Abstract

Improvement of cattle production can be obtained by the application of feed and breeding technology. One of the breeding technologies that are rapidly growing is in vitro fertilization, which is part of In vitro embryo production The source of oocytes influence the success of in vitro embryo production The quality of the oocyte were determined by Ovarian follicular environment and followed the follicular growth that were characterized by the presence of a dominant follicle (DF) and corpus lutem (CL). The purpose of this study was to determine the influe nced of dominant follicles and corpus luteum on oocyte morphology ofBali Timor cattle, that were collected by aspiration method. Oocytes were obtained postmortem from the ovaries of Bali Timor cows slaughtered at an Oeba slaughterhouse Ovarian then class ified into 3 groups : I ) ovarian with CL without DF; (II) ovarian with DF, without CL; and (III) ovarian without both DF and CL. The results showed that the number of oocytes with good quality from group III (12.5%) was higher than group I (9.83%), but no significant differently (P> 0.05). The number of oocytes from group II that showed several layers of cells and has a cytoplasm (good quality) was very low.

Abstract

The purpose of this study was to conduct an assessment on the quality of pig semen spermatozoa during storage and determine which diluent material should be used and how long pig semen can be stored. Freshly ejaculated boar semen was obtained from a two-years-old Duroc male and was diluted with Beltsville Thawing Solution (BTS) and Tris Citrate Fructose (TCF) supplemented with 5% egg yolk (KT) and extract of Borassus flabellifer Linn mesocarp (EM) 0.01% and stored at 13°C for up to 4 days. An experimental study with a completely randomized design with four groups and five repeats was applied. Parameters observed were motility, viability and abnormality of spermatozoa. Sperm quality was evaluated by examining motility and abnormality using a phase-contrast microscope with a magnification of 100X, and viability by eosin nigrosine staining. Parametric data were analyzed with ANOVA followed by Duncan test.  The results showed that the supplementation of 5% egg yolk in the BTS diluent-based treatment group showed a higher percentage of motility (P<0.05) than without egg yolk, while the TCF-based diluent group showed no differences (P<0.05). Modified-Tris Citrate Fructose diluent supplemented with 5% egg yolk and 0.01% mesocarp extract was able to maintain a significantly higher percentage of viability (67.50±2.0) (P<0.05) than other groups, and had a percentage of abnormalities (9.75±0.957), significantly lower (P<0.05) than other groups. It was concluded that the modification of diluent TCF with 5% egg yolk and mesocarp extract 0.01% provide the best results in maintaining sperm quality with the highest percentage of sperm motility and viability, and the lowest percentage of sperm abnormality compared to the other three diluents. Likewise, all groups of modified-BTS and TCF diluent supplemented with 5% egg yolk and extract of Borassus flabellifer Linn mesocarp in this study were able to maintain motility >40%, viability >45% and abnormalities <20% so they were appropriate for using in the AI procedure.

Abstract

Penelitian ini bertujuan mengetahui tingkat maturasi oosit sapi Simmental Peranakan Ongole (SimPO) dan Limousin Peranakan Ongole (LimPO) secara in vitro. Oosit dari Ovarium sapi yang berasal dari rumah potong hewan (RPH), dikelompokkan berdasarkan jenis sapi PO (kontrol), SimPO dan LimPO. Oosit diaspirasi dari ovarium dengan syringe 5 ml dan jarum 18 G. Oosit dengan kualitas A dan B yang digunakan dalam penelitian ini. Oosit dikultur dalam media maturasi TCM 199 100 µl drop dan dilapisi minyak mineral, diinkubasi pada suhu 38,5° C, CO2 5% dan kelembaban 95% selama 24 jam. Tingkat maturasi in vitro ditentukan dengan pewarnaan aceto orcein 1% untuk melihat tahapan maturasi dengan adanya perubahan konfigurasi kromosom dan membran inti berupa germinal vesicle, germinal vesicle breakdown, metafase I, anafase/telofase I, dan metafase II. Hasil penelitian menunjukkan tingkat maturasi in vitro oosit yang mencapai metafase II dari oosit sapi PO lebih tinggi secara signifikan dibandingkan sapi SimPO dan LimPO.

Abstract

The integrity of the deoxyribonucleic acid (DNA) in sperm plays a crucial role in the fertilization process and embryonic development. This study aimed to evaluate the level of the DNA fragmentation of boar sperm in tris-citrate-fructose extender preserved at temperatures between 16-18°C for 3 days. The level of sperm DNA damage was analysed using toluidine blue (TB) staining. Pig semen collected from 4 male pigs aged 1.3-1.8 years was used as the research sample. Semen collected was conducted twice a week. The experimental method using a Completely Randomized Design with 4 storage duration groups as follows: Group I (0 hours of storage), Group II (24 hours of storage), Group III (48 hours of storage), and Group IV (72 hours of storage), each with 5 replications. The data were analysed using analysis of variance (ANOVA) and, if significant differences were found, Duncan's test was performed. The results of the evaluation of the percentage of DNA damage during the storage process at 16-180C at 0 hours, 24 hours, 48 hours and 72 hours was: 0.33 ± 1.66%, 0.80 ± 0.447%, 1.60 ± 1.14% and 2.80 ± 0.447%, respectively. These results demonstrate that liquid boar semen preserved with tris citrate fructose extender at 16-18 °C for 3 days has spermatozoa DNA damage levels within the normal range, making it suitable for artificial insemination and ensuring optimal AI success.

