Abstract

BACKGROUNDLead acetate may inhibit the enzyme aminolevulinate dehydratase (ALAD) resulting in decreased heme synthesis (and consequently in anemia) but in increased number of reticulocyte cells. IR Bagendit paddy leaf water extract has a high metallothionein protein content which acts to bind to lead. The study objective was to determine whether aqueous IR Bagendit rice leaf extract dosage variations prior to lead exposure decreases reticulocyte count in lead-exposed rats. METHODSThe study was of randomized post test only control-group design involving a sample of 28 rats, that were randomized into 4 groups consisting of 1 control group and 3 treatment groups, daily administered with aqueous IR Bagendit rice leaf extract of respectively 0.2; 0.4; 0.8 mg using a feeding tube up to week 13. Lead exposure was also given using a feeding tube to both control and treatment groups at a dose of 0.5 g/kg BW/day, up to week 13. The reticulocyte count was then examined using supravital brilliant cresyl blue staining. The reticulocyte count was determined per 1000 erythrocytes and then converted into a percentage. Kruskal Wallis test followed with Bonferroni test was conducted to figure out the differences between groups. RESULTSMean reticulocyte count decreased significantly, starting from the control group up to the third treatment group (15.48 ± 3.41; 12.25 ± 03.28; 10.45 ±1.47; 9.10 ± 2.35 average per unit) (p=0.02). The Bonferroni test showed that the reticulocyte count was significantly decreased in the third treatment group (p=0.004). CONCLUSIONAqueous rice leaf extract significantly decreases reticulocytes in rats exposed to lead.

Abstract

Plumbum (Pb) asetat menyebabkan gangguan hematopoesis melalui hambatan biosintesis heme dan defisiensi enzim G-6PD yang menimbulkan penurunan hemoglobin dan jumlah eritrosit, serta peningkatan sel retikulosit. Ekstrak daun padi mengandung protein metallothionein yang mengikat Pb. Tujuan penelitian adalah membuktikan ekstrak daun padi memperbaiki gangguan hematopoesis pada tikus terpajan Pb. Metode penelitian adalah randomized post test only control-group design, 28 tikus dibagi ke dalam 4 kelompok yaitu 1 kelompok kontrol dan 3 kelompok perlakuan. Kelompok kontrol dan perlakuan dipajan Pb 0,5 g/kgBB/hari dan kelompok perlakuan diberi ekstrak daun padi 0,2; 0,4; 0,8 setiap hari melalui sonde sampai minggu ke-8. Hari terakhir  minggu ke-8 diperiksa jumlah kadar Hb menggunakan cyanmethemoglobin dan jumlah eritrosit diukur menggunakan haematology analyzer, serta retikulosit menggunakan pengecatan BCB. Tempat penelitian dilaksanakan di LPPT UGM dan laboratorium biomedik UNIMUS Semarang, Maret–Mei 2014. Perbedaan antarkelompok untuk kadar Hb dan jumlah eritrosit dilakukan uji one way enzyme linked immunosorbent assays (ANOVA) dan Kruskal-Wallis untuk jumlah retikulosit. Hasil penelitian menunjukkan kadar Hb rata-rata mengalami kenaikan tidak bermakna (p=0.81), jumlah eritrosit rata-rata tidak ada perbedaan antara kelompok kontrol dan perlakuan (p=0,83), jumlah retikulosit rata-rata menurun bermakna dari kelompok kontrol sampai perlakuan (p=0,00). Simpulan, ekstrak daun padi mampu memperbaiki hematopoesis pada tikus terpajan Pb. [MKB. 2015;47(2):84–90]Kata kunci: Ekstrak daun padi, hematopoesis, pajanan Pb, tikusRice Leaf Water Extract Improves Hematopoiesis in Lead-Exposed Rats  Abstract Plumbum ( Pb ) acetate leads to hematopoiesis disruption by inhibiting heme biosynthesis and creating G-6PD enzyme deficiency, leading to decreased hemoglobin concentration and red cell count as well as  increased reticulocyte cells. Rice leaf extract contains metallothionein proteins that bind Pb. This study aimed to prove the ability of rice leaf extract wa to improve hematopoiesis disorder conditions in rats exposed to Pb. The method used was the randomized post-test only control group design. Twenty eight rats were divided into 4 groups: 1 control group and 3 treatment groups . In the control group and the treatments groups, the experimental rats were treated with 0.5 g/kg/day Pb. The treatment groups were treated by 0.2, 0.4, and 0.8 rice leaf extract, respectively, every day for 8 weeks. On the last day of the 8th week, blood samples were collected for Hb concentration measurements using cyanmethemoglobin and the erythrocytes and reticulocytes number were counted using a hematology analyzer and BCB, respectively. This study was conducted in LPPT UGM and Biomedicine UNIMUS in March–May 2014. The differences between groups in hemoglobin and erythrocyte parameters were tasted  using one way ANOVA while  Kruskal-Wallis test was used for the reticulocyte count. The results show that rice leaf extract treatment to rats exposed to Pb did not affect Hb (p=0.81) and erythrocytes count  (p=0.83), However, the mean reticulocyte count decreased significantly in Pb-exposed rats treated with rice leaf extract (p=0.00). Abstract

