Abstract

In Vitro Somatic Embryogenesis and Plantlet Regeneration of Three Varieties of Alfalfa (Medicago sativa L.)ABSTRACTAlfalfa (Medicago sativa L.) is a valuable plant as a source of food for animal, forage, pharmaceutical, medicine, food supplement, and human consumption.  In vitro selection technology combined with induction or spontaneous mutagenesis has been effective in altering or isolating genetic variability for desirable characters.  Consequently, a reproducible in vitro propagation technique of that plant is mandatory. The aim of the research was to obtain information on the embryogenic callus induction, somatic embryogenesis, and plantlet regeneration of three varieties of alfalfa. The results showed that an optimum embryogenic callus induction (82%) was obtained on Murashige & Skoog (MS) basal medium containing 2 ppm 2,4-dichlorophenoxyacetic acid (2,4-D), 2 ppm kinetin and 2 ppm a-naphthaleneacetic acid (NAA). Those embryogenic calli could subsequently develop into somatic embryos, which germinated and regenerated into normal plantlets on R1 medium consisting of MS nutrients without the addition of plant growth regulator.Keywords: alfalfa, callus, embryogenic, plantlets, regeneration ABSTRAKAlfalfa (Medicago sativa) adalah tanaman berharga sebagai sumber makanan untuk hewan, yaitu hijauan pakan ternak, farmasi, obat-obatan, suplemen makanan dan konsumsi manusia. Teknologi seleksi in vitro yang dikombinasikan dengan induksi atau mutagenesis spontan telah terbukti efektif dalam mengubah atau mengisolasi variabilitas genetik untuk karakter yang diinginkan. Oleh sebab itu, keberhasilan teknik perbanyakan in vitro yang telah terbukti dapat direproduksi dari tanaman tersebut menjadi syarat yang harus terpenuhi. Tujuan dari penelitian ini adalah untuk mendapatkan informasi mengenai induksi kalus embriogenik, embriogenesis somatik dan regenerasi planlet dari tiga varietas alfalfa. Hasil penelitian menunjukkan bahwa induksi kalus embriogenik optimal (82%) didapat pada media Murashige & Skoog (MS) dengan  2 ppm 2,4-dichlorophenoxyacetic acid (2,4-D), 2 ppm kinetin dan 2 ppm a-naphthaleneacetic acid (NAA). Kalus embriogenik tersebut dapat membentuk embrio somatik, embrionya berkecambah dan beregenerasi membentuk planlet normal pada perlakuan media R1 yaitu nutrisi MS tanpa penambahan zat pengatur tumbuh.Kata Kunci: alfalfa, embriogenik, kalus, planlet, regenerasi

Abstract

GROWTH ABILITY OF Jatropha curcas L. EXPLANTS ON THE IN VITRO MEDIA CONTAINING IBA AND BAResearch on the growth ability of Jatropha curcas L. shoots and callus in solid and liquid media have been conducted. Explants were planted in the initiation MS medium. After ten weeks, the explants were subcultured into solid and liquid media containing combination of IBA and BA treatments. The number of combinations was 12 treatments, each with 6 replications. Observation was conducted from the first week after subculturing upto the fourth week. Parameters of observation were the percentage of explant forming shoots, the number of shoots, height, number of leaves, weight, color, and form of callus. The results showed that the explant which was subcultured in liquid media had higher growth rate than those subcultured in solid media. Treatment of 1 ppm IBA + 0.5 ppm BA gave a good result on the growth of shoots on solid and liquid media. For callus formation, treatment of 2 ppm IBA + 1 ppm BA gave the best result.Keywords: Callus, Jatropha, IBA and BA, solid and liquid media, hormone ABSTRAKPenelitian terhadap kemampuan tumbuh kalus dan tunas tanaman jarak pagar (Jatropha curcas L.) telah dilakukan pada media padat dan cair. Eksplan diinisiasi pada media MS dan setelah 10 minggu dipindahkan ke media padat dan cair yang mengandung perlakuan kombinasi hormon IBA dan BA. Jumlah kombinasi sebanyak 12 perlakuan dan setiap perlakuan dibuat 6 ulangan. Pengamatan dilakukan dari minggu pertama subkultur hingga minggu keempat. Peubah yang diamati adalah persentase eksplan yang membentuk tunas, jumlah dan tinggi tunas, terbentuknya daun pada tunas, perbedaan berat, bentuk, dan warna kalus. Hasil yang diperoleh menunjukkan bahwa eksplan yang disubkultur pada media cair memiliki laju pertumbuhan yang lebih tinggi daripada media padat. Perlakuan IBA 1 ppm + BA 0,5 ppm menghasilkan pertumbuhan tunas yang paling tinggi pada media padat dan cair. Pembentukan kalus yang terbaik diperoleh pada perlakuan IBA 2 ppm + BA 1 ppm.Kata kunci: Kalus, Jatropha, IBA dan BA, media padat dan cair, hormon

