Abstract

Abstract

Abstract

Abstract. Wounds that are not treated and are kept open will provide an entrance for microorganisms from outside that can cause infection. One of the medical needs whose demand continues to increase is wound dressings. Chitosan is known to have wound healing activity by stimulating the formation of new tissue, and silver nanoparticles have good antimicrobial activity. Silver nanoparticles and chitosan with cellulose nanofibers carrier are made in the form of patches with the ratio formula between cellulose nanofibers and chitosan/silver nanoparticles are 1:9, 2:8, 3:8, 4:7, 5:5, 6:4, 7:3, 8:2, 9:1, and 10:0 then tested the antifungal activity against Trichophyton rubrum and Pitysporum ovale for finding the best formula for antifungal activity. The analysis showed that the patch with a ratio of 6:4 had the best antifungal activity against Trichophyton rubrum (14.7±0.1 mm) and a 9:1 patch on the Pitysporum ovale (6.9±0.05 mm) gave a significant difference to negative control (p<0.05). It can be concluded that the Patches Silver Nanoparticle and Chitosan with Cellulose Nanofibers (SNCCN) carriers have good antifungal activity in the inhibitory category.

Abstract

Research on antidiabetic activity of ethanol leaf extracts Kluwih (Artocarpus camansi) has been carried out to determine the antidiabetic activity of ethanol extract of leaves kluwih (EELK) of mice using glucose tolerance test. The study began by dividing the mice into 6 groups. Group 1 was given 1% CMC, group 2 to group 4, given EELK 50, 100, 200, 400 mg / kg bw, respectively, and group 6  administered with glibenclamide 0.45 mg/kg bw. Thirty minutes later each group was given glucose 30 mg/kg bw. After 30 min of glucose loading, blood glucose levels of mice were measured, and measured again at minute 60, 90, 120 and 150 minutes. The results showed that the glucose tolerance test EELK a significant effect in lowering blood glucose levels compared to controls at 30 and 60 minutes after administration of glucose. At minute 90 to 150, the effect of blood glucose levels EELK restore back to normal. EELK demonstrated ability to reduce blood glucose levels, and EELK doses of 50 and 100 mg/kg bw showed antidiabetic effects better than a dose of 200 and 400 mg/kg bw. Increasing doses showed antagonistic effects: the ability to increase blood glucose levels.

Abstract

Abstract. Cyclophosphamide is an antineoplastic drug belonging to the alkylating agents commonly used in treating cancer. However, the use of cyclophosphamide causes a decrease in the body's immune system by reducing lymphocyte proliferation. The curent study was conducted to evaluate the immunomodulatory effect of ethyl acetate extracts of the leaf of the P. macrocarpa (EEADMD) and the ethanol extracts of the leaf of the P. macrocarpa (EEDMD) in normal and cyclophosphamide induced rats. This study used rats classified into two major groups: normal rats and rats induced by cyclophosphamide which were then given P. macrocarpa leaf extract until day 14. On day 4, the test animals were infected with 1% Staphylococcus aureus suspension in. Induced with cyclophosphamide 70 mg/kg BW was carried out on the 8th and 13th days, and then the immunomodulatory activity was tested using total leukocyte analysis, leukocyte differential, and delayed type hypersensitivity response. The results showed that EEDMD 400 mg/kg BW  in normal rats and EEDMD 100 mg/kg BW in cyclophosphamide induced rats could increase total leukocytes and leukocyte differential with a significant difference to the negative group (p <0.05). The results of the delayed hypersensitivity response test of EEADMD and EEDMD at a dose of 100 mg/kg BW with cyclophosphamide induction and normal rats could give an increase in rat paw volume with a significant difference to the negative group (p <0.05). This study shows that EEADMD and EEDMD have an immunomodulatory effect on increasing total and differential leukocytes and leukocyte differential and delayed type hypersensitivity response.

Abstract

One of the medicinal Zingiberaceae plants which is regularly used by the society as a traditional remedy is turmeric rhizome. The pharmacological properties of turmeric rhizome include anti-inflammatory, anti-immunodeficiency, anti-viral. Anti-bacterial, anti-fungal, anti-oxidant, anti-carcinogenic, and anti-infectious. This study aimed to identify the immunomodulator activity of curcuminoid extract of turmeric rhizome against the increase of leucocyte and its components, lymphocyte and neutrophil in white Galur Wistar rats that had been injected with Staphylococcus aureus. These test animals were given curcuminoid extract in VCO 100 mg/ml on a dosage of 1 ml, 0,5 ml, 0,25 ml curcuminoid powder 200 mg/kg body weight. Curcuminoid was suspended in Na CMC0,5%, Levamisole 25 mg/kg of body weight suspended in Na CMC 0,5%, VCO 1 ml. For the negative control, merely Na CMC 0,5% was given. The extract was orally given daily for 14 days. On the 4th day, Staphylococcus aureus was injected intra-peritoneally. The rats’ blood was then drawn from the heart and was observed to calculate the volume of the leucocytes, lymphocyte, and neutrophil which was conducted using flow cytometry. The findings suggested that there was a significant difference between the immunomodulatory activity of the curcuminoid extract in VCO at the doses of 1 ml, 0,5 ml, 0,25 ml on the increase in leucocyte, lymphocyte, and neutrophil compared to that of the negative control. Therefore, the findings of this study indicated that the extracts of curcuminoid in VCO could increase the number of leucocytes, lymphocytes, and neutrophils. Thus, the curcuminoid extract of turmeric rhizome in VCO potentially possessed immunomodulatory activity.

