Abstract

Backgrounds. Diabetes mellitus is a degenerative disease that has various complications. One of neglected diabetes mellitus complication is skeletal muscle atrophy. Turmeric and java turmeric extracts were need to be clarified due to those effect as antihyperglycemic agent which might affected muscle atrophy as one of diabetes mellitus complication. Aim. To compared the efficacy of turmeric and java turmeric extract on reducing the progression of skeletal muscle atrophy in streptozotocin-induced diabetes mellitus. Methods. We conducted experimental animal studies with a post test only control group design using 25-30 grams male Swiss mice aged 12 weeks. Diabetes mellitus was induced by streptozotocin (STZ) administration. All 28 samples were randomly divided into 4 groups : control + PBS, control + STZ, STZ + turmeric, and STZ + java turmeric groups. Before treatment, blood glucose and body weight were examined on 3rd day. Blood glucose, body weight, hindlimb muscle weight, and muscle diameter results were examined after treatment (on 21st day). Data was analyzed with statistical software after all procedures were done. Results. STZ + turmeric group showed insignificant decreased in blood sugar levels (p=0.107). Control + STZ group showed significantly reduced hindlimb muscle weight compared to other groups. Hindlimb muscle weight was significantly ameliorated by STZ + turmeric extract (p=0.048). There were no significant improvement on skeletal muscle diameter among groups. Conclusion. The administration of turmeric extract improved hindlimb muscle weight but may not mediated by glucose lowering effect in streptozotocin induced diabetic mice.

Abstract

Background: The risk of anemia in stunted toddlers is higher than in normal toddlers. There is limited research on anemia in children under 3 years old in the community and there is a need for supporting research to prove the possibility of controlling stunting and anemia together. Previous studies have shown that family characteristics and immunization status are associated with the stunting and anemia.Objective: This study aims to analyze the association between anemia and stunting in toddlers that are stratified by characteristics of family and Immunization status.Methods: This study used a cross-sectional design study and was conducted on September-October 2020 in the Mlonggo District, Jepara Regency. One hundred two respondents were selected by cluster random sampling technique. The questionnaire was applied to collect the data on stunting, anemia, immunization status, age, mother's occupation and education status, SCREEM score, and APGAR score. This study used the Chi-Square statistic test.Results: The anemia status was measured by the Hb meter. The characteristics of stunting status (stunting = 17.7%, no stunting= 82.3%), anemia status (Yes = 63.7%, No = 36.3%). The association between stunting and anemia was stratified by the child age (6-23 months) (PR=1.667; 95%; CI=1.103-2.519; P=0.123), employed mother (PR=1.593; 95%CI=1.265-2.005; P=0.154), incomplete and inappropriate immunization status (PR=1.636; 95%CI= 1.132-2.366; P=0.133) and highly internal family function; (PR=1.512; 95% CI=1.270-1.799; P=0.029).Conclusion: There was an association between stunting and anemia in toddlers from child age of 6-23 months, employed mother, incomplete and inappropriate immunization, and highly family function groups.

