Abstract

Information related to the prevalence and intensity of para-sites in an aquatic environment is crucial as preventive and responsive efforts to manage aquatic resources, especially for fish farming pur-poses. Up to now, information related to the prevalence and intensity of ectoparasites on catfish from aquaculture ponds in Aceh Besar Dis-trict is rare. This study aims to investigate the prevalence, intensity, and dominance of ectoparasites on catfish from aquaculture ponds in Aceh Besar district. The total of 200 catfish from four research stations at Sibreh, Kareung, Lambaro, and Limpok was observed in this study. The ectoparasites observation was focused on the skin, fins, and gill of the catfish. The observation parameter consists of prevalence, in-tensity, domination, length-weight relationship, and condition factor. Identification result showed that there were four types of ectoparasites infect the catfish in the aquaculture ponds in Aceh Besar District i.e Trichodina sp., Gyrodactylus sp., Dactylogyrus sp. and Ichthyopthyri-us multifiliis. Station 3 (Lambaro) had the highest prevalence and in-tensity, which were 64% and 5.87 ectoparasites/fish, respectively. In-fected catfish tended to have a lower weight growth than healthy catfish.

Abstract

Fruits are one of the sources of nutrition needed for health. Fruit quality is generally assessed by physical and chemical properties. Measurement of fruit internal quality is usually done by destructive techniques. Ultraviolet, visible and near-infrared (UV-Vis-NIR) spec-troscopy is a non-destructive technique to measure fruit quality. This technique can rapidly measure the fruit quality, the measured fruit still remains intact, and can be marketed. Besides, UV-Vis-NIR spectrosco-py can also be used to classify fruits. The study aimed to classify var-ious types of fruits using UV-Vis-NIR spectroscopy with wavelengths of 300-1041 nm and Principal Component Analysis (PCA). First de-rivative savitzky-golay with 9 smoothing points (dg1) and multiplica-tive scatter correction (MSC) were applied to correct the spectra. The results showed that the use of uv-vis-nir spectroscopy and PCA com-bined with spectra pre-treatment of the MSC method were able to clas-sify various types of fruits with 100% success rate in all fruit samples including sapodilla, ridge gourd, mango, guava, apple and zucchini.

Abstract

A commercial saponin as biosurfactant can reduce the surface tension of water and increase of hydrocarbon degradation. However, this saponin can be toxic to some hydrocarbonoclastic bac-teria. This study aimed to obtain bacterial isolates that were tolerant and incapable to degrade saponin, and to identify them based on 16S rDNA sequence. Bacteria were isolated from petroleum contaminated soil in Wonocolo Village, Bojonegoro Regency, East Java, Indonesia. The soil samples were acclimated using Bushnell-Haas (BH) broth with 0.5% crude oil at room temperature for 3 weeks. The culture was spread onto BH agar incubated at 30°C for 7 days. The first screened, isolates were grown in nutrient broth with addition of sap-onin 0%, 8%, and 12% (v/v) then incubated at 30°C for three days. The bacterial cell density was measured using a spectrophotometer. Second screened, the isolates were grown on BH broth with addition of 0.5% saponin as a sole carbon source, and their cell densities were measured. The selected isolates were identified based on 16S rDNA sequences. Among 34 bacterial isolates, nine isolates were tol-erant to 12% saponin. Three bacterial isolates IHT1.3, IHT1.5, and IHT3.24 tolerant to high concentration of saponin and did not use this substance as growth nutrition. The IHT1.3, IHT1.5, and IHT3.24 isolates were identified as Ochrobactrum pseudogrignonense (99% similarity), Pseudomonas mendocina (99%), and Ochrobactrum pi-tuitosum; (97%), respectively. Those three selected isolates are good candidates as hydrocarbon-degrading bacteria to bioremediation of soil contaminated crude oil. However, the combined activity of bacteria and saponin to degrade hydrocarbon needs further study.

