Abstract

Diabetes is a chronic disease caused by impaired insulin secretion. WHO has emphasized the use of ethnomedicine for diabetes control. More than 1200 plants have been traditionally used for hypoglycemic effects, of which 800 plants have been scientifically reported to have antidiabetic potential, one of which is clove. Previous research stated that the total flavonoid content in clove leaves was 73.08 mgRE/g extract with a percentage of 7.308%. These flavonoids are thought to be antidiabetic agents. The purpose of this study was to determine the antidiabetic activity of clove leaf ethanol extract and histology of the pancreas of the test rats. Clove leaf simplicia powder was extracted by maceration method using 70% ethanol as solvent. Antidiabetic testing of clove leaf ethanol extract using alloxan at a dose of 150 mg/kgBW. The results showed that clove leaf extract had antidiabetic activity. The percentage decrease in blood sugar levels extract dose 100; 200 and 300 mg/kgBW were 55.61%, 64.06%, and 73.06% and the positive control was 65.83%. The results of histology observations of the pancreas of mice in the negative control group showed that the structure of the islets of Langerhans changed, shrank, irregular, and degenerated cells and pyknosis were found. While the treatment group that was induced by alloxan and given clove leaf extract doses of 100, 200, and 300 mg/kgBW and the positive control showed a clear structure of the islets of Langerhans, more cell mass, cells regenerated and no cells that experienced degeneration and pyknosis were found. The conclusion of this study is that clove leaf extract can reduce blood sugar levels with an effective dose of 300 mg/kgBB and clove leaf extract can improve pancreatic islets of Langerhans cells.

Abstract

Tapak dara (Catharanthus roseus) is a plant belonging to the Apocynaceae family originating from Central America and is generally grown as an ornamental plant. The purpose of this study was to determine the percentage of mortality data and the LC50 value of tapak dara methanol extract (Catharanthus roseus) against Artemia salina Leach larvae using the Brine Shrimp Lethality Test (BSLT) method. The BSLT method used consisted of 6 concentration treatments, namely 20 ppm, 60 ppm, 100 ppm, 140 ppm, 180 ppm, and 0 ppm a negative control, each of which was repeated three times. At each concentration, 10 experimental animals of 48 hours old Artemia salina Leach larvae were used. The toxic effect of the extract was identified by the percentage of mortality of Artemia salina Leach larvae using probit analysis (LC50). The results showed the linear regression equation y = 3.0809x – 12.065. This shows that the mortality of the test animals reached 50% when the compound concentration reached 34,599 ppm. Based on the toxicity test of the methanol extract of the tapak dara flower (Catharanthus roseus) using the Brine Shrimp Lethality Test (BSLT) method, in this study it was toxic because LC50>1000 ppm so it has potential as an anticancer.

Abstract

Gagatan harimau leaves (Vitis gracilis BL) is a plant contains several secondary metabolite compounds, namely alkaloids, glycosides, tannins, saponins and flavonoids and has been used traditionally to treat diarrhea, infectious diseases, inflammation and diabetes. The aim of this study was to to determine data on the percentage of deaths and the LC50 value of the ethanol extract of the Gagatan harimau (Vitis gracilis BL) against Artemia Salina Leach using the Brine Shrimp Lethality Test (BSLT) method. The method used is the Brine Shrimp Lethality Test (BSLT) with concentrations of 180 ppm; 200 ppm; 220ppm; 240ppm; 260ppm; 280ppm; 300 ppm and 0 ppm as negative controls each replicated three times. At each concentration, 10 Artemia salina Leach larvae were used as test animals, each concentration was 48 hours old. The toxic effect of the extract was determined by the percentage mortality of Artemia salina Leach larvae using probit analysis (LC50). The results of this research gave the linear regression equation Y = 11.674x-68.227. Therefore, the LC50 value is 6.275 and the LC50 gets a value is 187.068 ppm. Research on the toxicity of Gagatan harimau (Vitis gracilis BL.) ethanol extract using the BSLT (Brine Shrimp Lethality Test) method in this study is toxic because the LC50 is < 1000 ppm so it has the potential to be anticancer.

