Abstract

Arumanis mango leaves (Mangifera indica L.) has antibacterial activity and antifungal, evidently. Escherichia coli and staphylococcus aureus are opprtunistic pathogen bacteria that can cause gastrointestinal and skin infection. Purification of extract was done to eliminate the presence of ballast substances that cannot produce therapeutic effects. This study aims to acknowledge the antibacterial activity of purified mango arumanis leaves extract (PMALE) against E. coli and S. aureus, flavonoid compound content by TLC. Arumanis Mango leaves powder was extracted with 96% ethanol solvent by maceration method, followed purification the extract was done with hot water-ethylacetate solvent by liquid-liquid extraction method. Antibacterial activity test was done by agar diffusion method against E. coli and S. aureus bacteria with the same serial concentration of 6.25; 12.5; 25; 50; 75 and 100% and Thin Layer Chromatography (TLC) tests for detection flavonoid compounds, the stationary phase used silica gel 60F 254 and the mobile phase used n-hexane-ethylacetate (1:3). The results of PMALE had antibacterial activity at lower concentrations 6,25% against both of E. coli and S. aureus. PMALE had flavonoid compounds with Rf values of 0.81.

Abstract

Infeksi kandidiasis meningkat selama dekade terakhir di dunia. Peningkatan ini menandai munculnya isolat Candida albicans resisten Flukonazol. Diperlukan obat baru untuk mengatasi permasalahan resistensi ini.  Daun pacar kuku (L. inermis L.) secara empiris mempunyai aktivitas sebagai antijamur. Kandungan kimia daun pacar kuku larut dalam etil asetat. Penelitian ini bertujuan untuk mengetahui aktivitas antifungi dari fraksi etil asetat ekstrak daun pacar kuku (L. inermis L.) terhadap strain C. albicans sensitif Flukonazol dan C. albicans resisten Flukonazol. Fraksi etil asetat ekstrak daun pacar kuku (L. inermis L.)dibuat dengan memfraksinasi ekstrak etanol dengan  pelarut etil asetat.Konsentrasi fraksi etil asetat ekstrak daun pacar kuku yang digunakan terhadap C. albicans sensitif Flukonazol dan C. albicans resisten Flukonazol sebesar 10%; 5%; 2,5%; 1,25% dan 0,625%. Pengujian aktivitas antifungi fraksi etil asetat ekstrak daun pacar kuku menggunakan metode mikrodilusi cair dengan parameter pengukuran Minimum Inhibitory Concentration (MIC) dan Minimum Fungicidal Concentration (MFC). Nilai MIC dan MFC yang diperoleh sama yaitu sebesar 1,25% untuk C. albicans resisten Flukonazol dan 0,625% untuk C. albicans sensitif Flukonazol. Rasio MFC terhadap MIC yang diperoleh sebesar 1 yang menyatakan bahwa fraksi etil asetat ekstrak daun pacar kuku memiliki daya fungicide. Skrining fitokimia fraksi etil asetat ekstrak daun pacar kuku mengandung fenolik dan kuinon.

Abstract

Post-harvest management is essential because it will affect the final results. Post-harvest management products can be classified as simplicia because the stages include wet sorting, washing, draining, changing shape, drying, packaging, and storage. These steps must be paid attention to to ensure the content of active compounds in simplicia. The Bejo Bintang Toedjoe herbal garden at the Plalangan Gunungpati sub-district is managed by women from the Plalangan sub-district, both housewives and workers. Community service was implemented by preparing and making leaflets/brochures, conducting site surveys, conducting pretests and posttests, and monitoring and evaluating the practical results. This activity was attended by 25 participants, not only participants from the managers of the Bejo Bintang Toedjoe herbal garden but also residents from representatives of several community associations. The discussion session was opened after the counseling had been delivered. After the discussion session is finished, it continues with practice. The results of the pretest analysis showed that most participants still need to learn the effect of direct heating with sunlight for drying medicinal plant materials. Some participants also needed to learn that the tools used to cut medicinal plant materials were stainless steel. The posttest results showed that there was an increase in knowledge after conducting counseling and post-harvest management practices of medicinal plants.

Abstract

Leaves and stem bark of orange jasmine have bioactive compounds such as phenol and flavonoids, which these compounds could be extracted by organic solvent and have antifungal activity against Malassezia furfur which cause excema, dandruff and tinea versicolor in humans. Fractionation of the extract would obtain a specific compound based on the polarity of the solvent. This research aims to know the active fraction of leaves and stem bark of orange jasmine which had antifungal activity against Malassezia furfur.  The simplicia powder of leaves and stem bark orange jasmine was macerated with 96% ethanol solvent and then fractionated as gradually with n-hexane, ethyl acetate, and water solvent. The antifungal activity of ethanol extract and fractions was tested against Malassezia furfur with 80; 90; 100% concentration by disk diffusion method at 37˚C for 48 hours of incubation. The data of antifungal activity was analyzed by two-way ANOVA. The result of this research showed antifungal activity from active fraction as water fraction of leaves and stem bark of orange jasmine against Malassezia furfur. The diameter of inhibition zones for the active fraction of leaves and stem bark of orange jasmine such as 8.6 – 14 mm and 12.3 – 15.8 mm respectively.
