Abstract

Tea is well known as favourite healthy drink for almost all people over the world. Tea propagation using conventional and modern ways are now developing rapidly. However, information regarding the protein profile of tissue culture of tea plant has not been revealed yet. This study aimed to determine the difference of protein profile of tea’s tissue culture using SDS-PAGE. This study was conducted using embryonic axes of TRI2025 tea clone cultured on MS media supplemented with 2,4-D for inducing somatic embryogenesis and globular-like structure (GLS) regeneration, and MS media supplemented with BAP for inducing shoot via organogenesis. The results revealed that proteins in the size of 37.69; 54.89; 60.77; 71.35; 87.34; and 92.99 KDa might be involved at somatic embryogenesis, and about 38.69 KDa, 69.27 KDa, and 55.76 KDa respectively for GLS, initiation of shoot, and initiation of GLS derived leaf. Predicted key protein for leaf initiation both directly or through GLS was about 31-33 KDa, while for callusing were about 27.56 and 52.73 KDa, and constitutive protein was about 22.75 KDa. This study provides the first report of protein profile of tea’s tissue culture. The information obtained can be beneficial as a marker for explant for somatic embryogenesis, GLS, or organogenesis pathway and as a scientific information for further biotechnology development.

Abstract

Actinomycetes SAE4034 isolates was isolated from Rhizophora apiculata rhizosphere mud showed some antibacterial properties. The antibacterial ability of this isolate has not been tested on antibiotic resistant bacterial pathogens. However, there was no research has been reported regarding actinomycetes from Segara Anakan mangrove area resulting compounds inhibit the growth of antibiotics-resistant bacteria. Therefore, it is important to investigate its capability against antibiotics resistant bacteria or multi drug resistant bacteria (MDR bacteria). The research aimed to know the ability of actinomycetes SAE4034 in inhibit MDR bacteria and to identify the species profiles. The research methods included isolate characterization involving morphology, physiology/enzymatic and molecular properties. MDR bacterial inhibition assay, antibacterial compound extraction and antibacterial compound test using thin layer chromatography (TLC) method, observation of morphological and biochemical properties, DNA isolation, amplification and analysis of 16SrRNA sequence, and phylogeny tree analysis. The methods of this study included MDR anti-bacterial assay and antibacterial compound test. Subsequent step was isolate characterization including observation of morphological and physiological / enzymatic properties, and 16S rRNA gene sequence. The results showed that culture extract was able to inhibit the growth of MDR bacteria i.e. Escherichia coli, Staphylococcus aureus, Klebsiella pneumoniae, Pseudomonas aeruginosa, Enterococcus sp., but no inhibition to Enterobacter cloacae. The bioactive compound showed 4 spots with Rf values of 0.36; 0.45; 0.54; and 0.6. Based on morphology, physiology / enzymatic and 16S rRNA gene sequences characteristics, actinomycetes SAE4034 isolate is Streptomyces sp. This research showed new Streptomyces strain that serves as a source of MDR antibacterial compounds and useful in development of antibiotic for combating infectious diseases caused by MDR bacteria

Abstract

The use of synthetic dyes has an impact on the possibility of disposing such dyes into the environment. Fungal decolorization is one promising approach due to its capability to degrade dyes, thus, exploring fungi that can be applied in dye decolorization is essential. We investigated our potential strain of Trametes hirsuta to decolorize Remazol brilliant blue R (RBBR). The enzyme activity of laccase in various conditions was observed using Syringaldazine as a substrate, while fungal immobilization was conducted using calcium alginate as a solid support. The results indicated that CuSO4 was the best inducer for the decolorization process. The fungus was able to perform 79.5% of RBBR decolorization for 48 hours in the presence of CuSO4. Laccase was the prominent detected ligninolytic enzyme when decolorization was performed. The immobilized cells were able to decolorize 85% RBBR under 0.8 mM CuSO4 andused for 3 cycles of decolorization. This study reveals the potential of fungal usage in the form of the immobilized and free cell to overcome the persistence of dye pollutants problem, as it is considered an effective, economic and eco-friendly approach for RBBR dye decolorization. These strategies can be suggested to encourage ecologically sustainable development for bioremediation.