Abstract

This study aims to develop and validate an advanced UV spectrophotometry- Classical Least Squares (CLS) for the simultaneous determination of Paracetamol (PA), Phenylpropanolamine HCl (PH), and Chlorpheniramine Maleate (CH) in tablet dosage form. The method involves the preparation of standard solutions of the three compounds across their respective concentration ranges 3-9 µg/mL for PA, 170-512 µg/mL for PH, and 7-19 µg/mL for CH. The CLS spectrophotometry UV method was applied to these spectra to construct calibration, which were then used to predict the concentrations of PA, PH, and CH in tablet dosage form. Validation of the method demonstrated excellent linearity, precision, and accuracy, with recovery rates of 99.26% for PA, 100.06% for PH, and 99.41% for CH. Sensitivity analysis revealed Limits of Detection (LOD) of 0.10 µg/mL for PA, 1.20 µg/mL for PH, and 0.30 µg/mL for CH, and Limits of Quantification (LOQ) of 0,30 µg/mL for PA, 4.00 µg/mL for PH, and 1.50 µg/mL for CH. The method’s robustness and reliability make it an excellent choice for routine quality control of these compounds in pharmaceutical tablet formulations.

Abstract

The study focused on the simultaneous quantification of Isoniazid (INH) and Pyridoxine Hydrochloride (PRD) in tablet form, commonly used in antituberculosis treatments. Assessing the accurate concentration of both INH and PRD in tablets is vital to ensure their effectiveness, safety, and quality. Using the ratio subtraction spectrophotometry method, the study analyzed INH and PRD in Pehadoxin forte® tablets (batch no. 36057007, PT. Phapros, Indonesia), with 0.1 N HCl as the solvent. The method involved obtaining ratio absorption spectra by dividing the absorption spectra of INH and PRD, respectively, to derive zero-order spectra for each drug. Method validation parameters included linearity, accuracy, precision, Limit of Detection (LOD), and Limit of Quantification (LOQ). The results showed linearity values of 0.9985 for INH and 0.9988 for PRD. Accuracy was 98.1838% for INH and 100.0205% for PRD, while precision was 1.8769% for INH and 0.2037% for PRD. LOD and LOQ for INH were 0.8116 µg/mL and 2.7053 µg/mL, respectively, and for PRD, 1.3127 µg/mL and 4.3757 µg/mL. The levels of INH and PRD in the tablets were found to be 102.1157% and 101.3874%, aligning with the Indonesian Pharmacopoeia's standards. This methodological approach provides a reliable analytical tool for the simultaneous assessment of INH and PRD in tablets, potentially extendable to other drug combinations and formulations, thereby contributing to pharmaceutical quality control processes.

Abstract

Gynura procumbens is a diuretic plant that has been used traditionally to cure kidney stones. In order to determine the calcium levels in the rats' kidneys, the anti-nephrolithiasis activity of male white rats was tested in this study. This research method was carried out experimentally in a laboratory which included sampling, plant identification, making simplicia, making ethanol extract of sambung nyawa leaves, characteristics of simplicia, phytochemical screening, administering animal test preparations, then the data was analyzed using the Anova test. Oral administration of succulent leaf extract is done at three different doses: 50, 100, and 200 mg/kg bw. A positive control is provided with Batugin elixir, which is 0.54 mL/125 g BW and induced at a volume of 7.5 mL/125 g BW/day using ethylene glycol 0.75% and ammonium chloride 2%. The study's findings demonstrated that, with a kidney stone inhibitor percentage of 31.71%, the ethanol extract of Sambung Nyawa leaves at a dose of 200 mg/kg BW was more successful in preventing the formation of kidney stones than doses of 50 mg/kg BW and 100 mg/kg BW. This proves that the ethanol extract from Sambung Nyawa leaves can be an alternative way to prevent kidney stones. The conclusion is that the ethanol extract of Gynura procumbens L. leaves has antinephrolithiasis activity in male white rats, especially at a dose of 200 mg/kg BW. It also reduced calcium oxalate levels in mice induced by ethylene glycol and ammonium chloride.
