Abstract

Talok (Muntingia calabura L.) leaf is a traditional medicinal plant which empirically used to treat diabetes mellitus. This research aimed to know hypoglycemic effect of combination dose of talak leaves infuse-metformin and glibenclamide on Na2EDTA induced-diabetic rats. Diabetic animal model were conducted by induction of Na2EDTA dose of 70 mg kg bw for 3 days. The test groups divided into six groups which were treated with: 50 mg kg bw CMC-Na suspension (negative control 1st group), 0.9 mg kg bw glibenclamide (2nd group), metformin 90 mg kg bb (3rd group), 900 mg kg bw talok leaves infuse (4th group), combination of talok leaves infuse-glibenclamide (450 mg : 0.45 mg/kg bw) (5th group), combination of talok leaves infuse-metformin (450 mg : 45 mg kg bw) (6th group). The levels of blood glucose were examined in all 6 experimental groups on day 0, 4th and 7th. The data obtained were analyzed by one way ANOVA (p < 0.05). The results showed that all of combination could decrease the blood glucose levels significantly. The combination treatment of talok leaves infuse-glibenclamide showed a higher hypoglycemic effect than either drug alone, while combination treatment of talok leaves infuse-metformin showed a similar hypoglycemic effect to either drug alone.

Abstract

Skopoletin diisolasi dari ekstrak metanol daun Macaranga hispida. Ekstrak daun diisolasi data spektroskopi dan dibandingkan dengan referensi. Uji sitotoksisitas skopoletin dengan menggunakan kromatografi dengan pelarut bergradien. Penentuan struktur kimia berdasarkan menggunakan metode assay MTT terhadap sel MCF-7 dan T47D menunjukkan aktivitas sitotoksik yang kuat dengan IC50 51,5 dan 66,5 μg/mL

Abstract

Cytotoxic and antioxidant activities of Typhonium divaricatum (L) Decne leaves extract have been explored. Preliminary phytocheinical screening on the extract were carried out for the compound`s profile. Biological activity was measured by using Brine Shrimp Lethality Test (BSLT), while cytotoxic activity was conducted on breast cancer cell line T-47D using MTT method, Antioxidant activity was measured by using DPPH free radical scavenging method. The results of phytochemical screening showed that the methanol extract contained Havonoid, saponin and steroid/triterpenoid compounds, meanwhile thc n-hexane extract contained steroid/triterpenoid compounds only. The highest biological activity was found in methanol extract (IC50 = 32.9 ppm). The highest cytotoxic activity was shown by n-hexane extract (IC50 = 32.50 ppm). The highest antioxidant activity using vitamin C as positive control was found in methanol extract (IC50 = 56.63 ppm). The preliminary results indicated that Typhonium divaricatum (L) Decne lcavcs is potential for anticancer and free radical scavenger.

Abstract

This research aimed to identify the cytotoxic compounds from Sarcophyton sp. which dominated the coral reefs area at Panggang Islands water, Seribu Islands National Parks. Preparative chromatography and bioassay MCF-7 cell lines guided fractionation had been used in isolation process of cytotoxic compounds. Moreover, UV-VIS, HR-ESI-MS, and 400 MHz NMR spectroscopy methods had been used as the technique to elucidated all of the active compound. The result of this study found three active cytotoxic compounds. These compounds were Sarcophytol (IC50 36,041 ppm), sarcophytol (IC50 71,03 ppm), and sarcophyhtolide (IC50 67,227 ppm). Ecologically, these results showed that Sarcophyton from Panggang Islands coral reefs waters produced a diversed bioactive compound so this organism can win the spatial competition, detterant predation, and dominate in this area.

Abstract

Mimba, Azadirachta indica is one of the family Meliaceae plants, which be used to lower blood sugar and contain terpenoids, fl avonoids, alkaloids, saponins, tannins, fatty acids, steroids and triterpenoids. The aim of this research is to isolate and identify one of chemical structure and to know the inhibition α-glucosidase enzyme from extract of leaves Mimba, Azadirachta indica. Extraction is done by maceration using n-hexane, ethyl acetate, methanol, water and then treatment for antidiabetic activity using the inhibition of the enzyme α-glucosidase. Purifi cation of n-hexane extract (the higher activity) is subjected by column chromatography (SiO2; (i).n-hexane-ethyl acetate = 10 : 1 ~ 1 : 1, ethyl acetate, (ii). n-hexane-ethyl acetate = 5 : 1) gave one pure compound as white powder. Identifi cation by interpretation of spectra [UV, IR and NMR (1H, 13C-NMR and DEPT)] the isolate compound is β-sitosterol has inhibion α-glucosidation enzyme as (IC50 208,1 μg/mL).

Abstract

Sambiloto (Andrographis paniculata Nees) is a medicinal plant containing andrographolide as its primary bioactive component, which has been shown to inhibit breast cancer cell proliferation. Andrographolide is a diterpene lacton and sparingly soluble in water. The aim of this study is to prepare microspheres of A . paniculata ethylacetate fraction by microencapsulation using spray drying method. The ethyl acetate fraction of A. paniculata herbs was microencapsulated using PVP K30 and HPMC as the coating polymer in different ratios of 1:5; 1:7,5 and 1:10. Microspheres containing the ethyl acetate fraction of A. paniculata herbs were characterized in term of recovery factor, morphology, particle size distribution, water content and entrapment efficiency. The results showed that the A. paniculata microspheres have the recovery factor of 19,50% to 31,64%, spherical shape varies from smooth surface to irregular shape, particles size of 1,44% to 19,23 µm, moisture content of 5,15% to 13,44%, and entrapment echiency between 94,83 to 112,15%.

