Abstract

Proteins and peptides are food components that have proven ability to prevent hypertensive disease through inhibition of angiotensin converting enzyme (ACE). Buffalo milk or fermented curds are naturally potentially developed as functional food as antihypertensive food. The purpose of this study were to determine the optimum fermentation time to obtain curd with the best ACE inhibition activity. First the curd protein was extracted using alcohol in stages for curd with fermentation length of 1, 2, 3, and 4 days. Nol days or buffallo milk without fermentation as control. Every time fermentation variation of were tested pH acidity level, chemical composition, protein extraction, dissolved protein content, degree of hydrolysis and ACE inhibitor activity. The value ACE inhibitor the highest was done fractionate using ultrafiltration membranes helping centrifugation. The protein fraction with optimal ACE inhibitor activity was analyzed amino acid composition by ultra performance liquid chromatography. The result of research are time fermentaion is one day and dissolved protein content is 912,73 ppm. The highest ACE inhibition activity was obtained at 87.52%, and 23.38% for degree of hydrolysis. Based on molecular weight was obtained the highest ACE inhibition value at fraction below 3 kDa, the ACE inhibitor activity value is 99,3%. Amino acid composition are threonine 0,0249%, proline 0,039%, alanine 0,045%, phenyl alanine smaller than 0,012% and acid glutamate 0.158%. In general, dadih was able to inhibit ACE activity and potentially as an antihypertensive agent in vitro.

Abstract

Diltiazem hydrochloride has a half life of 3-4 hours and a bioavailability of 40%. The influence of a mixture of carbopol 940 and hydroxypropyl methyl cellulose as a mucoadhesive matrix on tablets made by the wet granulation method, had been determined by the dissolution and “wash off method. The result showed that increasing the concentration of carbopol 940 decreased the dissolution rate. The concentration of carbopol 940 and hydroxypropyl methyl cellulose in formula I (0%; 40%), II (5%; 35%), III (10%; 30%), IV (15%; 25%), and V (20%; 20%) released respectively 75,99%; 74,08%; 73,07%; 72,04%; and 71,29% diltiazem hydrochloride. The higher the concentration of carbopol 940 in acid solution, the shorter is the mucoadhesive action: 2 hours in formula I and II, and 1 hour in formula III, IV, and V. All of the formulas (pH 7,2) did not show a difference in mucoadhesive duration; after 8 hours, still attached to the mucosa of the small intestines.

Abstract

Endophytic fungi of Pasak bumi (Eurycoma longifolia Jack). were inoculated to synthetic medium and incubated for 28 days. The fumated medium then were extracted with chloroform. The antioxidant activities by 1,1-diphenyl 2-picrylhidrazyl (DPPH) methods showed that isolated compound has scavenging effect at IC285.7 ppm. Identification by interpretation of infra red spectra showed that the isolated compound was a terpenoid.

Abstract

Averrhoa bilimbi L. fruits and Kalanchoe pinnata (Lam.) Per. leaves are useful as anti bacterial. The extracts containing the compound bufadienolides have the potential to be used as an anti bacterial. The purpose of this study to fi nd the best formula of the preparation facial wash gel with the active substance from the combination of extracts. Search methods extract concentration using the agar diff usion to obtain regional Diameter Inhibition (DDH) of each extract and the combinations. Research conducted with 3 forrmula containing extracts with diff erent concentrations. The results of the evaluation of the preparation at room temperature storage does not change the color, shape and scent. Retrieved best formula is the formula I containing a concentration of 0.7% resulting extract preparations light green and scented perfume. The results of the evaluation of the formula I at room temperature after 3 month storage include: high foam in hard water 17 mL, high foam in distilled water >25 mL, the specifi c gravity 1.0386 g/cm3, surface tension 19, 3627 dyne/cm, viscosity 853,33-1103,33 cPs, pseudoplastis fl ow properties, pH 5.16, anti bacterial test results using Diameter test Inhibitory Region shows constraints on Staphylococcus aureus by 5,0±0 mm.

Abstract

It has been proven that gotu kola (Centella asiatica (L.) Urban) have an antioxidant activity that make it possible to use in preventing degenerative illness. The aims of this study are to find the effect of n-hexane purification before maceration on asiaticoside content, free radical scavenging activity and total phenolic content of gotu kola ethanolic extract. The gotu kola extract was made by maceration process using the mix of ethanol and aquadest as solvents in different proportion, which are 100%:0% (E100), 75%:25% (E75), 50%:50% (E50), 25%:75% (E25), and 0%:100% (E0). The determination of asiaticoside content in gotu kola ethanolic extract was measured using Thin Layer Chromatography (TLC) Densitometry method, free radical scavenging activity was measured using diphenylpicryl hydrazyl (DPPH) and defined by parameter of IC , and total phenolic content was defined by equivalency of galic acid using Folin-Ciocalteau. The results show that the hexane purification would increasing total phenolic content significantly in ethanolic extract which extracted with solvents in proportion of ethanol:aquades 75%:25% (E75), 50%:50% (E50), and 25%:75% (E25), but not significant in decreasing of IC . Statistically, n-hexane purification was also not significant to increasing asiaticoside content in gotu kola ethanolic extract.

