Abstract

Abstract: Abnormality of lipid profile is one of cardiovascular risk factors which increases with aging.Anthropometric measurement is one of the predictors of abnormal lipid profile without invasive methodsand affordable cost. One of the anthropometric measurements that could be used as a predictor is waist tohip ratio (WHR). The aim of this study was to determine the correlation between WHR with abnormality oflipid profile which were related to total cholesterol, Low Density Lipoprotein (LDL), High DensityLipoprotein (HDL), and triglyceride (TG) in pre-geriatrics and geriatrics group. This study was analyticalobservational with cross sectional design. Statistical analysis used Kolmogorov-Smirnov followedbivariable analysis with confidence interval (CI) 95%. The correlation statistical test used Pearson andSpearman. The results showed no statistically significant correlation between WHR with total cholesterol(r=-0.054, p 0.05), LDL (r=-0.047, p 0.05), TG (r=0.018, p 0.05), and HDL (r=0.016, p 0.05).Keywords: lipid profile, waist-to-hip ratio (WHR), pre-geriatric, geriatri

Abstract

Nicotine, the active compound in cigarettes, can cause impaired glucose metabolism byincreasing insulin resistance as well as decreasing insulin secretion in ? cell pancreas. Thiscondition can increase the risk of type 2 diabetes in human. This study aims to evaluate the effectof smoking behaviour, determined by Cigarette per Day (CPD) and smoking duration, onglychohemoglobine (HbA1c) levels of Javanese Indonesian smokers. 30 smokers were studiedconsisting of 7 smokers with 10 CPD, 19 smokers with 11-20 CPD and 4 smokers with 21-30CPD. They had been smoking for more than 10 years. The whole blood sample was used toexamine the HbA1c levels. The HbA1c levels were tested at Bethesda Hospital's cliniclaboratories using Architect 600 instrument. The results showed that CPD and smoking durationsignificantly influenced HbA1c, in which F count was F table (370.541 3.354) withsignificance 0.05 (2.35. 10-20 0.05) and multiple correlation coefficient (R) of 0.982.Therefore, based on this research finding, it was concluded that longer smoking duration andhigher CPD caused higher smokers HbA1c level.

Abstract

Abstract: Mahkota dewa (Phaleria macrocarpa (Scheff) Boerl) is one of the plants frequentlyused as anticancer. But mahkota dewa extract have a high toxicity. One of certain effort thatcould reduces the toxic effect of medicine is nanoparticle decivery. The aims of this study were tomake nanoparticle from chitosan-mahkota dewa extract using TPP as a cross linker and todetermine it's particle size, zeta potential, loading capacity and loading efficiency. Chitosannanoparticles extract of Phaleria macrocarpa fruits with TPP as cross linker by ionic gelationmethod. Chitosan was dissolved in acetic buffer solution at pH 4 with various concentrations(0.045 and 0.09 % b/v), meanwhile tripolyphosphate (0.009 dan 0.018 % b/v) with volumecompare (5 : 1). The characterizations of chitosan nanoparticles of extract were determined bymeasure its particle size, zeta potential, loading capacity, and determining efficiency valuenanoparticle process formed. The results showed that the most stable extract were onconcentration 0.68 and 0.9 mg/ml (kitosan 0.09 % b/v, TPP 0.018 % b/v) with 350 rpm stirringspeed. The average of nanoparticle size were 190.9 and 162.87 nm. The zeta potential were 48.5mV and 60.86 mV, the loading capacity were 2.96 and 5.33 %, than the loading efficiency were35.75 and 45.26 %. Preparation of chitosan-extract of mahkota dewa fruits by ionic gelationmethod can produced nanoparticles and has a short range of size distribution, grade ofuniformity and good stability.Key words: Mahkota dewa, nanoparticles, chitosan, ionic gelation.

Abstract

Lemongrass was found to be a promising herb for anti-hyperglycemia treatment due to its activities to inhibit alpha glucosidase and alpha amylase in vitro activities and ability to improve blood glucose profile. Lemongrass potency through its anti-hyperglycemic ingredients requires evaluation of the functional stability during processing. In this study, the in vitro anti-hyperglycemic activities of spray-dried aqueous extract of lemongrass were determined by its inhibitory activity against rat intestinal glucosidase enzymatic hydrolysis of sucrose. In vivo activity was observed based on its ability to prevent blood glucose elevation in oral glucose, sucrose and maltose tolerance tests (OGTT, OSTT and OMTT). The in vitro evaluation showed that aqueous extraction, which involved stirring at 70 C for 40 min, successfully increased the glucosidase inhibitory activity of lemongrass extract, while spray drying with inlet 130 C had no significant impact to the activity tested in vitro. Spray-dried lemongrass powder was found to be effective for lowering blood glucose level in OGTT, OSTT and OMTT. This study provides support for further development of lemongrass extracts as functional ingredients for hyperglycemia treatment.

Abstract

A study on molecular docking-based virtual screening has been conducted to select virtual hit of compounds, reported its existence in fungal endophytes of Chaetomium sp. as cytotoxic agent of breast cancer. The ligands were docked into Human Estrogen Receptor alpha (HERa) as the protein which regulates the breast cancer growth via estradiol-estrogen receptor binding intervention. The results showed that two compounds bearing xanthone and two compounds bearing benzonaphtyridinedione scaffolds were selected as virtual hit ligands for HERa leading to the conclusion that these compounds were good to be developed as anti breast cancer.

