Abstract

Sianobakteri merupakan kelompok fitoplankton yang umum dijumpai di perairan tawar di seluruh dunia. Sianobakteri menghasilkan toksin mikrosistin yang dihasilkan oleh Microcystis aeruginosa yang menyebabkan kematian ikan. Penelitian ini bertujuan mengetahui potensi bakteri proteolitik yang berasal dari saluran pencernaan ikan nila GIFT dalam menghambat pertumbuhan M. aeruginosa BT-02. Isolat bakteri proteolitik asal saluran pencernaan ikan nila yang menghambat pertumbuhan M. aeruginosa BT-02, yaitu isolat i.e Aeromonas caviae NU-4 and Aeromonas sp. NU-8. Indeks penghambatan bakteri NU-4 (1,71) terhadap M. aeruginosa lebih besar daripada NU-8 (1,34). Mekanisme penghambatan belum diketahui. Aplikasi Microcystis aeruginosa BT-02 pada ikan mas tidak menyebabkan kematian ikan, tetapi menimbulkan beberapa perubahan histopatologi pada hati dan usus ikan mas.Kata kunci: Bakteri proteolitik, Aeromonas sp., Microcystis aeruginosa, ikan mas, tes toksisitas

Abstract

Utilization of soil bacteria as biocontrol agent is becoming popular due to its valuable and effective mechanisms to suppress plant pathogenic microbes. We have previously isolated Bacillus sp, designated as Bacillus sp CR64, which exhibited effective plant growth promoting and antifungal activities. In this study, CR64 was examined in inhibiting the growth of Rhizoctonia solani, the causing agent of root rot disease. Partial sequence analysis of 16S rRNA gene revealed that this isolate similar with Bacillus cereus (94%). Furthermore, a gene designated zmaR was detected by means of specific amplification of DNA fragment approximately 950 bp. This fragment was then cloned onto pCRII-TOPO (3.9 kb) and sequenced using DNA sequencer ABI PRISM 310. Sequence analysis revealed that it had highest homology with the ZmaR protein (89% identity; 90% similarity) of B. thuringiensis serovar kurstaki (AAF82729.2). Alignment analysis with other ZmaR sequences from other antibiotic-producing Bacilli exhibited an almost fully conserved region within ZmaR sequences.Key words : PGPR, Bacillus sp CR64, Zwitermicin A, Cloning, Antifungal.

Abstract

Some of the methanotrophic bacteria and N2O reducing bacteria have been proven to be able to support the plant growth and increase the productivity of paddy. However effect of the methanotrophic and N2O reducing bacteria application as a biofertilizer to indigenous N2O reducing bacteria is still not well known yet. The aim of this study was to analyze the diversity of N2O reducing bacteria in lowland paddy soil based on a nosZ gene. Soil samples were taken from lowland paddy soils in Pelabuhan Ratu Sukabumi, West Java, Indonesia. There were two treatments for the paddy field soil, ie. biofertilizer-treated field 20% fertilizer (50 kg/ha) with the addition of biofertilizer and 100% fertilizer. PCR amplification of nosZ gene was successfully conducted using nosZF and nosZR primer pair. Denaturing Gradient Gel Electrophoresis (DGGE) process was conducted at 150 V for 5.5h. There were three differences nosZ bands were sequenced. The phylogenetic analysis showed that they were close to uncultured bacteria. Microbial diversity in the biofertilizer-treated field was higher than that of in the 100% fertilizer-treated field. The biofertilizer treatment has higher in microbial diversity than that of applied non-biofertilizer paddy fields. This research might have impact in the application of biofertilizers due to the emission of N2O as a green house gas from paddy fields farming activity. The biofertilizer has great potential application in sustainable environmental friendly agriculture systems.

Abstract

Pyrene degradation and biosurfactant activity by a new strain identified as Gordonia cholesterolivorans AMP 10 were studied. The strain grew well and produced effective biosurfactants in the presence of glucose, sucrose, and crude oil. The biosurfactants production was detected by the decreased surface tension of the medium and emulsification activity. Analysis of microbial growth parameters showed that AMP10 grew best at 50gmL-1pyrene concentration, leading to 96% degradation of pyrene within 7days. The result of nested PCR analysis revealed that this isolate possessed the nahAc gene which encodes dioxygenase enzyme for initial degradation of Polycyclic Aromatic Hydrocarbon (PAH). Observation of both tensio-active and emulsifying activities indicated that biosurfactants which produced by AMP 10 when grown on glucose could lower the surface tension of medium from 71.3 mN/m to 24.7 mN/m and formed a stable emulsion in used lubricant oil with an emulsificationindex (E24) of 74%. According to the results, it is suggested that the bacterial isolates G. cholesterolivorans AMP10 are suitable candidates for bioremediation of PAH-contaminated environments.How to CiteKurniati, T. Abstract