Abstract

 An α-L-Rhamnosidase released by Aspergillus niger during solid-state fermentation (SSF) using coffee pulp (CP) wastes media has been investigated. The activity of α-L-Rhamnosidase based on reducing sugar production against 2% CP alkali extract substrate in 50 mM acetate buffer pH 5. The maximum activity of α-L-Rham-nosidase was obtained in sixth-day SSF with reducing sugar pro-duction of 13 μg/mL. The enzyme is actively hydrolyzed 0.1% p-ni-trophenyl-α-L-rhamnopyranoside (PNP-Rha) to 95% from initial concentration. Purification using DEAE-Toyopearl 650M increased hydrolysis activity ten times against the substrate, reaching 134 μg/mL of reducing sugar. Optimum enzyme activity at pH 4.5 and 50°C, while stable at pH and temperature in a pH range of 3.5-7 and below 50°C.

Abstract

  Cow urine contains urea as nitrogen source, therefore it can be expected to isolate the beneficial bacteria for plants, for example indole-3-acetic acid (IAA) or auxin producing bacteria. The objective of research was to obtain IAA producing bacteria from cow urine, to characterize bacterial isolate, and to measure its ability to stimulate the growth of green bean seedlings (Vigna radiata). The methods used in this study were collecting urine from cow cattle, obtaining IAA-pro-ducing bacteria from urine, measuring IAA using Salkowski method, and applying selected bacterial supernatants on green bean seedling plants. The number of IAA producing bacteria that was successfully purified was 18 isolates. There are five isolates, namely US 5, BS1, BS 2, BS 4 and BS 5 which have the ability to solubelize phosphate on Pikovskaya agar. The five isolates were also able to fix free nitro-gen on N Free media and did not show hypersensitivity on tobacco leaves. The results of the growth of isolates in blood agar showed positive for US 5 and BS 2 as beta hemolysin producers. Further-more, isolate BS 4 was chosen to produce exogenous IAA quantita-tively. Isolate BS 4 produced IAA 6.364 ppm at the 45 h incubation at stationary phase. The use of BS 4 supernatant on green bean seed-lings showed an effect on plant height and lateral root length better than control (without treatment) on 6 days after planting. Morpho-logical characteristic of isolate BS 4 was rod shape, Gram positive, endospore producing, aerobic, and had similarity with genus Bacillus

Abstract

Lemon peel and leaf  as well as lower grade of lemon contain a considerable amount of flavonoid which can be extracted to produce a valuable product. The presence of pectin in lemon hinders the extraction of flavonoid compound. This research was conducted to examine the effect of pectin biodegradation of lemon leaf, peel, and lower grade fruit using solid-state fermentation towards the yield of pectin, crude flavonoid, and total flavonoid content. Fermentation was carried out with the help of Aspergillus niger at 30℃ for 3, 5, 7, and 9 days of cultivation time. Pectin was extracted from the fermentation broth using citric acid. Crude flavonoid was extracted using maceration and stirred about 500 rpm at 50℃ for 40 minutes. Total flavonoid content was analyzed using a spectrophotometer. The optimum cultivation time of Aspergillus niger with leaf and peel was 9 days with a pectin yield from lemon leaf was 0.43% dry weight and the pectin yield from lemon peel was 0.04% dry weight. As for the lower grade fruit, a cultivation time for 7 days showed an optimal result with a pectin yield of 0.08% dry weight. The total flavonoid content in the leaf increased up to 94.3% (1.06 mg of the quercetin equivalents per g of substrate) whereas the total flavonoid content in the peel and lower grade fruit increased up to 42% (0.12 mg of the quercetin equivalents per g of substrate) and 48% (0.021 mg of the quercetin equivalents per g of substrate), respectively. These results highlighted the potential valorization of Citrus limon L. as an alternative source for producing flavonoid in food and pharmaceutical industry.