Abstract

Background: Bacterial resistance to antibiotics is still common due to unwise use. Urinary Tract Infections (UTI), 80% ofwhich are caused by E. coli and other bacteria such as Enterobactersp, Klebsiellasp, S. aureus. Phosphomycin is a first-line antibiotic for UTI. Combining natural compounds with antibiotics is one treatment strategy to increase the effectiveness of anti-bacterialt herapy. Andrographispaniculata has been reported to have strong anti-infectiveactivity. This study aimed to prove the differences in the germ number, leukocyte esterase levels,and urine procalcitonin levels in Rattus norvegicus UTI model given the phosphomycin, Sambilotoleaf extract, and Sambiloto leaf extract-phosphomycin combination. Methods: Thirty Rattus novergicus rats were divided into five groups. All groups were induced 50 µl of E. coli bacterial inoculum for 7 days, followed by standard feed (negative control), fosfomycin (Monuril®) 54 mg (positive control), Sambilotoleaf extract (S1 [100 mg/BW], S2 [200 mg/BW],Sambiloto leaf extract-phosphomycin combination (FS1 [sambiloto 100 mg/BW and fosfomycin 54 mg], and FS2 [sambiloto200 mg/BW and phosphomycin 54 mg]) for the next 7 days orally.The germ number, leukocyte esterase, and urine procalcitonin were measured after all rats were given treatment.  Results: The largest averagere duction in the germ number, levels of leukocyte esterase, and urinary procalcitonin (4.80 ± 3.70 CFU/ml [p<0.05], 3,00 ± 6,71 cells/µL [p<0.05], 4,66 ± 1,35 ng/L [p<0.05] respectively)was observed in the combination of 200 mg/BW Sambiloto leaf extract-phosphomycin combination group.  Conclusion: A combination of Sambiloto leaf extract and phosphomycin reduced germ number, levels of leukocyte esterase and urinary procalcitonin in rat model of UTI.

Abstract

Background: Type 2 diabetes mellitus (T2DM) in obese has been considered a risk factor for male infertility. Garcinia mangostana pericarp extract (GMPE) is known to have anti-hyperglycemic, anti-diabetic, and anti-inflammatory effects. This study aimed to evaluate the effect of GMPE therapy on reproductive function in obese T2DM rats by examining testosterone level, testicular histopathological features, and hs-CRP level.Methods: Thirty-six male Wistar rats, aged 2–3 months, were randomly divided into 6 groups and treated with a standard diet (NC), high-fat diet (HFD) with GMPE 200 mg/kgBW (obese GMPE control/OGC200), HFD with 45 mg/kgBW STZ-NA (obese-diabetic control/ODC), obese-diabetic rats with GMPE 100 (DG100); 200 (DG200); and 400 mg/kgBW (DG400). STZ-NA was administered after 8 weeks of HFD treatment and followed by GMPE for 8 weeks after T2DM was confirmed. The level of hs-CRP and testosterone were measured in the serum using an enzyme-linked immunosorbent assay. Testicular histopathological examination was measured after 8 weeks of treatment by using Modified Jonhson Score (MJS) with HE staining. Results: ODC rats significantly showed increased hs-CRP level compared to NC (8.76±0.27 vs 0.30±0.07 ng/mL, p<0.001) and reduced testosterone level and MJS compared to NC (73.69±2.22 vs 170.14±1.34 ng/dL, 4.57±0.93 vs 9.87±0.16 MJS, respectively, P<0.001). Testosterone and hs-CRP levels showed a negative and significant correlation (r=-0.974 and p<0.001). On the treatment group, GMPE significantly reduced hs-CRP and increased testosterone levels in a dose-dependent manner.Conclusion: GMPE effectively protects reproductive function in obese DM rats by increasing testosterone levels and advanced spermatogenesis, as well as decreasing hs-CRP level

Abstract

Background: Fosfomycin is the first-line of antibiotic for urinary tract infection (UTI). To prevent antibiotic resistance, antibiotics and herbal medicines combination was developed. Studies have shown that Andrographis paniculata has antimicrobial and anti-inflammatory agent.Objective: To investigate the effect of A. paniculata extract for treating UTI in animal modelMethods: Using post test only control group design, thirty females Rattus norvegicus rats were induced by 1 cc transurethral inoculation of E. coli and then randomly grouped into negative control (NC) received no treatment, positive control (PC) that had 54 mg fosfomycin, and treatment groups which were divided into T1 (A. paniculata 100 mg/kgBW), T2 (A. paniculata 200 mg/kgBW), T3 (fosfomycin 54 mg with A. paniculata 100 mg/kgBW), T4 (fosfomycin 54 mg with A. paniculata 200 mg/kgBW). Urinary samples were utilized to count leukocytes under microscope, nitrite levels with a dipstick, and lactoferrin using ELISA.Results: Combination of fosfomycin and A. paniculata led to a reduction in the number of leukocytes and nitrite levels compared to the single therapy group, particularly in T4 group (p<0.001). Abstract