Abstract

Colorectal carcinogenesis induced by cigarette smoke requires at least 30-40 years. This long time duration causes an animal research conducted becomes relevant. This research was carried out to observe colorectal carcinogenesis due to cigarette smoke exposure in Wistar Rat. The observations focused on changes in epithelial morphology and expression of APC, MSH2 and MLH1. Twenty male Wistar rats inbreed strain were randomly allocated into control group and experimental group exposure to cigarettes smoke for 14 weeks and 28 weeks sequentially.  Colorectal epithelial morphology was assessed on the histopathology examination, whereas the expression of APC, MSH2 and MLH1 was assessed on aspect of immunohistochemistry. The comparative analysis between the two groups was performed using non-parametric Mann-Whitney U test. Histology of colorectal epithelium showed pattern of colitis associated cancer that was significant both in 14 weeks and 28 weeks of treatment.  This research indicated negative expression of APC, MSH2 and MLH1 in the colorectal cancer that were significant at 28 weeks of exposure. This research implies that chronic exposure to cigarette smoke can induce colitis associated colorectal cancer through decreased expression of APC, MSH2 and MLH1.

Abstract

Gas buang yang dihasilkan oleh kendaraan bermotor pengguna bahan bakar bensin bertimbal mengemisikan Pb ke dalam lingkungan dan berpotensi terserap ke dalam jaringan daun tanaman peneduh jalan. Penelitian bertujuan untuk menguji kandungan timbal (Pb) dalam daun tanaman peneduh di jalan protokol Kota Semarang. Sampel penelitian adalah daun angsana (Pterocarpus indicus Willd), glodokan (Polyalthia longifolia Bent Hook. F) dan mahoni (Swietenia mahagoni (L.) Jacq.) yang dominan digunakan sebagai peneduh di lima jalan protokol Kota Semarang yaitu Jl. Kalibanteng, Jl. Pemuda, Jl. Brigjen Katamso, Jl. Kaligawe dan Jl. Setyabudi. Kandungan Pb dalam daun tanaman angsana, glodokan dan mahoni diuji menggunakan metode AAS (Atomic Absorption Spectrophotometery) di Laboratorium Balai Besar Teknologi Pencegahan Pencemaran Industri (BBTPPI) Semarang. Hasil penelitian ditemukan kandungan Pb dalam daun tanaman peneduh di jalan protokol Kota Semarang. Kandungan Pb dalam daun tertinggi 0,05 ppm/g daun basah terdapat pada tanaman angsana dan terendah sebesar 0,01 ppm/g terdapat pada tanaman glodokan dan mahoni. Dari hasil penelitian disimpulkan bahwa kandungan Pb dalam daun tanaman peneduh di jalan protokol Kota Semarang berkisar 0,01-0,05 ppm/g daun basah dan tergolong rendah dibawah kadar normal Pb dalam tanaman yaitu 0,5-3,0 ppm.Exhaust gases produced by motor vehicle users leaded gasoline emits Pb into the environment and potentially absorbed into the leaf tissue of plants along the roadside. The study aimed to examine the content of lead (Pb) in the shade plant leaves in the Semarang City road protocol. Samples were angsana leaves (Pterocarpus indicus Willd), glodokan (Polyalthia longifolia Bent Hook. F) and mahogany (Swietenia mahagoni (L.) Jacq.). They were predominantly used as a shade in five main streets of Semarang is Kalibanteng St, Youth St, Brig Katamso St, Kaligawe St and Setyabudi St. The content of Pb in the Angsana, glodokan and mahogany leaves were tested using AAS (Atomic Absorption Spectrophotometery) method in Great Hall of The Pollution Prevention Technology Laboratory Semarang. The results showed that the content of Pb was found in the shade plant leaves in the Semarang road protocol. The highest Pb content in leaves was 0.05 ppm/g wet leaves at Angsana plants and the lowest was 0.01 ppm/g contained in mahogany and glodokan plants. Based on the results of this study, it concluded that the content of Pb in the shade plant leaves in the Semarang protocol ranged from 0.01 to 0.05 ppm/g wet leaves and relatively low under normal levels of Pb in plants is 0.5 to 3.0 ppm.