Abstract

Barringtonia asiatica (L.) Kurz (nama local: keben) termasuk ke dalam suku Lecythidaceae, dan biasanya digunakan sebagai racun ikan. Tanarnan ini banyak tumbuh di sekitar pantai, sepanjang sungai, atau di hutan bakau. Secara tradisional, keben digunakan untuk pengobatan yang pemakaiannya berbeda-beda di setiap daerah, seperti obat sakit perut, hernia, dan sakit kepala. Biji tanaman ini mengandung saponin yang diperkirakan berperan sebagai zat bioaktifnya. Ekstrak metanol dari biji ini dipartisi dengan etilasetat- air (1:1) dan ekstrak air yang diperoleh dimurnikan dengan kromatografi kolom (LH-20; MeOH; MeOH 50%; air) hingga memberikan suatu serbuk amorf yang berwarna coklat. Dari hasil identifikasi struktur kimia dengan FT-IR, RMI (protondan karbon), diperkirakan komponen ini adalah senyawa triterpenoidsaponin 3-0-{[ β -D-galaktopiranosi1(1→3)-β-D-glukopiranosil(1→2)]-β-D-glukuronopiranosiloksi}-22- O-(2-metilbutiroiloksiy-15,16,28-trihidroksi-(3β,15α,16 α,22 α)-olean-12-en.

Abstract

The knowledge of teachers on asthma and its managements are very important, in order that teachers can provide the assistance required by the students in primary school who suffered an asthma attack while at school. This study aimed to examine the effect of an educational book to improve knowledge among primary school teachers on asthma and its management. This study was descriptive study, using questionnaires and educational book “Asthma: The Role of Teachers in the Management of Asthma in Schools”. The research was conducted in 3 phases: (1) asses the knowledge of teachers on asthma before being given an educational book, (2) administration of an educational book “Asthma: The Role of Teachers in the Management of Asthma in Schools”, (3) re-asses the knowledge of teachers on asthma after administration an educational book. A total of 48 teachers from three primary schools in the Medan Deli District involved in the study. In general, their knowledge of asthma before being given education is fairly low. After administering education the teacher knowledge increased, this can be seen from the increase in the number of teachers who answered correctly on any given question. An increase of almost 30% of teachers who correctly answer to questions about the causes of asthma after booklet administration and about 20% for questions about symptoms, and treatment of asthma. It can be concluded that the educational materials such as educational book has the effect to increase the knowledge of teachers about asthma and its treatment. It can be seen from the existence of increase in the number of teachers who answered correct on every the question given. This study concluded that the educational book entitle “Asthma: The Role of Teachers in the Management of Asthma in Schools “ has effective to improve knowledge among primary school teachers on asthma and its managements.

Abstract

Curcumin has been known to reduce cholesterol levels in hyperlipidemic state. Faculty of Pharmacy Universitas Gadjah Mada (UGM) synthesized curcumin analogues namely PGV-0 and GVT-0. Like curcumin, both PGV-0 and GVT-0 exhibit anti-inflammatory activities. The aims of this study were to determine the effect and the mechanism of curcumin analogues PGV-0 and GVT-0 on regulation of cholesterol levels in serum. This study was aimed to examine whether PGV-0 and GVT-0 affected cholesterol levels through cholesterol absorption which is regulated by lipase enzyme. To determine the reduction in cholesterol levels, rats were feed with high fat diet (HFD) for 45 days. PGV-0 and GVT-0 were given on day 31-45 at doses of 40 mg/kg bw and 60 mg/kg bw consecutively. Total cholesterol and lipase activity in serum were measured and then statistically analyzed using ANOVA and t-test. The increase of cholesterol levels was markedly reduced by the treatment with both curcumin analogues. Furthermore, lipase activity was clearly inhibited by the treatment with PGV-0 and GVT-0, suggesting that these compounds inhibit cholesterol levels through the reduction of lipase enzyme activity

Abstract

Diuretics use as first treatment in hypertension and heart failure. Empirically, community use belimbi as diuretic. Preliminary study to explore the potency of bilimbi ethanolic extract as a natural diuretic was conducted using ninety male mice which divided into 6 groups. Each group consist of three sub-group of 5 mice respectively for three replications. Group 1 was given orally saline solution of tween 80, Group 2 was given orally urea solution 500 mg/kg BW, Group 3 was given orally furosemide 3 mg/kg BW, Group 4 was given orally extract of bilimbi 2.5 g/kg BW, Group 5 was given orally extract of bilimbi 5 g/kg BW, and Group 6 was given orally aquadest. The diuretic activity was determined by measuring cumulative urine volume every 1 hour during 5 hours post administration. The result showed that 2.5 g/kg BW had a strong diuretics activity 2 hours post administration and reached its maximum activity 3 hours post administration. At the peak of diuretic activity, extract of bilimbi 2.5 g/kg BW increased diuretic effect by 219% as compared to the aquadest and 41,5% as compared to the furosemide. It is concluded that extract of bilimbi has a potency as a natural diuretic.

Abstract

Oyster mushroom (Pleurotus ostreatus) is one of the commonly consumed mushrooms due to its nutritious as well as its pharmaceutical value. It has been known that this mushroom contains several substances such as interferon inducer to help prevent several diseases i.e. hepatitis and many others. There are three cultivars of oyster mushroom based on the color of their fruit body, namely white oyster mushroom (Pleurotus floridae), grey oyster mushroom (Pleurotus sajur-caju) and brown oyster mushroom (Pleurotus cystidious). A possibility to increase its phamaceutical value is mutation using Gamma ray. The doses of mutation used are 0.25 KGy, 0.5 KGy, 0.75 KGy, and 1.0 KGy respectively. Since most of the pharmaceutical substances are secondary metabolite products of this mushroom, the mutation is intended to increase these metabolites. In this preliminary experiment, the mutation process is observed using isozyme analysis of the mutated mycelia as the indicator.