Abstract

Utilization of agroindustrial byproduct as cheap raw materials for aquafeed was hampered by its poor nutritional value as well as high antinutrition content which could be overcome through fermentation. Coconut testa (CT) and cassava bagasse (CB) were mixed, and fermented using Rhizopus oryzae for preparing aquafeed. Subsequent feeding test was carried out on tilapias (Oreochormis niloticus L.) using 5 feeding treatments: one unfermented feed (commercial feed 100%), and the other four feeds produced by fermentation using substrate mixture of CT and CB in 4 different ratios, namely 100%:0%, 75%:25%, 50%:50%, and 25%:75%. Feeding 100% commercial feed (true protein 22.64% and crude fibre 14.67%) showed the best results on the fish growth with body weight gain of 3.96 g and feed conversion ratio of 8.63. Meanwhile, feeding fermented feeds (true protein 7.96-20.27% and crude fiber 14.14-18.47%) resulted in body weight gain in the range of 2.22 to 2.75 g with feed conversion ratio of 10.89 to 13.62. Thus, the fermented feeds promoted growth in tested tilapias albeit less optimally than commercial feed did.Keywords: Rhizopus oryzae, Oreochromis niloticus, coconut testa, cassava bagasse, fermentation ABSTRAKPenggunaan hasil samping agroindustri sebagai bahan pakan murah ikan terkendala rendahnya nutrisi dan tingginya antinutrisi yang dapat diatasi melalui fermentasi. Dalam penelitian ini, kulit daging buah kelapa (KK) dan onggok singkong (OS) dicampur dengan perbandingan tertentu, lalu difermentasi menggunakan Rhizopus oryzae untuk pakan ikan. Uji pemberian pakan dilakukan terhadap ikan nila (Oreochormis niloticus L.) dengan 5 perlakuan: satu perlakuan pakan tanpa fermentasi (pakan komersial 100%), dan empat perlakuan pakan fermentasi substrat campuran KK dan OS dengan 4 perbandingan yang berbeda, yakni 100%:0%, 75%:25%, 50%:50%, dan 25%:75%. Pemberian pakan komersial 100% (protein sejati 22,64% dan serat kasar 14,67%) memperlihatkan hasil terbaik pada pertumbuhan ikan nila dengan pertambahan bobot badan 3,96 g dan rasio konversi pakan 8,63. Sebaliknya, pemberian pakan fermentasi (protein sejati berkisar 7,96-20,27% dan serat kasar 14,14-18,47%) menghasilkan pertambahan bobot badan ikan pada kisaran 2,22-2,75 g dengan rasio konversi pakan 10,89-13,62. Dengan demikian pakan fermentasi tersebut mendorong pertumbuhan ikan nila namun kurang optimal dibandingkan pakan komersial.Kata Kunci: Rhizopus oryzae, Oreochromis niloticus, kulit daging buah kelapa, onggok, fermentasi