Abstract

Typhonium flagelliforme atau yang dikenal dengan keladi tikus merupakan tumbuhan perdu yang banyak tumbuh di Indonesia. Tumbuhan ini mengandung lektin pada umbinya yang memiliki aktivitas antikanker. Namun hingga saat ini, tidak ada informasi ilmiah yang mengkaji mengenai lektin yang terdapat di umbi T. flagelliforme yang tumbuh diberbagai wilayah Indonesia. Tujuan dari penelitian ini adalah untuk mengidentifikasi dan mengkarakterisasi lektin umbi dari tujuh aksesi T. flagelliforme di Indonesia. Umbi yang digunakan dalam penelitian berumur 6 bulan tanam. Ekstrak protein umbi yang didapat dianalisis kandungan protein, toksisitas, dan aktivitas hemaglutinasinya. Aksesi yang memiliki aktivitas hemaglutinasi tertinggi, lektinnya dipurifikasi menggunakan kolom DEAE-Sepharose dan CM monolitic. Hasil menunjukkan bahwa protein umbi dari setiap aksesi memiliki toksisitas dan aktivitas hemaglutinasi. Aksesi Solok memiliki total protein dan aktivitas hemaglutinasi tertinggi dibanding aksesi lainnya. Aksesi ini memiliki kandungan lektin dengan bobot molekul 12,67 kDa. Lektin ini stabil pada temperatur 20 hingga 40 °C dan pada pH 5,0 hingga 7,2. Informasi karaktersitik ini dapat membantu dalam pengembangan obat antikanker berbasis lektin.

Abstract

Daun jambu mete memiliki daya antibakteri terhadap bakteri Gram positif dan negatif yang sensitif dan multiresisten. Tujuan penelitian ini adalah untuk mengetahui daya antibakteri ekstrak dan fraksi-fraksi daun jabu mete dan menentukan komponen senyawa yang terkandung di dalamnya. Metode ekstraksi dilakukan dengan maserasi menggunakan etanol 96%. Fraksinasi ekstrak etanol daun jambu mete dilakukan dengan metode partisi. Ekstrak dan fraksi-fraksi diuji daya antibakterinya menggunakan metode dilusi padat untuk menentukan konsentrasi hambat minimum (KHM). Hasil KHM dilanjutkan ke pengujian berikutnya untuk menentukan konsentrasi bunuh minimum (KBM). Analisis kandungan bahan aktif dilakukan dengan metode kromatografi lapis tipis (KLT). Hasil penelitian menunjukkan bahwa ekstrak etanol, fraksi kloroform dan fraksi etil asetat memiliki daya bunuh terhadap Staphylococcus sensitif dan multiresisten, sedangkan fraksi metanol-air hingga konsentrasi 2% tidak menunjukkan daya bunuh terhadap S. aureus sensitif, dan hingga 3% terhadap S. aureus multiresiten. Hasil pengujian KLT menunjukkan bhawa ekstrak dan fraksi-fraksi daun jambu mete mengandung alkaloid, flavonoid, minyak atsiri dan fenol.

Abstract

Cytotoxicity tests on ethanol extract of Aglaia elliptica Blume leaves and its fractions on cervical cancer cell line (HeLa) were carried out. Dried powder of Aglaia elliptica leaves was extracted using ethanol andthen fractionated by n-hexane, ethyl acetate, and buthanol solvents. The cytotoxicity tests using MTT method (prolipheration inhibiton test) on HeLa cell lines were conducted for ethanol extract, n-hexana, ethyl acetate, and buthanol lractions.The results showed that the ethanol extract of Aglaia elliptica and its fractions have inhibition effect against HeLa cell line with IC50 of 282.44 ppm, 177.64 ppm, 16.03 ppm, 211.77 ppm, 181.33 ppm for ethanol extract, n-hexane, ethyl acetate, buthanol, and Water fractions respectively.

Abstract

Polysaccharide from white turmeric [Curcuma zedoaria (Christm.) Roscoe)] rhizome has been found to have anti-tumor activity against sarcoma 180 cells and able to increase macrophage activity in vitro. Since the modulation of immune system also plays a role in eliminating tumor cell, this study was aimed to determine the immunomodulatory effect of C. zedoaria polysaccharide.Immunomodulatory activity of the polysaccharide fraction was studied in Swiss Webster mice through determination of phagocytic index from reticuloendothelial system, total antibody titers, delayed type hypersensitivity (DTH) reaction and splenocyte proliferation assay. Based on the carbon clearance test, polysaccharide fraction at dose 300 mg/kg bw has higher phagocytic index (1.34) which means it has immunostimulatory activity. At the same dose, polysaccharide fraction enhance humoral immune response shown by primary and secondary antibody titers (1:128 and 1:4096) higher than control. This dose also stimulated delayed type hypersensitivity reactions by 19.41% respectively. In splenocyte proliferation assay, polysaccharide fraction of C. zedoaria was significantly stimulated splenocyte proliferation at contentration 10-6 to 1 mg/mL (p < 0.01). Based on those results, polysaccharide fraction of C. zedoaria at dose 300 mg/kg bw has potency to be developed as immunomodulator.
