Abstract

Simultaneous determination of phenylpropanolamine hydrochloride and chlorpheniramine maleate in tablets by spectrophotometry was carried out. The method is based upon total absorption of the two compounds in a mixture which can be applied to quantify each ofthe component. The absorption of phenylpropanolamine hydrochloride and chlorpheniramine maleate in 0.1 N hydrochloric acid were measured at the maximum Wavelength of phenylpropanolamine hydrochloride (256.7 nm) and that of chlorpheniramine maleate (262.6 nm), with a correlation coefficient of 0.999. A high precision could be obtained as shown by the CV of 0.2139-0.4962% for phenylpropanolamine hydrochloride and 0.3182- 0.5289% for chlorpheniramine maleate, While the recoveries ofphenylpropanolamine hydrochloride and chlorpheniramine maleate were 99.93% ± 0.43% and 99.76% ± 0.39%, respectively. The proposed method is simple, with a high accuracy and suitable for the simultaneous determination ofphenylpropanolamine hydrochloride and chlorpheniramine maleate in tablets, Without prior separation.

Abstract

Type 2 diabetes is a metabolic disorder that results from insulin secretion disorder, insulin action or both. SUR1 gene is part of KATP channel in the pancreatic β-cell that plays an essential role related with cell membrane depolarization and glucose-induced insulin secretion. The polymorphism p.R1273R of SUR1 gene, causes KATP channel failed to give feedback to metabolic activation, failed of closing KATP channel, reduces insulin secretion, impaired glucose tolerance, and causes type 2 diabetes. This is case-control study using 80 subjects consist of 40 subjects with type 2 diabetes as case and 40 subjects of non diabetes as control. The SUR1 gene p.R1273R polymorphism genotyping were detected by PCRRFLP. Data were statistically analyzed by t-test, Chi-Square. The genotype frequency distribution of p.R1273R SUR1 gene in type 2 diabetes subjects were 90% of GG genotype and 10% of AG genotype. In non diabetes subjects, GG genotype was 87,5% and AG genotype was 12.5%. In type 2 diabetes and non diabetes subjects the AA genotype was not observed. The allele frequency distribution in type 2 diabetes subjects G allele was 95% and A allele was 5%, in non diabetes subjects G allele was 93.75% and A allele was 6.25%. All the genotype and allele frequency distributions in type 2 diabetes and non diabetes subjects were not statistically significant (p = .723 genotype frequency; p = 0.732 allel frequency).

Abstract

Fennel fruit (Foeniculum vulgare Mill.) is one of traditional plant medicine known having effects as anti-infl ammatory, analgesic and antioxidant. It also has been reputed to increase milk secretion, promote menstruation, facilitate birth, alleviates the symptoms of the male climacteric andincrease libido. Fennel fruit contains mainly with essential oils, such as anethole. It also contains lignan and flavonoids that are predicted having an estrogen-like eff ect or as phytoestrogen. Phytoestrogen is weak agonists for estrogen and can elicit statistic signifi cantly increases in uterine wet weight, at definite dose, in the uterothrophic bioassay. It is known that estrogen has eff ect on woman’s sexual organ, such as proliferation of uterine, mammary and ovary. The aim of this research was to investigate the estrogenic eff ect of fennel fruit’s ethanolic extract in immature, 19 days old, Wistar female rats. These models represent the climacteric/menopause phase, where estrogen level is very low because in this age, estrogen not yet produced by ovary. The testing animals were divided into fi ve groups, namely normal group, estradiol control group and three level doses of fennel extract (30 mg/200 g BW; 60 mg/200 g BW and 120 mg/200 g BW). The result indicated that start on 60 mg/200 g BW, fennel extract signifi cantly (α=0.05) increased wet weight of uterine. Fennel extract also lengthen the uterine and increased proliferation of uterine, such as uterine diameters, endometrium diameters and uterine thickness on dose 60mg/200gBW and 120mg/200gBW. Analysis of vaginal smear showed vaginal lubrication eff ect, surprisingly start on 60mg/200BW could induce estrous cycle.

Abstract

Assay of amoxycillin level in suspension form using spectrophotometry method at maximum wavelength around 291 nm is predicted to provide accurate and selective result because it is not interferred with its degradation products. This research aimed to obtain the predicted result. This research employed laboratory experimental method which included the process of measuring of maximum wavelength and operating time measurement, making of standard curve by measuring absorbance of amoxycillin standard solution at 40, 50, 80, 100 and 120 ppm at 290 nm and testing the selectivity and accuracy. Selectivity test was done by comparing UV spectrum and maximum absorbance of 1 mg/mL amoxycillin standard solution and 125 mg/5 mL amoxycillin suspension immediately after reconstitution with the one that was stored 9 days. Accuracy test was conducted by standard addition method using 1 mL amoxycillin suspension that was stored 9 days added by 1 mL 6260 ppm amoxycillin standard solution. Assay of amoxycillin on amoxycillin suspension by UV spectrophotometry at 290 nm using NaOH 0,1 N as solvent gave selective and accurate result with 97,36% recovery and it is not interfered with by its degradation product.

Abstract

Research on the antioxidative activity and toxicity of the bioactive substance of the green seaweed Ulva reticulata Forsskal extract had been carried out. The bioactive substance of the green seaweed Ulva reticulata Forsskal was extracted using methanol. The phytochemistry test on the methanol extract showed that the extract contained triterpenoid compound. The methanol extract was then partitioned using n-hexane-water (1:1) and chloroform-water (1:1). Each of the extract was then dried using freeze dryer. The antioxidative activity test of each fraction against free radical 1,1-diphenyl-2-picrilhydrazil (DPPH) showed that the water extract has the highest activity with IC 30 of 365,95 ug/ml. The toxicity test of each fraction against brine shrimp Artemia salina (Brine Shrimp Lethality Test, BSLT) showed that the chloroform extract showed the highest activity with LCs of 250,67 ug/ml. The water extract was then fractionated through silica column chromatography. The activity test of each fraction against DPPH and BSLT showed that fraction 1 has the highest activity with LCs of 100 ug/ml and ICs of 270,31ug/ ml. Identification using gas chromatography – mass spectrometric method showed that the antioxidative substance presence in the active fraction was probably nonyl phenol (C, H2,0).
