetes mellitus was induced by streptozotocin (STZ) administration. All 28 samples were randomly divided into 4 groups : control + PBS, control + STZ, STZ + turmeric, and STZ + java turmeric groups. Before treatment, blood glucose and body weight were examined on 3rd day. Blood glucose, body weight, hindlimb muscle weight, and muscle diameter results were examined after treatment (on 21st day). Data was analyzed with statistical software after all procedures were done. Results. STZ + turmeric group showed insignificant decreased in blood sugar levels (p=0.107). Control + STZ group showed significantly reduced hindlimb muscle weight compared to other groups. Hindlimb muscle weight was significantly ameliorated by STZ + turmeric extract (p=0.048). There were no significant improvement on skeletal muscle diameter among groups. Conclusion. The administration of turmeric extract improved hindlimb muscle weight but may not mediated by glucose lowering effect in streptozotocin induced diabetic mice.

Abstract

Background: The risk of anemia in stunted toddlers is higher than in normal toddlers. There is limited research on anemia in children under 3 years old in the community and there is a need for supporting research to prove the possibility of controlling stunting and anemia together. Previous studies have shown that family characteristics and immunization status are associated with the stunting and anemia.Objective: This study aims to analyze the association between anemia and stunting in toddlers that are stratified by characteristics of family and Immunization status.Methods: This study used a cross-sectional design study and was conducted on September-October 2020 in the Mlonggo District, Jepara Regency. One hundred two respondents were selected by cluster random sampling technique. The questionnaire was applied to collect the data on stunting, anemia, immunization status, age, mother's occupation and education status, SCREEM score, and APGAR score. This study used the Chi-Square statistic test.Results: The anemia status was measured by the Hb meter. The characteristics of stunting status (stunting = 17.7%, no stunting= 82.3%), anemia status (Yes = 63.7%, No = 36.3%). The association between stunting and anemia was stratified by the child age (6-23 months) (PR=1.667; 95%; CI=1.103-2.519; P=0.123), employed mother (PR=1.593; 95%CI=1.265-2.005; P=0.154), incomplete and inappropriate immunization status (PR=1.636; 95%CI= 1.132-2.366; P=0.133) and highly internal family function; (PR=1.512; 95% CI=1.270-1.799; P=0.029).Conclusion: There was an association between stunting and anemia in toddlers from child age of 6-23 months, employed mother, incomplete and inappropriate immunization, and highly family function groups.

Abstract

Background: Bacterial resistance to antibiotics is still common due to unwise use. Urinary Tract Infections (UTI), 80% ofwhich are caused by E. coli and other bacteria such as Enterobactersp, Klebsiellasp, S. aureus. Phosphomycin is a first-line antibiotic for UTI. Combining natural compounds with antibiotics is one treatment strategy to increase the effectiveness of anti-bacterialt herapy. Andrographispaniculata has been reported to have strong anti-infectiveactivity. This study aimed to prove the differences in the germ number, leukocyte esterase levels,and urine procalcitonin levels in Rattus norvegicus UTI model given the phosphomycin, Sambilotoleaf extract, and Sambiloto leaf extract-phosphomycin combination. Methods: Thirty Rattus novergicus rats were divided into five groups. All groups were induced 50 µl of E. coli bacterial inoculum for 7 days, followed by standard feed (negative control), fosfomycin (Monuril®) 54 mg (positive control), Sambilotoleaf extract (S1 [100 mg/BW], S2 [200 mg/BW],Sambiloto leaf extract-phosphomycin combination (FS1 [sambiloto 100 mg/BW and fosfomycin 54 mg], and FS2 [sambiloto200 mg/BW and phosphomycin 54 mg]) for the next 7 days orally.The germ number, leukocyte esterase, and urine procalcitonin were measured after all rats were given treatment.  Results: The largest averagere duction in the germ number, levels of leukocyte esterase, and urinary procalcitonin (4.80 ± 3.70 CFU/ml [p<0.05], 3,00 ± 6,71 cells/µL [p<0.05], 4,66 ± 1,35 ng/L [p<0.05] respectively)was observed in the combination of 200 mg/BW Sambiloto leaf extract-phosphomycin combination group.  Conclusion: A combination of Sambiloto leaf extract and phosphomycin reduced germ number, levels of leukocyte esterase and urinary procalcitonin in rat model of UTI.

Abstract

Background: Type 2 diabetes mellitus (T2DM) in obese has been considered a risk factor for male infertility. Garcinia mangostana pericarp extract (GMPE) is known to have anti-hyperglycemic, anti-diabetic, and anti-inflammatory effects. This study aimed to evaluate the effect of GMPE therapy on reproductive function in obese T2DM rats by examining testosterone level, testicular histopathological features, and hs-CRP level.Methods: Thirty-six male Wistar rats, aged 2–3 months, were randomly divided into 6 groups and treated with a standard diet (NC), high-fat diet (HFD) with GMPE 200 mg/kgBW (obese GMPE control/OGC200), HFD with 45 mg/kgBW STZ-NA (obese-diabetic control/ODC), obese-diabetic rats with GMPE 100 (DG100); 200 (DG200); and 400 mg/kgBW (DG400). STZ-NA was administered after 8 weeks of HFD treatment and followed by GMPE for 8 weeks after T2DM was confirmed. The level of hs-CRP and testosterone were measured in the serum using an enzyme-linked immunosorbent assay. Testicular histopathological examination was measured after 8 weeks of treatment by using Modified Jonhson Score (MJS) with HE staining. Results: ODC rats significantly showed increased hs-CRP level compared to NC (8.76±0.27 vs 0.30±0.07 ng/mL, p<0.001) and reduced testosterone level and MJS compared to NC (73.69±2.22 vs 170.14±1.34 ng/dL, 4.57±0.93 vs 9.87±0.16 MJS, respectively, P<0.001). Testosterone and hs-CRP levels showed a negative and significant correlation (r=-0.974 and p<0.001). On the treatment group, GMPE significantly reduced hs-CRP and increased testosterone levels in a dose-dependent manner.Conclusion: GMPE effectively protects reproductive function in obese DM rats by increasing testosterone levels and advanced spermatogenesis, as well as decreasing hs-CRP level

