Abstract

Betalains and anthocyanins are classes of natural and water soluble pigments. Now days, these pigments have been developed as a replacement colouring agents. One of betalain resource is dragon fruit and anthocyanin resource is tamarillo. These pigments are relative unstable in some condition, one of them is pH. This research aim is to determine stability of betalain and anthocyanin pigments by pH parameter. These pigments can be isolated by macerated methods, technically processed by chopping the fruit and than soaking in ethanol for ± 24 h. These extracts were condensed by rotary evaporator. These extracts were analysed by ultraviolet-visible spectrophotometer at wavelength (λ) 200-800 nm. The absorption spectrum showed two major absorption peaks at 269 nm and 536 nm for betalain and 531 nm for anthocyanins. The result showed the optimal pH for betalain stability was at pH 5 and pH 3 for anthocyanin

Abstract

Lantana camara Linn is a family of Verbenaceae that grows in tropical and subtropical areas and spread in about 50 countries in the world. This plant contains essential oils with different contents based on the difference in the place of growth. The purpose of this study was to isolate and determine the chemical content of the essential oil of L. camara Linn leaves obtained from Agam, Tanah Datar, Kampar Regency, Padang Panjang City and to determine its cytotoxic and antibacterial activity. Isolation was carried out by hydro distillation method and chemical content analysis using Gas Chromatography-Mass Spectrometry (GC-MS). The high yield from the isolation of essential oil from leaves of L. camara Linn obtained from Tanah Datar Regency was 0.095% (w/w). The results of the analysis of the essential oil content with the main compounds for all Regency and City being caryophyllene with the high area percent was Tanah Datar Regency (19.74%). Cytotoxic activity test using the Brine Shrimp Lethality Test (BSLT) method showed that L. camara Linn essential oil was highly toxic with an LC50 value of 28.34 µg/mL from Padang Panjang City. Antibacterial activity test by disc diffusion method showed that L. camara Linn essential oil had moderate antibacterial activity againts Staphylococcus aureus and Escherichia coli bacteria, with high clear zone from Agam Regency was 12.9 mm and Tanah Datar Regency was 7.8 mm, respectively, at concentration of 100%.

Abstract

The leaves of the temurui (Murraya koenigii (Linn.) Spreng) plant have long been known and used by Indonesian and even Asian people as a traditional medicine to treat stomach aches and diabetes. This study aimed to determine the secondary metabolite profile of the methanol extract of M. koenigii leaves. Murraya koenigii leaves fine powder was extracted in stages using n-hexane, ethyl acetate, and methanol. Each extract was prepared using a mixture of water, formic acid, acetonitrile, and formic acid, then injected into the UPLC-MS, then analyzed with MassLynx and ChemSpider. The results showed that the metabolite profile of the methanol extract of M. koenigii leaves contained 13 compounds, including phenolic, steroid, and alkaloid groups. Those compounds could be tested to identify their bioactivity.

Abstract

Toxicity and phytochemistry assay have been conducted on extracts and fractions of Aglaia odorata leaves using brine shrimp lethality test. Aglaia odorata is one of the plants that are found in Indonesia. To investigate Aglaia odorata leaves potential as anticancer, air-dried and powdered leaves were extracted with n-hexane, ethyl acetat, and metanol by using maceration method. Extract of ethyl acetat showed the highest toxicity with LC50 value is 125.89 mg/L

Abstract

Eusideroxylon zwageri Teijsm. & Binn T has been reported to have bioactivity, one of which is the potential as an excellent cytotoxic agent using the BSLT method. So far, stage tests using cancer cells and analysis of the content of secondary metabolites have not been reported, Therefore, it is essential to analyze the content of secondary metabolites using Liquid Chromatography – Tandem Mass Spectrometry (LC-MS/MS) and determination of cytotoxic activity using Brine Shrimp Lethality Test (BLST) mwthod with various solvent polarities. Then separation by bioassay-guided isolation using column chromatography of the active extract assay BSLT. Furthermore, the cytotoxic activity of the active extract and the active isolate was tested against T47D cancer cells with Microculture Tetrazolium (MTT) method. The results of the analysis of the content of secondary metabolites using LC-MS/MS identified senbusine B and 6,7-Dehydroartemisinic acid on hexane extract, while the ethyl acetate extract produced 4 compounds, and 15 compounds were identified in the methanol extract. The results of the cytotoxic activity test using the BSLT method on each extract showed that the hexane extract was the most active, with an LC50 of 17.56 mg/L. Furthermore, the cytotoxic activity of hexane extract against T47D cells showed weak activity with an IC50 value of 237.5 mg/L, while the AB1 sub-fraction showed moderate activity with an IC50 value of 138.4 mg/L. The results of the analysis using HPLC indicated that the active isolate in the form of sub-fraction AB1 contained the compound senbusine B.

