Abstract

The research was aimed to understand the composition and dominancy of aquatic microalgae, and to know the potential toxicity of Cyanobacteria occuring in three lakes of Jakarta-Bogor area. The study was done in 2006. The sampling sites were Lake Sunter 2 (North Jakarta), Lake Babakan (South Jakarta), and Lake Lido (Bogor). The water samples were taken using plankton-net (20 tan mesh) with horizontal tow. The results showed that. the number of microalgal genera in Sunter 2, Babakan, and Lido were different, i.e. 10 genera, 40 genera, and 14 genera, respectively. Cyanobacteria were dominant in Sunter 2 (96%) and Babakan (90.01%). The dominant Cyanobacteria in Sunter 2 was Planktothrix agardhii (53.48%), whereas in Babakan was Chroococcus dispersus (68.52%). The second abundant Cyanobacteria in Sunter 2 was Arthrospira (42.54%). Planktothrix agardhii and Arthrospira are known to produce harmful toxin.

Abstract

Cirebon has 18 rivers that flow into the Java Sea. The Cirebon rivers play an important role in several sectors such as industrial, agriculture, household, and also aquaculture. The increasing anthropogenic activities, may have affected the health and quality of the Cirebon rivers and the surrounding area. This study aimed to determine the water pollution level of Cirebon rivers based on the Storage and Retrieval (STORET) and Pollution Index approaches. The study was conducted at seven different rivers located in the districts (kabupaten) and municipals (kotamadya) of Cirebon. The observation was performed during the end of the west monsoon (March 2019) as well as the beginning (July 2019) and the end of the east monsoon (November 2019). The STORET score of the Cirebon district rivers were between -10 and -20, which is categorized as lightly to moderately polluted. Similarly, most of the Cirebon municipal rivers were moderately contaminated throughout the year (-18 to -30). In contrast, the Pollution Index (PI) values of all observed rivers were between 1.0 to 5.0 which are categorized as lightly polluted.

Abstract

This paper discusses the results of improved optical probe that works according to optical fluorescence and absorption phenomena for measuring the phytoplankton suspension concentrations. Measurements are made on the Scenedesmus sp. culture. The laboratory test has shown that range of concentration from 104 up to around 1 x 106 cells/mL; fluorescence intensity at λ = 685 nm; and logarithmic of transmission intensity at λ = 405 nm are proportional to the cell concentration linear with proportional constant γ = 4 × 10-5 and β = –2 × 10-7 mL/cell respectively.

Abstract

Exploratory research has been carried out on Indonesian indigenous Cyanobacteria in an effort to collect these isolates to be used as learning materials for microalgae biodiversity. Cyanobacteria are known as photosynthetic bacteria and prokaryotic microalgae. The members of the cyanobacteria found were isolated, cultured, identified, and collected. The location for sampling cyanobacteria is the UI Depok campus area which consists of water from small lakes (situ) and soil. The results showed that 22 strains of Cyanobacteria were isolated and cultured properly, and consisted of 12 genera/species, namely Merismopedia sp. (1 isolate), Synechococcus sp. (1 isolate), Limnothrix redekei (1 isolate), Oscillatoria sp.1 (1 isolate), Oscillatoria sp.2 (1 isolate), Pseudanabaena sp. (3 isolates), Nostoc sp. (4 isolates), Anabaena sp. (1 isolate), Scytonema sp. (1 isolate), Fischerella sp. (1 isolate), Stigonema sp. (1 isolate), and Hapalosiphon sp. (6 isolates). Twenty-two (22) strains were then collected and treated by subculture method. Of the 11 genera found, there are 3 genera that have been known as genera that have potential as sources of biobased materials, biofuels, biofertilizers, and others, namely Synechococcus, Nostoc, and Anabaena

Abstract

The cultivation of Subang Isolated Scenedesmus in tube-shaped photobioreactors (PBRs) with the exposure of audible sound (music) was done. The study was done to know the effect of different music to the growth and culture appearance of Subang Isolated Scenedesmus. Subang Isolated Scenedesmus itself is Indonesia’s indigenous microalgae from Subang, West Java. This study was using three groups of Subang Isolated Scenedesmus PBRs. One group was control group and not exposed to any sound (Control-PBR), one group was exposed to music called “Blues for Elle” (PBR-A), and another group was exposed to music called “Bloom” (PBR-B). Based on 8 days of observation, it is known that Subang isolated Scenedesmus in PBR-A has the highest average cell density at the peak phase (1.81±0.16 x 105 cells/mL), highest growth rate (26.22 % per day), and most intense green color compared to Subang isolated Scenedesmus in PBR-B and Control-PBR. Related-Samples Friedman’s Two-Way ANOVA by Ranks test has proven that the average cell density value of Subang isolated Scenedesmus in Control-PBR, PBR-A, and PBR-B are not the same (α = 95%, sig = 0.013). Therefore, different audible sound (music) with certain intensity and frequency may affect the growth of Subang Isolated Scenedesmus differently

