Abstract

Salam leaves (Syzygium polyanthum (Wight) Walp.) contains fl avonoids, volatile oil and phenolic acid that can inhibiting platelet aggregation induced by ADP (adenosine diphospate), arachidonic acid, epinephrine, thrombin, collagen and aff ecting the activity enzyme ectonucleotidase so it can be used as anti-platelet drug. The purpose of this study was to investigated 70% ethanol extract of salam leaves have anti-platelet aggregation eff ect and increase of dose can also increase anti-platelet aggregation. This study was performed in 5 groups, each group consisted of 5 mice there were: groups of positive control, normal control, 70% ethanol extract leaves doses (150, 300, 450) mg/ kg, each group is given orally once daily during 8 days and measurements of parameters at day-0 and day-9th. The results of this study was 70% ethanol extract of leaves of salam leaves doses (150,300, 450) mg/kgBW showed the eff ects prolong bleeding time, coagulation time, enhance the decreased plasma absorbance after the addition of ADP. From the measurement parameter bleeding time, coagulation time and measurement decreased plasma absorbance after the addition of ADP that 70% ethanol extract of salam leaves doses 150, 300 and 450 mg/kgBW have anti-platelet aggregation eff ect and increase of dose was able to increase the eff ect anti-platelet aggregation.

Abstract

Isolat P301 adalah bakteri penghasil antibiotik yang diisolasi dari rizosfer tanaman padi. Tujuan dari penelitian ini adalah untuk mengetahui aktivitas antibakteri dari cairan kultur isolat P301 terhadap Staphylococcus aureus ATCC 25923 dan mengetahui profil optimasi waktu produksi metabolit sekunder dari isolat P301 sebagai penghasil antibiotik. Aktivitas antibakteri ditetapkan dengan metode sumuran dengan mengukur zona hambat di sekitar sumuran. Penentuan profil optimasi waktu produksi metabolit sekunder yaitu dengan membuat grafik hubungan antara diameter zona hambat dengan waktu inkubasi. Hasil penelitian menunjukkan bahwa cairan kulturisolat P301 mampu menghambat pertumbuhan S. aureus. Berdasarkan uji aktivitas terhadap S. aureus, waktu produksi antibiotik yang optimal adalah setelah diinkubasi minimal selama 11 hari.

Abstract

Jamur tanduk (local name), Termitomyces eurrhizus Berk, is one of mushroome species from genus Termitomyces. The edible mushroom can be found in various areas in Indonesia. In this study, determination of chemical structure of components in the mushroom extract has been made. Based on interpretation of spectra data ultra-violet, infra red, nuclear magnetic resonance one dimension (proton, carbon and DEPT) and NMR two dimension (COSY, HMQC and HMBC), two compounds: mannitol and ergosterol, has been isolated from isopropanol fraction of the extract

Abstract

Trypsin is an enzyme which is important for research as well as for the pharmaceutical industry, medicine and health, which is especially developed as a raw material for digestive enzymes. This study aimed to characterize PST from Lactobacillus plantarum FNCC 0270 that have not been studied as a producer of trypsin. Fermentation for producing PST by L . plantarum FNCC 0270 was performed in the LKB fermentor with working volume of 3.5 liters, at temperature of 37 oC, pH value of 8, agitation rate of 77 rpm and aeration of 0.5 vvm. The media composition were 3.64% baker’s yeast, 1.21% glucose, 0.13% skim milk. The fementation resulted in enzyme activities of 0.00129 U/mL and protein content of 0.49 mg/mL. Purification of the PST conducted by ultrafiltration Hollow Fiber Catridge 5 kD, presipitated with ammonium sulfate saturated (30-70%), 30 kD membrane polyesthersulfone, ion exchange column chromatography Q-XL and HiTrap affinity column chromatography. All of system increased purity of the crude PST. SDS-PAGE of PST indicated the molecular weights of 47.35; 38.42;21.40 and 12.96 kD. The PST was stable at pH value of 8 and temperature range of 25-35 oC. It is proved that the same half-life was 693.2 minutes. Km and Vmax values of the PST were 0.231 mM and 0.001 U/mL, respectively using N-α-benzoyl-DL-arginine-p-nitroanilide as the substrate. PST was inhibited by EDTA, Ca2+, Zn2+, Mg2+, Mn2+ and specific substrates. Based on the result of characterization of PST from L. plantarum FNCC 0270 can be concluded that the trypsin–like protease (PST) was trypsin.

Abstract

This study was conducted to evaluate the analgesic and anti-inflammatory acitvities of Eurycoma longifolia root in mice. The analgesic effect was assessed using chemical stimuli (wrihting test) to induce peripheral pain in mice. The anti-inflammatory activity was determined using carrageenan-induced paw edema in mice. This study exhibites that Eurycoma longifolia root possesses analgesic and anti-inflammatory activities in mice.

