Abstract

The incidence of skin cancer in Indonesia reaches 6-8%, so the skin needs effective protection. Zinc Oxide (ZnO) is a sunscreen with Sun Protecting Factor (SPF) 50 which is able to reduce exposure to Ultraviolet rays. ZnO is difficult to dissolve in water making an obstacle if dispersed in a hydro gel matrix, so it is formulated into a Self-Nanoemulsifying Drug Delivery System (SNEDDS) preparation. SNEDDS formula was made using tween 80 as surfactant, propylene glycol as a co-surfactant, and olive oil. The optimum proportion of the three components was optimized with the D-Optimal method using Design Expert Stat-Ease 9 Trial software. Software obtained 16 formulas which were tested for physical stability response: transmittance value (%) and pH value. SNEDDS optimum formula was compared with the D-Optimal prediction formula using the student's t-test statistical analysis (p0.05), the loading dose of ZnO, Particle Size Analysis, and Zeta Potential. The optimum proportion of propylene glycol, tween 80, and olive oil making up SNEDDS were 9.9%: 81%: 9.1% respectively. The result of the percent transmittance response was 92.30% and the pH value was 7.20. Software prediction results: transmittance value was 92.59% and pH value was 7.37. Statistical analysis of one sample t-test showed no difference between observations and D-Optimal predictions. SNEDDS was able to load 2.0 mg ZnO/gram SNEDDS with a particle size of 150.2 nm; polydispersity index of 0.54 and zeta potential of -28.50 mV. The SPF value of SNEDDS ZnO was 16.

Abstract

The aim of this research is to evaluate the effect of pasak bumi roots (Eurycoma longifolia Jack) towards the blood glucose level in glucose-loaded mice. The blood glucose-lowering effects were tested using Oral Glucose Tolerance Test (OGTT) method. The mice were given with infusion of pasak bumi roots at the doses of 0.83; 1.67; 3.33 g/kgBW and methanol extract of pasak bumi roots at the doses of 102; 210; 420 mg/kgBW. All treatments were conducted orally, 30 minutes before the administration of glucose (2 g/kgBW). The blood glucose levels were measured at 0 minute before the administration of glucose and at 15, 30, 60, 90, and 120 minutes after the administration of glucose. Blood samples were obtained through the mice tails vena lateralis using glucometer. The blood glucose levels result which were obtained at the 0 until 120 minutes were calculated to obtain AUC. AUV values of each treatment group were analyzed statistically. Based on the results of the research, it can be concluded that the methanol extract of pasak bumi roots has blood glucose-lowering effect at the doses of 210 and 420 mg/kgBW, but infusion of pasak bumi roots does not have effects on lowering the blood glucose level in the glucose-loaded mice.

Abstract

The application of bacterial endotoxin test (BET) using TAL reagent on radiopharmaceutical kits is very important to conduct. The radiopharmaceutical kits that will be tested are macro aggregated albumin (MAA), tetrofosmin and ethambutol kits. Endotoxin testing stage was TAL 0.25 EU/mL verification test, inhibition/enhancement test, and endotoxin test for sample. Pyrogen testing using rabbits was also performed as a comparison test. The results of the TAL reagent verification test were all samples showed values corresponding to the standards of 2? = (+), 1? = (+), 1/2? = (-), 1/4? = (-), and negative water control (NWC) = (-). Furthermore, inhibition/enhancement tests for MAA, tetrofosmin, and ethambutol products show non-inhibiting or gel-inducing results, which are in accordance with acceptability standards, so that the samples can be tested using TAL reagents. The pH measurement results in each sample were MAA of 6.0, tetrofosmin of 7.0, and ethambutol of 8.0. The results of MAA, tetrofosmin, and ethambutol product testing were a sample = (-), positive product control (PPC) = (+), positive water control (PWC) = (+), and NWC = (-). In addition, the results of pyrogen testing also showed negative for MAA, tetrofosmin, and ethambutol.
