Abstract

Chalcone is a flavonoid derivative that has a variety of interesting biological activities. One of the compounds suspected of being an antibacterial agent is chalcone. The properties of chalcone compounds depend on the substituents that depend on the two aromatic rings, such as Cl, Br, OH groups and so on. The purpose of this study was to synthesize chalcone analogue compounds and determine the potential of these analog compounds as antibiotics. In this research, a chalcone analogue compound has been successfully synthesized, namely 3ꞌ-Bromo-4-methoxy-chalcone through the stirring method using KOH catalyst, ethanol solvent. The results of the synthesis carried out by recrystallization and the yield obtained were 64 %. Identification of compounds that have been carried out by TLC test, melting point, and HPLC analysis. The structure of the compound was characterized by UV-Vis spectroscopy, FTIR and 1H NMR, indicating that the analogue chalcone compound has a structure as expected. Testing the activity of chalcone analogue compounds was carried out on Staphylococcus aureus and Escherichia coli at a concentration of 1%, 5%, 10% and 20% test solutions. The test results showed that the antibacterial activity of the compound 3ꞌ-Bromo-4-methoxy-chalcone was categorized as weak.

Abstract

Dermatomyositis merupakan penyakit autoimun yang termasuk jenis idiopatik inflamasi miopati (IIM), penyakit ini dapat mempengaruhi kulit dan otot manusia. Gejala klinis Dermatomyositis pada sebagian besar pasien adalah kelemahan otot tubuh, ruam kulit dan kulit bersisik. Salah satu faktor penyebab Dermatomyositis yang sering dilaporkan adalah faktor genetik. Hingga kini,  penelitian terkait Dermatomyositis masih terbatas pada identifikasi jenis variasi gen yang mempengaruhi, namun tidak melaporkan variasi gen mana yang paling berkontribusi pada Dermatomyositis khususnya yang bersifat missense/nonsense. Sehingga pada penelitian ini kami memanfaatkan database genomik dan analisis bioinformatik  untuk mengidentifikasi variasi gen yang paling berhubungan dengan penyakit Dermatomyositis. Penelitian ini menggunakan beberapa database, termasuk GWAS catalog, PheWAS catalog, HaploReg (v41.), dan GTEx portal. Hasil dari penelitian ini ditemukan bahwa gen ZBP1 berkaitan erat dengan penyakit Dermatomyositis dan menunjukkan ekpresi yang tinggi pada beberapa jaringan seperti paru-paru, lambung, esophagus, kulit, jantung dan otot. Variasi gen berdasarkan frekuensi varian alel (rs59626664, rs60542959, rs2066807, rs1048661, rs745400, rs2305480, rs2305479) terkait Dermatomyositis menunjukkan ekspresi jaringan tertinggi di kulit suprapubic, kulit dibawah lengan, otot rangka, dan esofagus. Penelitian ini menekankan bahwa integrasi database genomik dan analisis bioinformatik menunjukkan variasi gen yang berperan dalam patogenesis Dermatomyositis khususnya yang bersifat missense/nonsense. Kami menyarankan untuk peneliti selanjutnya untuk fokus pada variasi gen tersebut untuk divalidasi di fase klinis khusunya di populasi Indonesia.

Abstract

Abstract. Multidrug resistance is still a major health problem worldwide. Biofilm formation was related to the resistance of bacteria mechanism. Acinetobacter baumannii, one of the bacteria that cause nosocomial infections, is given particular attention due to its high prevalence of resistance. The current study address observing the antibacterial activity and antibiofilm formation activity of Curcuma domestica rhizome extract against multidrug-resistant Acinetobacter baumannii (MDR A.baumannii). Curcuma domestica rhizome was macerated by ethanol 96%. Determination of Minimum Inhibitory Concentration (MIC) was carried out by the microdilution method. Antibiofilm formation analysis was determined by standard quantitative using crystal violet-stained assay. Ethanol extract of C. domestica possesses antibacterial activity against MDR A. baumannii with MIC value 250 µg/mL. Additionally, the ethanol extract of C. domestica could inhibit the biofilm formation of MDR A.baumannii, thus percentage of biofilm formed only 23.7% and significance different with control after analyzed using ANOVA post hoc test Tukey. According to these results, ethanol extract of C. domestica revealed antibacterial activity and had the potency in suppressing the biofilm formation of MDR A.baumannii, thus could be utilized in clinical application, especially in MDR A.baumannii infection.