Based on the raw materials, inasua consists of two types namely inasua with sap and inasua without sap. Research of inasua with sap has never been done and considered as the novelty of this research. The sensory characteristics and shelf life of two types of inasua were different. The research aims to analyze the microbiological and physicochemical characteristics of two types of inasua during fermentation. The microbiological analyzes include the total number of bacteria and lactic acid bacteria, while physicochemical analyzes include temperature, pH, water activity, proximate analysis, salt, alcohol, histamine, amino acids and fatty acids contents. The total number of bacteria and lactic acid bacteria has decreased during fermentation. At the end of the fermentation the total number of bacteria and lactic acid bacteria inasua with sap were 3.2x107 CFU/g and 3.0x107 CFU/g, while inasua without sap were 5.4x105CFU/g and 3.5x105 CFU/g, respectively. The moisture, protein, alcohol contents and water activity decreased, otherwise the salt, fat, ash, amino acids, and fatty acids contents increased during fermentation. Generally, microbiological and physicochemical characteristics of inasua with sap was better than inasua without sap. The results of this research to improve the quality of this fermentation product in the future.

Abstract

Antioxidant Activity of Endophytic Fungi Cb.Gm.B3 Extract from Cinnamon (Cinnamomum burmanni) TwigsABSTRACTThere are many degenerative diseases that are caused by a free radical effect. Cinnamon (Cinnamomum burmanni) contains cinnamaldehyde compounds that have activity as a powerful antioxidant and fight free radicals. Endophytic fungi can be found in cinnamon plants living symbiotically. Endophytic fungi produce a variety of bioactive metabolites including antioxidants. This research was conducted to isolate endophytic fungi from C. burmanni plant which is active as antioxidant. Endophytic fungi isolation was carried out using surface sterilization method and cultivated in PDA media. Antioxidant activity test was performed using free radical 2.2-diphenyl-1-picrylhydrazyl (DPPH) method. Selected isolates were then identified molecularly to determine their species. A total of nine fungi were isolated from cinnamon twigs. The result showed that the highest antioxidant activity was obtained from Cb.Gm.B3 with IC50 of 13.219 ± 0.755 µg/mL. The selected isolate Cb.Gm.B3 taxonomically has a high similarity with Neofusicoccum parvum isolate PEL23 (Accession no: KY053054.1).Keywords: antioxidant, Cinnamomum burmanni, 2.2-diphenyl-1-picrylhydrazyl, endophytic fungi, Neofusicoccum parvum ABSTRAKKayu manis (Cinnamomum burmanni) mengandung senyawa sinamaldehid yang memiliki aktivitas sebagai antioksidan kuat dan dapat menangkal radikal bebas. Dalam tanaman kayu manis terdapat kapang endofit yang hidup bersimbiosis. Kapang endofit dapat menghasilkan berbagai senyawa metabolit bioaktif termasuk antioksidan. Penelitian ini dilakukan untuk mengisolasi kapang endofit dari tanaman C. burmanni yang aktif sebagai antioksidan. Isolasi kapang endofit dilakukan menggunakan metode sterilisasi permukaan dan ditanam pada media PDA. Pengujian aktivitas antioksidan dilakukan menggunakan metode peredaman radikal bebas dengan reagen 2.2-difenil-1-pikrilhidrazil (DPPH). Isolat terpilih diidentifikasi secara molekuler untuk menentukan spesiesnya. Sebanyak 9 isolat kapang berhasil diisolasi dari jaringan ranting tanaman kayu manis. Aktivitas antioksidan tertinggi (IC50) didapatkan dari isolat Cb.Gm.B3 sebesar 13,219 ± 0,755 µg/mL. Abstract

