Abstract

This study aims to know the percent cover of mangrove and to assess the community structure. It was carried out in Gamtala village, Jailolo district, and west Halmahera regency, using line transect method with 10x10 m plot.. Data analysis covered the community structure and percent cover of mangrove canopy. This study found seven mangrove species, Bruguiera gymnorrhiza, Rhizophora apiculata, Sonneratia alba, Rhizophora mucronata, Avicennia alba, Bruguiera sexangula, dan  Xylocarpus granatum. Based on Importance Value Index (IVI), Bruguiera gymnorhiza had the highest, 149.06, and Avcsennia alba did the lowest, 9.3507. In addition, Gamtala village had manrove percent cover of 72.11 %.Keywords: canopy, community structure, mangrove, Gamtala.ABSTRAKTujuan dari penelitian ini adalah untuk mengetahui persentasi tutupan mangrove dan menghitung struktur. Penelitian ini dilakukan di Desa Gamtala Kecamatan Jailolo Kabupaten Halmahera Barat. Penelitian ini menggunakan metode transek garis dengan 10x10 m plot. Penelitian ini menemukan tujuh spesies mangrove, yaitu Bruguiera gymnorrhiza, Rhizophora apiculata, Sonneratia alba, Rhizophora mucronata, Avicennia alba, Bruguiera sexangula, dan  Xylocarpus granatum. Berdasarkan Indeks Nilai Penting, Bruguiera gymnorhiza memiliki nilai tertinggi, 149,06, dan Avcsennia alba memiliki nilai terendah 9,3507. Sebagai tambahan, desa Gamtala memiliki tutupan mangrove sebesar 72,11 %.Kata Kunci: kanopi, struktur komunitas, mangrove, Gamtala

Abstract

DNA isolation and gene amplification of algae are significantly influenced by various factors such as characteristics and components of the algae cell wall. Therefore techniques and methods of DNA isolation in certain algae, sometimes only succeed in one particular species and can not be applied to another algae species. Based on that issue, this study was conducted with the aims to determine the succeed of DNA isolation and amplify the rbcL gene as a target gene for identification. Algae DNA was isolated by using innuPrep Plant DNA commercial kit, and the second one with a modified conventional Cetyl Trimetyl Ammonium Bromide (CTAB) method,  for the amplification process was using rbcL gene (ribulose-1,5-bisphosphate carboxylase / oxygenase large subunit) with two pairs of primers : rbcL 7F-753R and rbcL 577F-rbcSR. The results showed that the DNA of Gracilaria sp was succeed isolated by using CTAB method and it was denoted by the presence of DNA bands in agarose gel. Meanwhile DNA amplification for Gracilaria sp., and Sargassum sp., were succeed amplified with the appearance of DNA bands. But in algae Caulerpa sp., was only succeed on 1 pair of primary rbcL 7F and 7.Keywords : DNA, gene rbcL, algae Caulerpa sp., Sargassum sp., Gracilaria sp;AbstrakIsolasi DNA dan amplifikasi gen pada alga sangat dipengaruhi oleh beberapa faktor seperti karakter dan komponen pada dinding sel alga. Oleh karena itu proses isolasi DNA terkadang bisa berhasil pada satu jenis alga, namun tidak berhasil pada jenis alga lainnya. Oleh karena alasan tersebut, maka penelitian ini dilakukan untuk menentukan keberhasilan Isolasi DNA dan mengamplifikasi gen rbcL sebagai gen target identifikasi. Penelitian ini dilakukan dengan tahapan awal Isolasi DNA yang menggunakan kit komersil innuPrep Plant DNA Kit, dan metode konvensional Cetyl Trimetyl Ammonium Bromide (CTAB) yang telah dimodifikasi. Sedangkan untuk proses amplifikasi, menggunakan gen rbcL (ribulose-1,5-bisphosphate carboxylase/oxygenase large subunit) digunakan dua pasang primer yaitu rbcL 7F-753R dan rbcL 577F-rbcSR. Hasil isolasi DNA dari alga Gracilaria sp berhasil diisolasi menggunakan metode CTAB yang ditandai dengan adanya pita DNA pada gel agarose. Amplifikasi DNA pada alga Gracilaria sp., dan Sargassum sp., berhasil diamplifikasi dengan munculnya pita DNA. Namun pada alga Caulerpa sp. hanya berhasil pada 1 pasang primer rbcL 7F dan753R.Kata kunci : DNA, gen rbcL, alga Caulerpa sp., Sargassum sp., Gracilaria sp.