Abstract

Abstract: Approximately 11.000 new child cancer cases per annum. An appropriate cytostaticdrug regimen can worse renal function. Aims of this research to determine the use of cytotoxic inchildren chemotherapy based on GFR according Schwartz and CB formulas in RSUP Dr.Sardjito Yogyakarta 2010 period. Cytotoxic drugs include antibiotic and non antibioticcytotoxics. An observational descriptive evaluative study with retrospective design was done.Inclusion criteria were patient 0 12 years with creatinine serum data and exclusion criteriawere acute or chronic kidney failure with creatinine serum 5 mg/dL. The data taken werepatient's age, height, weight, and creatinine serum. Adjustment doses was evaluated based on2GFR 60 mL/min/1.73 m for cytotoxic drugs. Dose adjustment was evaluated according to DIHand BSA guidelines. Using 171 patient's datas with 160 cases of antibiotics prescription and 704cases of non antibiotics prescription. 1,8%; 5,6% cases of antibiotics prescription and 1,0%;3,6% cases of non antibiotics prescription required adjusment doses according to the Schwartzand CB formulas. Unappropriate doses were found for doxorubicine and bleomycine as cytotoxicantibiotics usage and etoposide, L-asparginase, dan cyclophosphamid as non cytotoxicantibiotics usage.Keywords : cytotoxic drugs, children, glomerular filtration rate, Schwartz formula, CounahanBarratt formula.

Abstract

Sweet potato cultivar Cilembu, kind of rancing from Cilembu Village, Sumedang, West Java has really sweet taste due to the high content of reducing sugar. The amount of reducing sugar needs to be proven scientifically. The study aimed to compare Somogyi-Nelson method and Anthrone-Sulfate method in order to determine reducing sugar level in Cilembu sweet potato. Cilembu sweet potato must be stored in freezer at 15C for 2 weeks before used as the sample. Analytical method validation was performed for the two methods. The determination results between two methods were compared. The amount of reduction sugar (glucose) from Cilembu sweet potato determined by Somogyi-Nelson method was 3.42%, the value of %recovery was 99.8%, LOD = 0.00167 (mg/mL), LOQ = 0.00557 (mg/mL), and value of RSD was 3.7%. While the average amount of reduction sugar determined by Anthrone-Sulfate method was 14.9%, the value of %recovery was 74.7%, LOD = 0.0198 (mg/mL), LOQ = 0.066 (mg/mL), and value of RSD = 8.6%. The Somogyi-Nelson method was more recommended for the analysis of the reducing sugar in Cilembu sweet potato compared with Anthrone-Sulfate method.

Abstract

Abstract: Antioxidant plays a role in inhibiting oxidation by binding to free radicals. As a result,the cell damage that leads to degenerative diseases can be inhibited. This research wasconducted to determine the antioxidant activity and total phenolic content of ethyl acetatefraction of velvet aple (Diospyros blancoi A.DC.) juice. Previously known that other plants fromgenus Diospyros contain phenolic compounds such as quercetin. Antioxidant activity testperformed qualitatively and quantitatively using radical of 1,1-diphenyl-2 pikrilhidrazil(DPPH) and expressed as the value Inhibition Concentration 50 (IC50). Determination of totalphenolic using Folin-Ciocalteu method expressed as equivalent mass of gallic acid. Phenoliccompounds will be oxidized by the Folin-Ciocalteu under alkaline conditions, forming a bluesolution. The results showed that the ethyl acetate fraction of velvet apple juice has very strongantioxidant activity with IC50 of (30.0 0.09) mg / mL. Total phenolic content of (393.5 0.35) mggallic acid equivalents per gram of ethyl acetate fraction.Keywords: antioxidant, velvet apple (Diospyros blancoi A. Abstract

Parijoto (Medinilla speciosa Reinw. ex Blum) fruit is known to have pharmacological activity as cholesterol lowering levels. Its activity needs to be increased with nanoparticle system so that the active substance can bind 100% to the action target. This study aims to determine the formation of nanoparticles from parijoto fruit (NEBP) and activity test as a decrease in cholesterol levels. The formation of nanoparticles used variations of concentration and volume of chitosan and NaTPP. Anti-cholesterol testing is based on the amount of free cholesterol in the sample that reacted with Lieberman-Burchard into complex green compounds. The best formation of NEBP was 0.2% chitosan, 0.1% NaTPP and volume ratio 5:1. The particle size showed an average size of 269.3 nm (10-1000 nm). The result of the percent transmittance and polydispersity index were 99,379 (close to 100%) and 0.378 (PDI 0.5). The functional group-specific of NEBP was OH, N-H, PO3. The morphology was round and non-uniform particles. NEBP can decrease 50% cholesterol levels with a smaller EC50 value was 89.08 compared to the extract (EC50 259.98 ppm). Nanoparticles of parijoto fruit is a potential candidate for anti-cholesterol drug.

Abstract

Epigallocatechingallate (EGCG) in green tea extract has activity as an anti-inflammatory agent. On the other hand the stability of EGCG is poor because of the oxidation. The aim of this study was to determine the influence of Vitamine C and Vitamine E in formulation of green tea extract cream to the stabiliy of EGCG. The green tea extract was obtained from the extraction process by infundation followed by fractination with ethyl acetate as the solvent. The three formulas were compiled in similar composition with the concentration of vitamine C 1% (FI), Vitamine E 1% (FII) and there was no Vitamine C and Vitamine E (FIII) as a control. The EGCG level was determinated by TLC-densitometry methode. The stability parameter was determinated by calculated of the Q10 of each formula. The result of this study showed that the parameter of t90 of EGCG with Vitamine C 1%, Vitamine E 1% and control addition were 0.0108 hours, 0.0087 hours, 0.0084 hours, respectively. Stability of EGCG in green tea leaf extract cream with addition of the vitamin C 1% was higher than it stability with the addition of vitamin E 1%. The concentration of Vitamin C 1% was the optimum concentration as antioxidant in formulation of green tea extract cream.
