Abstract

The process of producing, refining and transporting petroleum can cause pollutants that are harmful to the environment and the living things that surround them. Indigenous microorganisms can be a solution to degrade hydrocarbons that are difficult to degrade. This study aims to isolate the indigenous bacteria from pollutant-contaminated soil and test its effectiveness in degrading hydrocarbons. The research began with soil sampling at the Joint Operating Agency of PT Siak Bumi Pusako-Pertamina Hulu Siak Regency, Riau Province with a purposive sampling method. The next step is the isolation of indigenous bacteria, testing the parameters of pH, Optical density (OD) and CO2 levels during the 16 incubation periods and determining the highest percentage of biodegradation. The results showed that Pseudomonas sp. with a code BTM2 (Bakteri Tanah Minyak 2) had the largest percentage (52.20%) after 16 days of incubation under optimum conditions to degrade the total hydrocarbon of petroleum. Pseudomonas sp. BTM2 is the most effective isolate to degrade hydrocarbons.

Abstract

Depression is a common neuropsychiatric disorder that occurs in humans regardless of gender and age. Areca catechu, as a local plant, can be utilized as an antidepressant to treat depression. The current investigation aimed to determine the antidepressant action from an n-hexane extract of young areca nut using the Forced Swim Test (FST) as a standard method in screening antidepressants validated by the behavior of animals. Short-term medication or acute FST was performed by injecting mice with extract at doses 10 and 50 mg/kg, while the mice were treated with the best dose in long-term medication or sub-chronic FST. A day after the re-test on sub-chronic FST, toxicity test was performed. The extract at the dose of 50 mg/kg possessed antidepressant action during acute and sub-chronic FST. The results of the phytochemical analysis stated that the saponins and steroids present in the young areca nut n-hexane extract may be the active constituents that act as antidepressants. No toxic effects were detected from the young areca nut n-hexane extract in mice that had been injected for 7 consecutive days. n-Hexane extract from young areca nut presents antidepressant potential.

Abstract

Era modern menyebabkan teknologi semakin canggih dan mampu memanjakan masyarakat global terutama remaja, sebagai pengguna terbanyak, dapat memenuhi berbagai kebutuhannya melalui internet. Mereka dapat mencari berbagai informasi, bersosialisasi, hingga memenuhi kebutuhan hiburan dengan menggunakan fasilitas internet. Akan tetapi, penggunaan teknologi digital secara berlebihan dapat menimbulkan kesenangan instan dan berujung pada tingkat kecanduan. Hal inilah yang kemudian menyebabkan produktivitas menurun. Para pemuda yang tergabung ke dalam Barisan Muda Riau mengaku sulit untuk menentukan skala prioritas dan menjadi kurang produktif dalam berbagai kegiatan organisasi maupun perkuliahan karena mulai kecanduan bermain game online, bersosial media, serta kegiatan adiksi lainnya. Oleh karena itu, tim pengabdian Universitas Riau yang melibatkan dosen-dosen yang ahli di bidang Biokimia, Kedokteran (Spesialis Kedokteran Jiwa), dan Biologi serta mahasiswa Kuliah Kerja Nyata Terintegrasi, menginisiasi kegiatan seminar yang bertajuk “Kesehatan Mental yang berkaitan dengan Adiksi dan Dopamin”. Kegiatan ini melibatkan Barisan Muda Riau, yang diharapkan setelah dibekalkan ilmu terkait tips untuk membangun kebiasaan baik, para pemuda dapat emnjadi trendsetter bagi masyarakat.

Abstract

Depression is a global mental health issue and herbal antidepressants are increasingly sought due to the side effects of synthetic drugs. The present study aimed to evaluate the potential of ethyl acetate extract of old areca nut in treating depression of mice subjected to acute and sub-chronic Forced Swim Test (FST). Ethyl acetate extract was injected intraperitoneally 1 h before the FST at a dose of 50 and 100 mg/kg. The 100 mg/kg dose showed the greatest reduction in immobility time (22.29%) during acute testing. Following seven days of treatment, it was discovered that the extract significantly alleviated the immobility time in sub-chronic FST. The results of the phytochemical analysis showed that phenolics present in the ethyl acetate extract are probably active constituents with antidepressant properties. After prolonged administration, the substance has no toxic effects on the bodies of test animals.

