Abstract

The use of alcohol-based hand sanitizers can cause dry skin. Centella asiatica (L.) extract can be used as an alternative to alcohol because it contains triterpenoids, especially asiatic acid, potentially an antibacterial and inhibitor of the SARS-CoV-2 virus. Cream is a preparation that can moisturize and protect the skin. Glycerin and sorbitol are humectants often used in cream preparations and have a large percentage in the cream preparation's formula so that they can affect the physical properties of the cream. This study aims to obtain the optimum composition range of glycerin and sorbitol and determine the effect of glycerin and sorbitol as humectants on the physical properties and stability of the hand sanitizer cream with Centella asiatica (L.) extract. The composition optimization was done using a factorial design method on two factors and two levels using the Design Expert 13 (free trial) application. The data on the physical properties and stability of the cream were statistically analyzed using a two-way ANOVA test at a 95% confidence level. The results of qualitative phytochemistry screening showed that the extract contained triterpenoid. The average test of physical properties and stability of physical properties meet the desired criteria: cream type O/W, viscosity 120–170 dPa.s., spreadability of 5-7 cm, shift of spreadability, and shift of viscosity below 10%. The optimum area shows that the formulas F1, FB, and FAB meet the desired criteria.

Abstract

Burns represent a critical global health issue, contributing to considerable morbidity and mortality rates, particularly within the Southeast Asian region. The administration of appropriate burn therapy is essential to prevent infections and promote effective wound healing. The binahong leaf (Anredera cordifolia) represents a highly promising natural substance for burn therapy, attributed to its ursolic acid content, which is acknowledged for its wound healing properties. However, the limitation caused by its limited solubility and bioavailibility requires the use of nanoparticle technology, such as liposomes, to enhance its efficacy. The aim of this research was to determine an optimal formulation of gel containing liposome-encapsulated binahong leaf extract, with the goal of promoting burn wound healing and examining its in vivo wound healing activity. Histological analysis was employed to provide additional insights into the activity of the gel. The formulation was assessed through a factorial design, exploring various amounts of Na-CMC utilized as a gelling agent alongside glycerine employed as a humectant. The results were subsequently analyzed utilizing Design Expert v13 software. The gel’s viscosity, spreadability, and uniformity were assessed. Na-CMC increased viscosity while reducing spreadability, whereas glycerine had the opposite effect. The optimal formulation contained 2.78–4 g Na-CMC and 5–10 g glycerine. Statistical validation confirmed the model's accuracy. In vivo studies demonstrated that liposomal binahong gel significantly accelerated burn wound healing compared to controls. The results suggest that 10% binahong liposomal gel is a promising alternative for burn treatment.

Abstract

B12 injections are one of the most widely used vitamins in Indonesia. The nature of vitamin B12, which is easily degraded by environmental factors, has been widely reported. This research aimed to determine the effect of stability optimization model predictions with post-market sampling of vitamin B12 injection preparations by considering the variables of temperature, humidity, and distribution time. Factorial design was used to determine the stability optimization model. Validation of the analysis method using ultraviolet (UV) spectrophotometry showed valid results for testing. Stable product outcomes were seen during the research period based on the results of post-market sampling, optimization of stability conditions by evaluating pH parameters, and content determination. Post-market sampling data showed a pH decrease of 3.46% in the Palembang distribution area and an increase in pH of 4.54% and 5.33% in the Cirebon and Denpasar distribution areas. In the Palembang distribution area, the content of cyanocobalamin decreased by 0.90%, while in the Cirebon distribution area, it decreased by 0.94%. A paired t test for statistical analysis was used to determine the significance of the model. The results obtained show that there was no significant difference between the prediction model and the results of post-market sampling (p-values of 0.065 and 0.491) or H0 received

Abstract

Burn injuries are a significant global health concern due to their profound impact on morbidity and mortality rates. Immediate medical care is essential given the potential for bacterial infections to complicated burn injuries. Binahong leaves (Anredera cordifolia (Ten.) Steenis) contain vitexin compounds that demonstrate wound healing and antibacterial potential. However, vitexin has low solubility, prompting the use of ethanol for extraction and the introduction of nanoparticle technology to enhance solubility, namely transfersomes. The present study explored the development of a gel using binahong extract transfersomes with the composition of carbopol 940 and Na-CMC to determine the effect of carbopol 940, Na-CMC, and the interaction of the two on the gel’s physical properties and stability as well as obtain the optimum composition of the two. This study demonstrates positive results in terms of gel physical properties, including pH, spreadability, and viscosity. A Factorial design method is used for optimization. The results showed that Na-CMC had the most effect on increasing viscosity, while carbopol 940 had the most effect on reducing spreadability. The optimal formula obtained in this research was to use 0.653 grams of Na-CMC and 4.391 grams of Carbopol as gelling agents.

Abstract

Goroho banana (Musa acuminata L.) peel extract has been traditionally used in Minahasa for wound healing. This study aimed to develop and optimize a spray gel formulation containing Goroho banana peel extract and evaluate its wound healing activity. A factorial design was employed with two factors (Carbopol 940 and HPMC) at two levels. Preliminary testing determined 3% as the optimal extract concentration through wound healing evaluation in rats. Four spray gel formulations were developed and characterized for physicochemical properties, including viscosity, pH, and drying time. Results showed Formula B (Carbopol 0.5% and HPMC 0.6%) exhibited optimal characteristics with viscosity 223.8 ± 7.6 cPs, pH 5.13 ± 0.05, and drying time 4.22 ± 0.05 minutes. The HET-CAM irritation test revealed mild irritation scores (2.5-3.8) for all formulations, with Formula B showing the lowest score (2.5). Design Expert optimization generated 100 solutions, with the optimal formula containing Carbopol 0.670% and HPMC 0.517%. The wound healing activity showed no significant differences between spray gel formulations and Bioplacenton (p0.05), indicating comparable efficacy. This study demonstrates the successful development of a safe and effective spray gel formulation of Goroho banana peel extract for wound healing applications.