Abstract

Metarhizium anisopliae is a parasitic fungus on insects, and thus called entomopathogenic fungus. This fungus is used as a biological control agent for insect pests. Fungal propagation can be done using a variety of media. The purpose of this study was to analyze the growth of M. anisopliae on four types of media, with conidial density and viability as the growth parameters. This research was conducted at the Laboratory of Microbiology, Universitas Negeri Semarang. This study was an experimental research used a Completely Randomized Design (CRD) with one factor and four treatments: Control (PDA/Potato Dextrose Agar medium), Treatment I (ELSA/Extract Larvae Sucrose Agar medium), Treatment II (CWSA/Coconut Water Sucrose Agar medium), Treatment III (CWELSA/Coconut Water and Extract Larvae Sucrose Agar medium). The and results showed that there was an effect of growth media on the density and viability of M. anisopliae conidia. CWELSA media had the highest conidial density (2.91 x 108 cfu/mL) and viability (97.17%). CWSA media had  2.82 x 108 cfu/mL and 95.33%. PDA media had 2.25 x 108 cfu/mL and 92.83%. ELSA media had 1.64 x 108 cfu/mL and 90.83%. The high conidial density and viability of M. anisopliae is CWELSA  medium. This study is as an alternative growth medium to improve the quality of M. anisopliae propagation.

Abstract

In the plant breeding program, selection is one process which aimed to obtain some varieties with superior traits. Within this process, one of criteria that must be consider is the wide genetic diversity of the plant which was selected, and how these traits are inherited. The purpose of this research was to determine (1) the influence of cultivar differences and the colchicine concentration towards the growth and production of local corn cultivar from Kisar Island Maluku, (2) to determine the heritability of growth and production variables after induced with colchicine. Completely randomized block design  was used in this research with factorial design that consist of two factors i.e corn cultivar and colchicine concentration. The research procedure starts from field and seed preparation, immersion of seed into the colchicine solution, transplantation of seed into the field, plant maintenance, and  harvesting. The variable measured including growth and production variables of corn. The analysis of variance showed a significant effect of cultivar differences on all growth and production variables (p value 0.05), while colchicine concentration only showed a significant effect toward the plant height and number of seed per ear.  The heritability analysis result showed that all variables observed were in high category. The different corn cultivars have a significant effect on all the growth and production, while the colchicine concentration only affect the plant height and number of seed per ear, and the heritability were in high category.  This research provides useful information to farmers how to use colchicine to trigger plant growth and production. It can be used as an information in selecting corn cultivars with high yield potential.

Abstract

Physiological stress for a long time can lead to impaired kidney organ function. Snakehead fish meat are known to contain important nutrients such as albumin, glutamine, glycine, cysteine, and various minerals that can improve kidney function. This study was conducted to examine and analyze the effect of snakehead fish supplementation in feed towards body weight, kidney weight, diameter and number of glomerulus in Wistar rats with physiological stress. This study used 20 male Wistar rats divided into 5 groups consisted of 1 control group (P0) and 4 treatment groups (P1: 5%, P2: 10%, P3: 15%, P4: 20%). The treatments were performed with 4 times repetition. This study used Completely Randomized Design (CRD). The variables measured were body weight, kidney weight, diameter and number of renal glomerulus. The data obtained were analyzed using Analysis of Variance (ANOVA) at 5% significance level and continued with Duncan test with 5% significance level. The results of this study were: the supplementation of snakehead fish with 20% of concentration can increase the body weight; concentration 10; 15; and 20% can increase the weight of the kidney; concentrations of 5%, 10%, 15%, and 20% can increase the glomerular diameter in Wistar rats with physiological stress. The supplementation of snakehead fish with concentration of 5% to 20% did not affect the number of glomerulus of Wistar rats with physiological stress. Results of this study is useful as information for people who study the mechanism of kidney repair due to physiological stress.