Latar belakang: Prevalensi penyakit tidak menular termasuk tekanan darah tinggi dan Diabetes Mellitus (DM) terus meningkat di kalangan masyarakat. Namun, kesadaran dan pemahaman mengenai pentingnya pemeriksaan tekanan darah dan gula darah masih rendah di wilayah pedesaan, termasuk di Desa Tambakbulusan di Kabupaten Demak, Provinsi Jawa Tengah. Karena itu, peningkatan kesadaran tentang pentingnya pemeriksaan tekanan dan gula darah bagi masyarakat desa melalui penyuluhan relevan untuk dilakukan. Tujuan: Penyuluhan bertujuan meningkatkan kesadaran masyarakat Desa Tambakbulusan tentang pentingnya pemeriksaan kesehatan untuk menurunkan prevalensi penyakit hipertensi dan DM. Metode: Pelaksanaan kegiatan pengabdian kepada masyarakat dalam platform Pembangunan Kesehatan Masyarakat Desa (PKMD) diawali dengan identifikasi kebutuhan masyarakat akan penyuluhan tekanan darah tinggi dan DM melalui survei pendahuluan. Tahap perencanaan mencakup penentuan tujuan, materi penyuluhan, strategi penyampaian, lokasi, dan waktu kegiatan. Pra-pelaksanaan kegiatan meliputi pembuatan materi penyuluhan (poster). Dalam pelaksanaan kegiatan inti dilakukan diskusi, tanya jawab, studi kasus serta pembagian kuesioner pre- dan post-test. Kegiatan penyuluhan diakhiri dengan evaluasi untuk mendapatkan umpan balik dari peserta mengenai kegiatan yang telah dilakukan. Hasil: Setelah dilakukan kegiatan penyuluhan, pengetahuan masyarakat desa tentang penyakit tekanan darah dan DM meningkat berdasarkan skor pre-dan post-test, masing-masing dari rata-rata 61,7 menjadi 96,2 (55,6%) dan dari rata-rata 60,5 menjadi 95,4 (57,7%). Kesimpulan: Penyuluhan dengan media poster yang melibatkan pengisian kuesioner dapat meningkatkan kesadaran masyarakat Desa Tambakbulusan Kabupaten Demak tentang penyakit tekanan darah dan DM. Hasil penyuluhan ini perlu ditindaklanjuti dengan kegiatan pemeriksaan tekanan darah dan gula darah bagi masyarakat Desa Tambakbulusan. Kata kunci: Desa Tambakbulusan, diabetes mellitus, hipertensi, penyuluhan, poster ___________________________________________________________________________________________ Abstract Background: The prevalence of non-communicable diseases including high blood pressure and Diabetes Mellitus (DM) continues to increase in society. However, awareness and understanding of the importance of checking blood pressure and blood sugar is still low in rural areas, including Tambakbulusan Village in Demak District, Central Java Province. Therefore, increasing awareness about the importance of checking blood pressure and sugar in rural communities through counseling is relevant to do. Objective: Counseling aims to increase the awareness of the Tambakbulusan Village community about the importance of health checks to reduce the prevalence of hypertension and DM. Method: The implementation of community service activities in the Village Community Health Development (PKMD) platform was initiated by identifying community needs for counseling on high blood pressure and DM through a preliminary survey. The planning stage included determining objectives, extension materials, delivery strategy, location, and time of activity. Pre-implementation of activities included making counseling materials (posters). In the core activities discussions, questions and answers, case studies, and distribution of pre-and post-test questionnaires were carried out. The counseling activity ended with an evaluation to get feedback from the participants regarding the activities that had been carried out. Result: After the counseling activities were carried out, the knowledge of the villagers about blood pressure and DM increased based on the pre-and post-test scores, respectively from an average of 61.7 to 96.2 (55.6%) and from an average of 60.5 to 95.4 (57.7%). Conclusion: Counseling with posters involving the use of questionnaires can increase the awareness of the people of Tambakbulusan Village, Demak Regency about blood pressure and DM. The results of this counseling need to be followed up with activities to check blood pressure and blood sugar for the people of Tambakbulusan Village.  Keywords: Tambakbulusan Village, diabetes mellitus, hypertension, counseling, posters

