Abstract

Peel of purple and yellow passion fruit has the potential to be an antioxidant. The flavonoid and total phenol compound groups are two examples of natural antioxidant compounds that can potentially be antioxidants. Many studies have used 70% methanol solvent to extract flavonoids and total phenolics from plants. The aim of this study is to analyze the potential of 70% methanol extract from the peel of purple and yellow passion fruit as an in vitro antioxidant using the DPPH method, total flavonoid, and phenolic levels. The passion fruit peel powder was macerated with 70% methanol for four days. The total flavonoid contents were determined using quercetin, and total phenols were determined using gallic acid. The 2.2-diphenyl-1-picrylhydrazyl (DPPH) was used for in vitro antioxidant activity by determining the percent inhibition of DPPH radicals. The flavonoid content in the 70% methanol extract of purple and yellow passion fruit peel is 1.2824 and 1.9775 mg QE/g extract, respectively. In contrast, the total phenolic content in 70% methanol extract of purple and yellow passion fruit peel is 15.8 and 3.6 mg GAE/g extract, respectively. The 70% methanol extract of purple passion fruit peel at a concentration of 80 mg/L resulted in a more significant percent inhibition of DPPH free radicals (72.80 %) than yellow passion fruit (64.91 %). In conclusion, the results suggest that 70 % methanol extract from purple and yellow passion fruit peel can decrease free radicals and be a natural antioxidant.Keyword: Antioxidant activity, Flavonoid, Methanol extract, Passion fruit peel, Total phenol

Abstract

Hyperlipidemia, especially elevated Low-Density Lipoprotein (LDL) cholesterol, is a global health problem that increases the risk of cardiovascular disease. Conventional treatment with statins is effective, but it often causes side effects. This study aims to evaluate the potential of sandalwood (Santalum album L.) leaf, biduri (Calotropis gigantea) leaf, and arabic bidara (Ziziphus spina-christi L.) leaf as an antihypercholesterolemia agent using ethanol extract to lower cholesterol levels in mice.  Extraction is carried out through maceration with 96% ethanol for 3 x 24 hours, then evaporated to produce a thick extract.  The samples used in the antihyperocholesterolterol test were extracts of Santalum album L., Calotropis gigantea, and Ziziphus spina-christi L. Six groups, six mice each, consisted of: negative control (I), positive control (II), simvastatin drug control (III), and three test dose groups (IV: 3.5 mg/20 g BW, V: 7 mg/20 g BW, VI: 14 mg/20 g BW). Cholesterol levels were measured by the Point of Care Testing (POCT) Method using a cholesterol strip test after a blood draw.  The results showed that simvastatin can lower cholesterol by 59 mg/dl. Moreover, Santalum album L. leaf extracts provided the most significant reduction in cholesterol (33 mg/dl), followed by Ziziphus spina-christi L. (31 mg/dl) and Calotropis gigantea (28 mg/dl).  Although simvastatin showed a greater decrease, the plant extract still exerted significant effects as an alternative to antihypercholesterolemia therapy. This decrease in cholesterol is related to the content of active compounds, such as flavonoids and phenolics, that reduce LDL oxidation. These findings suggest that Santalum album L. has promising potential as an herbal antihypercholesterolemic agent

Abstract

Hypercholesterolemia is a lipid metabolism disorder that increases the risk of cardiovascular disease. Although statins are effective, long-term use may cause side effects, prompting the exploration of natural alternatives that contain bioactive flavonoids, phenolics, and terpenoids with hypocholesterolemic activity. This study aimed to evaluate the cholesterol-lowering effects of ethanol extracts of Brotowali (Tinospora crispa L.), Lemongrass Stem (Cymbopogon citratus), Red Galangal (Alpinia purpurata K. Schum.), and White Galangal (Alpinia galanga L.) in hypercholesterolemia-induced male mice. A true experimental study was conducted using 36 male mice divided into six groups: Negative Control (I), Positive Control (II), Drug Control (Simvastatin) (III), and three extract groups (IV–VI) receiving doses of 14, 28, and 56 mg/20 g BW. After a seven-day adaptation period, the animals were administered propylthiouracil for 14 days to induce hypercholesterolemia, which was confirmed by blood cholesterol measurement on Day 15. Subsequently, simvastatin or the extracts were administered for three days, and cholesterol levels were measured on Day 18 using a spectrophotometric method. The results showed that all extracts reduced cholesterol levels compared to the Positive Control (194.65 ± 8.26 mg/dL), but were less effective than simvastatin (103.15 ± 15.54 mg/dL). Brotowali reduced levels to 140.90 ± 10.10 mg/dL; Lemongrass Stem to 143.47 ± 21.65 mg/dL. Red Galangal reached 138.79 ± 10.69 mg/dL and White Galangal 139.03 ± 16.53 mg/dL at the highest dose (56 mg/20 g BW). In conclusion, the four ethanol extracts showed potential as natural antihypercholesterolemic agents, with Red and White Galangal demonstrating the greatest cholesterol-lowering effects at a dose of 56 mg/20 g BW, although their efficacy remained lower than simvastatin.