Abstract

Benalu mangga mengandung senyawa kuersetin yang bertindak sebagai hepatoprotektor. Kodein merupakan jenis alkaloid dan banyak digunakan sebagai obat. Oversdosis kodein dapat menyebabkan kerusakan hepar. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak benalu mangga terhadap perubahan histologis hepar tikus yang diinduksi kodein. Sebanyak 20 ekor tikus wistar jantan dibagi menjadi 5 kelompok (K0, K, KB1, KB2, dan KB3). Kelompok K0 hanya diberi minum dan pakan standar, kelompok K diberi kodein 12mg/ekor selama 7 hari, selanjutnya hanya diberi pakan dan minum standar. Kelompok KB1 diberi kodein 12mg/ekor selama 7 hari dilanjutkan ekstrak benalu mangga 22mg/ekor sampai hari ke 21, kelompok KB2 diberi kodein 12mg/ekor selama 7 hari dilanjutkan ekstrak benalu mangga 44mg/ekor sampai hari ke 21, dan kelompok KB3 diberi kodein 12mg/ekor selama 7 hari dilanjutkan ekstrak benalu mangga 88mg/ekor sampai hari ke 21.Hari ke-27 semua tikus dibedah, diambil heparnya dan dibuat preparat histologi dengan pengecatan HE.Hasil penelitian kelompok K0 menunjukkan perubahan histologis hepar bermakna (p0,05) antara K,KB1, KB2, dan KB3, sedangkan kelompok K tetapi tidak menunjukan perubahan bermakna (p0,05) antara K,KB1, KB2, dan KB3. Pada kelompok K kerusakan struktur hepar meliputi inflamasi perportal dan degenerasi sedangkan pada kelompok KB1, KB2, dan KB3 kerusakan meliputi inflamasi periportal,degenerasi, dan nekrosis.Mango’s mistletoes contain quercetin, a hepatoprotector. Codein is a type of alkaloids widely used as drug. Codein overdose can damage the liver. The objective of the study was know the effect of the administration of extract of mango’s mistletoes on the changes of liver histology of codein-induced rats. As many as 20 male wistar rats were divided into 5 groups (K0, K, KB1, KB2, and KB3). K0 group were given only standard diet and water daily. K group were given codeine at 12mg/rats for 7 days, and afterwards they were given only standard diet and water daily. KB1 group were given codeine at 12mg/rats for 7 days and then were given extract of mango’s mistletoes at 22 mg/rats until day 21, KB2 group were given codeine at 12mg/rats for 7 days and then were given extract of mango’s mistletoes at 44 mg/rats until day 21, and KB3 group were given codeine at 12mg/rats for 7 days and were given extract of mango’s mistletoes at  88 mg/rats until day 21. At day 27, all rats were killed, the livers were taken out for preparations with HE staining for histology examination. Research showed  that K0  group was significantly different (p 0.05) to K, KB1, KB2, and KB3 groups, whereas K group was not significantly different to KB1, KB2 and KB3 groups. Rats in K group showed the changes in histological appearance such as periportal inflammation and degeneration, whereas rats in KB1, KB2, KB3 groups showed similar histological appearance but with necrosis.

Abstract

The present study aims at expressing and partially purifying PtpB in active form. To achieve this, Mtb PtpB gene has been cloned into pET30a vector and overexpressed in Escherichia coli BL 21(DE3) under IPTG induction in the form of an inclusion body. Following resolubilization by urea and dialysis, the resulted PtpB has been shown to be active against para-Nitrophenyl phosphate.  It is concluded that the resulted PtpB has had been recovered from inclusion body to give the active form of the enzyme, and thus the success in overexpressing PtpB provides the required material to investigate the biochemical properties of the pathogen virulence factor further.