Abstract

The Improvement of Cacao Beans Quality through Fermentation by Using Lactobacillus sp. and Pichia kudriavzeviiABSTRACTIndonesia is one of the main cacao producers in the world. Indonesian cacao product is, however, relatively of low quality. Quality improvement of cacao beans is thus needed to increase added value of the product through such method as fermentation using bacteria and yeast. This study was conducted using four fermentation treatments, namely F1 (spontaneous fermentation without the addition of inoculum), F2 (addition of lactic acid bacteria inoculum), F3 (addition of yeast inoculum), F4 (addition of mixed lactic acid bacteria and yeast inoculum). The fermentation was carried out for 5 days. The parameters measured were the microbial cell number, pH, ethanol, total reducing sugar, and total acid concentration, as well as cacao seed quality. Results showed that, compared to the other treatments, the F4 treatment gave the best result, namely 83% of the cacao seeds being fermented, 2% non-fermented, 14% unfermented, 1% moldy, and 2% germinated. The liquid produced during the fermentation contained the highest reducing sugar of 123.38 mg·mL-1, the highest total acid of 24.42 mg·mL-1, and 3.57% ethanol.Keywords: cacao beans, fermentation, lactic acid bacteria, starter, yeast ABSTRAKIndonesia adalah salah satu penghasil kakao utama di dunia. Namun berdasarkan mutu, produk kakao Indonesia masih relatif tergolong rendah. Peningkatan kualitas biji kakao diperlukan untuk memberikan nilai tambah pada produk melalui metode seperti fermentasi menggunakan bakteri dan khamir. Penelitian ini dilakukan dengan empat perlakuan fermentasi yaitu F1 (fermentasi secara spontan tanpa penambahan inokulum), F2 (dengan penambahan inokulum bakteri asam laktat (BAL)), F3 (dengan penambahan inokulum khamir), F4 (dengan penambahan inokulum campuran bakteri asam laktat dan khamir). Fermentasi dilakukan selama 5 hari, dan parameter yang diukur selama fermentasi adalah jumlah mikroba, pH, kadar etanol, gula pereduksi, total asam serta kualitas biji. Hasil menunjukkan bahwa, dibandingkan perlakuan lainnya, perlakuan F4 memberikan hasil terbaik yaitu 83% biji terfermentasi, 2% tidak terfermentasi, 14% terfermentasi sebagian, 1% berjamur, dan 2% berkecambah. Cairan fermentasi tersebut mengandung gula reduksi yang paling tinggi 123,38 mg·mL-1, total asam tertinggi 24,42 mg·mL-1, dan kadar etanol mencapai 3,57%.Kata Kunci: bakteri asam laktat (BAL), biji kakao, fermentasi, khamir, starter

Abstract

Tanaman sagu (Metroxylon sagu Rottb.) memiliki potensi yang besar sebagai sumber pangan, energi dan bahan baku industri. Kultur jaringan tanaman sagu telah dilakukan di Balai Pengkajian Bioteknologi BPPT dalam rangka perbanyakan genotipe atau aksesi unggul secara massal. Namun demikian, kendala utama yang dihadapi pada perbanyakan in vitro tanaman sagu adalah sulitnya pembentukan akar. Penelitian ini bertujuan untuk mendapatkan kombinasi hormon yang tepat dalam menginduksi perakaran tanaman sagu in vitro. Tunas anakan muda (15-20 cm) yang diperoleh dari daerah Rangkasbitung, Provinsi Banten digunakan sebagai eksplan. Dalam penelitian ini perakaran in vitro diinduksi dengan berbagai perlakuan jenis dan konsentrasi hormon auksin, konsentrasi medium dan jenis agar. Sebagai medium dasar digunakan medium Gamborg. Hasil penelitian menunjukkan bahwa konsentrasi IBA dan NAA yang terbaik adalah pada taraf 35 ppm. Selanjutnya Gelrite memberikan respon yang positif dengan munculnya perakaran pada pangkal eksplan.Kata Kunci: Induksi perakaran,  jenis agar, kultur in vitro, auksin, sagu ABSTRACTSago palm (Metroxylon sagu Rottb) has huge potential as food, energy and industrial bioresources. In vitro culture of sago palm was performed in Biotech Center, BPPT in order to obtain a large-scale of mass clonal propagation of superior genotypes. Nevertheless, the main obstacle for the sago palm in vitro propagation was rooting formation. The purpose of our study was to obtain the best hormones combination for root induction on sago palm shoots in vitro. The young suckers (15-20 cm) obtained from Rangkasbitung area, Banten Province, were used as explants. In our study, in vitro rooting was induced by different types and concentrations of auxin, medium strength and solidifying agents. The shoots were cultured on Gamborg media. The result showed that the best level of both hormones IBA and NAA for root induction was 35 ppm. Moreover the solidifying agent of Gelrite gave positive response by stimulating root at the basal-end.Keywords: Rooting induction, solidifying agent, in vitro cultures, auxin, sago palm