Abstract

Sharks are a group of cartilaginous fish that are vulnerable to overfishing.  Genetics approaches play an important role in shark conservation. Shark fishing has become the main activity of fishermen in several areas, including in North Sulawesi. This research is focused on the molecular aspects and conservation status of shark species obtained from the shark fin trade in Manado, North Sulawesi. COI gene was amplified using Fish BCL5 (for) and HCO219 (rev) primers. Nucleotide sequences of each sample were aligned with the closest sequences in the GenBank database using the BLAST (Basic Local Alignment and Search Tool) method. The conservation status of the shark species is carried through the IUCN (International Union for the conservation of nature red list) Red list website. Molecular identification results showed that the shark’s fins from Manado had high similarity with Carcharhinus falciformis (HM1 and HM2) and Carcharhinus melanopterus (HM3). According to IUCN red list data, the C. falciformis and C. melanopterus were categorized as vulnerable to extinction (VU).Keywords:  Molekuler; COI; Shark; Manado and IUCN Red list . AbstrakHiu merupakan kelompok ikan bertulang rawan yang sangat rentan terhadap dampak penangkapan secara berlebihan.  Informasi terkait genetik hiu  yang semakin terancam populasinya sangat berperan penting dalam upaya konservasi hiu. Penangkapan hiu telah menjadi aktivitas utama nelayan di beberapa daerah, termasuk di Sulawesi Utara. Penelitian ini difokuskan pada aspek  molekuler dan penentuan status konservasi spesies hiu menggunakan sampel sirip yang di dapatkan  dari perdagangan sirip  hiu, di kota Manado.  Amplifikasi gen COI dilakukan dengan menggunakan primer Fish BCL5 (for) dan HCO219 (rev). Sekuens nukleotida masing-masing sampel disejajarkan dengan nukleotida terdekat yang ada dalam database genbank menggunakan metode BLAST (Basic Local Aligment and Search Tool)  Penentuan status konservasi dilakukan melalui penelusuran spesies rujukan di situs IUCN (International Union for the Conservation of Nature) Red list. Hasil identifikasi molekuler menunjukan bahwa sampel sirip hiu dari  kota Manado  memiliki kemiripan yang tinggi dengan spesies : Carcharhinus falciformis  (HM1 dan HM2) dan C. melanopterus (HM3).  Menurut data IUCN Red list, C. falciformis dan C. Melanopterus merupakan jenis hiu dalam status konservasi rentan punah (VU). Kata Kunci: Molekuler; COI; hiu; Manado dan IUCN Red list

Abstract

The implementation of the ship maintenance management system on Rear Admiral Jhon Lie's training ship did not run optimally, which impacted the less-than-optimal condition of various training ship equipment because the crew did not carry out the ship maintenance system according to the procedure, resulting in the incident that the ship's anchor winch operation did not function optimally when the ship was anchored in Belang Port, Southeast Minahasa. This study aims to identify and analyze the positive and significant effect of the planned maintenance system on shipping safety. The primary data collection methods used in this study were questionnaires and interviews. In this study, data processing and presentation were in the form of diagrams and path analysis with structural equation modeling (SEM-PLS) using SmartPLS. From the results of the data analysis, the R-square value of shipping safety (Y) is 0.307, which means that the planned maintenance system (X1) and ship maintenance (X2) can affect shipping safety (Y) by 30.7%. Ship maintenance (X2) has a significant effect on sailing safety (Y), with P-values = 0.033 0.05 (hypothesis accepted). The planned maintenance system (X1) has a significant effect on sailing safety (Y), with a P-value of 0.037 0.05 (hypothesis accepted). As a result of the analysis, it was found that the planned maintenance system has a significant effect on shipping safety.
