Abstract

Staphylococcus aureus is a species of bacteria that is often identified as a source of infection and has increased the number of cases of resistance. Resistance to Penicillin-class antibiotics in S. aureus is associated with plasmids containing the blah-Z gene encoding ß-Lactamase. Some factors that can be modified to obtain optimal test results in PCR are annealing temperature and primary concentration. This study aims to identify the primary concentration and annealing temperature that produce PCR products with the best band in detecting the blaZ gene in Metichilin Resistant Staphylococcus aureus (MRSA) bacteria. The research design used used experimental methods. The results of the study obtained were PCR products at primary concentrations of 10 μM were clearly visible, thick and clean, compared to primary concentrations of 5 μM. Annealing temperature with the best band results at temperatures of 61.8 °C and 58.7°C. This study concluded the primary concentration to obtain a clearly visible band result of 10 μM. The annealing temperature of 61.8°C produced the best band in the sample code R.PG. U4, R.V.U2, and R.151.U2 temperatures. While the sample code R.IGD. U5 band results best at annealing temperature 58.7°C.

Abstract

Kyuri (Cucumis sativus L) plants have good economic value and are very much needed for health. In its cultivation, good media and pruning are needed to stimulate the growth of new productive branches which are very much needed in increasing production. This study aims to compare the filter cake planting media and pruning techniques to increase the growth and yield of kyuri cucumber plants. This study used a factorial randomized block design consisting of 2 factors, repeated three times. M1: filter cake and kettle ash (BK), M2 filter cake and rice husk charcoal (BS), M3 Soil, rice husk charcoal and manure (TSK), and M4: Soil and Cocopeat and Manure (TCK). Factor 2: Pruning System, where P1 without pruning, P2 prunes the shoots on the main stem and the shoots of three lateral branches (PP3L), and P3 prunes the shoots on the main stem and the shoots of five lateral branches (PP5L). The variables measured include plant height, stem diameter, number of leaves, number of fruits, fruit weight, and fruit length. Data analysis using analysis of variance (ANOVA) with a significance level of 5%. If the ANOVA results show a significant difference, further testing is carried out using the Duncan test with a level of 5%. The best treatment combination is M2P2, namely the use of filter cake planting media: rice husk charcoal 5: 2 and pruning the main stem and three lateral branches which can produce an average plant height of 166.2 cm, Stem Diameter 12.1 mm, Number of Leaves 22.3, Number of Fruits 7.1, Fruit Weight 337.3 kg, Fruit Length 28.4

Abstract

This study analyzes the influence of motivation and compensation on the performance of shallot farmers in Randuputih Village, Probolinggo. Quantitative methods were used with a sample of 40 farmers who are members of the local Shallot Farmers Group Association. Data were collected through questionnaires and interviews, then analyzed using multiple linear regression. The results showed: Motivation (X1) and compensation (X2) have a significant effect simultaneously on performance (Y) (sig. <0.05; R²=0.459) Partially, both variabels have a positive effect with compensation as the dominant factor (coefficient 0.452). This finding indicates that increasing compensation has a greater impact on farmer performance than motivation. This study recommends the need for policies that focus on improving the compensation system to encourage the productivity of shallot farmers.Keywords: Motivation, Compensation and Farmer Performance.

Abstract

Demam tifoid merupakan penyakit infeksi akut usus halus yang disebabkan oleh bakteri S. typhi dan S. paratyphi. Penyakit demam tifoid termasuk penyakit menular. Penularan disebabkan karena makanan atau minuman yang disajikan tidak bersih dan tingkat higenis individu sanitasi lingkungan. Pemeriksaan widal merupakan suatu pemeriksaan serologis untuk mendeteksi antibodi terhadap bakteri S. typhi yang berdasarkan reaksi aglutinasi antara antigen dengan antibodi yang disebut aglutinin, dengan mengukur aglutinasi titer antibodi terhadap antigen O dan H dalam sampel. Penelitian ini bertujuan untuk mengetahui ada tidaknya perbedaan nilai titer pada pemeriksaan widal yaitu metode slide sampel serum dan plasma EDTA dengan rancangan cros sectional sampel yang digunakan sebanyak 26. Pada Mahasiswa Teknologi Laboratorium Medik. Data yang dihasilkan dianalisis dengan uji univariat dan uji pearson chi square. Hasil penelitian ini didapatkan hasil pada serum terbanyak yaitu pada titer 1/320 (61,5%) sedangkan pada plasma EDTA terbanyak yaitu pada titer 1/80 (46,2%) yang menunjukan nilai p velue 0,0247 (p >0,05), maka berdasarkan hasil tersebut dapat disimpulkan bahwa tidak terdapat perbedaan antara pemeriksaan widal menggunakan sampel serum dan plasma EDTA antigen O dan H.

Abstract

Di desa Tegalrejo nasi basi biasanya dibuang percuma tanpa di manfaatkan bahkan dibuang, akibatnya dapat terjadi pencemaran tanah, air maupun udara, menyebabkan bau tidak sedap dan untuk mengurangi limbah tersebut, agar lingkungan terbebas untuk itu kami berupaya mengelola limbah tersebut menjadi pupuk organik melaluipembetukan MOL (Mikro-Organisme Lokal). Tujuan kegiatan pengabdian kepada masyarakat ini yaitu untuk membuat MOL dengan bahan baku dari limbah nasi basi sebagai pupuk organik ramah lingkungan. Metode yang digunakan yaitu metode ceramah dan demontrasi pembuatan MOL kepada petani dan ibu-ibu kader Desa Tegalrejo.Kegiatan ini dilakukan dengan pemberian materi melalui power point lalu melakukan demontrasi sehingga penyuluhan ini memiliki keberhasilan yaitu petani dan ibu-ibu kader memiliki pengetahuan dan kemampuan dalam membuat MOL dari nasi basi untuk menjadi pupuk organik yang ramah lingkungan.

Abstract

Typhoid fever is an acute infection of the small intestine caused by the bacteria S. typhi and S. paratyphi. This study aims to evaluate the results of typhoid fever testing using the Widal slide method and the Rapid test IgG IgM method at Dr. R. Goeteng Taroenadibrata Hospital in Purbalingga. Conducted in May 2024 with an observational analytic design and a cross-sectional approach, the sample consisted of 18 patients with typhoid fever. Widal slide examination results using O antigen reagent at RSUD dr. R.Goeteng Taroenadibrata, Purbalingga Regency, there titer 1/160 from 16 respondents (100%). Results of the IgG IgM Rapid test using O antigen at RSUD dr. R. Goeteng Taroenadibrata, Purbalingga Regency, there were 13 respondents (81.2%) IgG negative cases, 3 respondents (18.8%) had IgG positive cases, and 3 respondents (18.8%) had IgM negative cases, 13 IgM positive cases. respondents (81.2%). There was no difference in the results of the Widal slide examination and the Rapid IgG IgM test using O antigen in respondents at RSUD dr. R. Goeteng Taroenadibrata, Purbalingga Regency with p rate = 0.01 (P= < 0.05). Similarly, Fisher's test for IgG IgM yielded a P value of 0.04, So it can be concluded that there is a difference in the results of the Widal slide examination and the results of the IgM Rapid test.