Abstract

Being the world’s third largest producer of cocoa (Theobroma cacao), Indonesia provides abundant cocoa pod husk byproduct. Despite its high content of biological materials, its use as animal feed, however, has been limited due to its low nutritive values and significant content of antinutritive substances. Thus, this study was aimed to investigate the changes of selected amino acids glutamate, aspartate, valine, alanine, and proline, as well as the antinutritional compound theobromine in cocoa byproduct-rice bran mixed substrate following fermentation using Rhizopus oryzae. The fermented substrate obtained had its true protein content increased from 1.95% to 23.16%. After analyses using ultra-performance liquid chromatography quadrupole time of flight mass spectrometry, the following amino acids, namely: total and free glutamates, total and free valine, total proline, as well as free alanine underwent increase, while the others decreased. The concentration of the antinutritional factor theobromine was below the limit detectable by HPLC.Keywords: Rhizopus oryzae, Theobroma cacao, theobromine, fermentation, amino acidsABSTRAKSebagai penghasil kakao (Theobroma cacao) terbesar ketiga di dunia, Indonesia mempunyai hasil samping melimpah berupa kulit cangkang kakao. Meskipun kandungan bahan biologisnya tinggi, penggunaan produk samping ini sebagai pakan ternak masih terbatas karena nilai gizi yang rendah serta kandungan zat antinutrisi yang tinggi. Oleh karenanya, penelitian ini bertujuan mengetahui perubahan kandungan asam amino glutamat, aspartat, valin, alanin, dan prolin, serta senyawa antinutrisi teobromin dalam campuran hasil samping coklat-dedak padi pasca fermentasi menggunakan Rhizopus oryzae. Substrat hasil fermentasi mengalami peningkatan kandungan protein sejati dari 1,95% menjadi 23,16%. Hasil analisis menggunakan ultra-performance liquid chromatography quadrupole time of flight mass spectrometry, menunjukkan bahwa kandungan asam amino: glutamat total dan glutamat bebas, valin total dan valin bebas, prolin total, serta alanin bebas mengalami peningkatan, sementara asam amino selainnya mengalami penurunan. Kandungan antinutrisi teobromin berada di bawah ambang batas deteksi oleh HPLC.Kata kunci: Rhizopus oryzae, Theobroma cacao, teobromin, fermentasi, asam amino