Abstract

Diapers are made by cotton and pulp containing cellulose that can be used as a substrate in the production of cellulose enzymes. The purpose of this research is to determine the ability of cellulolytic fungi Trichoderma asperellum LBKURCC1 and cellulolytic bacteria S-22 to degrade the used diapers containing urine by solid fermentation for 10, 20, and 30 days. The activity of crude extract of cellulose enzyme was observed with CMC 2% as substrate at pH 5,5 (Trichoderma asperellum LBKURCC1) and pH 7 (bacteria S-22 isolate), incubation temperature 40ºC during 30 minutes by using Nelson Somogyi’s method. The result showed that used diapers can be used as substrate for cellulose enzyme production by fungi Trichoderma asperellum LBKURCC1 with the highest activity of cruide extract cellulose enzyme obtained at 10th days solid fermentation of (1,891 ± 1,453) × 10-3 U / mL and by bacteria S-22 isolate at 20th days solid fermentation of (2,854 ± 0,019) × 10-3 U/mL. It can be concluded that bacteria S-22 isolate would degrade the used diapers better than fungi Trichoderma asperellum LBKURCC1. The result of this research compared to quality standards compost of SNI: 19-7030-2004 have not fulfilled, so it can not be used as compost.

Abstract

Naphthalene is one of the pollutants that is difficult to degrade because it belongs to the PAH group and it was carcinogenic. The purpose of this study was to test the effectiveness of an indigenous fungi isolate from Riau Province to degrade naphthalene toxic compounds with glucose as growth cosubstrate. Naphthalene degradation by fungi isolates was influenced by several parameters, including biomass, pH and percentage degradation measured at 0, 4, 8, 12 and 16 days of incubation. Based on biomass parameters, the isolate of Penicillium sp. LBKURCC153 experienced an exponential phase until the 16th day. In the exponential phase, the maximum number of cells increases which shows a positive response to the use of glucose and naphthalene as the carbon and energy source. Isolate decreased their pH from 7 to 3,8, that mean proves the growth activity produce organic acid metabolites of degrade naphthalene. Isolate were also able to degrade naphthalene respectively 27,5% by Penicillium sp. LBKURCC153 in medium with glucose at optimation time until 4 th day incubation. In conclusion, glucose cosubstrate can increase naphthalene degradation percentage by Penicillium sp. LBKURCC153.

Abstract

This study aims to screen the potential of five active compounds in areca nut L-phenylalanine, protocatechuic acid, ocanine, nicotinic acid, L-tyrosine as antidepressants through pharmacokinetic analysis and drug likeness analysis using the SwissADME online application, then predict toxicity and brain penetration through 2D radar bioavailability results. using the ADMETLab online application. Compounds that met all parameters were L-phenylalanine and L-tyrosine with high GI absorption results, no P-gp inhibitors, no CYP1A2 inhibitors, no CYP2D6 inhibitors, yes BBB permeant and had a logKp value <2.5cm/s, no hepatotoxicity and no AMES toxicity, complies with Lipinski, Ghose, Veber regulations, bioavailability score 0.55, and showed the optimal range of values as an oral drug.

Abstract

This study aims to investigate the potential of methanolic extract from aged areca nut as an antidepressant using acute and sub-chronic Forced Swim Test (FST) methods. In the acute FST, mice were injected with methanolic extract doses of 10, 50, and 100 mg/kg intraperitoneally as a single dose to determine the dose with the best activity. Meanwhile, in the sub-chronic FST, the best dose was administered for 7 consecutive days, and a re-test was conducted on the seventh day after acute FST. A significant reduction in immobility time after acute treatment was only observed with a dose of 100 mg/kg. Therefore, mice were injected with this dose for the next 7 days. However, this dose did not significantly decrease immobility time based on the results of sub-chronic FST. Thus, the methanolic extract at a dose of 100 mg/kg was considered not potentially effective for continuous use. Phenolic compounds, steroids, and saponins as the main constituents in the extract did not induce toxic effects after treatment for 7 days.
