Abstract

Orchid flowers are believed to be an antioxidant source, however, the antioxidant activity of the Cymbidium Golden Boy (CGB) orchid has not been identified. The antioxidant potential can be applied to flower leather products, and a main reason behind the idea is the valorization of agricultural waste. The aim of this research is to study phenol content and antioxidant capacity of ethanol extracts of CGB orchid petals. Additionally, this study aimed to determine the best formulation for flower leather products with a combination of carrageenan and gum arabic based on organoleptic acceptance. This research was conducted by extracting CGB orchid flowers using ethanol, and total phenol and antioxidant activity of the extracts were determined. Moreover, the flower leather products were mad from CGB orchid flowers as the raw material, accroding to a Factorial Randomized Block Design using two factors as follows: carrageenan concentration (0, 0.3, 0.5, and 0.7%) and gum arabic concentration (0.9, 1.2, and 1.5%). Flower leather samples were analyzed for moisture content, tensile strength, total dissolved solid, color intensity, and sensory. The ethanol extract of CGB orchid flowers showed antioxidant capacity up to 23.88 ppm and high phenolic content which is equal to 15.32 mg GAE/g. The differences between carrageenan and arabic gum concentration showed significant effect on water content and dissolve solid, but they did not affect on tensile strength and color intensity. The best treatment of flower leather according to panelist evaluation was those prepared from carrageenan 0.5% and arabic gum 0.9%, reaching tensile strength 4.86 N, brightness level (L) 40.1, (a+) +5.7, (+b) 23.7, water content 19.18%, aroma score 3.76 (slight flowery), taste 4.84 (pleasant), and texture 4.56 (approaching chewiness).

Abstract

Kacang tanah (Arachis hypogaea  L) memiliki banyak manfaat seperti mencegah peningkatan kadar kolesterol, pencegahan keriput, kandungan serat tinggi, sumber energi dan pencegahan kanker. Akan tetapi jika dalam proses pemanenan, penyimpanan serta pengolahannya yang tidak sesuai, maka kacang tanah terkontaminasi aflatoksin yang berbahaya bagi kesehatan. Batas kadar cemaran aflatoksin pada kacang tanah dan hasil olahannya diatur dalam Peraturan Badan POM Nomor 18 Tahun 2018. Penelitian ini bertujuan menetapkan kadar kontaminasi aflatoksin B1, B2, G1 dan G2 pada olahan kacang tanah dengan prinsip ekstraksi menggunakan immunoaffinity column dengan instrumen High Performance Liquid Cromatogram (HPLC) detektor fluoresen. Hasil menunjukkan bahwa kromatogram baku kerja Aflatoksin G1, G2, B1, dan B2 berturut-turut menunjukkan respon terbentuknya peak pada waktu retensi 6,966 menit, 8,395 menit, 12,806 menit dan 10,395 menit. Hasil kromatogram sampel yang diuji tersebut memberikan respon terbentuknya peak pada waktu retensi yang berbeda dengan baku aflatoksin. Sehingga kedua sampel tersebut dapat disimpulkan tidak terdeteksi mengandung aflatoksin B1, B2, G1 dan G2 dengan nilai LOD aflatoksin G1 sebesar 0,8311 ng/mL, aflatoksin G2 sebesar 0,0941 ng/mL, aflatoksin B1 sebesar 0,3046 ng/mL dan aflatoksin B2 sebesar 0,3143 ng/mL.

Abstract

Bajakah Tampala extracted with ethanol was indicated to contain phenolic and flavonoids. So far, the ethanol extract of Bajakah Tampala wood has only been analyzed for its antioxidant activity. Whereas flavonoids not only act as antioxidants but also play a role in inhibiting -amylase which is related to antidiabetic properties. The ethanol extract of Bajakah Tampala wood has never been used as a fortificant in honey. The purpose of this study was to determine the levels of phenols, flavonoids, antioxidant activity, and inhibitory activity of -amylase from the ethanol extract of Bajakah Tampala Wood (EEBTW) at various solvent concentrations. In addition, to investigate the effect of adding EEBTW to Kapok honey on its bioactivity. This study was conducted using Post Test Only Group Design that was applied to 1% fortification of the three extracts in Kapok honey. Extraction used three concentrations of ethanol (70%, 80%, and 90%). Parameters tested in the study were total phenol, total flavonoid, antioxidant activity, and % inhibition of -amylase. The result showed the higher the ethanol concentration, the lower the total phenolic, flavonoid, % DPPH scavenging activity, and -amylase inhibition. The addition of 1% EEBKT has synergism with brown honey in increasing phenol, antioxidant activity, and % inhibition of -amylase. EEBTW has the potential as an antidiabetic candidate and Kapok honey with EEBTW has the potential as functional honey