Abstract

The natural occurrence of solar eclipses can cause various impact on bird species. The purpose of this study was to examine the effect of the partial solar eclipses on changes in behavior of three species of Ardeidae in the Rancabayawak, Gedebage, Bandung, West Java. The research method used was qualitative which direct observation sampling technique was applied.  The results of study showed that three bird species of Ardeidae namely Javan Pond-Heron (Ardeola speciosa Horsfield 1821), Buffalo Egret (Bubulcus ibis Linnaeus 1758, and small egrets (Egretta garzetta Linnaeus 1766) with total population 566 individuals were recorded in the resting and breeding place of bamboo trees and surrounding area of Rancabayawak during the partial solar eclipse.The behavior of these tree bird species, including sound of chicks, sunbathing, preening the feather, and flying of individual adults went out the resting and breeding place of bamboo trees observed tend to be normal before occurring the partial solar eclipse. During the partial solar eclipse, however, the sound of the chick stopped for a second. Some individual adult birds stopped involve in preening their feather. While, some adult individual birds were flying go back to the nest of bamboo trees that may be assumed it has already late afternoon due to dark of sun shine. Implication of this research have been considered to be very important for contributing the scientific knowledge on bird behavior changes caused by natural phenomena of the partial solar eclipse.

Abstract

Red dragon fruit (Hylocereus polyrhizus Britton & Rose) is one type of cactus plant which is very potential as an ornamental plant and edible fruit. Flower is organ that play an important role in the process of breeding plants generatively. This reasearch aimed to study the anatomical structure of sepals and petals of red dragon fruit plants during flower development. The research stages included: sepals and petals sampling that held at various stages of flowering ; morphological observation (measurement length of sepals and petals); anatomical slides of sepals and petals cross section using the embedding method, anatomical observation and image capture of sepals and petals. The parameters observed were bud size, sepal length, petal length, sepal thickness, petal thickness, and tissue description composed. The results of this study indicated that buds have an increased development pattern. The increase in bud size is directly proportional to the stage of the bud. Sepal and petal are composed of epidermal tissue which form papillae, cryptophore stomata, secretory parenchyma space containing mucus, and tissues transport system is closed collateral.

Abstract

The growth of Pak Choy (Brassica rapa var. chinensis) using nutrients from catfish “Sangkuriang” (Clarias gariepinus) wastes was investigated in the household scale aquaponics systems by comparing the growth parameters value among the grow beds. The growth parameter of Pak Choy was fresh weight, leaf number, and leaf area. Meanwhile, catfish growth parameters were body weight and length, and feed conversion ratio (FCR). The experiments were arranged in a Completely Randomized Design (CRD) with 4 treatments and 3 replications. The data were analyzed using Analysis of Variance (Anova) followed by Duncan Multiple Range Test (DMRT) at 95% level of significant The result of the study showed that there were not significantly different in the growth parameter of Pak Choy among the grow beds in the vertical system. However, those in grow beds “Ä” were significantly higher than the rest of the grow beds in the horizontal aquaponics system. In general, all the growth parameter of Pak Choy in the vertical system was higher than in the horizontal one, although, it was not statistically significantly different. Nevertheless, the horizontal aquaponics system gave better results in the body weight and FCR of catfish.

Abstract

The process of forest regeneration can take place within a certain period of time. Sprouting is a form of strategy for in adapting to its environment for success in regenerating. The Research on  the diversity and diameter growth rate of sprouting-type tree in Bukit Pinang-pinang forest permanent plots, West Sumatra was carried out from October 2012 to December 2016 in the forest of Bukit Pinang-Pinang, West Sumatra and Herbarium Department of Biology, Universitas Andalas, Padang. This research was conducted using a survey method by census. The parameters analyzed were plant species composition, Morishita Index, Mann-Whitney test, Regression and Correlation test as well as RDGR (Relative Diameter of Growth Rate). The result of experiment was found 40 species of trees sprouting type in forest permanent plot Bukit Pinang-Pinang. The pattern of sprouting type trees distribution in the study locations was found to have a clustered, random and uniform pattern. Relative Diameter Growth Rate (RDGR) of 40 sprouting types of trees in the Bukit Pinang-Pinang forest permanent plots ranged from 0.003 cm/cm/year to 0.0288 cm/cm/year. Altitude did not affect  RDGR of an individual tree sprouting type for 31 years