Abstract

Anemia is a global health issue that affects people of all ages, particularly adolescent girls. Adolescent girls require nutritional intake, both from food and water. According to the balanced nutrition guidelines, people (particularly adolescents and adults) require 2000 liters (or eight glasses) of water daily. Most adolescent girls barely consider their water consumption. A lack of water in the body affects the balance of minerals (salt and sugar), disrupting regular metabolic processes such as hemoglobin (Hb) level regulation. This study investigates the correlation between adolescent girls' water consumption and hemoglobin levels. This quantitative study uses a cross-sectional design to collect variable data at a certain time. The study used blood with anti-coagulants EDTA from 30 adolescent girls from Universitas Muhammadiyah Semarang. The primary data for this study are water consumption and hemoglobin levels. Results of the research show that adolescent girls' water consumption was adequate (66.7%) and low (33.3%). The average Hb level among adolescent girls was 12.4 g/dL. Adequate water consumption resulted in 3 respondents with low Hb levels and 7 with normal Hb levels. In contrast, low water consumption resulted in 2 respondents with low Hb levels and 18 with normal Hb levels. The Rank-Spearman correlation test showed sig 0.177 (p>0.05). The study concluded that there was no correlation between adolescent girl water consumption and hemoglobin levels. Food fluid intake can compensate for a lack of water consumption.

Abstract

Anemic patients often have low hemoglobin and red blood cell level, which on the slide method and ABO system blood type test is used to obtain low degree of agglutination. Addition of treatment of a centrifugation was used to obtain a stronger degree of agglutination. The purpose of this research was to determine the difference degree of agglutination between centrifuged and uncentrifuged blood specimens in anemic patients. Quasi experimental research with non-equivalent control group design was conducted in March 2022 at the Hematology Laboratory, Universitas Muhammadiyah Semarang. The research sample were 16 specimens of anemic patients for slide method and ABO system blood type test. All samples were checked for blood type testbefore being centrifuged, then re-checked after the centrifugation there only concentrated red blood cell deposit were taken with only a small amount of plasma. The data from the test were analyzed using the Kruskal Wallis method. Comparison anti-A, anti-B, and anti-AB between centrifuged and uncentrifuged blood specimens there was a significant difference (p value < 0,005 means p = 0,002). The result showed that there were effect by centrifugation for the degree of agglutination improved better in centrifuged if compared with uncentrifugedanemicpatients blood specimens on blood type test slide method.

Abstract

Laboratory examination consists of three stages, namely pre-analytical, analytical, and post-analytic. The biggest error occurred in the pre-analytical stage, which was around 61%. Homogenization including the pre-analytical stage is the process of mixing blood and anticoagulants which are included in the pre-analytical stage. Homogenization has two ways namely manual and automatic. Inadequate homogenization can cause clots or rupture of erythrocytes/lysis and shrinkage of erythrocytes, leading to a falsely low hematocrit value. The study aimed to find out and analyze differences in hematocrit values by homogenizing the inversion method 2, 4, and 10 times. This type of research is observational analytic. The research was carried out in June - August 2021 at the Clinical Laboratory of Muhammadiyah University of Palangkaraya. The research sample consisted of 9 EDTA blood from 9 people according to the inclusion criteria. All samples were examined for hematocrit using a Hematology Analyzer Sysmex XP-300. The results showed that the average hematocrit value in 2 times inversion homogenization was 38.40%, 4 times homogenization was 38.78%, and 10 times homogenization was 39.14%. The conclusion of this study was that there was no significant effect of the inversion homogenization technique 2, 4, and 10 times on the hematocrit value (p=0.584 > 0.05).