Telemedicine menjadi solusi bagi pasien dan tenaga medis untuk mempermudah akses pelayanan kesehatan. Dokter dan dokter gigi perlu memiliki pengetahuan, sikap, dan praktik yang baik terhadap telemedicine sehingga mengoptimalkan manfaat telemedicine. Tujuan penelitian ini untuk memperoleh gambaran skor knowledge, attitude, dan practice dari dokter dan dokter gigi yang berpraktik di wilayah yang berbeda (Grobogan dan Kudus). Penelitian ini merupakan penelitian observasional analitik cross sectional study menggunakan intrumen kuesioner KAP. Subjek penelitian ini yaitu dokter dan dokter gigi di rumah sakit, puskesmas, dan praktik mandiri di Kabupaten Grobogan dan Kudus. Uji statistik menggunakan Mann-Whitney test (p<0,05). Hasil penelitian menunjukkan bahwa dokter dan dokter gigi memiliki skor knowledge (p=0,317) dan attitude (p=0,319) yang tidak berbeda secara signifikan, baik di wilayah kerja Kabupaten Grobogan maupun Kudus dengan skor yang tergolong sangat baik di kedua wilayah. Namun, terdapat perbedaan yang signifikan (p=0,007) pada skor practice terhadap telemedicine antara wilayah kerja Kabupaten Grobogan dan Kudus. Dokter dan dokter gigi di Kabupaten Grobogan memiliki skor practice yang tergolong tidak baik, sedangkan di Kabupaten Kudus tergolong kurang baik. Faktor lingkungan kerja, karakteristik pasien di wilayah kerja, usia dan lama praktik dapat mempengaruhi keterampilan dan praktik profesional kesehatan dalam menggunakan telemedicine sehingga perlu adanya dukungan dari berbagai pihak.

Abstract

Leprosy ulcer is one of the worsening manifestations of leprosy that often causes disability if not appropriately handled. Healing of ulcers requires a long enough duration so that a good immunological response is needed to encourage the wound healing process, one of which is transforming growth factor-  (TGF-), which is necessary of the plants that plays a role in the expression of TGF-  to spur wound healing is Curcuma longa rhizome or turmeric rhizome. This study is expected to prove that administering Curcuma longa rhizome extract can increase serum TGF-β levels in leprosy ulcer patients. This study is an experimental study with a pre and post-randomised single-blind controlled trial design involving 22 subjects with leprosy ulcers. The control group obtained standard therapy for leprosy ulcers, and the treatment group received standard therapy and adjuvant Curcuma longa rhizome extract at a dose of 1 g per day for 30 days. TGF-β levels were measured by the ELISA method. Data analysis used the Wilcoxon test and the Mann-Whitney test. The results showed no significant difference in TGF-β levels after and before the intervention in the control group (p=0.79) and the treatment group (p=0.14). However, there was a significant difference in TGF-β levels after the intervention in the control group and treatment (p=0.01). Curcuma longa rhizome extract did not show an important role in changes in serum TGF-β levels in leprosy ulcer patients. Abstract

