Abstract

Inflammation is a manifestation of the immune response to eliminate antigens and a process to respond to tissue damage. There are various drugs to control and suppress inflammation, referred to as anti-inflammatory agents. One method to test anti-inflammatory activity is the inhibition of Bovine Serum Albumin (BSA) protein denaturation. This review article will discuss the stages and optimal conditions in the inhibition of BSA protein denaturation method using various leaf extract samples. Literature was gathered from several journal database websites such as PubMed, ScienceDirect, and Google Scholar with keywords "anti-inflammatory," "inhibition of BSA protein denaturation," and "leaf extract," spanning publication years from 2002 to 2023. The results of anti-inflammatory activity tests can be expressed as either percentage inhibition or IC50 values. Optimal conditions were obtained by preparing a 0.2% BSA solution with Tris Buffer Saline (TBS), maintaining a 9:1 composition ratio between the BSA solution and the sample, and heating at an optimal denaturation temperature of 70 °C. Before conducting tests using this method, it is crucial to ensure that the suspected active compounds with anti-inflammatory activity are not degraded at this heating temperature.

Abstract

Consuming healthy foods such as nutraceuticals or functional foods is very important to prevent and reduce the risk of degenerative and chronic diseases. Efforts to improve community knowledge and skills related to nutraceuticals are very important, so community service activities are carried out with the aim of providing socialization and training on the production of nutraceutical drinks from local natural ingredients, as well as health checks including blood pressure, blood sugar, cholesterol and uric acid. The partners of this community service activity are PKK women and the community in Kongok Hamlet, Meninting Village, Batulayar District, West Lombok Regency, NTB. The community service activities consist of several stages, namely activity preparation, location survey and coordination with partners, socialization of nutraceutical drinks using PowerPoint slides and leaflets, training on the production of nutraceutical drinks, health checks, and ending with post-tests and activity evaluations using questionnaires. The 56 residents who attended were very enthusiastic in participating in the activities. The results of the questionnaire showed that 35.7% of the participants who attended were able to answer questions related to the socialization material with an average score of 74.3. The results of the activity evaluation questionnaire stated that activities like this are very important to do and the selection and provision of material are very good and clear. Thus, it can be concluded that this community service activity is very beneficial for the community and the socialization and training materials can be understood well by the participants of the community service activity.

Abstract

Abstract Fennel (Foeniculum vulgare) is a plant that has been used empirically to treat insomnia. This study aims to compare the hypnotic-sedative effects of fennel seed decoction with herbal drug Lelap® by in vivo in Swiss Webster mice. Hypnotic and sedative effects were determined by motor coordination using a rotarod test and counted as latency times. This study used 12 healthy female mice which divided into 2 control groups (negative control, water 25 mL/kgBW and  positive control, Lelap® 78 mg/kgBW) and 2 treatment groups (fennel seed decoction dose 325 mg/kgBW and 650 mg/kgBW). The results showed that at the 30th minute, the average latency time for the negative control, positive control, fennel seed decoction dose 325 mg/kgBW and 650 mg/kgBW respectively were 328.33±206.99; 43.33±21.85; 50.67±33.98 and 51.33±4.19 seconds. Statistical analysis (Kruskal Wallis followed by Mann Whitney) showed that the latency time of fennel seed decoction was not significantly different from the positive control (p>0.05) and significantly different from the negative control (p<0.05). Based on the results, concluded that the fennel seed decoction dose of 325 mg/kgBW and 650 mg/kgBW have a hypnotic-sedative effect equivalent to the drug Lelap®, and the dose of 325 mg/kgBW is the most effective dose.   Keywords:          Fennel Seed, Hypnotic-Sedative, Insomnia, Rotarod Test, Latency Time   Abstrak Adas (Foeniculum vulgare) merupakan tanaman yang dimanfaatkan secara empiris sebagai ramuan untuk mengatasi gejala susah tidur (insomnia). Penelitian ini bertujuan untuk mengetahui efek hipnotik-sedatif rebusan biji adas dengan pembanding obat herbal Lelap® secara in vivo pada mencit Swiss Webster. Efek hipnotik dan sedatif ditentukan dengan uji koordinasi motorik menggunakan alat rotarod test dan dinyatakan sebagai waktu latensi. Penelitian ini menggunakan 12 ekor mencit betina sehat yang dibagi menjadi 2 kelompok kontrol (kontrol negatif air 25 mL/kgBB dan kontrol positif Lelap® 78 mg/kgBB) serta 2 kelompok perlakuan (rebusan biji adas dosis 325 mg/kgBB dan dosis 650 mg/kgBB). Hasil penelitian menunjukkan bahwa pada menit ke-30, rata-rata waktu latensi kelompok kontrol negatif, kontrol positif, rebusan biji adas dosis 325 mg/kgBB dan dosis 650 mg/kgBB secara berturut-turut yaitu 328,33 ± 206,99; 43,33 ± 21,85; 50,67 ± 33,98 dan 51,33 ± 4,19 detik. Analisis statistik (Kruskal Wallis diikuti Mann Whitney) menunjukkan waktu latensi rebusan biji adas tidak berbeda signifikan dengan kontrol positif Lelap® (p>0,05) dan berbeda signifikan dengan kontrol negatif air (p<0,05). Berdasarkan hasil penelitian ini, disimpulkan bahwa rebusan biji adas dosis 325 mg/kgBB dan 650 mg/kgBB memiliki efek hipnotik-sedatif yang setara dengan obat Lelap®, dan dosis 325 mg/kgBB merupakan dosis paling efektif.   Kata Kunci:         Biji Adas, Hipnotik-Sedatif,  Insomnia, Uji Rotarod, Waktu Latensi