Abstract

The influence of temperature and time on ripening of Noni fruits (Morinda citrifolia L.) for the production of Noni juice was determined using mature, but unripe Noni fruits. The design of this experiment was a split-plot. The ripening times used as the main plot were 1, 2 or 3 months, and the ripening temperatures used as the sub-plot were 30°C, 40°C, and 50°C. The results showed that the difference of Noni juice properties at 50°C as ripening temperature during 1, 2 or 3 month were not significant, as were those at 40°C during 2 and 3 months. The concentration of antraquinon increases with the ripening time of Noni fruits.

Abstract

The rind of the mangosteen (Garcinia mangostana L.) contains many xanthones which have anti-infl ammatory and antioxidant activity that can counteract free radicals and prevent or inhibit cell degeneration (inhibiting aging). Utilization of mangosteen rind generally in the form of powder capsule. The lack of utilization in topical dosage forms makes the researcher interested in making a body butter of ethanol extract of mangosteen rind and evaluating its physical quality. Simplicia of mangosteen rind were dried using oven at 50 °C. Mangosteen rind extract was obtained from the maceration of simplicia powder with 96% ethanol solvent. The formula of body butter mangosteen rind extract is prepared based on the literature study. The body butter was evaluated for organoleptic, pH, adherence, spreading, homogeneity test, and physical stability test for five months. Besides using literature standards, researchers also using body butter that has been market legally as a comparison in evaluated its physical quality. Based on the statistical analysis, it can be concluded that the preparation of body butter extract of mangosteen rind has a good physical quality compared with the body butter that has been market legally. Physical stability for five months showed that the body butter of mangosteen rind extract was stable.

Abstract

Saat ini, jumlah perokok di Indonesia masih tinggi. Asap rokok mengandung radikalbebas. Penelitian ini bertujuan untuk menganalisis dampak pemberian ekstrak rosella merah terhadappencegahan kenaikan malondialdehid (MDA) serum tikus wistar yang dipapar asap rokok. Total sampel20 tikus jantan dan dibagi menjadi empat kelompok: kontrol negatif, kontrol positif, perlakuan ekstrakkelopak rosella merah dosis 270 mg/bb dan ekstrak kelopak rosella merah dosis 540 mg/bb. Kontrolnegatif hanya diberi pakan standar. Kontrol positif diberi pakan standar + dipapar 2 rokok per hari.Kelompok perlakuan diberi pakan standar + ekstrak kelopak rosella merah di pagi hari dan setelah itudipapar 2 rokok. Penelitian ini dilakukan selama 28 hari. Di akhir penelitian, serum darah dianalisisuntuk menentukan nilai MDA. Hasil penelitian uji MDA serum dianalisis dengan One Way ANOVA dandiikuti oleh uji Tukey HSD pada level 5%. Pemberian ekstrak kelopak rosella merah dosis 270 mg/kgbb dan ekstrak kelopak rosella merah dosis 540 mg/bb dapat mencegah peningkatan MDA serum secarasignifikan pada tikus wistar yang dipapar asap rokok.

Abstract

Two fruits, Guava (Psidium guajava L.) and Carambola (Averrhoa cammbola L.), Which are frequently consumed by Indonesian people, have been evaluated for its activities as 2,2-diphenyl-1-picrylhidrazyl (DPPH) radical scavenging. The fruits were macerated, evaporated, dissolved in Water, partitioned with chloroform and then with aethyl acetate to afford methanolic extract and fractions of chloroform, aethyl acetate, and Water respectively. Total phenolic and total flavonoid contents of each extract and fraction Were determined by spectrophotometric technique and used to evaluate the correlation between the natural chemical contents and their activities as DPPH radical scavengers. The results showed that extracts and fractions of R guajava L. fruit have more ability to scavenge 2,2-diphenyl- 1-picrylhidrazyl (DPPH) radical than those of A. carambola L. The total phenolic and total flavonoid contents of extracts and fractions of R guajava L. fruit are also higher than those of A. cammbola L.

Abstract

Isolation of phenolic polymers from plants are challenging because the low yield and time consuming. Synthesis of phenolic polymer is urgently needed to overcome the lack of the compounds. Phenolic oxidation with H2O2 by peroxidase, results oxidative coupling reaction and finally produces phenolic polymers. Peroxidase (e.g guaiacol) catalyzes oxidative reaction by hydrogen peroxide with substrate acted as hydrogen donor. In this study, peroxidase from sawi hijau (Brassica juncea) was isolated. The obtained crude peroxidase had specific activity 5.618U/mg protein. Reaction between guiacol, H2O2 and crude peroxsidase produced guaiacol dimer. The new dimer was identified by NMR spectrometry, and the product showed coupling in para-para of 4,4’-biguaiakol, with 17% yield. In summary, crude peroxidase obtained from sawi hijau (Brassica juncea) was able to act as a catalyst for the oxidative coupling reaction of guiacol into guaicol dimer compounds.