Abstract

Tuberculosis (TBC) is a bacterial infection caused by Mycobacterium tuberculosis. The duration of TBC treatment is routinely 6 months; otherwise, resistance will develop. TBC can increase leukocytes, including monocytosis as an inflammatory response and an elevated ESR. The aim of this study was to determine the relationship between the number of monocytes and the ESR value on the duration of tuberculosis treatment in patients at the Respira Lung Hospital, Yogyakarta City. The study was conducted in June and July 2023 at the Respira Pulmonary Hospital in Yogyakarta City. The study sample was 32 tuberculosis patients and blood was drawn to determine the monocyte number and ESR value. The results showed that the average number of monocytes was 7% (normal), the ESR value was 41.59 mm/hour (high) with the average duration of tuberculosis treatment for respondents was 3 months. Spearman test obtained Sig p<0.05. This study concluded that there is a correlation between the number of monocytes and the ESR value on the duration of TB treatment in patients at the Respira Lung Hospital in Yogyakarta City, with the direction of the relationship reversed.

Abstract

Diabetes Mellitus (DM) disease is among the top 10 causes of death in the world. It is a metabolic disease causing unusual high blood sugar level in the body. Fortunately, controlling risk factors through early detection can prevent the abnormality. Dian Darat is among villages in Southeast Maluku Regency, where DM screening is rarely carried due to its relatively remote location. This community activity aimed to provide DM disease education through blood sugar screening and counselling for village’s community. Methods included early detection of DM and counselling based on blood sugar level screening results supported by poster media. Screening test results showed that 9 out of 26 respondents (35%) had blood sugar level higher than normal. Based on results of pre- and post-test scores before and after the education, there was an increase in participants’ knowledge regarding DM disease by 62 %. (from average score of 56,15 ± 13,88 to 90,77 ± 9,77, n=26, p ≤ 0.01). Future community services are recommended on similar and more sustainable community service activities by cooperation with other relevant stakeholders to optimize results.

Abstract

Blood is a body fluid whose stability can be influenced by both internal and external factors such as storage period. Storing blood specimens for a long time can cause changes in the physiological condition of blood cells such as color, size, shape and cell formation. This study aims to determine the effect of the storage period for donor blood specimens at UTD RSUP dr. Kariadi Semarang on hemoglobin levels and erythrocyte morphology. The research design is an experiment using a modified Time Series design with a pre and posttest control group design. The samples in this study were CPDA-1 (Citrate Phosphate Dextrose Adenine) donor blood specimens at the Blood Transfusion Unit of RSUP dr. Kariadi Semarang. Observation of erythrocyte cell morphology using blood smear preparations (SHD). Primary data was taken from the results of measurements of hemoglobin levels and observations of erythrocyte cell morphology with blood specimens examined directly (day 0) and storage days 1, 2, and 3. Analysis of research data used the One-way Anova statistical test. The results of the One-Way Anova test showed that there was a significant difference in hemoglobin levels on the 3rd day of donor blood storage (p-value 0.000). The results of the evaluation of erythrocyte cell morphology showed that there were abnormalities in crenation morphology on the 3rd day of storage. Blood specimens should be processed immediately after they leave the body because the condition of the blood cells is still stable and optimal for use in laboratory tests.

Abstract

Background: One of the genetic diseases that is usually detected in children is thalassemia. World Health Organization (WHO) states that thalassemia is the most common hereditary disease in the world and has been recognized as a world health problem. Indonesia is one of the countries in the world's thalassemia belt. Thalassemia major is the most severe clinical condition of thalassemia. Thalassemia minor can also be called a carrier trait and does not show clinical symptoms. Supporting examinations can be carried out with simple laboratory tests, one of which is measuring hemoglobin levels. This study aimed to determine the differences in hemoglobin levels in patients with thalassemia major and thalassemia minor at RSI Sultan Agung Semarang. Method: The type of research used was cross-sectional, with a sample size of 48 people. Hemoglobin level data was obtained from the medical records of Sultan Agung Semarang Islamic Hospital. The data distribution is normal, so the difference test is tested using the independent t-test. Result: The average hemoglobin levels in patients with thalassemia major and thalassemia minor are 5.7 mg/dL and 10.6 mg/dL. The independent t-test shows a significance value of 0.00 (<0.05). Conclusion: There are differences in hemoglobin levels in thalassemia major and thalassemia minor patients at Sultan Agung Semarang Islamic Hospital, although hemoglobin levels in both thalassemia major and minor patients are still below the reference value.