Abstract

Physical Characteristics of Non-Extruded Floating Fish Feed Produced through Rhizopus oryzae Fermentation ABSTRACTTo reduce the feed production cost of floating fish feed, an innovative method which is cheaper, easier, and simpler compared to the use of extruder machine is needed to provide buoyancy to fish feed. This research was conducted to determine the physical characteristics of floating fish feed prepared through fermention using Rhizopus oryzae on the mixed substrate consisting of commercial sinking fish feed, cassava bagasse, and Lemna minor. The resulting fermented feed was subsequently compared with commercial sinking fish feed in terms of its water stability, absorption, and floatability. The water stability curves of both feed types showed a similar pattern for 20 hours. Both decreased dramatically in the first 0.5 hours, from 100% to 81% for the fermented feed, and to 83% for the commercial feed. The shape of a 40-minute water absorption curve for both feed types was very similar. The fermented feed absorbed water nearly 4 times its initial dry weight, while commercial feed up to about twice as much as its initial dry weight. For three hours, both fermented and commercial floating pellets stayed afloat 100% without sinking. Thus, fermentation using R. oryzae deserves further development as an alternative way to provide buoyancy to fish feed.Keywords: floatability, floating feed, sinking feed, water absorption, water stability ABSTRAKUntuk menekan biaya produksi pakan ikan apung, diperlukan inovasi pengapungan pakan ikan yang lebih murah, mudah, dan sederhana dibandingkan dengan penggunaan mesin ekstruder. Penelitian ini dilakukan untuk mengetahui karakteristik fisik pakan apung hasil fermentasi menggunakan kapang Rhizopus oryzae pada substrat campuran pakan tenggelam komersial, onggok, dan Lemna minor. Pakan fermentasi ini lalu dibandingkan dengan pakan apung komersial dalam hal stabilitas dalam air, absorbsi air, dan daya apung. Stabilitas dalam air kedua jenis pakan memiliki pola yang mirip selama 20 jam. Stabilitas keduanya menurun drastis pada 0,5 jam pertama, dari 100% ke 81% untuk pakan apung fermentasi, dan ke 83% untuk pakan apung komersial. Bentuk kurva absorpsi air selama 40 menit untuk kedua jenis pakan sangatlah mirip. Pakan apung fermentasi menyerap air hampir 4 kali bobot kering awalnya, sedangkan pakan apung komersial hingga sekitar 2 kali bobot kering awalnya. Selama tiga jam, baik pakan apung fermentasi maupun komersial mengapung 100% tanpa tenggelam. Jadi, fermentasi menggunakan R. oryzae layak dikembangkan lebih lanjut sebagai cara pengapungan alternatif pakan ikan.Kata Kunci: daya apung, daya serap air, stabilitas dalam air, pakan apung, pakan tenggelam

Abstract

Cocoa black pod rot is caused by pathogenic fungi, Phytophtora palmivora, which decrease the cocoa production up to 90%. The use of biological control agents, Trichoderma spp., is one of the promising P. palmivora controllers since it is low-cost, easily found and safe for the environment. The aims of this research were to understand the compatibility, antagonistic and effectiveness of biofungicide containing active ingredient of Trichoderma spp. against P. palmivora in vitro and to test the effective concentration of biofungicide containing active ingredient of T. asperellum to control P. palmivora in vitro and in vivo. T. asperellum, T. harzianum, and T. viride were grown together on PDA medium to test their compatibility. Antagonistic and effectiveness test of Trichoderma spp. against P. palmivora were tested using the in vitro dual culture method. The effectiveness of T. asperellum biofungicide was tested in vivo on cocoa pot. Compatibility test showed that all three species were compatible and the best effectiveness showed by the combination of T. asperellum and T. viride. The result also showed that T. asperellum biofungicide had an ability to inhibit P. palmivora.   Keywords: Trichoderma spp., effectivity, compatibility, antagonistic, biofungicide  ABSTRAKPenyakit busuk buah kakao disebabkan cendawan patogen Phytophtora palmivora, yang dapat menurunkan produksi kakao sampai 90%. Penggunaan agensia pengendali hayati (APH), Trichoderma spp., merupakan salah satu pengendalian P. palmivora yang menjanjikan karena murah, mudah didapat dan aman terhadap lingkungan. Penelitian ini bertujuan mengetahui kesesuaian, antagonistik, dan efektivitas biofungisida berbahan aktif Trichoderma spp. secara in vitro. Di samping itu juga bertujuan mengetahui konsentrasi efektif biofungisida berbahan aktif T. asperellum untuk mengendalikan P. palmivora secara in vitro dan in vivo. T. asperellum, T. harzianum, dan T. viride, ditumbuhkan bersama pada media PDA untuk mengetahui kesesuaian antarspesies. Antagonistik dan efektivitas Trichoderma spp. terhadap P. palmivora secara in vitro diuji menggunakan metode dual culture. Biofungisida berbahan aktif T. asperellum diuji efektivitasnya secara in vivo pada buah kakao. Hasil uji kesesuaian menunjukkan bahwa ketiga spesies yang diuji berkesesuaian dan efektifitas terbaik ditunjukkan pada kombinasi T. asperellum dan T. viride. Hasil penelitian juga menunjukkan bahwa biofungisida berbahan aktif T. asperellum dengan konsentrasi tertinggi terbukti dapat menghambat pertumbuhan P. palmivora.Kata kunci: Trichoderma spp., efektivitas, kesesuaian, antagonis, biofungisida