Abstract

Infections caused by multidrug-resistant (MDR) bacteria are on the rise globally. MDR is facilitated by overexpression of efflux pump and permeability changes of membrane. Acinetobacter baumannii is a pathogenic germ that causes a major problem in infection, also, there has been an increase in the incidence of resistance to various antibiotics. The present study highlights the synergistic of ethanolic extract of Curcuma domestica (EECD) rhizome with tetracycline against multidrug-resistant A. baumannii (MDR-Ab). Assessment of Minimum Inhibitory Concentration (MIC) was determined by microdilution using 96-well plates. The synergistic effect of EECD and tetracycline was determined by checkerboard method. The effect of EECD and tetracycline combination was investigated by bacteriolytic activity and inhibition of efflux pump by Ethidium Bromide (EtBr) accumulation assay. EECD presented the MIC value 250 µg/mL against MDR-Ab. Fractional Inhibitory Concentration Index (FICI) value of EECD and tetracycline combination was 0.4, which showed their synergistic effect. Additionally, the combination of EECD and tetracycline could inhibit the efflux pump in MDR-Ab. This combination can also compromise cell integrity by altering membrane permeability thus lysing the bacteria cells. According to these results, EECD and tetracycline combination has synergistic effects at some sites of action, and thus could be used as a breakthrough to overcome infection problems due to MDR-Ab. Abstract

The natural human environment is inhabited by various organisms, including those harmful to the body such as bacteria, viruses, fungi, and other pathogens. This condition weakens the immune system's response to pathogens, making the body more susceptible to diseases such as rheumatoid arthritis, cancer, and other inflammations. In some cases, this condition can be fatal, leading to death. This study aims to examine the immunomodulatory effects of Ethanol Extract of Lime Peel on phagocytosis activity and delayed-type hypersensitivity response in experimental animals. The research methods include sample collection and processing, plant identification, extract preparation, and immunomodulatory effect testing. Phytochemical screening results showed the presence of alkaloids, glycosides, flavonoids, tannins, and steroids in the extract of the lime peel. Simplicia and ethanol extract of lime peel characterization yielded results meet the standards of the Indonesian Herbal Pharmacopoeia 2nd edition. Immunomodulatory effect testing was conducted through phagocytosis activity and delayed-type hypersensitivity response assays. The results showed that the extract at doses of 25, 50, and 100 mg/kgBW had immunostimulatory effects in enhancing phagocytosis activity, while doses of 17.5 mg/kgBW, 35 mg/kgBW, and 70 mg/kgBW significantly increased delayed-type hypersensitivity response compared to the negative control. These findings indicate the potential of ethanol extract of Citrus aurantifolia as an immunomodulator that can enhance the body's immune system. This study provides an important contribution to the development of immunomodulatory therapy derived from natural ingredients such as lime peel for managing immunity-related diseases with lower side effects.

Abstract

This study was carried out to investigate the protective effect of ethanol extract of Curcuma mangga rhizomes on paracetamol-induced hepatotoxicity. High dose of paracetamol (1.35g/kg bw) was used to induce hepatic necrosis of mice liver. The male mice received ethanol extract of C. mangga rhizomes (100, 200 and 400 mg/kg BW) for 7 days. The hepatoprotective actvity of extract was compared to normal, positive (curcuma) and negative control. The liver function was evaluated by measuring the biochemistry parameters which include alanine aminotransferase (ALT) and aspartate aminotransferase (AST). In addition, histophatological study on hepatic tissue section was also carried out. The C. mangga extract displayed hepatoprotective effect except at dose of 100 mg/kg bw. The increasing of serum levels of AST and ALT were inhibited after treatment with ethanol extract at doses of 200 and 400 mg/kb bw which was comparable with normal and curcuma as postive control (p>0.05). In addition, histological assessment of hepatic tissue demonstrated no liver damage, specially at dose of 400 mg/kb BW. The result indicate that ethanol extract of C. mangga rhizomes has hepatoprotective effect, especially at doses of 200 and 400 mg/kg bw .