Abstract

Penyakit kulit berbenjol (Lumpy Skin Disease) pada sapi disebabkan oleh virus genus Capripoxvirus dan tidak termasuk penyakit zoonosis. Gejala utama berupa nodul pada kulit di area genital, punggung, area kepala, leher, ekor, dan abdomen. Infeksi LSD dapat menggangu bahkan menurunkan peforma produksi dan reproduksi sapi hingga penurunan produksi susu. Penyakit ini dapat ditularkan melalui vektor seperti nyamuk, kutu, dan lalat. Diagnosa dapat dilakukan melalui pengamatan gejala klinis serta uji laboratorium meliputi Polymerase Chain Reaction (PCR), mikroskop electron transmisi, uji fluorescent antibody, imunohistokimia dan isothermal genome amplification. Pencegahan dan pengendalian LSD dapat dilakukan dengan cara vaksinasi, sanitasi kandang, biosekuriti, dan terapi suportif.   Kata kunci: Capripoxvirus, Diagnosa, Gejala klinis, Penyakit kulit berbenjol, Pencegahan.

Abstract

Kualitas Domba Ekor Tipis jantan dapat diketahui dari performa produktivitas berupa bobot badan dan kualitas reproduksi berupa lingkar skrotum dan volume semen yang merupakan indikator kualitas reproduksi untuk keberhasilan fertilisasi. Penelitian ini mengnalisis korelasi dan regresi antara bobot badan (BB), lingkar skrotum (LS), dan volume semen (VS) pada sepuluh ekor Domba Ekor Tipis jantan (Ovis aries) berumur 8–18 bulan di Teaching Farm, Jurusan Peternakan, Fakultas Pertanian, Universitas Lampung. Metode purposive sampling digunakan untuk memilih hewan percobaan. Pengukuran BB (kg) dilakukan sebelum pemberian pakan pagi, LS (cm) diperoleh dari rerata dua kali pengukuran, dan koleksi VS (ml) menggunakan vagina buatan. Setiap parameter diukur sebanyak tiga kali dengan selang waktu empat hari, menghasilkan total 30 data (n=30). Data dianalisis menggunakan perangkat lunak R dengan uji korelasi Pearson untuk menentukan kekuatan dan arah hubungan antarvariabel, serta regresi linear untuk memodelkan hubungan prediktif antara: (1) BB dan LS, (2) BB dan VS, dan (3) LS dan VS. Hasil penelitian menyajikan korelasi positif kuat antara BB dan LS (r = 0,623; Y = 17,689 + 0,242X), korelasi positif sedang antara BB dan VS (r = 0,505; Y = -0,153 + 0,056X), serta korelasi positif sedang antara LS dan VS (r = 0,406; Y = 21,010 + 1,428X). Kesimpulan penelitian ini menunjukkan bahwa bobot badan dan lingkar skrotum dapat digunakan sebagai indikator fenotipik untuk memprediksi volume semen, sehingga dapat menjadi kriteria praktis dalam seleksi Domba Ekor Tipis jantan unggul.

Abstract

Keberhasilan reproduksi merupakan faktor kunci dalam meningkatkan efisiensi dan produktivitas usaha peternakan babi. Salah satu strategi yang digunakan untuk mendukung keberhasilan tersebut adalah inseminasi buatan (IB), yang sangat bergantung pada kualitas semen selama penyimpanan. Penelitian ini bertujuan untuk mengevaluasi integritas membran plasma (IMP), fragmentasi DNA spermatozoa, tingkat keberhasilan IB, dan litter size menggunakan semen babi yang diencerkan dengan berbagai jenis pengencer berbasis tris sitrat fruktosa (TCF), selanjutnya disimpan pada suhu 16 °C hingga 72 jam. Penelitian dilakukan dalam dua tahap. Tahap pertama menilai kualitas semen, yaitu PMI dan fragmentasi DNA, menggunakan rancangan acak lengkap dengan lima ulangan. Tiga jenis pengencer yang diuji adalah TCF + ekstrak mesokarp + kuning telur (TCF+EM+KT), TCF + ekstrak mesokarp (TCF+EM) dan TCF, masing-masing pada penyimpanan 24, 48, dan 72 jam. Tahap kedua mengevaluasi keberhasilan IB pada 36 ekor induk babi hasil persilangan Duroc-Landrace berumur 1–3 tahun, menggunakan semen yang disimpan selama 72 jam. Parameter yang diamati meliputi conception rate (CR), service per conception (S/C), non-return rate (NRR), dan ukuran litter. Hasil penelitian menunjukkan bahwa pengencer TCF+EM+KT memberikan hasil terbaik, dengan nilai IMP tertinggi (97,4±0,43%), fragmentasi DNA terendah (0,40±0,55%), CR tertinggi (83%), S/C terendah (1,17), serta rata-rata litter size terbesar (7,42), secara signifikan berbeda (P<0,05) dibandingkan pengencer lainnya. Kesimpulan penelitian ini menunjukkan pengencer TCF+EM+KT paling efektif dalam mempertahankan kualitas semen dan meningkatkan performa reproduksi pada IB babi.