Lipase Production by Mutant Fungal Isolates for Transesterification ABSTRACTLipase is used amongst others in biodiesel production, namely in the transesterification reaction. Kernel B (KB) was a fungus isolated from the waste of palm kernel and seed. The fungus produced lipase that catalysed the transesterification reaction with a lower activity compared to that of AK Amano commercial lipase. The purpose of this study was to obtain mutant fungi with higher transesterification activities than the wild type (KB). The mutation process was carried out using ultraviolet (UV) light, ethyl methane sulfonate (EMS), and N-methyl-N’-nitro-N-nitrosoguanidine (NMNG) on KB fungus. The mutations using UV light produced 11 isolates, of which isolate m4.1KB1 produced a higher transesterification activity (0.172 U·mg-1) compared to the wild type. Mutant m5.7KB, which was generated from mutant m4.1KB1 treated using EMS, had its transesterification activity decreased to only 0.051 U·mg-1. Mutant m6.0,3KB2, which was resulted through NMNG treatment, experienced an increase in transesterification activity which was 91.2% higher than that of KB.Keywords: ethyl methane sulfonate, lipase, mutant fungi, N-methyl-N’-nitro-N-nitrosoguanidine, ultraviolet ABSTRAKLipase dimanfaatkan salah satunya dalam produksi biodiesel, yaitu dalam reaksi transesterifikasi. Kernel B (KB) merupakan kapang yang diisolasi dari limbah inti dan biji kelapa sawit, yang menghasilkan lipase sebagai katalis dalam reaksi transesterifikasi. Namun aktivitas transesterifikasi yang dihasilkan oleh lipase dari KB lebih rendah dibandingkan dengan lipase komersial AK Amano. Tujuan penelitian ini adalah mendapatkan mutan kapang dengan aktivitas transesterifikasi yang lebih tinggi dibandingkan tipe liarnya (KB). Proses mutasi dilakukan dengan menggunakan sinar ultraviolet (UV), ethyl methane sulfonate (EMS), dan N-methyl-N’-nitro-N-nitrosoguanidine (NMNG) terhadap kapang KB. Mutasi KB dengan menggunakan sinar UV menghasilkan 11 isolat, dimana isolat dengan kode m4.1KB1 menghasilkan aktivitas transesterifikasi yang lebih tinggi dibandingkan tipe liar, yaitu 0,172 U·mg-1. Mutan m5.7KB, yang dihasilkan dari mutan m4.1KB1 dengan perlakuan EMS, mengalami penurunan aktivitas transesterifikasi hingga hanya sebesar 0,051 U·mg-1. Abstract

Isolate RA05 has the highest mannanolytic index and mannanase activity which isolated from copra soil waste from Pasaman, West Sumatra. The best growth condition that produces best mannanase activity of isolate RA05 was achieved from 500 ml flask containing 100 ml medium with 100 rpm agitation. Isolate RA05 showed its mannanase activity in medium containing Locust Bean Gum and coconut meal but not in medium containing kolang kaling. This mannanase had the highest activity on medium containing 2% of coconut meal with optimum condition temperatur 800C and pH 2.5. Adding of 5 mM MnCl2 on the crude enzym increased the activity near 300%. Other kation (Ca2+, Zn2+, Cu2+, Mg2+, Fe2+ dan Co2+) did not display great effect on the activity.

Abstract

Alkalotolerant bacteria identified as Bacillus firmus KH.9.4 was isolated from tapioca liquid waste from Kedung Halang, Bogor. The isolate showed optimum a-amylase activity after 15 hours of cultivation on liquid media. Optimum enzyme activity occurred at pH 6.0 and temperature 800C. Upon heat treatment at pH 6.0 and 800C the enzyme was still 100% active after 6 hours incubation without substrate. The enzyme activity was enhanced by supplement 10 mM MgSO4.7H2O. The presence of 1, 5, and 10 mM ethylenediaminetetraacetic acid demonstrated the inhibitory effect to this enzyme. Zymogram analysis revealed one translucent zone corresponding to soluble starch activities after 60 minutes of incubation at 400C in the 0.05 M phoshate buffer pH 7.0. Whereas SDS-PAGE analysis showed that there are 5 bands of proteins.

Abstract

A total of 6 fungi isolates of growing at pH 9 and 2 isolates on pH 10 with amylolytic indexes 0f 0.07-1.42 have been isolated from cassava starch liquid waste. Two isolates having the highest amylolytic index were identified as Aspergillus sydowii K10 (1.42) and Aspergillus versicolor L30 (1.4). Both A. sydowii K10 and A. versicolor L30 were described as alcalotolerant for being able to grow with range pH 5-10. The optimal -amylases production of A. sydowii K10 and A. versicolor L30 was obtained after 4 and 3 days of incubation at 300C. The optimum of -amylase activity from A. sydowii K10 was at 400C and 700C, and pH 6; while those from A. versicolor L30 was at 500C and pH 6 respectively. Both A. sydowii K10 and A. versicolor L30 could produce glucoamylase. The optimum of glucoamylase activity from A. sydowii K10 was at 400C and pH 5, while those from A. versicolor L30 was at 500C and pH 5 respectively.