Abstract

One of Rawa Besar Small Lake’s roles for the surrounding community is as a fishing spot. Locals would often go fishing at the edges of the small lake. The ability of a lake to provide fishes to catch is partly due to the availability of food in the ecosystem, most importantly the microalgae community as the main producer in the freshwater food web. This concept is used in aquaculture as a way to obtain a more sustainable ingredient for feedant. Microalgae are used for aquaculture feedant due to its nutritional value. However, it is important to assess the safety and suitability of microalgae used for feedant due to the difference in toxicity and nutritional value of various microalgae species. Observation of the water samples taken from Rawa Besar Small Lake shown that potential microalgae species like Kirchneriella and Arthrospira lives in Rawa Besar Small Lake. However, toxic microalgae species like Planktothrix has been found in said water sample as well. Isolation of potential species is necessary in order to produce a healthy feedant for aquaculture out of Rawa Besar Small Lake’s microalgae community

Abstract

Diatoms are cosmopolite microalgae with unique characteristics caused of their cells are covered by silica which is called a frustule. Diatom identification is carried out using morphological features that are contained in the frustules. There are some methods for diatom preservation including using Lugol’s iodine, ethanol and formaldehyde solution. This study aims to compare those solutions. Those solutions had prepared with one control and four observations. Lugol’s iodine was prepared with a dose of 1%. Ethanol was used at doses of 20% while the formaldehyde solution was used at 4%. The observed diatom characteristics including the presence of cytoplasm and ornamentation of the diatom frustules (including striae and raphes) were observed to find the distinction in those methods. the presence of cytoplasm in each cell was not too different in the treatment, with the percentage of cytoplasmic cells being 73% (ethanol and formalin) while in Lugol the percentage of cytoplasmic cells was 65% and in the control was 76%.  Based on the results, it was found that there was no significant effect of differences in preservation methods on cells with the cytoplasm or not with a p-value> 0.05.

Abstract

The research aims to know the effect of variation temperature to the growth of Nostoc HS (Hot Spring)-5 and HS-20. Strain of Nostoc HS-5 was isolated from Ciseeng hot spring which has habitat temperature range of 30-43 °C, and Nostoc HS-20 was isolated from Pancar Mountain hot spring which has temperature range of 46-69 °C. The research was done by measuring biomass weight and chlorophyll content on day-1, 2, 3, 4, 7, 10, 14, 17, and 21. The temperatures used were 20 °C, 35 °C, and 50 °C. The growth medium used was Bold Basal Medium (BBM) with pH 6.6. Each treatment was made in four replications. Non-parametric statistical analysis used were the Friedman test (a=0.05) and Spearman test (a=0.01).The result showed there were significant differences on the biomass weight of Nostoc HS-5 and HS-20 grown at temperature of 20 °C, 35 °C, and 50 °C. The average amount of biomass highest weight for Nostoc HS-5 and HS-20 occurred in both strains were grown at 35 °C.  Besides that, there was no correlation between the weight of biomass and chlorophyll content of Nostoc HS-5 and HS-20.

Abstract

Study about the effect of hydrocarbon-polluted seawater on the cell density of microalgae Scenedesmus vacuolatus has been done. Hydrocarbon pollution derived from oil in the sea can inhibit photosynthesis process of microalgaes. This might impact the density of microalgae cells. The purposes of this study are to determine the effect of the concentration of hydrocarbon-polluted seawater on the density of Scenedesmus vacuolatus microalgae cells and to determine the optimum treatment to reduce total petroleum hydrocarbons (TPH) levels. Sampling of hydrocarbon-polluted seawater was taken at Kali Adem port, Jakarta. The treatment done in this research used Walne medium with the addition of 25% hydrocarbon-polluted seawater (A), 50% (B), 75% (C), and 100% (D). Control is Walne medium with sterile seawater that was not from the Kali Adem port. The results showed the highest average density of Scenedesmus vacuolatus cells was in the control sample. This can be seen from the results of the average cell density at peak time of 29.48 x 105 cells / mL, as well as the log phase length of Scenedesmus vacuolatus. Measurement of TPH levels showed decreases of TPH in all treatments. The optimum treatment to reduce TPH levels is on treatment B with reduction percentage of 70.62%. Abstract

Biodiversity of Cyanobacteria from several ponds/lakes of Jakarta-Depok-Bogor, Indonesia. Research about the exploration on Cyanobacteria diversity from several ponds/lakes of Jakarta-Depok-Bogor area was done. Five research locations were Sunter 2 Lake of North Jakarta, Babakan Pond of South Jakarta, Ulin-Salam dan Agathis Pond at UI Campus of Depok, dan Lido Lake of Bogor. Samples were taken by purposive sampling, and samples analysis using subsample methode. The sampling times were on August to September 2006. Three types of samples which were taken were planktonic samples, epifitic samples, and waters bank soil samples. The results showed that several toxic potential Cyanobacteria were found in several waters, even they were dominant at Sunter 2 Lake dan Agathis Pond (UI Campuss). Based on similarity levels of Cyanobacteria, the five of waters could be classied into 3 group, i.e. Sunter 2 Lake and Agathis Pond, Babakan dan Ulin-Salam Pond, dan Lido Lake. Lido Lake waters was very unique on distribution and diversity of Cyanobacteria. Based on diversity Index (H’) of Cyanobacteria, several waters condition could be catagories as moderate waters, i.e. Babakan, Situ Ulin-Salam, and Situ Agathis Pond. The epifitic and waters bank soil Cyanobacteria which were found were also found as planktonik species except Oscillatoria sp. 3. At least 26 isolate of Cyanobacteria were acquired such as Miicrocystis, Oscillatoria, Gloeocapsa, Merismopedia, Planktothrix, dan Arthrospira.