Abstract

Background: Ulcers are one form of chronic wounds as one form of complication experienced by leprosy sufferers. In the process of healing wounds such as ulcers, TGF-β has a broad role. Several studies have stated that in chronic wound conditions there is a decrease in TGF-β levels so that the wound is increasingly difficult to heal. This study aims to determine the relationship between TGF-β serum levels and the degree of ulcer wound healing. Methods: This study is an observational analysis study with a cross-sectional design and involved 33 leprosy ulcer patients undergoing treatment at the dr. Rehatta Jepara Hospital in the period March-April 2024. Serum TGF-β levels were measured using the Enzyme-linked immunosorbent assay (ELISA) method while the degree of wound healing was assessed by scoring the Pressure Ulcer Scale for Healing (PUSH). Bivariate analysis using the Spearman Correlation Rank Test. Results: The correlation test between serum TGF-β levels and the degree of wound healing (p = 0.807) and duration of leprosy as covariate (p = 0.96) showed insignificant results. Conclusion: There is no relationship between serum TGF-β levels and the degree of wound healing and duration of leprosy.

Abstract

Background: Purine metabolism, carried out in the liver, results in the production of uric acid. Hyperuricemia is a condition of elevated level of uric acid in the blood that can lead to gout if untreated. Lemon (Citrus Limon.) contains flavonoids which act as antioxidants and have the potential role as antihyperuricemia. Aim: To determine the effect of lemon peel extract on uric acid levels. Methods: Thirty-six male wistar rats from Farmhouse Ungaran were randomly divided into 6 groups, consists of healthy control group (K0), hyperuricemic control (K1), allopurinol control (K2), and 3 treatment groups (P1, P2, P3). Hyperuricemia induced with administration of block broth and potassium oxonate for 4 weeks. Lemon peel extract doses of 17,5, 35, and 70 mg/kg BW, and allopurinol 90 mg/kg BW was given orally for 2 weeks. Statistical analysis was conducted to evaluate differences among groups before and after the intervention. Results: There was a significant difference (p<0,05) in uric acid levels before and after intervention of lemon peel extract in groups K1, K2, and P3. Meanwhile, in groups K0, P1, and P2, there were no significant differences observed. In the pre-test uric acid levels, there was a significant difference (p<0,05) between the K0 group and K1, K2, P1, P2, and P3. Conclusion: There were no significant differences observed in the uric level acid of the treatment and control group after intervention, but there were significant differences in the effects of graded dosages between the treatment groups.

Abstract

Background: Leprosy is an infectious disease caused by Mycobacterium leprae bacteria which is still the focus of a global problem and causes disability in patients. The degree of disability in leprosy is assessed as mild and severe as a result of the inflammatory process. This study aims to determine the relationship between the degree of disability in leprosy and IL-6 serum levels as an inflammatory biomarker in leprosy patients. Methods: A Cross-sectional observational analytical study on leprosy patients aged 20-60 years, not pregnant, and undergoing treatment at Donorojo Jepara Hospital from March to April 2024. Serum IL-6 levels were analyzed using the ELISA method and tested for correlation with the degree of disability leprosy. Results: Statistical analysis of Spearman's correlation rank test shows a p value = 0.279 and the results of the line pattern on the Scatter dot graph are not linear. Conclusion: There is no correlation between serum IL-6 levels and the degree of disability in leprosy patients at Donorojo Jepara Hospital.Keywords: Degree of Disability in Leprosy, Leprosy, Serum IL-6

Abstract

Background: Fosfomycin is the first-line of antibiotic for urinary tract infection (UTI). To prevent antibiotic resistance, antibiotics and herbal medicines combination was developed. Studies have shown that Andrographis paniculata has antimicrobial and anti-inflammatory agent.Objective: To investigate the effect of A. paniculata extract for treating UTI in animal modelMethods: Using post test only control group design, thirty females Rattus norvegicus rats were induced by 1 cc transurethral inoculation of E. coli and then randomly grouped into negative control (NC) received no treatment, positive control (PC) that had 54 mg fosfomycin, and treatment groups which were divided into T1 (A. paniculata 100 mg/kgBW), T2 (A. paniculata 200 mg/kgBW), T3 (fosfomycin 54 mg with A. paniculata 100 mg/kgBW), T4 (fosfomycin 54 mg with A. paniculata 200 mg/kgBW). Urinary samples were utilized to count leukocytes under microscope, nitrite levels with a dipstick, and lactoferrin using ELISA.Results: Combination of fosfomycin and A. paniculata led to a reduction in the number of leukocytes and nitrite levels compared to the single therapy group, particularly in T4 group (p<0.001).
