Abstract

Anggrek Tebu (Grammatophyllum scriptum) propagation can be improved optimally by adding plant growth regulators such as BAP and NAA through in vitro technique. Murashige and Skoog (MS) medium is an essential medium for plant tissue growth that needs another complex mineral and vitamin to develop the protocorm-like bodies (PBL) of Anggrek Tebu. Hence, this study aimed to gain the appropriate composition of modified MS by adding BAP and various coconut water concentrations and to evaluate their effect on PBL of Anggrek Tebu growth (budding time, number of buds, and leaves). The research was conducted using a completely randomized design with two factorials such as modified MS medium (MS (M1), ½ MS (M2), and ¼ MS (M3))and coconut water concentration (0 ml/L (C0), 100 ml/L (C1), 200 ml /L (C2), and 300 ml/L (C3)) with triplication. The result showed that the fastest budding time was remarkable growth from M2C1 offered the bud at three weeks after planting (WAP). In the number of budding, M3C2 showed excellent results and significantly increased the budding growth at 8.33 ± 0.88  in 10 WAP. Meanwhile, the highest leave number was M2C1 at 23.00 ± 3.60, followed by M2C2 and M3C2, respectively, at 20.67 ± 1.45 and 20.67 ± 1.33. This result indicated that modifying MS medium composition by adding the coconut water could assist the PBL of Anggrek Tebu growth in a specific concentration.

Abstract

Coffee (Coffea spp.) consumption increased faster than five years ago. However, coffee production availability declined due to the limited field, soil nutrient deficiency, and chemical fertilizer. The aimed research was to obtain a bacteria-producing IAA hormone isolate and to evaluate its density and diversity in improving soil quality and fertility for coffee growth. The rhizosphere soil samples were collected around the coffee plantation in UBF at Sumberwangi, Malang. The sample was isolated using serial dilution on 0.85% NaCl and spread on a Tryptic soy agar medium. The result revealed that the total number of bacteria-producing IAA was 36 isolates, and two isolates (TAS1.2 and TAS 2.6) had higher IAA hormone production at 60 ppm. The selected bacteria can be optimized for a candidate of bacteria-based fertilizer coffee production. Keywords: IAA, density, diversity index, PGPR

Abstract

Student empowerment through the acclimatization of tissue culture–derived orchids represents a strategic approach that integrates biodiversity conservation efforts with the enhancement of human resource capacity. This community service program aimed to improve the knowledge and skills of final-year Biology students in acclimatizing two orchid species of high conservation and economic value, namely Coelogyne pandurata and Phalaenopsis gigantea. The program employed a combination of theoretical instruction, demonstrations of acclimatization techniques, hands-on training, and interactive discussions on business opportunities in orchid cultivation. A total of 15 students participated in the program, which was preceded by a pre-test and concluded with a post-test to assess improvements in participants’ understanding. The evaluation results showed a significant increase in post-test scores (85.33%) compared to pre-test scores (58.67%), with an average N-Gain value of 0.65, classified as moderate. In addition, participants were able to independently perform all stages of the acclimatization process, including the removal of plantlets from culture bottles, washing off residual media, planting in sterile substrates, and the covering process. Questionnaire results indicated a high level of participant satisfaction with the materials, methods, and overall benefits of the program. Overall, this community service activity successfully enhanced students’ knowledge and technical skills in the acclimatization of tissue culture–derived orchids, supported the conservation of high-value orchid species, and strengthened students’ capacity in horticulture and bioresource-based entrepreneurial development. Pemberdayaan mahasiswa melalui kegiatan aklimatisasi anggrek hasil kultur jaringan merupakan pendekatan strategis yang mengintegrasikan upaya konservasi keanekaragaman hayati dengan peningkatan kapasitas sumber daya manusia. Kegiatan pengabdian kepada masyarakat ini bertujuan untuk meningkatkan pengetahuan dan keterampilan mahasiswa tingkat akhir Program Studi Biologi dalam melakukan aklimatisasi dua spesies anggrek bernilai konservasi dan ekonomi tinggi, yaitu Coelogyne pandurata dan Phalaenopsis gigantea. Metode pelaksanaan kegiatan meliputi pemberian materi teoritis, demonstrasi teknik aklimatisasi, praktik langsung (hands-on training), serta diskusi interaktif terkait peluang usaha budidaya anggrek. Kegiatan diikuti oleh 15 mahasiswa dan diawali dengan pre-test serta diakhiri dengan post-test untuk mengukur peningkatan pemahaman peserta. Hasil evaluasi menunjukkan adanya peningkatan signifikan nilai post-test sebesar 85,33% dibandingkan nilai pre-test sebesar 58,67%, dengan nilai rata-rata N-Gain sebesar 0,65 yang tergolong dalam kategori sedang. Selain itu, peserta mampu melaksanakan seluruh tahapan aklimatisasi secara mandiri, mulai dari pelepasan planlet dari botol kultur, pencucian sisa media, penanaman pada media steril, hingga proses penyungkupan. Hasil angket menunjukkan tingkat kepuasan peserta yang tinggi terhadap materi, metode, dan manfaat kegiatan. Secara keseluruhan, kegiatan pengabdian ini berhasil meningkatkan kompetensi pengetahuan dan keterampilan mahasiswa dalam aklimatisasi anggrek hasil kultur jaringan, mendukung upaya konservasi anggrek bernilai tinggi, serta memperkuat kapasitas mahasiswa dalam bidang hortikultura dan pengembangan kewirausahaan berbasis sumber daya hayati.
