Abstract

The binding of Plasmodium falciparum erythrocyte membrane protein 1 (PfEMP1) to Intracellular Adhesion Molecule-1 (ICAM-1) is a major pathological mechanism in severe malaria including cerebral malaria. The binding is mediated by PfEMP1-DBL2β domain. The study aimed to explore there combinant protein of PfEMP1-DBL2β domain of P. falciparum isolated from Indonesia. DNA was isolated from a severe malaria patient. The DBL2β domain was amplified using Polymerase Chain Reaction (PCR) with specific primer and cloned into the pJET1 cloning vector. The DBL2β recombinant protein was constructed from DBL2β-pJET1 clone using pET-30a expression vector and expressed in Escherichia coli BL21-DE3. PCR colony and digestion of plasmid clones using restriction enzymes were conducted to confirm cloning result, and the expression of recombinant protein was analyzed using Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis (SDS-PAGE). The expression of DBL2β-PfEMP1 domain is higher in pellet than in supernatant fraction. In conclusion, the DBL2β-PfEMP1 domain recombinant protein of P. falciparum isolated from Indonesia expressed as a ~66 kDa protein in full length. Keywords: DBL2β domain, Indonesia, PfEMP1, Plasmodium falciparum, recombinant protein.

Abstract

It is hoped that the healthy eating pattern approach will not be carried out with angry or violent actions but rather with a good approach so that children can enjoy eating and support their growth and development. An approach that can be taken to encourage children to consume healthy food is introducing various types of food and their contents as a basis for children's knowledge. With this knowledge, children can assess whether the food content on their dinner plate includes four healthy, five perfect, or not. Based on this, it is important to carry out Community Service (PkM) activities targeting elementary school students at SDN Karanganyar Village, Wates District, Kediri Regency. This PkM activity aims to invite students to become familiar with the contents of foods that are often found in the daily diet as an effort to maintain a healthy diet for children's growth and development. Based on the observations and evaluations, the students were enthusiastic when receiving material about "My Dinner Plate" and guessing the nutritional content of various foods. The results of the T-test show a value of 0.001, which means there is a significant difference between the pre-test and post-test scores and there is an increase in students' knowledge of 70%.

Abstract

Diabetes mellitus (DM) is a metabolic disorder that is accompanied  by an increased glucose in the blood or called hyperglycemia. Diabetes can be caused by problems with the pancreas not being able to produce insulin, insufficient insulin secretion, imperfect insulin action, or both. When blood glucose is left elevated for a long time, it can cause complications such as gangrene. Diabetic gangrene is one of the complications caused by tissue damage. Gangrene occurs due to neuropathy and vascular disorders, usually in the peripheral areas of the body. Bacteria found in diabetic wounds are bacteria that produce biofilms such as Staphylococcus aureus. Staphylococcus aureus can release exotoxins such as leukocidin. Leucosidine lyses PMNs and releases ROS which will then induce a cytokine response such as interleukin-6 which will increase CRP levels. hs-CRP is a high precision CRP measurement even at low concentrations. This study aims to determine the correlation between hs-CRP levels and the presence of Staphylococcus aureus in type II diabetes mellitus gangrene wounds. The research design used is observational analytical using a cross sectional approach and accidental sampling. There were 16 research respondents with increased hs-CRP levels in 15 respondents and 9 respondents with Staphylococcus aureus bacteria. Correlation test with a value of p = <0.05 (p=0.032) means that there is a correlation between hs-CRP and the presence of Staphylococcus aureus in type II diabetes mellitus gangrene wounds.

Abstract

Type 2 Diabetes Mellitus (DM type 2) is associated with low-grade systemic inflammation. Neutrophil-Lymphocyte Ratio (NLR) is a pro-inflammatory and oxidative stress biomarker in cardiometabolic diseases such as T2DM. This study aims to analyze the relationship of NLR with leukocyte count, random blood glucose, creatinine, total cholesterol, Serum Glutamic Pyruvic Transaminase (SGPT), and Serum Glutamic Oxaloacetic Transaminase (SGOT). The study design is a retrospective analysis with respondents of 162 uncomplicated T2DM patients treated at Mojowarno Christian General Hospital, Jombang. The results of the study showed that the average number of leukocytes was 12,688 cells/µl, neutrophils 75.3%, lymphocytes 17.2%, NLR 8.5, blood glucose 273.2 mg/dl, SGOT 40.88 U/l, SGPT 54.64 U/l, total cholesterol 196.3 mg/dl (<200), and creatinine 1.13 mg/dl. Correlation tests showed that NLR was significantly associated with leukocyte count (p = 0.000 < 0.05) and total cholesterol (p = 0.000 < 0.05), but not significantly correlated with random blood glucose (p = 0.064 > 0.05), SGOT (p = 0.089 > 0.05), SGPT (p = 0.929 > 0.05), or creatinine (p = 0.625 > 0.05). In conclusion, in patients with type 2 diabetes, NLR was significantly correlated with leukocyte count and total cholesterol, but not with random blood glucose, SGOT, SGPT, or creatinine.