Abstract

There is no consensus on the optimal dosage and mechanism of action of saga leaf ethanol extract in lowering cholesterol levels in test animals; therefore, further research is needed to understand the effectiveness and optimal dose of this plant as a hyperlipidemia therapy. This study aims to determine the optimal dose of ethanol extract from the leaves of the saga tree (Adenanthera pavonina L.) in reducing cholesterol levels in hyperlipidemic mice. The study was conducted on 36 male mice that were acclimatized for 7 days and induced hyperlipidemia using Propylthiouracil. The test animals were divided into six treatment groups, including a negative control, a positive control, a drug control (simvastatin), and three treatment groups with extract doses of 14, 28, and 56 mg/20 g body weight (BW). Cholesterol levels were measured using the spectrophotometry method, and the data were analyzed using the ANOVA test. The results of the ANOVA test showed a significance value of 0.000 (p < 0.05), indicating a difference in cholesterol levels between groups. A dose of 56 mg/20 g BW showed the highest reduction in cholesterol levels of 57.87 mg/dL. It was concluded that the ethanol extract of the leaves of the saga tree was effective in reducing the cholesterol levels of hyperlipidemic mice at an optimal dose of 56 mg/20 g BW. These results show the potential of the extract as a natural antihyperlipidemia agent that can be developed into an alternative cholesterol-lowering therapy.

Abstract

Tests on the dosage and effectiveness of turmeric leaf ethanol extract in lowering cholesterol levels in animal models have never been conducted to explore its therapeutic potential. This study aims to determine the effect of turmeric leaf ethanol extract (Curcuma longa) in reducing hypercholesterolemia levels in mice. This study employed an experimental design, dividing 36 mice into six treatment groups. The treatment group consisted of a negative control, a positive control, a drug control, and three doses of turmeric leaf ethanol extract (14 mg/20 g body weight, 28 mg/20 g body weight, and 56 mg/20 g body weight). Cholesterol levels were measured using a spectrophotometer, and the results were analyzed with the ANOVA test. The results showed a significance value of 0.000, indicating a significant difference in the reduction of cholesterol levels between treatment groups (p < 0.05). Turmeric leaf ethanol extract at a dose of 56 mg/20 g body weight resulted in a reduction of 33.6 mg/dL in mouse cholesterol levels, which is higher than that observed with other doses. Turmeric leaf ethanol extract can lower cholesterol levels in mice, with a dose of 56 mg/20 g body weight providing the most significant reduction. This study demonstrates that turmeric leaf ethanol extract has the potential to serve as a natural therapeutic alternative for managing hypercholesterolemia.

Abstract

Hypercholesterolemia is a significant risk factor for cardiovascular disease, and its prevalence continues to increase. Pharmacological therapy using statins is effective but may cause side effects when used long-term. Therefore, exploring natural ingredients as an alternative antihypercholesterolemic therapy is essential. This study aimed to evaluate the activity of a combination of ethanol extracts of Calotropis gigantea L. leaves and Adenanthera pavonina L. on the cholesterol levels of male mice induced with propylthiouracil (PTU). The study was conducted experimentally using a post-test-only control-group design with 36 male mice divided into six groups: negative control, positive control, drug control (simvastatin), and three treatment groups receiving extract combinations at doses of 14, 28, and 56 mg/g BW. The research stages included seven days of animal adaptation, induction of hypercholesterolemia with PTU for 14 days, administration of the extract combination at doses of 14, 28, and 56 mg/g BW for four days, and blood sampling for total cholesterol measurement using an automated chemical spectrophotometer. The results showed a significant increase in cholesterol levels in the positive control group, while simvastatin produced the most significant reduction. The extract combination gradually decreased cholesterol levels with increasing dose, with the highest dose approaching the effect of simvastatin. The paired t-test showed a p-value < 0.05, indicating significant differences between treatments. The combination of Calotropis gigantea L. and Adenanthera pavonina L. leaf extracts has potential as a herbal agent to support antihypercholesterolemic therapy.