Type 1 leprosy reaction / Reversal reaction (RR) is an inflammatory episode in existing leprosy lesions and aggravates the pain rate. Long-term therapy with steroids causes a variety of side effects, so an adjuvant therapy alternative is needed to alleviate steroid use. Curcuma longa rhizome extract has become an anti-inflammatory therapy choice in various previous inflammatory diseases. This study aims to determine the anti-inflammatory effect of Curcuma longa rhizome extract as an adjuvant therapy for reversal reaction patients by analyzing serum levels of IL-6 as a marker of inflammation. The pre and post-test randomized single-blinded controlled trial was conducted on two groups, namely control (RR patients with steroid therapy & placebo) and treatment (steroid therapy & Curcuma longa rhizome extract 1 gram/day). IL-6 serum levels were analyzed from sampling before and after the intervention for one month. The serum levels of IL-6 post-test in the control group were significantly higher than those of the pre-test. The treatment group showed lower serum levels of IL-6 post-test compared to pre-test, although there was no significant difference. Curcuma longa rhizome extract 1 gram/day for one month as adjuvant therapy for patients with type 1 leprosy reaction did not significantly reduce serum IL-6 levels.

Abstract

The maximum physical activity led to generate of free radical levels in the human body called oxidative stress. Red cabbage is well known as the vegetable containing highest anthocyanin among various other head cabbage plants. Anthocyanin is an antioxidant which is proven can prevent oxidative stress through reduction of MDA levels. The study aimed to investigate the effect of red cabbage extract (RCE) on MDA levels in rat after doing maximum physical activity. It was a true experimental study with post-test only control group design. Twenty-four male albino Wistar rats were used in this study. The rats were randomly divided into four groups i.e. the rats receiving maximum physical activity without and with RCE administration at dose of 86 mg/kg BW for eight days and the rats not receiving maximum physical activity without and with RCE administration. A swim test was used for maximum physical activity intervention. Serum MDA levels were measured using TBARS method. Data were analysed using one-way Anova followed by post-hoc LSD. A p value <0.005 was considered to be significant. The results showed that the rats treated with RCE both receiving maximum physical activity or not showed significant MDA reduction compared to rats without treated RCE (p=0.011 and p=0.027, respectively). Meanwhile, there was no significant difference in serum MDA levels between rats treated with RCE receiving maximum physical activity and rats not treated with RCE and not receiving maximum physical activity (p=0.540). Supplementation of RCE can reduce serum MDA levels in rats with and without maximum physical activity. Further study to investigate effective dose of RCE is recommended.

Abstract

BACKGROUND: Hydroxyapatite (HA) is essential for bone regeneration and healing. Green mussel (Perna viridis) shell is a potential choice for preparing HA because they are abundant, widely available, have a smaller particle size, and have a higher HA content. Rather than using HA in powder or granule form, bioscrew has been fabricated as a composite of green mussel shell-derived HA, polylactid acid and polycaprolactone. However, its dynamic bone healing process has not been clearly disclosed, therefore Procollagen 1 Intact N-Terminal Propeptide (P1NP) and bone Alkaline Phosphatase (ALP) were investigated.METHODS: Male New Zealand white rabbits (Oryctolagus cuniculus) were used as animal model. The rabbits were anesthetized and prepared for surgery. A standardized defect was created in the metaphyseal region. For the treatment group, the defect was filled with a bioscrew implant, whereas the control group did not receive any implant. At week-2, -4 and -6 post-surgery, about 3 mL of blood was collected from rabbits' marginal ear vein to collect blood serum. The serum was used to quantify P1NP and ALP levels using Enzyme-linked Immunosorbent Assay (ELISA). Data of P1NP and ALP levels were then statistical analyzed.RESULTS: P1NP level of the treatment group was significantly (p<0.05) higher than the one of control group since the first monitor, at week-2. At the next monitor (week 4 and 6), P1NP levels of the treatment group were also significantly (p<0.05) higher than the ones of control group. In accordance with the P1NP results, the ALP level of the treatment group was significantly (p<0.05) higher than the one of control group.CONCLUSION: Since bioscrew of green mussel shell-derived HA, PCL and PLA could increase the PINP associated early matrix synthesis, and ALP associated with later-stage mineralization, it can be concluded that bioscrew of green mussel shell-derived HA, PCL and PLA can be a promising material to promote bone repair.KEYWORDS: bone, HA, green mussel, bioscrew, P1NP, ALP, PCL, PLA