Abstract

Semambu (C. surinamense L) plant is a shrub plant that is easy to find. Several terpenoid compounds have been isolated from this plant, previous studies have shown cytotoxic activity of terpenoid class compounds. Terpenoid compounds in a plant are mostly found in essential oils (monoterpenes and sesquiterpenes). So far, there has been no report on the cytotoxic potential of essential oils from the leaves of this plant. It is necessary to isolate the essential oils from C. surinamense L leaves and test their cytotoxic potential. Isolation of essential oil of C. surinamense L leaves was carried out by hydrodistillation method, the oil was obtained in the form of a light yellow liquid with a specific gravity of 0.968 g/mL. Analysis of chemical components with Gas Chromatography-Mass-Spectrometry (GC-MS) through comparison of data from the National Institute of Standards and Technologies (NIST) found that there were 55 compounds (monoterpene and sesquiterpene groups) with six main compounds, namely β-caryophyllene (30.4%), β-sesquiphellandrene (8.46%), 3 carene (8.16%), α-bisabolene (4.05%), α-humulene (4.0%), and epi- bicyclosesquiphellandrene (4.0%). The potential cytotoxic test of essential oil from isolation showed highly cytotoxic activity with the Brine Shrimp Lethality Test (BSLT) method against Artemia salina L shrimp larvae with LC50 value of 0.9261 μg/mL and Microculture tetrazolium test (MTT) method against T47D breast cancer cells and HeLa cervix with IC50 values of 12.72 μg/mL and 30.14 μg/mL.

Abstract

Cervical cancer is a life-threatening disease. The use of synthetic drugs for its treatment can have serious adverse effects on the body. Therefore, it is necessary to search for natural drugs that are safe for the body. One of the secondary metabolites contained in plants is essential oils (EO), EO are known to have activity as anti-cervical cancer. Genus Citrus is known for containing EO compounds, one of the species of the genus is Citrus×lemon (L) Osbeck. The purpose of this study is to identify the chemical components of the EO isolated from the leaves of C×limon and to evaluate its cytotoxic activity against cervical cancer. EO was isolated from C×limon leaves collected in Padang City, Indonesia using the hydrodistillation method. EO was then analyzed by GC-MS. To determine its cytotoxicity, BSLT was conducted, followed by molecular docking and MTT assays. The experiment yielded a yellow liquid with a density of 0,8684 g/mL and a yield of 0,181%. GC-MS analysis of the EO identified 56 chemical components, with the main compounds are (-)-β-pinene (7.32%), (-)-limonene (28.40%), geranial (5.54%), caryophyllene (5.22%). The EO showed high toxicity against Artemia salina L larvae, with an LC50 value of 3,697 µg/mL. Through molecular docking, it is known the (-)-limonene, geranial, (-)β-pinene and caryophyllene compounds as the main compounds in of EO are known to bind to form complexes with cervical cancer proteins (HPV18E6). MTT assay showed weak cytotoxic activity of EO against HeLa cells, with an IC50 of 218.9 µg/mL.

Abstract

Activated carbon from corn stalks (Zea mays L.) has been successfully synthesized by the dehydration method and used as a supercapacitor electrode. Corn stalk powder was dehydrated with 1 MH2SO4 and activated with 5 M NaOH at a carbon and activator mass ratio of 1:4 with a heating temperature of 800°C and an N2 gas heating rate of 10°C/minute for 1 hour. SEM characterization results show that heterogeneous pores have formed on the surface of corn stalk-activated carbon. EDX results showed a carbon percentage of 54.82%; this data is supported by FTIR results, which show several functional groups containing C and O atoms on the surface of activated carbon. Based on desorption adsorption isotherm analysis, the activated carbon shows H4 type with a specific surface area of 117.973m2/g, average pore diameter of 1.730 nm and total pore volume of 0.1021 cm3/g. Measurement of the electrochemical properties of corn stalk activated carbon with a three-electrode system and 3 M KOH electrolyte showed a specific capacitance value of 133.9 F/g, power density 225.2 W/kg and energy density 0.863 Wh/kg. The synthesis of activated carbon through the dehydration method offers advantages in terms of simplicity in preparation, low cost, shorter synthesis time and being environmentally friendly.

Abstract

Toona sinensis has been used as a traditional medicine to treat diarrhea, dysentery, and fever. In the present study, the stem bark extracts of T. sinensis were investigated to determine the antibacterial activity, total phenolic content, 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity, and chemical constituent using gradual extraction by maceration method with hexane, ethyl acetate, and methanol respectively. In vitro, antibacterial activity was evaluated against Staphylococcus aureus and Escherichia coli by agar well diffusion method. The hexane fraction was more sensitive to S. aureus and the ethyl acetate and methanol fractions were more sensitive to E. coli. The methanol extract showed the highest total phenolic content and antioxidant activity. Phytochemically, Toona sinensis stem bark extract contained phenolic, flavonoid, steroid, triterpenoid, and coumarin. Investigation of ethyl acetate extract produced 5,7,3',4'-tetrahydroxyflavan-3-ol (catechin). Keywords: Antibacterial, antioxidant, catechin, Toona sinensis, total phenolic content

Abstract

Piper porphyrophyllum or known as tiger’s betel, is a medicinal plant of the Piper genus that is quite widespread in Indonesia, especially on Java, Sumatra, and Kalimantan island. Tiger’s betel is widely used traditionally to treat headaches, bone pain, chest tightness, skin diseases, relieve inflammation, and diabetes mellitus. This study aims to determine the type of secondary metabolite compounds contained in the ethyl acetate fraction of tiger’s betel and determine its antidiabetic activity. Determination of the type of compound is conducted by phytochemical screening by a qualitative test using various reagents to identify flavonoids, phenolics, saponins, triterpenoids, steroids, alkaloids, and coumarins. The antidiabetic assay was carried out by inhibiting α-glucosidase activity and the results were reported as IC50 values. The ethyl acetate fraction of tiger’s betel positively contains alkaloids, flavonoids, and phenolics. The antidiabetic activity is classified as very strong, with an IC50 value of 14.06 µg/mL.
